Ecological Studies on Togyo Reservoir in Chulwon, Korea. 7. The Colonization of Epilithic Algae on Artificial Substrata (Tiles) at Mesocosm

  • Patterns of epilithic algal colonization on artificial substrata (unglazed ceramic tiles) were investigated from 23rd April to 3rd July 1999 at weekly intervals over a 10 weeks period outside and inside the mesocosm in Togyo reservoir within the Civilian Passage Restriction Line near Demilitarized Zone (DMZ) in Korea. The highest standing crops of epilithic algae was 1,798$\cdot$10³ cells$\cdot$$cm^{-2}$ outside the mesocosm on 26th June and also inside the mesocosm those was 2,391$\cdot$10³ cells$\cdot$$cm^{-2}$ on 26th June, 9 weeks after the experiment began. The dominants outside the mesocosm were Achnanthes minutissima, Navicula bicephala, Oscillatoria angusta, Synedra delicastissima, S. tenuissima, S. ulna v. danica and Tabellaria flocculosa, and those inside the mesocosm were Achnanthes minitissima, Coenochloris polycocca, Fragilaria crotonenesis, Peridinium cinctum, Synedra delicatissima, Tabellaria flocculosa and Ulothrix subtilissima. Diatoms were most abundant and Achnanthes minutissima was the most important species colonizing on the tiles. Chlorophyll-a content was highest value of 5.4 mg$\cdot$$m^{-2}$ on 19th June after 8 weeks growth outside the mesocosm and was 24.4 mg$\cdot$$m^{-2}$ on 26th June, 9 weeks after the experiment began on tiles inside the mesocosm. It was also shown that unglazed ceramic tiles were a more suitable substratum for colonization than the glass slides. Consequently the substratum selection plays an important role in the colonization by the epilithic algal community.

Invasion potential of Kappaphycus alvarezii on corals at Kurusadai Island, Gulf of Mannar, India

  • The marine red alga Kappaphycus alvarezii is a major source of $\kappa$-carrageenan. It has been introduced in 20 countries including India. Recently, several reports have expressed concerns about Kappaphycus invasion on Acropora corals at Kurusadai Island in the Gulf of Mannar, India, which is part of a marine bioreserve. To understand the extent of the Kappaphycus invasion, 27 randomly selected locations around Kurusadai Island and the mainland coast were surveyed during May-August 2008 and July 2009. Our rigorous sampling revealed that K. alvarezii was confined to two different patches of 105 m $\times$ 55 m and 8 m $\times$ 9 m located at the southeastern part of Kurusadai Island. The actual extent of the K. alvarezii canopy coverage was 76.7 $m^2$, accounting for less than 0.0035% of the total coral reef area. The daily growth rate of the K. alvarezii at Kurusadai was 0.7%. K. alvarezii was not observed in the coral reef area of the adjoining Pullivasal and Poomarichan Islands or the Palk Bay area cultivation sites. The lack of functional reproductive cycle, low spore viability, and the absence of microscopic phases in the life cycle of this alga coupled with the abundance presence of herbivores may restrict the further spread of this alga, so its invasive potential at Kurusadai Island is considered remote.

Polypropylene Bundle Attached Multilayered Stigeoclonium Biofilms Cultivated in Untreated Sewage Generate High Biomass and Lipid Productivity

  • The potential of microalgae biofuel has not been realized because of the low productivity and high costs associated with the current cultivation systems. In this study, a new low-cost and transparent attachment material was tested for cultivation of a filamentous algal strain, Stigeoclonium sp., isolated from wastewater. Initially, the different materials tested for Stigeoclonium cultivation in untreated wastewater were nylon mesh, polyethylene mesh, polypropylene bundle (PB), polycarbonate plate, and viscose rayon. Among the materials tested, PB led to a firm attachment, high biomass (53.22 g/m2, dry cell weight), and total lipid yield (5.8 g/m2) with no perceivable change in FAME profile. The Stigeoclonium-dominated biofilm consisted of bacteria and extracellular polysaccharide, which helped in biofilm formation and for effective wastewater treatment (viz., removal efficiency of total nitrogen and total phosphorus corresponded to ~38% and ~90%, respectively). PB also demonstrated high yields under multilayered cultivation in a single reactor treating wastewater. Hence, this system has several advantages over traditional suspended and attached systems, with possibility of increasing areal productivity three times using Stigeoclonium sp. Therefore, multilayered attached growth algal cultivation systems seem to be the future cultivation model for large-scale biodiesel production and wastewater treatment.

