One of the real issues of the recent years is water contamination because of harmful synthetic dyes. Liquid Membranes (LM) resemble a promising alternative to the current separation processes, demonstrating various points of interest as far as effectiveness, selectivity, and operational expenses. The improvement of various Liquid Membranes designs has been a matter of examination by few researchers, particularly for the expulsion of dyes from aqueous solutions. The choice of organic surfactants plays an essential role in the efficiency of the dye removal. In LM design, the most significant step towards productivity is the decision of the surfactant type and its concentration. Liquid emulsion membrane (LEM) was used to remove safranin from aqueous solutions in which the emulsion was made with the help of D2EHPA as carrier, kerosene was used as a diluent and Span 80 (Sorbiton monooleate) was used as an emulsifying agent or surfactant. Various sorts of internal stages were utilized, to be specific sulphuric acid and sodium hydroxide. The impact of parameters influencing extraction efficiency such as pH of feed solution, concentrations of surfactant and emulsifying agent in membrane phase, volume ratio of internal phase to membrane phase, internal phase concentration, agitation speed and time of extraction were analyzed.

This study examined the construction method shown in literatures and materials discovered in rampart, barbican and guard platform for Cheongdo-Eupseong castle which was constructed immediately before Japanese Invasions of Korea (1592-98). According to the result of the examination, the construction method used in the coastal areas and inland areas of Gyeongsang province in the early Joseon period was applied to the rampart, barbican and guard platform in the east and north castle walls, which was constructed earlier than other parts of Cheongdo-Eupseong castle. However, from the western end of the northern castle wall, which was later constructed, to the western castle wall was rebuilt using a construction method different from the method applied to the parts that were constructed earlier after Japanese Invasions of Korea. Cheongdo-Eupseong castle was constructed on a flatland on a large scale and at a low height under unreasonable pressures to construct the castle in a short period of time for military defense as well as civil agitation in the imminent situation just before the outbreak of Japanese Invasions of Korea. In addition, various methods of constructing Eupseong castle utilized at that time were used together, and barbican and guard platform were built generally focusing on rapid completion and convenience, using other methods than the one mainly used at that time. This is the reason why there was a difference in the characteristics of the method and period of castle construction between Cheongdo-Eupseong castle and other Eupseong castles in the coastal areas and inland areas of Gyeongsang province.
Using response surface methodology(RSM), the various conditions(agitation speed, air flow, glucose concentration) in jar fermentor culture were investigated to find the optimum conditions for maximum enzyme production. Central-composite-design was used to control the variable constant in the experiment. The glucose isomerase production of Steptomyces chibaensis J-59 was mostly affected by the air flow rate and glucose concentration. The estimated optimum conditions were as follows: 1% birchwood xylan, 1.5% CSL, 0.1% $MgSO_4{\cdot}7H_2O$, 0.012% $CoCl_2{\cdot}6H_2O$, pH 7.0; air flow, 2.2vvm; agitation speed, 587rpm; glucose concentration, 0.586%. Experimental values(7.43GIU/ml) for the enzyme production obtained from the given optimum conditions had a almost resemblane to response values(7.67GIU/ml) predicted by the RSM. The jar fermentor culture by the RSM produced xylose isomerase about 2.7 times as much as the baffled flask culture.
Process variables are factors in an experiment that are not mixture components but could affect the blending properties of the mixture ingredients. For example, the effectiveness of an etching solution which is measured as an etch rate is not only a function of the proportions of the three acids that are combined to form the mixture, but also depends on the temperature of the solution and the agitation rate. Efficient designs for the mixture components-process variables experiments depend on the mixture components-process variables model which is called a combined model. We often use the product model between the canonical polynomial model for the mixture and process variables model as a combined model. In this paper we propose three starting models for the mixture components-process variables experiments. One of the starting model we are considering is the model which includes product terms up to cubic order interactions between mixture effects and the linear & pure quadratic effect of the process variables from the product model. In this paper, we propose a method for finding robust designs and practical designs with respect to D-, G-, and I-optimality for the various starting combined models and then, we find practically efficient and robust designs for estimating the regression coefficients for those models. We find the prediction capability of those recommended designs in the case of three components and three process variables to be good by checking FDS(Fraction of Design Space) plots.
During the last few decades, it has become evident that the compatibility of the yeast biochemical environment with the ability to process and translate the RNA transcript, along with its capacity to modify a translated protein, are relevant requirements for selecting this host cell for protein expression in several pharmaceutical and clinical applications. In particular, Pichia pastoris is used as an industrial host for recombinant protein and metabolite production, showing a powerful capacity to meet required biomolecular target production levels in high-throughput assays for functional genomics and drug screening. In addition, there is a great advantage to using P. pastoris for protein secretion, even at high molecular weights, since the recovery and purification steps are simplified owing to relatively low levels of endogenous proteins in the extracellular medium. Clearly, no single microexpression system can provide all of the desired properties for human protein production. Moreover, chemical and physical bioprocess parameters, including culture medium formulation, temperature, pH, agitation, aeration rates, induction, and feeding strategies, can highly influence product yield and quality. In order to benefit from the currently available wide range of biosynthesis strategies using P. pastoris, this mini review focuses on the developments and technological fermentation achievements, providing both a comparative and an overall integration analysis. The main aim is to highlight the relevance and versatility of the P. pastoris biosystem to the design of more cost-effective microfactories to meet the increasing demands for recombinant membrane proteins and clinical antibodies for several therapeutic applications.
