The effect of herbal mixture on alcohol metabolism in Sprague Dawley rats (알코올을 투여한 흰쥐의 알코올 대사에 미치는 한약재 혼합추출물의 영향)

  • Objectives : Alcohol hangover is a common phenomenon which basically occurs after heavy drinking. Moreover, heavy alcohol consumption leads to acute and chronic diseases. We investigated the effect of herbal mixture (SJ) on alcohol metabolism in serum or/and liver. Methods : 5W-old Sprague Dawley rats were used for the study. To overnight fasted rats, 0.9% saline or SJ extract was administrated per os before alcohol treatment. Then, 40% alcohol was orally administrated to all rats in 30 mins. Ethanol, acetaldehyde concentrations, alcohol dehydrogenase (ADH), acetaldehyde dehydrogenase (ALDH) activities were measured by assay kits. Aspartate aminotransferase (AST) and alanine transaminase (ALT) were measured by analyzer. Results : Ethanol and acetaldehyde concentrations lowered in SJ groups compared with CON group. Especially, acetaldehyde concentration significantly decreased in SJ groups compared with CON group. AST and ALT levels tended to increase in SJ groups compared with CON group but there was no significant difference between CON group and SJ groups. ADH activity in serum was higher in SJ groups than CON group but no significant difference in liver. ALDH activity in both serum and liver showed significantly increased in SJ groups compared with CON group. Conclusions : Treatment of SJ extract showed not only reducing concentration of alcohol and acetaldehyde but also increasing activities of ADH and ALDH. These results suggest SJ may influence in alcohol metabolism via control of metabolic enzyme activities and metabolite. Therefore, SJ, herbal mixture, might have a function of preventing hangover after drinking alcohol.

Activity of Glutathione S-transferase and Effect of Alcohol Decomposition on the Fruit of Hovenia dulcis Thunb (지구자의 Glutathione S-transferase 활성과 알코올 분해효과)

  • For the determination of glutathione S-transferase activity, a new method was established by using HPLC system. Moreover, amount of enzyme for a optimum reaction was determined by a comparative study with a variety concentration of enzyme. Using a established method, activity of glutathione S-transferase that is alcohol metabolizing enzyme was investigated on the fruit of Hovenia dulcis Thunb. As the result of experiment, EtOH and $H_2O$ extracts of the fruit of Hovenia dulcis Thunb showed visible a synergistic effect of glutathione S-transferase activity. On a continuous experiment, EtOH and $H_2O$ extracts of the fruit of Hovenia dulcis Thunb showed alcohol decomposition activity on the in vivo test using rat. These results suggest that the fruit of Hovenia dulcis Thunb may be useful in the prevention of hangover.

Effects of mixed supplementation on Hoveni dulcis Thunb extracts and Ginseng-Berry extracts on hangover curves (헛개과병추출물과 인삼열매추출물의 혼합 음료 섭취가 숙취해소에 미치는 효과)

  • The purpose of this study was to investigate the effects of ingestion of rabies and ginseng fruit extracts on alcohol hangover, liver damage protection, fatigue recovery, and physical strength improvement. A total of 64 volunteers aged over 20 were participated in this study and the randomized and repeated measures design method was used to divide a group of participants with a random assignment. All participants were divided into 4 groups (n=16) treated with hoveni dulcis thunb extract + ginseng berry extract (ARI 1000), hoveni dulcis thunb extract, ginseng berry extract, and placebo. As a result of respiratory alcohol concentration change, the group treated with ARI 1000 was significantly lower than the group treated with hoveni dulcis thunb extract, ginseng berry extract, and placebo in 1 hour of drinking, and significantly lower than the placebo group in 2 hours and 3 hours of drinking (p<0.05). After 2 and 3 hours of alcohol consumption, blood alcohol concentration of the group treated with rabies ARI 1000 was significantly lower than those of the other 3 groups (p <0.05). In conclusion, ingestion of ARI 1000 before drinking may significantly reduce the respiratory and blood alcohol concentrations, which may induce an effect on the hangover effect.

