Effects of Various Levels of Cassava Hay on Rumen Ecology and Digestibility in Swamp Buffaloes

  • Four, mature rumen fistulated male swamp buffaloes with an average initial weight of $426{\pm}25kg$ were randomly allocated to receive dietary treatments according to a $4{\times}4$Latin square design. Four dietary treatments with varying proportions of ureatreated rice straw (UTRS) and cassava hay (CH) were offered (100:0, 75:25, 50:50, 0:100). Each feeding period lasted 21 days, the first 14 days for feed adjustment and intake measurement and the final 7 days for rumen fluid and faecal collections. The results demonstrated the potential use of both UTRS and CH as roughage sources. As levels of CH increased in the diets pH values were maintained (6.5-7.0). Ruminal $NH_3-N$ concentrations were significantly (p<0.05) raised as higher levels of CH were incorporated into the diets. Moreover, cellulolytic and proteolytic bacterial populations were enhanced while total protozoal counts were decreased (p<0.05). In addition, DM, OM and CP digestibilities and their digestible intakes as well as estimated energy increased with increasing levels of CH in the diets. The results suggest a favorable effect of CH substituting for UTRS.

Supplementing Maize or Soybean Hulls to Cattle Fed Rice Straw:Intake, Apparent Digestion, In situ Disappearance and Ruminal Dynamics

  • Steers with ad libitum access to rice straw were assigned to four diets to evaluate the effects of maize or soybean hull supplementation on intake, in vivo digestibility, ruminal pH, VFA, ammonia-nitrogen ($NH_3-N$) and in situ ruminal disappearance of feed nutrients by cattle consuming rice straw. Supplement treatments were: no supplement (RS); soybean meal at 0.127% BW (SBM); cracked maize at 0.415% BW plus 0.044% BW soybean meal (MAIZE); or soybean hulls at 0.415% BW plus 0.044% BW soybean meal (HULLS). The MAIZE and HULLS diets were formulated to provide approximately 4 MJ of $NE_m$ per kg of diet. Rice straw DMI was not affected (p = 0.34) by supplement. Apparent dry matter (DM) digestibility was greater (p<0.001) for MAIZE and HULLS (56.6 and 60.0%, respectively) than for steers consuming SBM or RS (51.8 and 44.4%, respectively). Apparent NDF digestibility was greater (p<0.0004) for HULLS than MAIZE (61.7 vs. 58.0%, respectively) and apparent ADF digestibility was greater (p<0.0008) for HULLS than MAIZE (61.1 vs. 49.2%, respectively). There was no difference in apparent hemicellulose digestibility (p = 0.43). Analysis of ruminal fluid collected 0, 2, 4, 6, and 8 h post-feeding revealed ammonia-nitrogen was greatest (p<0.05) for steers on SBM and HULLS diets at 2 h (24.08 and 22.57 mg/dl, respectively) and total volatile fatty acids was greatest (p<0.05) for HULLS at 4 h (230 mM/L). In situ disappearance, measured at 0, 2, 4, 6, 8, 16 and 24 h, indicated that SBM, MAIZE and HULLS tended to enhance the digestibility of DM and fiber components of rice straw. In situ disappearance of rice straw DM was greatest for SBM and/or HULLS from 4 to 24 h (p = 0.03). Rice straw NDF and ADF disappearance was enhanced by supplementation from 16 to 24 h (p<0.02). Rice straw DM, NDF and ADF disappearances at 24 h were similar for MAIZE and HULLS treatments. When feeding cattle rice straw diets, energy and protein-based supplements are essential. This study showed that fiber-based supplements are just as, if not more, effective as starch-based supplements in rice straw utilization. This study shows that soybean hulls, in spite of their high fiber content, are as efficient as maize for supplementing rice straw primarily because fiber in soybean hulls is highly digestible as shown by in vivo digestibility and in situ disappearance.