The flow characteristics in a stirred tank are very useful in a wide variety of industrial applications. Generally, the flow pattern, power consumption and mixing time in stirred vessels depend not only on the design of the impeller, but also on the tanks' geometry and internal structure. In this study, the analysis of an unstable and unsteady complicated flow characteristics generated by the interaction between the baffle shape and impeller were performed using the ANSYS FLUENT LES Turbulence Model. The study compared the predictions of CFD with the interaction between two types of rotating impellers (axial and radial flows) and the shapes of three baffles. The results of the comparison verified that the design model showed a relatively efficient trend in the mixing flow fields and characteristics around the impeller and baffles during agitation.
Strain improvement and bioprocess development were undertaken to enhance hyaluronic acid(HA) production by Streptococcus zooepidemicus cells. Using a high-yielding mutant strain, statistical medium optimization was carried out in shake flask cultures, resulting in 52% increase in HA production (5.38 g/l) at the optimal medium composition relative to the parallel control cultures. For sufficient supply of dissolved oxygen (DO), which turned out to be crucial for enhanced production of HA, agitation system and speed were intensively investigated in 5 L bioreactor cultures. Increase in oxygen mass transfer coefficient (kLa) through increment of agitation speed (rpm) and 35% expansion of diameter of the newly-designed impellers showed significantly positive effects on HA production. By installing an expanded Rushton-turbine impeller for efficient break-down of sparged air, and an extended marine impeller above the Rushton-turbine impeller for efficient mixing of the air-born viscous fermentation broth, maximum amount of HA (9.79 g/l) was obtained at 450 rpm, 1.8 times higher level than that of the corresponding flask culture. Subsequently, the possibility of bioprocess scale-up to a 50 L bioreactor was investigated. Despite almost identical maximum HA production (9.11 vs 9.25 g/l), the average HA volumetric productivity (rp) of the 50 L culture turned out only 74% compared to the corresponding 5 L culture during the exponential phase, possibly caused by shear damages imposed on the producing cells at the high stirring in the 50 L culture. The scale-up process could be successfully achieved if a scale-up criterion of constant oxygen mass transfer coefficient (kLa) is applied to the 50 L pilot-scale bioreactor system.

In this study, culture conditions were optimized to confirm the feasibility of Acetobacter pasteurianus as a starter for fermentation vinegar. Acetobacter pasteurianus strain can be used as a food ingredient. The optimal temperature and pH conditions of the selected Acetobacter pasteurianus SRCM101388 were 28℃ and pH 6.00, respectively. The response surface methodology (RSM) was used to optimize the composition of the medium, and Plackett-Burman design (PBD) was used to obtain the effective selection of culture medium, resulting in that glucose, sucrose, and yeast extract had the highest effect on increasing biomass. The optimal concentration, which was performed by central composite design (CCD), were determined to be 10.73 g/L of glucose, 3.98 g/L of sucrose, and 18.73 g/L of yeast extract, respectively. The optimal concentrations of trace elements for the production of biomass were found to be 1 g/L of ammonium sulfate, 0.5 g/L of magnesium sulfate, 2 g/L of sodium phosphate monobasic, 2 g/L of sodium phosphate dibasic, and the final optimized medium was pH 6.10. When incubated in a 5 L jar fermenter, the SRCM101388 strain showed a faster-dissolved oxygen (DO) reduction at a lower agitation rate (rpm), and it was able to grow even at reduced DO level when aeration was maintained. The amount of final biomass produced was 2.53±0.12×109 CFU/mL (9.40±0.02 log CFU/mL) when incubated for 18 hours at 150 rpm, 0.5 vvm, pH 6.0, and 28℃.
The present study was carried out to investigate the effect of ultrasonic bath in tissue preparation for transmission electron microscopy. The method used standard reagents and media, and employed ultrasonic bath agitation to accelerate fluid exchange. The liver kidney, stomach and cardiac muscle tissues of male Sprague-Dawley rats were used for the experiment, and the experimental design was divided into 4 groups; The control group using rotators (Traditional method, 1,625 mins) and the three experimental groups using ultrasonic bath (UB) in the primary fixation through the infiltration processes (UB I; 62.5 mins, UB II; 125 mins, UB III; 250 mins). The results were as follows; 1. In the control group, tissues were easily sectioned, and showed well preserved intact membranes, and cell organelles such as mitochondria, lysosome, peroxisome, rough endoplasmic reticulum and smooth endoplasmic reticulum. 2. In the UB treated group I, tissues showed holes due to the inadequate removal of both water and fluids used in the dehydration process. Also the mitochondria of cell organelles, especially, showed swollen intracristal spaces and dense matrices due to poor fixation. 3. In the UB treated group II, tissues showed good preservation of cell organelles and specimen slice sections. Also, no holes were observed. 4. In the UB treated group III, tissues showed leaching of structural components in the cytoplasm, but no holes were observed. In conclusion, the ultrasonic bath procedure takes approximately 120 minutes from specimen fixation to resin infiltration and gives excellent results.
Non-Newtonian fluid mechanics takes charge of an important role in the oil industries. Especially in the oil well drilling process, the drilling fluid such as mud keeps the drill bit cool and clean during drilling, with suspending drill cuttings and lubricating a drill bit. The purpose of this study is to examine the effect of fluid mud rheological properties to predict different characteristics of non-Newtonian fluid in the mud mixing tank on offshore drilling platforms. In this paper, ANSYS fluent package was used for the simulation to solve the hydrodynamic force and to evaluate mud mixing time. Prediction of the power consumption and the pumping effectiveness has been presented with different operating fluid models as Newtonian and non-Newtonian fluid. The comparison between Newtonain mud model and non-Newtonian mud model is confirmed by the CFD simulation method of drilling mud mixing tank. The results present useful information for the design of the drilling mud mixing tanks and provide some guidance on the use of CFD tool for such non-Newtonian fluid flow.