Effect of Microalgal Species on Nauplii Production in the Benthic Copepod Tigriopus japonicus (저서성 요각류 Tigriopus japonicus의 nauplii 생산에 미치는 미세조류의 영향)

  • The survival and growth of marine benthic invertebrate larvae such as abalone depend on the nutritional value of micro algae. However, it is difficult to determine the dietary value of the many microalgal species used for food by benthic larvae. Therefore, we tested the benthic copepod, Tigriopus japonicus, which grazes microalgae on substrata in a manner similar to abalone larvae. It also has short generation time and is easy to rear which makes to be easier to examine the dietary value of each micro algal species. We measured the daily production of nauplii from gravid females of T. japonicus fed 26 microalgal species separately. Amino acid and fatty acid content of the micro algae and the copepod was also analyzed. The nauplii production of T. japonicus was the highest (10.7) when they were fed Navicula sp. (B-394) and the lowest (0.8) when they were fed Scrippsiella trochoidea. In Tetraselmis suecica the nauplii production was so high (8.2), which was not significantly different with the diatom group. We determined that Navicula sp. (B-394), Rhaphoneis sp. and T. suecica were good sources of food for T. japonicus. We suggest that a diet of with a mixture of these three micro algal species may be also good for invertebrate larvae such as abalone.

Spatio-temporal variabilities of nutrients and chlorophyll, and the trophic state index deviations on the relation of nutrients-chlorophyll-light availability

  • The object of this study was to determine long-term temporal and spatial patterns of nutrients (nitrogen and phosphorus), suspended solids, and chlorophyll (Chl) in Chungju Reservoir, based on the dataset of 1992 - 2013, and then to develop the empirical models of nutrient-Chl for predicting the eutrophication of the reservoir. Concentrations of total nitrogen (TN) and total phosphorus (TP) were largely affected by an intensity of Asian monsoon and the longitudinal structure of riverine (Rz), transition (Tz), and lacustrine zone (Lz). This system was nitrogen-rich system and phosphorus contents in the water were relatively low, implying a P-limiting system. Regression analysis for empirical model, however, showed that Chl had a weak linear relation with TP or TN, and this was mainly associated with turbid, and nutrient-rich inflows in the system. The weak relation was associated with non-algal light attenuation coefficients (Kna), which is inversely related water residence time. Thus, values of Chl had negative functional relation (R2 = 0.25, p < 0.001) with nonalgal light attenuation. Thus, the low chlorophyll at a given TP indicated a light-limiting for phytoplankton growth and total suspended solids (TSS) was highly correlated (R2 = 0.94, p < 0.001) with non-algal light attenuation. The relations of Trophic State Index (TSI) indicated that phosphorus limitation was weak [TSI (Chl) - TSI (TP) < 0; TSI (SD) - TSI (Chl) > 0] and the effects of zooplankton grazing were also minor [TSI (Chl) - TSI (TP) > 0; TSI (SD) - TSI (Chl) > 0].

Distribution of Alexandrium tamarense in Drake Passage and the Threat of Harmful Algal Blooms in the Antarctic Ocean

  • While phytoplankton diversity and productivity in the Southern Ocean has been widely studied in recent years, most attention has been given to elucidating environmental factors that affect the dynamics of micro-plankton (mainly diatoms) and nano-plankton (mainly Phaeocystis antarctica). Only limited effects have been given to studying the occurrence and the potential risks associated with the blooming of dinoflagellates in the relevant waters. This study focused on the appearance and toxicological characteristics of a toxic dinoflagellate, Alexandrium tamarense, identified and isolated from the Drake Passage in a research cruise from November to December 2001 The appearance of A. tamarense in the Southern Ocean indicates the risk of a paralytic shellfish poisoning (PSP) outbreak there and is therefore of scientific concern. Results showed that while the overall quantity of A. tamarense in water samples from 30meters below the sea surface often comprised less than 0.1% of the total population of phytoplankton, the highest concentration of A. tamarense (20 cells $L^{-1}$) was recorded in the portion of the Southern Ocean between the southern end of South America and the Falkland Islands. Waters near the Polar Front contained the second highest concentrations of 10-15 cells $L^{-1}$. A. tamarense was however rarely found in waters near the southern side of the Polar Front, indicating that cold sea temperatures near the Antarctic ice does not favor the growth of this dinoflagellate. One strain of A. tamarense from this cruise was isolated and cultured for further study in the laboratory. Experiments showed that this strain of A. tamarense has a high tolerance to temperature variations and could survive at temperatures ranging from $5-26^{\circ}C$. This shows the cosmopolitan nature off. tamarense. With regard to the algal toxins produced, this strain of A. tamarense produced mainly C-2 toxins but very little saxitoxin and gonyailtoxin. The toxicological property of this A. tamarense strain coincided with a massive death of penguins in the Falkland Islands in December 2002 to January 2003.

