Evaluation of Acute Toxicity of Black Pepper extracts, Clove bud, Rosemary and Origanum Essential oils (식물추출물 후추, 클로브버드, 로즈마리 및 오리가늄오일의 급성독성평가)

  • Highly bio-active plant essential extracts and oils from pepper, clove bud, rosemary and origanum which are selected to develop environment-friendly insecticides was studied for their acute toxicity. The results of acute oral toxicity using rats showed $LD_{50}$ value of over 2,000 mg/kg bw for pepper, clove bud, rosemary and origanum oils. The calculated acute dermal $LD_{50}$ value of pepper was over 4,000 mg/kg bw and anther testing materials was over 4,000 mg/kg bw. The skin irritation test showed that pepper, clove bud and rosemary oils had no irritation while origanum oil had a moderate irritation. According to the eye irritation test, it showed that there was no irritation for pepper and rosemary oils, while there were irritation for clove bud and origanum oils. Consequently, pepper and rosemary oils were shown to be low in toxicity whereas clove bud oil was indicated to cause a mild eye irritability and origanum oil, causing a moderate skin and eye irritability.

A Study on Antigencity (Immunotoxicity Study) to the Expressed Proteins of ${\beta}$-Carotene Biofortified Rice (베타카로틴강화미 발현단백질에 대한 항원성연구)

  • As part of safety evaluation of 2A (amono acid), PAT (phosphinotricin Acetyl-transferase), CtrI (Carotene desaturase) and PSY (phytoene synthase), the expressed proteins inserted to ${\beta}$-carotene Biofortified rice were tested for antigencity test. As a result, the group of administering high-concentration PAT, the expressed protein, showed a great content of total WBC; however, other expressed proteins did not show much difference. Against ASA (Active Systemic Anaphylaxis) test, the group of administering high-concentration PAT, the expressed protein, showed mild or medium degree of symptoms, but there was no dead entity. According to the result of the PCA (Passive Cutaneous Anaphylaxis test), the group of administering high-concentration PAT, 2A, PSY, and mixture of expressed proteins indicated positive response in low anti-serum concentration, and the group of administering the clinical concentration of mixture indicated mild positive response. However, because the group of administering the clinical concentration of expressed proteins, PAT, 2A, PSY, and CtrI, did not show positive response, it is thought that IgE is not generated. Further studies are needed to verify the safety of ${\beta}$-carotene Biofortified rice.

Whitening improvement effect of Hermetia illucens larvae extracts (아메리카동애등에(Hermetia illucens) 유충 추출물의 미백개선 효과)

  • The present study investigated the feasibility of using the ethanolic extract of Hermetia illucens larvae (HIE) as a whitening improvement material. In cell viability assays using B16F1 melanoma cells, no cytotoxicity was recorded up to 200 ㎍·mL-1 of HIE. Moreover, while tyrosinase inhibitory activity increased, melanin content decreased in a dose-dependent manner, indicating that HIE likely inhibited tyrosine-induced intracellular melanin biosynthesis in B16F1 melanoma cells. Therefore, HIE is expected to serve as a potent whitening improvement material.

Acute and Chronic Exposure Assessment of Organophosphate Pesticides through the Consumption of Fruit Vegetables (과채류 섭취를 통한 유기인계 농약의 급성 및 만성노출평가)

  • In this study, we monitored the residues of organophosphate pesticides (OP) in eight fruit vegetables grown in Korea, and assessed risk levels of acute and chronic exposure of OP through the consumption of fruit vegetables. Chlorpyrifos, EPN, methidathion and phosphamidon in eight fruit vegetables were detected in this study. The results of cumulative assessment of national estimated short term intake for acute exposure of OP were due to the following order; eggplant, tomato, squash and cucumber. Total theoretical maximum daily intake calculated at first step of chronic exposure assessment for registered OP in Korea was 76.14%, compared with acceptable daily intake ADI) based on chlorpyrifos. In addition, total national estimated maximum daily intake calculated at second step of chronic exposure assessment was 13.949%, compared with ADI. Third chronic assessment was conducted by probabilistic approach using OP residues detected in eight fruit vegetables and showed that total exposure risk was very low, corresponding to 0.0001% compared to ADI. Based on those finding, the risk of organophosphate pesticides in fruit vegetables was considered quantitatively negligible. In future, further investigation to expand the target should be followed to do more accurate and detailed risk assessment.