Effects of Saccharomyces cerevisiae Supplementation and Anhydrous Ammonia Treatment of Wheat Straw on In-situ Degradability and, Rumen Fermentation and Growth Performance of Yearling Lambs

  • The effects of Saccharomyces cerevisiae supplementation ($6.6{\times}10^8cfu$) and anhydrous ammonia treatment (3%) of wheat straw (WS) were investigated on in-situ dry matter (DM) degradability, and on rumen fermentation and growth performance of lambs. Rumen-fistulated Menemen sheep fed a diet with and without live yeast were used to assess the DM degradability characteristics of WS and ammonia-treated wheat straw ($WS_{NH3}$). Twenty-six yearling Menemen male lambs were fed in four groups. Lambs of control group (WS) received untreated WS without supplemental yeast, whereas other three groups were fed WS treated with anhydrous ammonia ($WS_{NH3}$ group), untreated WS and yeast (WS+YEAST group) or WS treated with anhydrous ammonia and yeast ($WS_{NH3}$+YEAST group). Supplemented live yeast (4 g/d) was added in the diet. Lambs were offered untreated or ammonia treated WS ad-libitum and concentrate was fed at 1% of live body weight. The degradability of the water-insoluble (fraction B) was significantly increased by all of the treatment groups. Potential degradability (A+B), effective DM degradability's (pe2, pe5, and pe8) and average daily weight gain increased only in $WS_{NH3}$+YEAST group (p<0.05). Voluntary DM intake was not increased by the treatments (p>0.05), but voluntary metabolizable energy and crude protein intake were increased by $WS_{NH3}$ and by $WS_{NH3}$+YEAST (p<0.05). Average daily rumen pH was not affected by any of the treatments, but average daily $NH_3$-N was significantly higher in the $WS_{NH3}$ and $WS_{NH3}$+YEAST groups, and total volatile fatty acids were significantly higher in the WS+YEAST and $WS_{NH3}$+YEAST groups. In conclusion, the improvement of feed value of WS was better by the combination of ammonia-treatment and yeast supplementation compared to either treatment alone.

생쥐 수정란의 유리화 동결보존에 있어서 동결전 처리방법에 관한 연구 (Effects of Equilibration Time, Precooling and Straw Loading Method on Survival of Mouse Embryos Frozen by Vitrification)

  • 생쥐 수정란의 동결보존법을 개선하고자 유리화 동결과정에서 제1보존액에서의 평형시간, 제2보존액에서의 전냉각 여부 및 straw loading 방법에 따른 동결보존 후 수정란의 생존율을 조사하고, 또한 유리화 동결보존 과정에서 수정란의 부분적인 손상 여부를 확인하기 위하여 동결보존 후 배양하여 수정란의 할구수를 Hoechst 33342로 염색하여 조사하였던 바 그 결과는 다음과 같다. 1. 동결융해 후 생존율은 Medium-1에서 10분간 평형시간을 준 경우(81.0%)는 5분간(40.0%) 및 15분간(74.1%)의 평형시간을 준 경우보다 유의적(P<0.05)으로 높았다. 2. Double Medium-2 column방법으로 얻은 생존율(81.0%)은 single Medium-2 column방법으로 얻은 생존율(62.8%)보다 유의적(P<0.01)으로 높았다. 그리고 Double Medium-2 column방법에서는 유리화 용액이 희석액에 오염되지 않아 순수하게 투명한 유리화를 이룰 수 있었다. 3. 전냉각을 않은 경우에 비하여 전냉각한 Medium-2로 동결한 수정란은 생존율에 있어서 다소 높은 성적을 보였으나 유의적인 효과는 없었다. 4. 상실배기의 수정란을 24~28시간동안 배양하여 얻은 후기 배반포를 Hoechst 33342로 염색하여 할구수를 조사하였던 바 신선란(53.3${\pm}$1.6)보다 동결란(41.4${\pm}$1.5)에서 유의적(P<0.05)으로할구수가 적었다. 이는 동결융해 과정에서의 부분적인 손상에 의하였던 것으로 사료된다.

볏짚과 규회석(珪灰石) 시용(施用)이 수도(水稻)의 생육(生育) 및 수량(收量)에 미치는 효과(效果) (Effects of Rice Straw and Wollastonite Application on the Growth and Yield of the Rice Plant)

