Production Method of Biochar-bead from Biochar Powder and Its Application for the Removal of Heavy Metal (분말 바이오-숯으로부터 중금속 오염수 처리용 바이오-숯 비드 제조 및 적용)

  • In this study, biochar-bead, prepared from biochar powder derived from woody biomass, was used for removal cadmium ion in aqueous solution. Various mixing ratios of alginate solution and biochar powder were used for the production of round shape biochar-bead. An optimum mixing ratio was selected as 1.5% alginate solution and 20 wt% biochar. The produced biochar-bead was characterized by SEM, FT-IR, and XRD analyses. The adsorption capacity of Cd(II) by biochar-bead was found to be 9.72 mg/g which was higher than that by GAC and PAC. According to this study, round shape biochar-bead is expected to be used as a media for reactive barrier or water filtration.

Enhancement of Cultivation Efficiency of Bifidobacterium longum Using Calcium Carbonate Buffer System (Calcium Carbonate Buffer System을 이용한 Bifidobacterium longum의 배양 효율 증대에 관한 연구)

  • Calcium carbonate ($CaCO_3$) immobilized with alginate was studied as buffer system to enhance the cultivation efficiency of Bifidobacterium longum (ATCC 15707) which is inhibited at low pH. To test the bufferring effect of the immobilized $CaCO_3$ beads, pH value in each modified trypticase-proteose peptone-yeast (TPY) broth which is adjusted to pH 4.0 with acetic acid, lactic acid and complex solution of acetic and lactic acid, 3:2 (M:M) was tested by concentration of $CaCO_3$ bead and reaction time. The bufferring effect of $CaCO_3$ bead became higher with increasing the amount of $CaCO_3$ bead in the acidic solution. The growth rate of bifidobacteria and bufferring effect were examined in relation to the amount of $CaCO_3$ bead and concentration of glucose in the modified TPY media. The growth rate of bifidobacteria and bufferring effect were increased with increasing the amount of $CaCO_3$ bead and concentration of glucose. Also, the exponential time of bifidobacteria became longer with increasing the amount of $CaCO_3$ bead and concentration of glucose in the modified TPY media. When we observed the growth rate of bifidobacteria by the method of pH-controlled culture and $CaCO_3$ buffer system, the $CaCO_3$ buffer system was more effective than that of pH-controlled culture. Therefore, this $CaCO_3$ buffer system may be useful as a method to enhance of the cultivation efficiency of bifidobacteria.

Removal of ammonia by packed bed bioreactor using immobilized nitrifiers (고정화 생물 반응기에 의한 암모니아 제거)

  • Nitrifier consortium entrapped in Ca-alginate bead were packed into aerated packed bed bioreactor and non aerated packed bed bioreactor and the performances of two bioreactors were evaluated for the removal of ammonia nitrogen from synthetic aquaculture water. Total ammonia nitrogen(TAN) removal rate was decrease in aerated packed bed bioreactor below 0.3hr of hydraulic residence time(HRT), but increased in non aerated packed bed bioreactor until 0.5hr of HRT. At HRT of 0.05hr, TAN removal rate of non aerated packed bed bioreactor was about 335g TAN/㎥/day and the optimum ratio of packing height and inside diameter of reactor (H/D) was 4. The performance of two bioreactors indicated that non aerated packed bed bioreactor was better than aerated packed bed bioreactor in ammonia removal from synthetic aquaculture water.

Ammonium Ion Effects and Its In Situ Removal by Using Immobilized Adsorbent in Hybridoma Cell Culture (하이브리도마 세포배양에서 암모늄 이온의 영향 및 고정화 흡착제에 의한 암모늄 이온의 동시제거)

  • The effects of ammonium ion on cell growth kinetics, monoclonal antibody productivity, and cell metabolism of hybridoma cells were investigated. The mouse-mouse hybridoma cell line VlIIH-8 producing mouse IgG2a was used as a model system. Ammonium ion showed an inhibitory effect on cell growth and monoclonal antibody production. New immobilized adsorbents were developed for the reduction of the inhibitory effect of ammonium ion. The ammonium ion selective zeolite, Phillipsite-Gismondine was entrapped in calcium alginate bead or in dialysis membrane and applied to the hybridoma cell culture system for the in situ removal of ammonium ion from culture media. The effects of ammonium the both serum supplemented and serum free media on the cell growth were studied by applying immobilized adsorbents of calcium alginate bead type. The results demonstrated a substantial enhancement in cell growth. Applying immobilized adsorbents of dialysis membrane type to serum supplemented media also resulted in the stimulation of cell growth, cell viability and monoclonal antibody production.