Anti-thrombosis Activity of Drone Apis mellifera Pupae Extracts

  • Drones of honeybee (Apis mellifera) have been regarded as a useful value only when mating with queen bee. However, the drone pupae have been reported to be nutritionally valuable, and a potential beekeeping product. In this study, drone pupae extracted with 5% acetic acid were used to measure anti-thrombosis related fibrinolytic activity using Strup and Mullertz fibrin plate method. As a result, the drone pupae extract showed higher effect of fibrinolytic activity(clear zone diameter 20.83mm) compared to the human plasmin (clear zone diameter 12.93mm) used as a positive control. It was suggested that the extract of drone pupae can be developed as a functional material helping prevention or treatment of various vascular diseases.

A 90-day Safety Study (Repeated-Dose Oral Toxicity Study) of Genetically Modified ${\beta}$-Carotene Biofortified rice in Sprague-Dawley Rats (SD 랫드에서 베타카로틴강화미의 90일 반복투여 경구독성시험)

  • This study was conducted to evaluate the safety of ${\beta}$-carotene biofortified rice, a genetically modified organism (GMO) developed by Rural Development Administration. ${\beta}$-carotene biofortified rice were exposed on Sprague-Dawley rats for 13 weeks. All rats survived until the end of the exposure period. There were no biologically significant differences in body weight, feed and water consumption, weight gains and feed efficiency. There were no clinical signs of toxicity attributable to exposure to GM rice. Mild decreases in AST, ALT, TG levels were observed in Group II (25% GM rice (w/w) and Group III (50% GM rice (w/w), both in females and males. Results of histopathological changes treated with the ${\beta}$-carotene biofortified rice had no significant differences between the control and treatment groups. Based on these results, we deemed that genetically modified ${\beta}$-carotene biofortified rice was as safe as conventional rice.

Removal of Arsenic from Leachate of Tailing using Laboratory-synthesized Zerovalent Iron

  • Feasibility of laboratory-synthesized zerovalent iron was investigated to remove arsenic from leachates of tailings taken from an Au-Ag abandoned mine. The tailings were seriously contaminated with arsenic, and its potential adverse effect on the ecosystems around the mine seems to be significantly high. Long-term column experiments were conducted for about 3.5 months to evaluate the effectiveness of the synthesized zerovalent iron for removal of arsenic. Over than 95% removal efficiency of As was observed in the zerovalent iron mediated tests. In addition, the XRD data suggest that the corrosion products of ZVI were identified magnetite, maghemite, goethite, and lepidocrocite, all of which support Fe(II) oxidation as an intermediate step in the zerovalent iron corrosion process. The results indicate that arsenic can be removed from the tailing-leachate by the mechanism of coprecipitation and/or adsorption onto those iron oxides formed from ZVI corrosion.

  • The performances of three same type of vibrating potato diggers were estimated by observing the potato separation and material flow on the bottom plate. Four-bar mechanism were adopted for three diggers and pairs of eccentric cams on both sides of driving shaft were used as driving link of the diggers. Each machine was tested with different amplitudes , frequencies, and travels speeds. Blade performance were observed in three categories : Impossible forward travel , acceptable operation, and unsatisfactory potato digging , but good material flow. Three parameters were used to set marginal values that enable the machines operate for potato digging, and the parameters were compared to select best one. Three parameters are λ, $\rho$, and K.λ is the ratio of vibrating speed to travel speed, $\rho$ is the ratio of blade acceleration to travel speed, and K is the ratio of blade acceleration to gravitational acceleration. K value of 2 or more is suggested to be used as design and evalu tion criterion of the vibrating digger.

Aspergillus Associated with Meju, a Fermented Soybean Starting Material for Traditional Soy Sauce and Soybean Paste in Korea

  • Aspergillus is an important fungal genus used for the fermentation of Asian foods; this genus is referred to as koji mold in Japan and China. A. oryzae, A. sojae, and A. tamari are used in the production of miso and shoyu in Japan, but a comprehensive taxonomic study of Aspergillus isolated from Meju, a fermented soybean starting material for traditional soy sauce and soybean paste in Korea, has not been conducted. In this study, various Aspergillus species were isolated during a study of the mycobiota of Meju, and the aspergilli were identified based on phenotypic characteristics and sequencing of the ${\beta}$-tubulin gene. Most strains of Aspergillus were found to belong to the following sections: Aspergillus (n = 220), Flavi (n = 213), and Nigri (n = 54). The most commonly identified species were A. oryzae (n = 183), A. pseudoglaucus (Eurotium repens) (n = 81), A. chevalieri (E. chevalieri) (n = 62), A. montevidensis (E. amstelodami) (n = 34), A. niger (n = 21), A. tamari (n = 15), A. ruber (E. rubrum) (n = 15), A. proliferans (n = 14), and A. luchuensis (n = 14); 25 species were identified from 533 Aspergillus strains. Aspergillus strains were mainly found during the high temperature fermentation period in the later steps of Meju fermentation.
