Culture study on the hybrid by interspecific crossing between Porphyra pseudolinearis and P. dentata (Bangiales, Rhodophyta), two dioecious species in culture

  • Physiological studies on the hybrid by crossing between two dioecious species, Porphyra pseudolinearis and P. dentata from Korea were conducted at constant temperatures (5, 10, 15, 20, and $25^{\circ}C$), at photon flux densities (10, 20, 40, and $80\;{\mu}mol\;m^{-2}s^{-1}$) under photoperiods (14 L : 10 D and 10 L : 14 D). In the hybrid, higher growth of conchocelis was observed at 20 and $40\;{\mu}mol\;m^{-2}s^{-1}$ under 14 L : 10 D. Conchosporangial branches were produced under $10-80\;{\mu}mol\;m^{-2}s^{-1}$ at only $25^{\circ}C$, and were abundant when the conchocelis was cultured under 10 L : 14 D. Foliose thalli of the hybrid grew well at the conditions of $10-20^{\circ}C$, 10 L : 14 D and $15-20^{\circ}C$, 14 L : 10 D. The foliose thalli grew very slowly at $5^{\circ}C$ and $30^{\circ}C$, respectively. No archeospores were observed at any culture conditions. Spermatangial and zygotosporangial sori were formed at the marginal portion of mature thallus. Zygotospores from the hybrid were released at $10-2^{\circ}C$ under both photoperiods, and gave rise to form conchocelis filament. Monoecious thalli were observed at $10^{\circ}C$ under 14 L : 10 D. Neither monospores nor protothalli were produced from the conchocelis in culture.

Development of a tide-simulating apparatus for macroalgae

  • A tide-simulating apparatus was developed for culturing marine macroalgae. The objective of this study was to introduce a novel tide-simulating apparatus that can simulate a diurnal or semi-diurnal tidal cycle in the laboratory. In this apparatus, the seaweeds are move up and down and the water level remains the same during the simulated tidal cycle. The apparatus consists of 18 cylindrical culture tanks (3 blocks $\times$ 6 culture tanks) with 12 cm diameter and 24.5 cm long containing up to 2.5 L of seawater. There is a horizontal plate which covered all 18 culture tanks, and it is raised and lowered by a programmable motor that can regulate exposure time. In one application, seaweeds are attached to braided twine hung on Plexiglas air-tubing. The air-tubing is attached to a lid that is set on a horizontal plate. This apparatus is made of colorless Plexiglas to maximize light transmittance. This apparatus is easily disassembled and transportable to any indoor laboratory, wet laboratory, greenhouse, etc. This apparatus also offers considerable flexibility in terms of design. The size of culture tank can be redesigned by either increasing the height of cylinder or/and using a different diameter of cylindrical Plexiglas, therefore, larger/taller thalli can be cultivated. Growth rates of three eulittoral Porphyra species from different tidal elevations have been compared using this device.

초분광영상을 이용한 서낙동강 조류 발생현황 분석에 관한 연구 (A study on the analysis of current status of Seonakdong River algae using hyperspectral imaging)

  • 조류는 수생태계에서 소비자의 에너지를 공급하여 생태계 내 없어서는 안 될 1차 생산자로서 크게 녹조류, 남조류, 규조류로 나뉘어진다. 남조류의 경우 수온이 상승하여 여름철 발생하여 과대 증식하여 녹조현상의 주원인이 되며, 최근 기후 변화로 인해 녹조현상의 발생시기의 변화와 빈도수가 늘고 있는 추세이다. 기존의 조류 조사 방식은 채수 및 센서를 통한 측정으로 이루어지고 있으며 시간, 비용 및 인력의 한계가 나타난다. 이러한 기존 모니터링 방법의 한계를 극복 하기위해 위성영상이나 무인항공기(Unmanned Aearial Vehicles, UAV), 등 탑제체를 운용한 다중분광 및 초분광과 같은 분광기기를 이용하여 원격 모니터링을 수행하는 연구가 진행되어 왔다. 본 연구에서는 조류 배양액 및 하천수 채수를 통한 실험실 규모의 실험을 통해 원격 모니터링의 종 구분에 대한 가능성에 대하여 확인 해보고자 하였다. 초분광 영상을 취득하기 위해 400-1000 nm에서 분석할 수 있는 초분광 센서를 활용하였다. 채수한 하천수의 조류 종 구분을 위한 분광특성을 추출하기 위해 GF/C필터를 이용하여 여과를 진행하여 시료를 제조하여 영상을 수집하였다. 수집된 영상을 방사보정 및 Base (하천수 및 배양보존액 통칭) 제거를 진행하였고 조류의 분광 정보 추출 과정을 통해 시료별 분광 정보를 추출, 분석하여 조류의 분광특성을 파악, 비교분석하여 하천·호소에서의 초분광영상 기반 원격탐사 모니터링의 적용성을 검토하고자 하였다.

