Fundamental Study on Solvent Sublation Using Salphen and Its Application for Separative Determination of Trace Ni(II), Co(II) and Cu(II) in Water Samples

  • A solvent sublation using salphen as a ligand was studied and applied for the determination of trace Ni(II), Co(II) and Cu(II) in water samples. The fundamental study was investigated by a solvent extraction process because the solvent sublation was done by extracting the floated analytes into an organic solvent from the aqueous solution. The salphen complexes of Ni(II), Co(II) and Cu(II) ions were formed in an alkaline solution of more than pH 8 and then they were extracted into m-xylene. It was known that the each metallic ion formed 1 : 1 complex with the salphen and the logarithmic values of extraction constants for the complexes were 3.3 5.1 as an average value. Based on the preliminary study, the procedure was fixed for the separation and concentration of the analytes in samples. Various conditions such as the pH of solutions, the influence of $NaClO_4$, the bubbling rate and time of $N_2$ gas, and the type of organic solvent were optimized. The metal-salphen complexes could be extracted into m-xylene from the solution of more than pH 8, but the pH could be shifted to acidic solution of pH 6 by the addition of $NaClO_4$. In addition, the solvent sublation efficiency of the analytes was increased by adding $NaClO_4$. The recovery of 97-115% was obtained in the spiked samples in which given amounts of 0.3 mg/L Ni(II), 0.8 mg/L Co(II) and 0.04 mg/L Cu(II) were added.

S5 Lipase : An Organic Solvent Tolerant Enzyme

  • In this study, an organic solvent tolerant bacterial strain was isolated. This strain was identified as Pseudomonas sp. strain S5, and was shown to degrade BTEX (Benzene, Toluene, Ethyl-Benzene, and Xylene). Strain S5 generates an organic solvent-tolerant lipase in the late logarithmic phase of growth. Maximum lipase production was exhibited when peptone was utilized as the sole nitrogen source. Addition of any of the selected carbon sources to the medium resulted in a significant reduction of enzyme production. Lower lipase generation was noted when an inorganic nitrogen source was used as the sole nitrogen source. This bacterium hydrolyzed all tested triglycerides and the highest levels of pro-duction were observed when olive oil was used as a natural triglyceride. Basal medium containing Tween 60 enhanced lipase production to the most significant degree. The absence of magnesium ions ($Mg^{2+}$) in the basal medium was also shown to stimulate lipase production. Meanwhile, an alkaline earth metal ion, $Na^+$, was found to stimulate the production of S5 lipase.

Monte Carlo Simulation Study of Solvent Effect on Selectivity of 18-Crown-6 to between La3+ and Nd3+ Ion

  • We have investigated the solvent effects on Δlog $K_s$ (the difference of stability constant of binding) and the relative free energies of binding of $La^{3+} and Nd^{3+}$ ions to 18-crown-6 by a Monte Carlo simulation of statistical perturbation theory (SPT) in diverse solvents. We compared relative binding Gibbs free energies and the differences in stability constant (Δlog $K_s$) of binding of $La^{3+} and Nd^{3+}$ ions to 18-crown-6 in $CH_3OH$ in this study with the experimental. There is a good agreement between our study and the experimental. We noted that Borns function of the solvents, the electron pair donor properties of the solvent, the radii of host and guest and the differences in solvation dominate the differences in the stability constant (Δlog $K_s$) as well as the relative free energies of binding of TEX>$La^{3+} to Nd^{3+}$ ions to 18-crown-6. The results of this study appear promising for providing the association properties of crown ethers with alkaline earth metals among polar solvents and the less polar or non-polar solvents.

  • 인쇄회로기판(Printed Circuit Board) 생산 공정에서 발생하는 동폐액을 재활용, 재이용하기 위하여 유기 용매 추출법을 이용하였으며 유기용매로 Lix 64 N을 사용하였다. 산성인 동폐액과 염기성인 동폐액을 혼합하여 pH=2에서 부피 비율로 30%인 Lix 64 N은 17.1gr/l의 동을 추출하였다. 벤치 규모의 연속공정에서 추출단 4단, 세척단 4단, 역추출단 2단이 사용되었다. 회수된 동은 유산동으로 재활용되고 추잔액은 동부식액으로 재이용되었으며 동의 회수율과 유산동의 순도는 모두 99.9% 이상이었다.

  • 방사성의 $Ba^{++}, Sr^{++}, Ca^{++}$ 이온과 양이온 교환수지를 이용한 평형교환법에 의하여 알칼리토류 금속이온과 2염기 유기산 이온사이에 어떤 착물이 형성되는가를 실온에서 조사하였다. 유기산으로써 썩신산과 타르타르산을 사용하였고, 용매로서는 $H_2O$, 20% 아세톤-$H_2O$ 및 20% 에탄올-$H_2O$를 사용하였다. 용액의 pH 는 7.2∼7.4로 조절하였고 용액의 이온 강도는 약 0.1로 조절하였다. 본 연구결과 알칼리토류 금속이온과 썩신산 또는 타르타르산 사이에 1:1 착물이 용액내에서 형성됨을 알았고 또 착물의 상대적 안정도가 다음 순으로 증가함을 알았다. $Ba^{++}; 썩신산<타르타르산; 수용액<혼합용매

