택사의 생약학적 연구 (Pharmacognostical Studies on Alisma Plants)

  • In order to clarify the originality of several Alisma Rhizomes, we investigated the differences of their morphological, anatomical and physiochemical characteristics. Morphological observation shows the variability of Alisma shape and quality. The diversities in morphological shapes and qualities of tuber were observed among different origins. Chinese product looked like corm shape, but korean's irregular shape. The microscopic observation of korean Alisma revealed the presence of irregular vascular bundle which was scattered transversely and longitudinaly, which was different from that of the rhizome. This facts suggest that the portion of crude drug in Alisma should be the corm rather than the rhizome. In the physicochemical differences, chinese Alisma contains larger amount of mineral elements such as K, Na and Ca than korean Alisma. On the contrary, protein contents in lyophilized powder of aqueous extracts in korean Alisma rhizome was significantly higher than chinese Alisma rhizome. The leaves and stems contained larger amount of Ca and Na than tuber, and may be used as the osmotic diuretics for chinese traditional medicine.

Hepatoprotective and Antioxidative Effects of Alisma orientale

  • The rhizome of Alisma orientale Juzep (Alismataceae) has been used as a crude drug for diabetes, edema, inflammation and urinary disturbances in oriental medicine. Recent animal studies have shown that the extract of Alisma orientale rhizome (AOR) can potently lower high levels of serum lipids and improve insulin resistance, which are usually detected in patients and animals with non-alcoholic fatty liver disease. So, we studied the antioxidative effects of AOR extracts and fraction in vitro and their protective effects against acute hepatotoxicity induced by $CCl_4$ in vivo.. We then investigated the effects of each fraction on hepatotoxicity induced by tert-butyl hydroperoxide (t-BHP). DAOR (dichloromethane fraction of the Alisma orientale rhizome) scavenged free radicals and superoxide anions. DAOR protected against $CCl_4$ induced hepatotoxicity. DAOR had hepatoprotective and antioxidative effects against t-BHP-induced hepatotoxicity in HepG2 cells and in rats.

택사에서 Alisol B 23-Acetate의 분리 및 함량분석 (Isolation and Quantitative Analysis of Alisol B 23-Acetate from the Rhizome of Alisma orientale)

  • 택사 MeOH 추출물에서 얻은 dichlorornethane 분획물을 칼럼 크로마토그라피를 실시하여 alisol B 23-acetate를 순수히 분리하였다. Alisol B 23-acetate를 여러가지 용매와 추출 조건을 달리하여 HPLC를 실시하여 이들의 분리능을 검토한 결과 70% $CH_3$CN 혼합용매를 사용할 때 분리능이 가장 양호하였다. 국내 각지에서 수집한 택사 33종과 중국 현지에서 수집한 중국택사 5종에 대해 HPLC를 이용하여 함량분석을 하였다. 국내구입 검체중 지표성분 alisol B 23-acetate의 함량은 0.25%에서 0.67%까지의 범위를 보였으며 평균값 및 표준편차가 0.47$\pm$0.11%이었다. 실험에 사용한 검체중 재배산지에서 직접 구입한 순천시 해룡산 택사가 alisol B 23-acetate의 함량이 0.67%로서 가장 많았다. 중국에서 구입한 택사에는 alisol B 23-acetate가 포함되어 있지 않거나 0.27%에서 0.33%함유하였다. 이 검체들은 한국에서 구입한 택사와 종이 다르거나 또는 재배조건이 상이하여 이러한 결과가 나온 것으로 추정된다. 택사의 표준화를 위해서는 지표성 분 alisol B 23-acetate의 함량은 0.25% 이상으로 규정함이 타당하다고 사료된다.

  • Alisol B(II) and alisol B monoacetate (III) isolated from the rhizome of Alisma orientale Jazepczuk (Alismataceae) showed antagonistic effects against the contractions induced by angiotensin and bradykinin in rat ileum. However, they seem to be nonspecific antagonism.

  • Objectives : The study was conducted to evaluate antioxidative, anti-atherosclerotic and anti-hypertensive effects of natural remedies. Alisma Rhizome (AR) has been used for a long time in Asia in folk remedies for treatment of hypertension and stroke and has been used in Korean traditional medicine for the treatment of glycosuria, gonorrhea, hypercholesterolemia, hypertension, and jaundice and its diuretic effect. These pharmacological effects of AR might come from antioxidant properties of phytochemicals in these materials. Methods : In this study, the antioxidant activity of extract from AR was studied with in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$ induced human LDL oxidation and on ACE. Results : The AR extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation and on ACE against all of the reactive species tested, with the water extract showing particularly strong antioxidant activities. Conculsions : The AR extracts have antioxidative, anti-atherosclerotic and anti-hypertensive effects in an in vitro system, which can be used for developing pharmaceutical drugs against oxidative stress and atherosclerosis.