  • 수도(水稻)의 생육(生育)과 수량(收量)에 미치는 볏짚과 규회석(珪灰石)의 효과(效果)를 규명코져 볏짚은 무시용(無施用)과 88g/pot(1000kg/10a)로 2수준, 규회석(珪灰石)은 무시용(無施用)(50ppm), 27(150ppm), 85(300ppm), 135g/pot(450ppm조절량) 4수준을 각각 다르게 처리하고 pot 시험(試驗)을 한 결과(結果)는 다음과 같다. 1. 볏짚이나 규회석(珪灰石) 단용(單用)을 수도(水稻)의 초기생육(初期生育)을 억제(抑制)시켰으나 이들의 병용구(倂用區)에서 이상적인 초형(草型)으로 발달(發達)되어 특히 최고분운기(最高分芸期) 이후(以後)에 효과(效果)가 매우 커서 정조수량(正租收量)이 증가(增加)되었다. 2. 볏짚 무시용구(無施用區)는 규회석(珪灰石) 27g/pot구(區)에서 모두 정조수량(正租收量)이 많았고 볏짚 시용구(施用區)에서는 시용당년(施用當年)에는 규회석(珪灰石) 85g/pot구(區), 그리고 다음해의 잔효구(殘效區)에서는 규회석(珪灰石) 135g/pot구(區)에서 정조수량(正租收量)이 가장 많아 볏짚시용(施用)은 규산(硅酸)의 요구량(要求量)을 증가(增加)시키는 것으로 나타났다. 3. 처리간(處理間)에 최고수량(最高收量)을 낸 구(區)는 시비년도(施肥年度)에는 볏짚+규회석(珪灰石) 85g/pot구(區)로서 대조구(對照區)보다 25%의 정조수량(正租收量)이 증대(增大)되었다. 볏짚과 규회석(珪灰石)의 잔효(殘效)를 본 익년도(翌年度)에서는 볏짚+규회석(珪灰石) 135g/pot구(區)가 대조구(對照區)보다 30%의 증가(增加)를 보여 볏짚이나 규회석(珪灰石) 단용(單用)보다 병용효과(倂用效果)가 컸었다.

  • Forty cross bred ($168\;{\pm}\;28\;kg$) bull calves were offered either untreated (US) or urea ammonia treated (TS) rice straw at five levels of feeding, namely; 60, 80, 100, 120 140% of ad libitum. Ad libitum level for each animal was estimated over a period of one month, which was followed by a preliminary period of 21 days and a collection period of 15 days. The maximum organic matter intake (OMI-kg/100 kg BW) were 2.08 and 3.35 for US and TS, respectively and urea treatment increased the maximum OMI by 61% than for US. In order to reach maximum intake the amount feed refusal should be 25% for US, but with TS maximum intake was not reached even when the amount of feed refused was 39% of that consumed. The OMD of US significantly decreased with increase in OMI, whereas with TS the decrease was not significant. The inability of animals to select between stems and leaves particularly in TS might be one of the reasons for its constant digestibility. Considering the substantial increases in intake and the negative effect on digestibility of US, further trials are warranted, as well as studies to determine the practical implications.

Effects of lactic acid bacteria and molasses on fermentation dynamics, structural and nonstructural carbohydrate composition and in vitro ruminal fermentation of rice straw silage

  • Objective: This study was to evaluate the fermentation dynamics, structural and nonstructural carbohydrate composition and in vitro gas production of rice straw ensiled with lactic acid bacteria and molasses. Methods: Fresh rice straw was ensiled in 1-L laboratory silos with no additive control (C), Lactobacillus plantarum (L), molasses (M) and molasses+Lactobacillus plantarum (ML) for 6, 15, 30, and 60 days. After storage, the silages were subjected to microbial and chemical analyses as well as the further in vitro fermentation trial. Results: All additives increased lactic acid concentration, and reduced pH, dry matter (DM) loss and structural carbohydrate content relative to the control (p<0.05). The highest organic acid and residual sugar contents and lignocellulose reduction were observed in ML silage. L silage had the highest V-score with 88.10 followed by ML silage. L and ML silage improved in vitro DM digestibility as compared with other treatments, while in vitro neutral detergent fibre degradability (IVNDFD) was increased in M and ML silage (p<0.05). M silage significantly (p<0.05) increased propionic acid (PA) content and decreased butyric acid content and acetic acid/PA as well as 72-h cumulative gas production. Conclusion: The application of ML was effective for improving both the fermentation quality and in vitro digestibility of rice straw silage. Inclusion with molasses to rice straw could reduce in vitro ruminal gas production.

