Effect of humidity on the quality characteristics of the 3rd day of 5th instar silkworm powder

  • Freeze-dried 3rd day of 5th instar silkworm powder (FDSP) has been produced and consumed as functional food ingredient in Korea. Generally, FDSP is distributed in the dehumidification state. In this study, the effect of humid condition (30%, 50%, 70%, and 90% RH) on the quality characteristics of FDSP was examined. The color of FDSP, greenish yellow, becomes dark with the change of redness and yellowness. When FDSP is exposed for 4 h at 90% RH, the lightness was changed from 56 to 51, redness from -0.8 to 3.7, and yellowness from 26.5 to 21.6. The proximate composition of FDSP was not significantly changed except 90% RH condition. At that condition, moisture content (5.7%) was increased to 26.8% and crude fat (11.3%) decreased to 1.9% (3 wk 90% RH). Mineral content change during storage can be classified to 3 types; sulfur rapidly decreased, potassium mostly stable with some decreased with humidity, and sodium increased with high humid condition. Vitamins were not significantly affected. However, DNJ was abruptly decreased to 0.03 g/100g in humid condition.

Antibacterial and antifungal effects of Korean propolis against ginseng disease

  • We investigated the anti-microbial activity of propolis against the pathogenic bacteria and fungi on ginseng. We selected six microbials that caused postharvest root rots in ginseng. Propolis extracts were prepared by using the ethanol extraction method. We seeded the bacteria and fungi related to ginseng disease on a specific culture medium, and treated it with propolis extracts by using the paper disc method. Propolis extracts indicate the anti-microbial activity against Paenibacillus polymyxa, Fusarium solani, Rhizoctonia solani AG-1 and Pythium ultimum. However, the anti-fungal activity of propolis is weak on Pseudomonas fluorescens subsp. Cellulosa and Colletotrichum gloeosporioides. As a result, the antimicrobial effects of propolis against microbial that prevent ginseng growth were confirmed. The antimicrobial effects are shown according to the concentration of propolis against root rot. The fungi also showed antibacterial effects in a dose-dependent manner.

Sterilization effect and fatty acid composition of silkworm powder (Bombyx mori L.) by heat treatment

  • The effect of heat treatment on the sterilization and fatty acid compositions of silkworm powder was carried out under the guidance of Ministry of Food and Drug Safety. Food borne pathogens or microorganisms including E. coli, Salmonella spp., Listeria monocytogenes, Staphylococus aureus, Clostridium perfringens and aflatoxin, were not detected. The fatty acid composition was hardly changed after the silkworm powder was treated to a high temperature of $121^{\circ}C$. The low temperature sterilization of silkworm powder at $63^{\circ}C$ decreased the concentration of E. coli while high temperature sterilization at $121^{\circ}C$ has found no traces of microorganism.

  • Heavy metals and residual agricultural chemicals in Bombyx mori silkworm cocoon for nontextile usages were examined. Proximate composition of silkworm cocoon was examined; contents of moisture, crude protein, crude fat, and ash were about 5 ~ 6%, 93 ~ 94%, 0.4 ~ 0.5%, and 0.3 ~ 0.4%, respectively. The Heavy metals were analyzed by ICP/MS after acid digestion. Arsenic, cadmium, and lead were not detected. The average contents of mercury, zinc, and cupric were 0.317, 3.99, and 2.06 mg/kg, respectively. Agricultural chemicals, such as dichlorvos and paraquart also were not detected. Silkworm cocoon may be safe used for nontextile applications.

  • In order to the feasibility of silk materials as protein delivery system, silk beads incorporated with bovine serum albumin (BSA) were prepared by dropping silk fibroin extract into dope solution composed of ethanol and dichloromethane. Structural and morphological characteristics of silk beads were examined using scanning electron microscopy (SEM), infrared spectrometry, and X-ray diffractometry. Swelling ratio of silk beads was also measured. Release behavior of prototypical protein, BSA, was studied by observing the electropheretic phenomenon and release profile. SEM showed that silk beads are spherical with porous interior structure. Infrared spectrometry and X-ray diffraction confirm that the silk beads have a ${\beta}$-sheet conformation. The swelling capability of silk beads increased with the incorporation of the protein. The protein was released from the beads with slow release following an initial burst release. Therefore, silk beads show promise as materials for encasing protein drugs to be delivered to targets in the human body.

  • Transparency and insolubility of eardrum patch against exudates are important for otolaryngological surgery. The author prepared silk fibroin (SF) films with various concentrations and temperature and then examined solubility and conformation of SF films. SF films were transparent regardless of the various preparation conditions. Although most SF films are soluble in 1X PBS solution at $37^{\circ}C$ for 1 h, the SF film with 3.4% with $60^{\circ}C$ was insoluble. Scanning electron microscopy (SEM) showed that the SF films have solid and smooth surface. Fourier transform infrared spectroscopy (FT-IR) and differential scanning calorimetry (DSC) results showed that the conformation of SF films was influenced by the preparation conditions including SF concentration and casting temperature. In conclusion, SF membrane with transparence and insolubility against exudates could be considered as eardrum patch resources.

  • Silk polymer has been focused recently on medical applications as a novel biomaterials. The factors for biomaterials were considered and reviewed recent research on surgical suture, wound dressing, tympanic regeneration patch, guided bone regeneration membrane, bone implant and ligament. Silk polymer is good mechanical properties and biocompatibility. Therefore it might be used as one of promising materials for medical application.

  • To establish a control strategy for the gypsy moth (Lymantria dispar asiatica), a sporadic pest, the insecticidal activity of 21 eco-friendly agricultural materials was tested at recommended concentrations under laboratory conditions. Among these, 11 eco-friendly agricultural materials showed more than 90% insecticidal activity. In response to the PLS(positive list system), the results of this study can be used to control of gypsy moth and development of eco-friendly agricultural materials.

  • Dynamic parameters of biochemistry including the POD (peroxidase) activity, contents of DNA and RNA, isozymes of POD and ES (esterase) in American send (Panax quinquefolium L.) seed are reported in the present paper. The dynamic changes of POD activity proved that the PAS (physiological afterripening stage) is a stage in which some substances are prepared for seed germination. The POD activity correlated with ER (embryo ratio) significantly DNA content changed little only within 0.0036∼0.013 mg/ml, which did not correlate with ER. RNA content changed from 0.1539 to 1.0313 mg/ml and correlated significantly with RE during all of the embryo afterripening. None of the POD isozyme band was obtained in ESGS (embryo slowly growth stage), but five bands in ERGS (embryo rapidly growth stage) and six bands in PAS. Four bands of ES isozymes were obtained in ESGS, but six bands in ERGS, particularly, the content of ES isozymes increased in PAS. All of these may provide some information for understanding the dormancy mechanisms of American song seed.