Ethanol metabolism in liver involves both liver alcohol dehydrogenase(ADH) which actalyzes the oxidation of ethanol to acetaldehyde, and liver aldehyde dehydrogenase(ALDH) which metabilized rapidly acetaldehyde, product of ethanol oxidation, to acetate. Effect of a Soy-Sprout Beverage Prepared with High-Concentrated Oxygen Water on Alcohol Metabolism in Rats (고농도 산소수로 재배한 콩나물 추출 음료의 알코올 분해 효능)

  • This study was conducted to investigate the detoxification effect of extract from soy-sprout grown using high concentrated oxygen water extract (SE) against alcohol-induced hangover in male Sprague-Dawley (SD) rats. The rats were orally administered with different concentrations of SE beverage [26, 260 and 2,000 mg/kg body weight (b.w.)] and, after 30 min, with alcohol at a dose of 3 g/kg b.w. After 1 and 3 h of alcohol administration, blood was collected from the retro-orbital plexus, while after 5 h, blood was collected from the heart. In the 2,000 mg/kg b.w. SE group, the concentration of blood alcohol was significantly reduced after 1-5 h of alcohol loading as compared with that in the other groups. In addition, the blood acetaldehyde concentration was reduced by SE (2,000 mg/kg b.w.). These results suggest that SE beverage can alleviate alcohol hangover symptoms by stimulating the activities related to hepatic alcohol-metabolizing enzymes.

Characteristics of Volatile Compound Adsorption from Alcoholic Model Solution onto Various Activated Carbons (알코올모델용액을 이용한 여러 종류 활성탄의 휘발성화합물 흡착특성)

  • Ten commercial activated carbons (ACs) prepared from four different sources (bamboo, wood, peat, and coal) were evaluated for their adsorptive efficiency of six volatile compounds (isoamyl alcohol, hexanal, furfural, ethyl lactate, ethyl octanoate, 2-phenyl ethanol) which were dissolved in a 30% alcoholic model solution. These six volatile compounds are frequently found in alcoholic beverages and possibly contribute to physiological hangover due to their high concentrations. They are also generally regarded as off-flavor compounds at certain levels in alcoholic beverages such as whisky and vodka. Two hundred mL of 30% alcoholic solutions containing these six volatile compounds were treated with 0.2 g of ACs while stirring for 16 hr; the treated solutions were then measured for their adsorptive efficiencies (or removal efficiencies) by gas chromatographic analysis using two different sampling methods (direct liquid injection and headspace-solid phase microextraction). The adsorptive efficiencies of the ACs varied depending on the identity of the volatile compounds and the source material used for making the ACs. Ethyl octanoate, 2-phenyl ethanol, and hexanal were removed at high efficiencies (34-100%), whereas isoamyl alcohol, ethyl lactate, and furfural were removed at low efficiencies (5-13%). AC prepared from bamboo showed a high removal efficiency for isoamyl alcohol, aldehydes (hexanal and furfural), and 2-phenyl ethanol; these major fusel oils have been implicated as congeners responsible for alcohol hangover.

  • Hangover is associated with alcohol metabolism in body after the ingestion of an alcoholic beverage. It has been known that hangover is caused by increasing blood acetaldehyde concentration. This study was carried out to evaluate effect against blood ethanol(EtOH) and acetaidehyde(AcH) on the seven natural products samples(Lotus seed, Sweet chest nut rose, Kohki, Gurume-K, Gurume-J, Phytic acid and Chlorophyll). Also, samples which were selected as good products were mixed. Then, effect against blood EtOH and AcH formulas were tested in vivo. (omitted)

  • The side effects, such as belching and headache, after Makgeolli intake are an obstacle to the development of the Makgeolli industry. The side effects of drinking Makgeolli have many causes. The possibility of BA production by incorrect storage conditions cannot be excluded. This study analyzed the BA contents produced in non-sterilized Makgeolli after 5 days storage at 4 and $20^{\circ}C$. BA was not detected in Makgeolli stored at $4^{\circ}C$, but putrescine and tyramine were detected in $20^{\circ}C$. A drinking test was performed to determine the relevance of the BA contents and hangover symptoms. The results revealed no significant difference in the taste score and symptoms score between 4 and $20^{\circ}C$ storage. Therefore, the results suggest that the presence of BA in Makgeolli produced during room temperature storage for 5 days does not induce or strengthen hangover symptoms. On the other hand, the alcohol and BA dose may be insufficient to represent the normal symptoms of a hangover.