The Solubility Characteristics of Organic Compounds in Urban Aerosol Samples

  • The solubility characteristics of organic compounds were studied in terms of the extraction efficiency as a function of the polarity of the organic solvent, and the acidity of water in urban aerosol samples collected in University of East Anglia (UEA), Norwich, England. The extraction efficiency of organic compounds were evaluated with respect to the organic carbon, -nitrogen and -hydrogen by means of a wide range of solvent which include polar and nonpolar organic solvents as well as acids and alkaline water. In addition, after being dissolved in aqueous solution, the aqueous chemistry of organic compounds were studied in terms of the organic metal complexes in aerosol, which were studied with oxalic acid, copper, and zinc. The results of this study indicate that solubility characteristics of organic compounds depend on the polarity of the solvents and the acidity of the solvents. In particular, some organic compounds are water soluble, even though they are much smaller than acetone soluble fractions. In the comparison between polar organic solvent extraction and non- polar organic solvent extraction, it can be thought that significant fraction of organic compounds analysed in the aerosol samples, are polar organic compounds because of the higher extraction efficiencies of organic compounds in polar organic solvent extraction than in nonpolar organic solvent extraction. Regarding the study of the oxalic -metal complexes, it can be thought that most oxalic acids are present in the form of oxalic -copper complexes in the aerosols collected at UEA.

알칼리막 연료전지용 전극의 제조방법에 따른 전기화학적 특성 분석 (Electrochemical Characteristics of Electrode by Various Preparation Methods for Alkaline Membrane Fuel Cell)

  • 최근에 알칼리막연료전지의 막전극접합체에서 이오노머에 의한 촉매 피독에 대한 연구 결과들이 보고되고 있다. 본 연구에서는 이를 해결하기 위해서 전극 제조 시에 사용되는 유기용매의 성분을 조절하여 막전극접합체의 성능을 향상시키고자 하였다. Fuma-Tech사의 상용 이오노머를 사용하여 N-Methyl-2-pyrrolidone (NMP)와 Ethylene glycol (EG)를 이용한 4가지의 혼합용매를 제조하였다. 혼합용액을 이용하여 제조된 캐소드 전극은 NMP기반의 상용 이오노머에 비해서 약 36%의 향상된 분극성능을 나타내었다. 이것은 용매의 종류에 따른 이오노머의 분산성 차이에 따른 결과로 추측되며 비균일성 분포의 이오노머가 전극의 성능을 향상시키는 것으로 관찰되었다. 이에 관한 원인분석을 위해서 막전극 접합체의 고주파 저항, 내부저항 보정 분극곡선, Tafel 기울기, Mass activity 및 임피던스 분광법을 사용하여 특성 분석을 실시하였다. 이오노머의 비용매의 비율 증가에 따라서 캐소드 전극 성능이 개선되는 것을 확인하였고, 이것은 이오노머의 입도 분포에 따라서 촉매의 피독이 감소되는 결과로 판단된다.

  • The Purpose of this study was to investigate spectrophotometric characteristics of Korean knotweed extracts. The properties were evaluated by it′s extracting solvent, effect of metallic ion, variations of pH values and effect of light exposure. The results were as following; The highest absorbance was found in methanol extract of Korean knotweed, while the lowest absorbance was carbon tetrachloride extract. The UV-vis. spectra of Korean knotweed colors in several solvents showed hypsochromic shift of n-π/sup */ transition with the polarity of solvent. Absorbance and peak of UV-vis. spectra of Korean knotweed colors became lower and broader by addition of metallic ion. The light stability in irradiation with xenon ]amp of the color solution with Cu were higher than those of Al and Fe. The UV-vis. spectra of Korean knotweed extracts in various pH values showed bathochromic shift under alkaline condition, and their peaks disappeared after 5 hours exposure.

내포체 단백질 재생을 위한 용해 및 재접힘공정의 비교분석 (Comparative Analysis of Dissolution and Refolding Processes for Inclusion Body Protein Renaturation)

  • Using rlFN-$\alpha$ and rhGH as the model proteins, the refolding performances of the published processes were evaluated and compared. Key engineering parameters such as the type of denaturant and this concentration, protein concentration in the refolding buffer, and pH and ionic strength of the buffer were experimentally investigated. Furthermore, the role of a co-solvent of surfactant type in aggregation reduction was also studied. Of the denaturants tested (8M urea, 6M guanidine HCI, 0.5% SDS), SDS at alkaline pH (9.5) and ambient temperature gave the highest recovery yield. The SDS process was effective in the refolding of observed where dissolution proceeded better under lower strength (10 mM) but aggregation was suppressed under higher strength (>50 mM.) When PEG-4000 and/or Tween were added as co-solvent or refolding-enhancing additive, 1.6-2 times higher yield was realized. The‘masking’of the hyrophobic patches located on the surface of the protein with the surfactant molecules was believed to be responsible for the considerable reduction in aggregation during refolding.