  • The green colonial algae Botryococcus braunii is characterized by unusual high hydrocarbon contents, ranging from 15 to 75% of dry weight, as long-chain unsaturated hydrocarbons. In two-phase bubble column using various organic solvents, poor recovery 08 - 32%) of hydrocarbon seems to be caused by insufficient mixing between two phases, which was operated using only aeration on the narrow interface between hydrophobic solvent and cell suspension. In addition, hydrocarbon was entrapped tightly in cell-matrix (formed by exopolysaccharide) of algal colony, which make difficult to extract using two-phase system. To improve recovery efficiency, mixed-solvent of extractive solvent (octane) and biocompatible solvent (octane) was tested in two-phase column for in situ extraction. In two-phase extraction culture using mixed-solvent, the algal growth was intensely inhibited even at low concentration of polar octanol solvent. the hydrocarbon recovery in two-stage cell-recycle extraction showed a 2.9 fold increase (57%) over that in two-phase extraction. Up to 60 % of hydrocarbon could be recovered without serious cell-damage in the case of downstream separation for 6 h at the high recycle flow rate using this process after batch culture.

Annual Fluctuation (2000 ${\sim}$ 2003) of Water Quality and Cyanobacterial Abundance in the Lower Part of Han-River (한강 하류의 남조류 및 환경요인의 연간 (2000 ${\sim}$ 2003) 변화에 대하여)

  • For 4 years (2000 ${\sim}$ 2003), annual fluctuations of standing crops of cyanobacteria and physicochemical factors were examined at five sites from Bridge of Seungsoo to Bridge of Seungsan in the lower part of Han River. The cyanobacterial abundance (ND to 4,167 cells $mL^{-1}$) was strongly decreased during the heavy rains in every year. During the similar periods in 2003, cyanobacteria hardly observed, and comprised below of 10 percentage of total phytoplankton. In the period of little cyanobacteria, some green algae and diatom dominated the phytoplankton community, while the concentration of chlorophyll a has not largely change. These results indicate that heavy frequent precipitation strongly limited the growth of cyanobacteria, and lead an algal succession by the appearance of new algal groups.

Retreatment of Arificial Wastewater by using Microalgae (조류를 이용한 인공하수의 재처리)

  • This study was performed in lab scaled oxidation pond. The removal efficiency of pollutant on the influence of changes of hydraulic retention time and pond style was investigated. The correlation between organic removal efficiency and dissolved oxygen concentration on algal photosynthesis showed the light time revealed a higher relationship more than the dark time, and the squares of the correlation coefficient of 15 days retention time were higher than that of 5 days in single pond. The variation of dissolved oxygen concentration of a series pond was from 4.2 to 19.8 mg/l under 5 days retention time, the concentration of dissolved oxygen increased with increasing step of series pond. Between the single pond and a series of pond system, a series of pond system showed better organic removal efficiency. Average removal efficiency range of $TBOD_5$ and $SBOD_5$ was $49{\sim}83%$ and $87{\sim}92%$, respectively. Algae should be removed appropriately to increase the removal efficiency of organic matter.

Differential Responses of Three Cyanobacteria to UV-B and Cd

  • Interactive effects of UV-B and Cd on growth, pigment, photosynthesis, and lipid peroxidation have been studied in Anabaena, Microcystis, and Nostoc; all the tested cyanobacteria showed differential sensitivity to different dosage of UV-B and Cd alone as well as in combination. Phycocyanin was severely affected by UV-B and Cd by all the strains; the degree of pigment bleaching was most pronounced in Anabaena followed by Microcystis and Nostoc. $UV-B_2+Cd_2$ produced nearly 83, 78, and 65% inhibition of phycocyanin in Anabaena, Microcystis, and Nostoc, respectively. The above treatment also significantly decreased the contents of Chl ${\alpha}$ and carotenoid. Presence of capsule in Microcystis protected the phycocyanin bleaching as compared to decapsulated cells. Laboratory-grown Microcystis revealed about 75 and 80% inhibition, following $UV-B_2+Cd_2$ treatment, respectively. in capsulated and decapsulated cells. Damage caused by Cd was more pronounced than UV-B. Inhibition of photosynthesis did occur in all the test strains, being maximum in Anabaena. PS II was the most sensitive component of the electron transport chain, showing 84, 80, and 70% inhibition in Anabaena, Microcystis, and Nostoc, respectively. As compared to control, significant lipid peroxidation (6.5-fold higher) was observed in Anabaena with $UV-B_2+Cd_2$, $^{14}C-uptake$ was more susceptible to Cd and Uv-B than oxygen-evolution. Approximately 84, 80, and 76% inhibition of $^{14}C-uptake$ was observed in Anabaena, Microcystis, and Nostoc, respectively. Similarly, $UV-B_2+Cd_2$ inhibited APT content of Anabaena by 87%. This ,study suggests that inhibition of carbon fixation was due to decreased ATP content of the test cyanobacteria by UV-B+Cd, where Anabaena was the most sensitive and Nostoc the most tolerant.