Effects of Improving Alcohol Metabolism of Yeast Extract Mixtures and Hovenia dulcis Mixtures in Healthy Men - A Double-Blind, Randomized Crossover, Placebo-Controlled Trial - (효모추출복합물과 헛개나무 열매추출 복합물이 알코올 대사에 미치는 영향 - 무작위, 이중맹검, 위약 대조군, 교차 인체적용시험 -)

  • This study was conducted to investigate if the supplement formula may improve alcohol metabolism in healthy adult men. In a double-blinded, randomized, crossover study, subjects were administrated yeast extract mixtures (YEM, n=15), Hovenia dulcis mixtures (HDM, n=15), placebo (PLA, n=15), and control (CON, n=15) in an oral dose followed by one week washout periods. At each visit (0, 1, 2, 3, 4 week), subjects drank 450 mL, 20.1 percent alcohol after administered mixtures. Blood was drawn periodically (0, 0.25, 0.5, 1, 2, 4, 6, 15 hours). Fifteen subjects completed the protocol and were included in the analysis. Plasma ethanol concentration was lower in YEM (10 percent) and the HDM (5 percent) groups. The area under the curves (AUC) and $C_{max}$ for plasma ethanol were significantly decreased only in the YEM group, when compared with the CON group. The AUC and $C_{max}$ for plasma acetaldehyde concentration were significantly decreased in the YEM (45 and 54 percent) and the HDM (35 and 53 percent) groups respectively, when compared with PLA (p<0.01). Together, these findings validate that YEM or HDM improved alcohol metabolism and hangover syndromes, leading to reduce alcohol concentration and acetaldehyde concentration without adverse effects.

Anti-inflammation and hangover relief effects of Schisandra chinensis (SC) and Lycium chinense (LC) water extracts depending on drug processing and fermentation (포제 및 발효 가공에 따른 오미자와 구기자 물 추출물의 항염증 및 숙취해소 효과)

  • Schisandra chinensis (SC) and Lycium chinense (LC) were widely distributed in Asia and the fruit has been used traditionally for medicinal herbs. The processing method was solid-state fermentation using Aspergillus oryzae for 48 h after stir-frying treatment at $220^{\circ}C$ for 12 min. In this study, in vitro the anti-inflammatory effect and in vivo hangover reduction were compared to unprocessed SC and LC water extract. Anti-inflammatory effects have been evaluated in pro-inflammatory mediators which were secreted by lipopolysaccharide (LPS)-induced RAW 264.7 macrophages. Nitric oxide (NO) was determined using Griess reaction. Proinflammatory cytokines such as tumor necrosis factor $(TNF)-{\alpha}$ and interleukin $(IL)-1{\beta}$ were measured by enzyme-linked immunosorbent assays (ELISA). Alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities were compared to processed SC or LC and mixtures thereof (1:1). In vivo study was compared to hangover relief in alcohol-fed mice. After administering a mixture of SC and LC (300 mg/kg) water extract (1:1), mice were fed 3 g/kg of ethanol. Serum was collected at 1, 3, and 5 h intervals to analyze ethanol and acetaldehyde levels using a colorimetric assay kit. The processed SC and LC water extracts compared to raw materials significantly inhibited LPS-induced NO and inflammatory cytokine production in RAW 264.7 cells. The results of the hangover mouse model are also consistent with anti-inflammatory effects. These results suggest that processed SC and LC extracts may be functional materials for the treatment of inflammation and hangover.