  • Alismatis Rhizoma is originated from the rhizome of Alisma plantago-aquatica L. var. orientale Samuelson or A. canaliculatum A. Br. et Bouche (Alismataceae). Prostane-type triterpenes, guaiane-type sesquiterpenes, and kaurane-type diterpenes have been reported as the main canstituents from these plants. Four prostane-type triterpenes. alisol B 23-acetate (1). alisol C 23-acetate (2), alisol B (3). and alisol A 24-actate (4). were isolated from the EtOAc-soluble fraction of this dried rhizome.   • 택사로부터 황산암모늄 분별 침전, DEAE-cellulose ion exchange chromatography, Sephadex G-150 chromatography 등의 방법을 이용하여 렉틴을 분리, 정제하였다. 정제한 렉틴의 분자량은 gel filtration법에 의해 측정한 결과 90,500 dalton이었으며, SDS polyacrylamide gel 전기영동을 실시한 결과 subunit 분자량은 각각 42,000, 27,000, 22,500 dalton으로 나타나므로 택사 렉틴이 heterotrimer임을 알았다. 정제된 택사 렉틴은 사람 적혈구의 경우 모든 혈액형에 대하여 응집현상을 나타내었으며 돼지, 쥐 및 개 등의 적혈구에 대해서도 모두 응집현상이 나타나 혈구 비특이성임을 보여주었다. 또한 이 렉틴은 sialic acid, glucose, ribose, sucrose, lactose, galactose 등 당에 의해서, 그리고 $Hg^{2+},\;Fe^{2+},\;Cu^{2+}$ 이온 등에 의해 적혈구 응집력이 저해되었다.

Alisol B 23-Acetate Ameliorates Ovalbumin-Induced Allergic Asthma during Sensitization and Challenge Periods

  • Rhizome of Alisma orientale has been used as a traditional medicine for treating kidney diseases in East Asian countries. Its inhibitory effects on hypersensitivity responses have been reported for methanol extracts, with alisol B 23-acetate (AB23Ac) being the most active constituent among six terpenes in inhibiting the direct passive Arthus reaction. However, whether AB23Ac has efficacy against allergic asthma has not been tested to date. The in vivo efficacy of AB23Ac in an ovalbumin (OVA)-induced allergic asthma mouse model was evaluated by administrating AB23Ac before OVA sensitization or OVA challenge in BALB/c mice. AB23Ac suppressed antigen-induced degranulation of RBL-2H3 mast cells in a concentration-dependent manner. The administration of AB23Ac both before OVA sensitization and OVA challenge greatly lowered pulmonary resistance and the increase in immune cell counts and inflammatory responses around the peribronchial and perivascular regions. In addition, the inflammatory cytokine levels of Th1/Th2/Th17 cells in the bronchoalveolar lavage fluid decreased in the AB23Ac-treated groups. AB23Ac reduced the number of PAS-stained cells in the lungs. Furthermore, a computer modeling study indicated that AB23Ac can bind tightly to spleen tyrosine kinase (Syk). These results suggest that AB23Ac may ameliorate allergic asthma by suppressing immune responses in dendritic cells during sensitization and in mast cells during challenge periods.

  • Alismatis rhizoma (AR), the dried rhizome of Alisma orientale (Sam.) Juzep, is a well-known, traditional medicine that is used for the various biological activities including as a diuretic, to lower cholesterol and as an anti-inflammatory agent. The present study was carried out to investigate the potential toxicity of the Alismatis rhizoma aqueous extract (ARAE) following 90-day repeated oral administration to Sprague-Dawley rats. ARAE was administered orally to male and female rats for 90 days at 0 (control), 500, 1,000 and 2,000 mg/kg/day (n = 10 for male and female rats for each dose). Additional recovery groups from the control group and high dose group were observed for a 28-day recovery period. Chromatograms of ARAE detected main compounds with four peaks. Treatment-related effects including an increase in the red blood cells, hemoglobin, hematocrit, albumin, total protein, and urine volume were observed in males of the 2,000 mg/kg/day group (p < 0.05). However, the diuretic effect of ARAE was considered, a major cause of hematological and serum biochemical changes. The oral no-observed-adverse-effect level (NOAEL) of the ARAE was > 2,000 mg/kg/day in both genders, and no target organs were identified.