  • In this work, to develop soil inoculant which maintains stable viable cells and normalized quality, studies on micro-encapsulation with bacteria and yeast cells were performed by investigating materials and methods for micro-encapsulation as well as variation and stability of encapsulated cells. Preparation of capsule was conducted by application of extrusion system using micro-nozzle and peristaltic pump. K-carragenan and Na-alginate were selected as best carrier for gelation among K-carageenan, Na-alginate, locust bean gum, cellulose acetate phthalate (CAP), chitosan and gelatin tested. Comparing the gels prepared with Bacillus sp. KSIA-9 and carriers of 1.5% concentration, although viable cell of K-carragenan and Na-alginate was six times higher than those of other, Na-alginate was finally selected as carrier for gelation because it is seven times cheaper than K-carragenan. The gel of 1.5% Na-alginate was also observed to have the best morphology with circular hardness polymatrix and highest viable cell. When investigating the stability of encapsulated cells and the stabilizer effect, free cells were almost dead within 30 or 40 days whereas encapsulated cells decreased in 10% after 30 days and 15-30% even after 120 days. As stabilizer for maintaining viable cell, both 1% starch and zeolite appeared to possess the level of 70-80% cell for bacteria and yeast until after 120 days.

Preparation and Release Characteristics of Polymer-Reinforced and Coated Alginate Beads

  • Polymeric reinforcement and coatings of alginate beads were carried out to control the release rate of drug from alginate beads. A poorly water-soluble ibuprofen (IPF) was selected as a model drug. A commercially available $Eudragit^{\circledR}$ RS100 was also used as a polymer. Effects of polymeric contents, the presence of plasticizers and amount of drug loading on the release rate of drug were investigated. The release rate of drug from alginate beads in the simulated gastric fluid did not occur within 2 h but released immediately when dissolution media were switched to the simulated intestinal fluid. No significant difference of release rate from polymer-reinforced alginate bead without plasticizers was observed when compared to plain (simple) beads. However, the release rate of drug from polymer-reinforced alginate beads was further sustained and retarded when aluminium tristearate (AT) as a plasticizer was added to polymer. However, polyethylene glycol 400 (PEG400) did not change the release rate of drug from alginate beads although PEG400 was used to improve dispersion of polymer and sodium alginate, and plasticize $Eudragit^{\circledR}$ RS100 polymer. The presence of plasticizer was crucial to reinforce alginate gel matrices using a polymer. As the amount of drug loading increased, the release rate of drug increased as a result of decreasing effects of polymer contents in matrices. The significantly sustained release of drug from polymer-coated alginate beads occurred as the amount of polymer increased because the thickness of coated membrane increased so that cracks and pores of the outer surface of alginate beads could be reduced. The sustained and retarded action of polymer-reinforced and coated beads may result from the disturbance of swelling and erosion (disintegration) of alginate beads. From these findings, polymeric-reinforcement and coatings of alginate gel beads can provide an advanced delivery system by retarding the release rate of various drugs.

Preparation and Stability of Sodium Alginate Beads Containing ${\beta}-Carotene$ (베타카로틴 함유 알긴산 나트륨 비드의 제조 및 안정성)

  • To protect ${\beta}-carotene$ at the stomach and to release rapidly at the intestine we prepared alginate beads containing ${\beta}-carotene$. ${\beta}-carotene$ and alginate solution was homogenized and prepared o/w emulsion was prepared. It was poured into $Ca^{2+}$ solution through syringe needle. The gel was formed spontaneously and alginate beads containing ${\beta}-carotene$ were prepared. ${\beta}-Carotene$ was incorporated into the beads more than 95%. The release rate of ${\beta}-carotene$ was dependent on the concentration of $Ca^{2+}$, ${\beta}-carotene$ and surfactants. However, the concentration of alginate did not affect the release rate of ${\beta}-carotene$. The high concentration of $Ca^{2+}$ slowed down the release rate of ${\beta}-carotene$. The addition of surfactants in the ${\beta}-carotene$beads increased the release rate of ${\beta}-carotene$ in the order of Tween 80 > Cremophor > Span 20. The contents of ${\beta}-carotene$ and diameter of ${\beta}-carotene$ beads did not change significantly at $50^{\circ}C$ for 20 days.

Citric Acid Production from Glucose and Pumpkin by Using Immobilized Bead of Aspergillus niger (고정화 Aspergillus niger Bead를 이용한 포도당과 호박으로부터 구연산 생성)

  • The spore of Aspergillus niger KCTC-6144 was immobilized on alginate gel beads. When pumpkin powder was used with glucose for a medium of citric acid fermentation by Aspergillus niger beads, the beaded Aspergilus niger grew up inside the bead and mycelia penetrated through the pore of the bead membrane. The bead size became largely from 2.0∼2.5mm to 6∼8mm after growing at 30$\^{C}$ for 4 days. Studies of optimum culture conditions on citric acid fermentation using Aspergillus niger beads on pumpkin medium (pumpkin powder 1% +glucose 7%, pH 6.0) were carried out in submerged cultures on 250m1 Erlenmeyer flask. As a result, it was found that to reinforce 12% as carbon source was good for citric acid production and that 1% pumpkin powder was good as nitrogen and mineral source in orbital shaker (150rpm) at 30$\^{C}$ for 5 days. The optimum initial pH on citric acid production was pH 6.0 and it was found that 100 beads of immobilized Aspergillus niger was adequate for citric acid production in a 250ml Erlenmeyer flask containing 50m3 of pumpkin medium solution with orbital shaker at 30$\^{C}$ for 5 days. We also found that maximal production of citric acid was 23.5g/ℓ at optimal condition (at 30$\^{C}$ for 5 days, pH 6.0, and 100 beads and medium containing 1% pumpkin powder plus 12% glucose).
