Using proteases, we produced a peptide mixture from fibroin of the silkworm $Bombyx$$mori$. Mice received D-galactose by subcutaneous injection for 8 weeks to accelerate aging, and then received the peptide mixture orally for 7 weeks. In the mice aged with D-galactose, the coefficient of friction of hair increased significantly up to 1.6-fold, but in the mice subsequently given the peptide mixture, it was normal. Scanning electron microscopy showed improved hair cuticles in the latter mice too. These results indicate that oral administration with peptides from $Bombyx$ fibroin counters the aging of hair cuticles.
Purpose: The goal of this study was to see how different aerobic exercise intensities affected AMP-activated protein kinase (AMPK), reactive oxygen, and antioxidant enzymes in young mice during an 8-week period. Methods: Forty male C57BL/6 mice, aged seven weeks, were randomly assigned to one of four groups: control (n=10), low-intensity exercise (n=10), moderate-intensity exercise (n=10), and high-intensity exercise (n=10). For eight weeks, aerobic activity was performed once a day for 35-40 minutes, five days a week. The data were analyzed using descriptive statistics, analysis of variance (ANOVA), chi-squared tests, and the Tukey test in the SPSS/WIN 25.0 program. Results: Weight (p=.001) was substantially different between the moderate-intensity exercise group and the control group in AMPK (p<.001). In addition, there were no significant differences between the moderate-intensity exercise group and the control group in reactive oxygen malondialdehyde (MDA) levels (p=.136) and antioxidant enzyme superoxide dismutase (SOD) levels (p=.521). Conclusion: These findings suggest that moderate-intensity aerobic exercise increased AMPK activation and helped young mice shed weight.

Alterations in sulfur amino acid metabolism are associated with an increased risk of a number of common late-life diseases, which raises the possibility that metabolism of sulfur amino acids may change with age. The present study was conducted to understand the age-related changes in hepatic metabolism of sulfur amino acids in 2-, 6-, 18- and 30-month-old male C57BL/6 mice. For this purpose, metabolite profiling of sulfur amino acids from methionine to taurine or glutathione (GSH) was performed. The levels of sulfur amino acids and their metabolites were not significantly different among 2-, 6- and 18-month-old mice, except for plasma GSH and hepatic homocysteine. Plasma total GSH and hepatic total homocysteine levels were significantly higher in 2-month-old mice than those in the other age groups. In contrast, 30-month-old mice exhibited increased hepatic methionine and cysteine, compared with all other groups, but decreased hepatic S-adenosylmethionine (SAM), S-adenosylhomocysteine and homocysteine, relative to 2-month-old mice. No differences in hepatic reduced GSH, GSH disulfide, or taurine were observed. The hepatic changes in homocysteine and cysteine may be attributed to upregulation of cystathionine ${\beta}-synthase$ and down-regulation of ${\gamma}-glutamylcysteine$ ligase in the aged mice. The elevation of hepatic cysteine levels may be involved in the maintenance of hepatic GSH levels. The opposite changes of methionine and SAM suggest that the regulatory role of SAM in hepatic sulfur amino acid metabolism may be impaired in 30-month-old mice.
Although many studies have reported that the breakdown of the blood-brain barrier (BBB) represents one of the major pathological changes in aging, the mechanism underlying this process remains relatively unexplored. In this study, we described that acid sphingomyelinase (ASM) derived from endothelial cells plays a critical role in BBB disruption in aging. ASM levels were elevated in the brain endothelium and plasma of aged humans and mice, resulting in BBB leakage through an increase in caveolae-mediated transcytosis. Moreover, ASM caused damage to the caveolae-cytoskeleton via protein phosphatase 1-mediated ezrin/radixin/moesin dephosphorylation in primary mouse brain endothelial cells. Mice overexpressing brain endothelial cell-specific ASM exhibited acceleration of BBB impairment and neuronal dysfunction. However, genetic inhibition and endothelial specific knock-down of ASM in mice improved BBB disruption and neurocognitive impairment during aging. Results of this study revealed a novel role of ASM in the regulation of BBB integrity and neuronal function in aging, thus highlighting the potential of ASM as a new therapeutic target for anti-aging.
This study was done to examine immunomodulative effects of aged doraji (AD) in the immune-suppressed mice induced by cyclophosphamide. The immune-stimulating effects of ethanol AD extract in in vivo at 75 and 150 mg/kg body weight (BW) for AD and 2AD groups were evaluated and compared to the normal doraji group (2ND, 150 mg/kg BW) treated with a doraji without aging process. After the 10 days of oral supplement, body and immune related organ weights, serum immunoglobulin G (IgG) and cytokines levels, splenocytes proliferation rate, and splenic NK cell activity were measured as immune-related biomarkers. Body weight and serum IgG level increased in the 2AD group. But, the serum Th2 cytokine (IL-6, $TNF-{\alpha}$) levels were lower in the AD and 2AD groups, respectively. Splenic T cell and B cell proliferation and NK cell activity increased in the doraji groups and the significant increases were found only in the 2AD group. Thus, the aged doraji extract may affect body weight, serum IgG level, splenocytes proliferation, and splenic NK cell activity, and normalize the Th2 cytokine levels in the immune-suppressed mice. The results suggest that the aged doraji improves effectively immune system rather than the normal one.
Acetylbenzoylaconine, 2-aminoethylisothiouronium bromide hydrobromide (AET), $\beta$-mercaptoethylamine HCI (MEA), L-thiazolidine-4-carboxylic acid (thioproline)의 Co-60 감마선에 대한 방어효과를 5-6주된 백색마우스를 이용하여 연구하였다. 백색마우스에 대한 acetylbenzoylaconine의 독성실험 결과 LD$_{50}$은 2.5mg/kg이었다. 위의 4종 실험약제를 백색마우스 복강내에 주사한 후 Co-60 감마선 800 rad를 전신조사하고 30일간 계속 관찰하여 실험군들에 대한 생존계수를 계산하였다. 이 생존계수치로 방사선장해에 대한 방어계수를 계산한 결과 acetylbenzoylaconine AET, MEA, thioproline에 대해 각각 2.24, 2.95, 2.78, 1.23 으로, acetylbenzoylaconine은 백색마우스를 Co-60 감마선장해로 부터 방어할 수 있는 능력이 있음을 알았다.
A new herbal composition. HemoHIM, was developed using irradiated animal models and was successfully applied as an immune-improving agent. In a view that the protection and recovery of immune, hematopoietic and self-renewal tissues are essential for radioprotective agents, HemoHIM was developed based on a novel combination of three edible herbs (Angelica Radix, Cnidii Rhizoma. Paeonin Radix) that meet all those requirements. HemoHIM significantly protected the immune and hematopoietic system and enhanced their recovery in y-irradiated mice. For the application of HemoHIM as a health functional food and a supplementary agent for the cancer patients, the efficacy of HemoHIM to improve the immune functions was further evaluated in immune-depressed animals and humans. Animal studies demonstrated that HemoHIM significantly improved the immune functions in cyclophosphamide-treated mice, aged mice, and dexamethasone-treated mice. In human studies, HemoHIM enhanced the immune activity and cytokine secretion in sub-healthy volunteers, and alleviated the severe leukocyre depression in cancer patients during radiation and chemotherapy. Based on these results, HemoHIM was approved by Korea FDA as a material of health functional food for immune function improvement and will be commercially available soon. This case of HemoHIM research and development suggested that irradiated animals can be good models for biological degenerations such as immune depression, self-renewal tissue damage, and aging for the development of biological modulators.