Caloglossa beccarii (Delesseriaceae, Rhodophyta) from freshwater rivers in Kerala, India, a critical new record

  • Caloglossa species occurs in freshwater streams around Southest Asia. We report it from 2 different riverine sites in Kerala, India. Tetrasporangiate plants were observed in field collections from the Periyar River and Chalakkudy River. The Chalakkudy isolate did not reproduce in culture but the Periyar isolate developed abundant tetrasporangial sori in culture. Many spores were discharged and most were abortive, but some germinated normally, sporelings forming male gametophytes with numerous spermatangial sori and females with many procarps, viable carposporophytes and some nonfunctional (no carpospores) pseudocystocarps. Some carpospores germinated forming new tetrasporophytes. Molecular evidence (28S rDNA and rbcL) placed the Indian specimens close to C. beccarii and C. fluviatilis. Considering the freshwater habitat and morphology of vegetative thalli (blade shape, rhizoid arrangement, and number of rhizoid filament per cell), the Indian specimens should be assigned to C. beccarii.

Laboratory, Field and Deep Seawater Culture of Eucheuma serra-a High Lectin Yielding Red Alga

  • The red seaweed Eucheuma serra is a high yielding source of lectins. The plants were collected from a depth of 5-6 meters and cultured in the laboratory, field and deep seawater. A Daily Growht Rate (DGR) of 3.5% was observed at 18℃ with a low light of 30μmol photon $ m^{-2} · s^{_1}$ in the laboratory. When the plants were cultured in the field at different depths during winter onths of December and January, best growth was observed at 1 m depth and a DGR of 2.14±0.04% was recorded. The plants grown in the tank with a continuous supply of deep seawater showed a DGR of 8.2% The results indicate that E. serra can be cultivated in large scale both in deep seawater in the tank and in the field for the extraction of lectins at a commercial scale.

Reproduction of Rhodochorton purpureum from Jeju Island, Korea and SanJuan Island, Washington, USA in Laboratory Culture

  • Rhodochorton purpureum 4187 from Jeju Island, Korea may have a sexual life history similar to that seen by other investigators working on other strains around the world. In culture short days (8:16, 11:13, 12:12 LD) at 10-15°C induced tetrasporogenesis. Discharged spores were observed with time lapse videomicroscopy. They showed a slight amoeboid movement for 2-3 minutes before rounding up and settling. Tetrasporelings develop into male and female gametophytes. No fertilisation was observed. Tetrasporangia often were borne on carpogonial clusters of females but no discharged spores were seen. Isolate 4241 from San Juan I., Washington, USA grew well in most conditions tested but did not reproduce in short days (8:16, 11:13, 12:12 LD) at 10-15°C.

Unusual Rhizoidal Development in Bangia (Bangiales, Rhodophyta) -Another Form of Vegetative Reproduction?

  • The gametophytic filaments of two genetically distinct taxa of Bangia from New Zealand showed unusual rhizoidal development in comparative culture experiments. In the past Bangia has been reported to possess simple, colourless rhizoids that extend from the basal cells of the unbranched filaments, whereas in this study the rhizoids observed became pigmented and multicellular. A reversal of growth direction occurred and filamentous extensions developed from the rhizoids under some culture conditions. These extensions were either prostrate or resembled new gametophytic filaments. This is the first report for filamentous members of the Bangiales of the development of such stolon-like rhizoids, apparently serving as a form of vegetative reproduction.

Observations on some mangrove-associated algae from the western Pacific (Guam, Chuuk, Kosrae, and Pohnpei)

  • The mangrove algal flora of Guam and the Federated States of Micronesia has been poorly explored. We add to our knowledge of this region by observations of collections from these regions. This paper presents new and additional records of: Rhodophyta-Acrochaetium globosum, Colaconema sp., Caulacanthus indicus, Bostrychia moritziana / B. radicans, B. radicosa, B. simpliciuscula, B. kelanensis and B. tenella, Murrayella periclados, and Caloglossa ogasawaraensis; Chlorophyta-Boodleopsis carolinensis; and Phaeophyceae-Dictyota adnata, Dictyotopsis propagulifera, and Canistrocarpus cervicornis. Most specimens were cultured to investigate their reproductive biology and many specimens were further identified using molecular data. Low molecular weight carbohydrates (dulcitol, sorbitol, and digeneaside) were identified in samples of B. radicosa and B. simpliciuscula. We also present data on manganese-rich deposits found on B. simpliciuscula and B. tenella in culture, possibly formed by epiphytic bacteria.