  • 소규모(小規模) 축산농가(畜産農家)에서 배출(排出)되는 돼지(돈(豚))분뇨폐수(糞尿廢水)를 처리(處理)함과 동시(同時)에 재활용(再活用)할 수 있는 방법(方法)을 찾기 위하여 볏짚을 살수여상법(撒水濾床法)의 충전재(充塡材)로 사용(使用)하였다. 폐수처리후(廢水處理後) 볏짚을 부숙(腐熟), 건조(乾燥), 분쇄(粉碎)하여 토양(土壤)에 첨가(添加)한 후(後) 오그라기 상치를 재배(栽培)한 결과(結果)는 다음과 같다. 1. 농가퇴비(農家堆肥)에 비(比)해 부숙(腐熟)된 짚을 처리(處理)했을 시(時) 수량지수(收量指數)가 월등(越等)히 높았으며 N.P.K. 기본(基本) 시비(施肥)에 부숙(腐熟)된 짚 1750 kg/10a로 시용(施用)했을 때 수량지수(收量指數)가 가장 높았다. 2. 부숙(腐熟)된 짚을 시용(施用)한 토양(土壤)의 화학적(化學的)인 성질(性質) 중(中) pH, T-N, ava.P2O5, OM, 그리고 Ex.K, Ca, Mg이 증가(增加)했는데 반(反)하여 CEC은 본래(本來)의 값으로 귀착(歸着)되는 경향(傾向)이었다. 3. 짚을 살수여상법(撒水濾床法)의 충전재(充塡材)로 사용(使用)했을 경우(境遇)의 BOD, COD, SS의 제거(除去) 효율(效率)이 통상적(通常的)인 충전재(充塡材)와 별(別) 차이(差異)가 없었다.

저품위 석탄을 충전한 칼럼실험에서의 바이오가스 생산에 관한 연구 (A Study on Biogas Production from Low Rank Coal in a Column Experiment)

  • 저급탄을 충전한 칼럼실험에서 미생물과 영양물질을 공급한 상태에서 미생물에 의한 메탄가스 발생을 유도하기 위하여 볏짚을 추가로 공급하여 장기간 바이오 가스 발생 특성을 관찰하였다. 충전한 석탄 대비 볏짚의 무게비가 0.04 이하에서는 유의미한 가스의 발생이 없었으며, 0.08에서는 약 90일간 바이오 가스가 발생되었으나, 최적 조건에서의 vial 실험 결과와 비교하면 투입한 볏짚 무게당 바이오 가스의 발생량이 5 % 수준에 불과하였다. 따라서 원위치에서 석탄을 채굴하지 않은 상태에서 바이오 가스를 생산하기 위해서는 볏짚 : 석탄의 비율이 1:3 이상의 조건에서 분해액의 COD 농도가 2000 mg/L 이상이 되도록 운전하는 반응조를 지상 혹은 지중에 설치할 필요가 있다. 또한 석탄과 볏짚을 충전한 칼럼실험 후의 지중수는 볏짚만을 투입한 vial 내 용액과 함께 파이로시퀀싱 방법으로 미생물 군집 분석을 실시하여 두 시료 간의 우점하는 미생물 종을 비교하였다. Bacteria의 균일도와 다양성 측면에서 석탄 충전탑이 보다 다양한 종분포를 이루고 있으며, 이는 결과적으로 메탄 생성에는 불리한 미생물 분포임을 나타낸다.

  • 중소규모(中小規模) 축산농가(畜産農家)에서 배출(排出)되는 돼지(돈(豚)) 분뇨폐수(糞尿廢水)를 효과적(效果的)으로 처리(處理)함과 동시(同時)에 재활용(再活用)할 수 있는 방법(方法)의 하나로 볏짚을 살수여상법(撒水濾床法)의 충전재(充塡材)로 이용(利用)하였다. 폐수처리(廢水處理) 후(後) 볏짚을 건조(乾燥) 분쇄(粉碎)하여 토양(土壤)에 첨가(添加)한 후 orchard grass를 재배(栽培)하여 다음과 같은 결과(結果)를 얻었다. 1. 볏짚 충전재(充塡材)를 시용(施用)했을 경우(境遇) 일반적(一般的)인 농가퇴비(農家堆肥)에 못지 않은 수량(收量) 향상(向上) 효과(效果)가 있었다. 기본시비(基本施肥)(NPK)에 처리(處理)한 짚 40 ton/ha의 비율(比率)로 혼합(混合) 시용 했을 때 가장 효과(效果)가 컸다. 2. Orchard grass 1차 예취(刈取) 후의 수량은 기본시비(基本施肥) 전량 시용구가 반량(半量) 시용구보다 많았다. 3. Orchard grass 1차, 2차 예취(刈取) 후(後) 볏짚충진제 처리구 토양(土壤)의 화학적 특성은, 공시토양에 비해 pH 의 증가와 함께 Av. $P_2O_5$, Ex. K, Ca, Na, Mg 와 유기물함량(有機物含量)이 증가(增加)했는데 특(特)히 Exc. Mg 의 현저한 증가(增加)가 있었다. 4. 볏짚 충진제를 시용한 결과 Orchard grass에 의해 흡수된 질소, 인산, 양이온등의 영양소는 증가되었고, 증가량은 처리량에 비례했다.