Objective: This study aimed to examine the effect of laser-assisted zona thinning (LAZT) at one or four-points on the blastocyst formation and hatching process in mice with respect to female age. Methods: Eight-cell or morula embryos collected from superovulated C57BL female mice with different ages (6-11 and 28-31 weeks) were treated with LAZT at one-point (LAZT1) or four-points (LAZT4). The zona pellucida was thinned to more than 70% of its initial thickness by making two holes of $15-20{\mu}m$. Results: In the young mice, LAZT resulted in a significant increase in early hatching and hatching rates compared to the control group (p<0.05). However, in the old mice, LAZT significantly increased blastocyst formation as well as early hatching and hatching compared to the controls (p<0.05). These effects were more remarkable in LAZT4 than in LAZT1 and in aged mice than in young ones. Conclusion: These results show that multi-point LAZT leads to a significant improvement of blastocyst formation and hatching in mice compared to controls.
Objectives : The application of DNCB (1-chloro-2,4-dinitrochlorobenzene) can cause cellular immunity allergic reaction such as erythema or edema on NC/Nga mice and combinational function of cells participating in immunity increase inflammatory mediator. In this study, the effects of cypress essential oil on NC/Nga mice have been assessed. Methods : Male SPF NC/Nga mice aged 8 weeks have been used for atopic dermatitis induction skin. 1% DNCB was applied on ears and backs of which hair was removed using clipper on $1^{st}$ day and 0.4% DNCB was applied three times a week for 3 weeks. In this study, cypress essential oil has been treated 1 time a day for 3 weeks after application of DNCB to induce atopic dermatitis on skin for further experiments. Results : This study shows that inhalation or application of cypress essential oil reduced edema in ears and the thickness of epidermis induced by DNCB treatment. And it can be known that treatment of cypress essential oil inhibited mast cell proliferation and reduced IgE level similar to that of the negative control especially when cypress essential oil was inhaled by the mice. Synthetic oil showed the effects lower than those of cypress essential oil. Conclusions : Inhalation or direct application of cypress essential oil on skin reduced IgE level in blood and prohibits the proliferation of mast cell, from which it can be known that cypress essential oil can be effectively used to reduce the symptoms of atopic dermatitis.

Toll-like receptor 5 (TLR5) is a specific receptor for microbial flagellin and is one of the most well-known receptors in the TLR family. We reported previously that TLR5 signaling is well maintained during aging and that caveolin-1 may be involved in TLR5 signaling in aged macrophages through direct interactions. Therefore, it is important to clarify whether caveolin-1/TLR5 interactions affect TLR5 expression during aging. To assess the effect of caveolin-1 on TLR5, we analyzed TLR5 expression in senescent fibroblasts and aged tissues expressing high levels of caveolin-1. As expected, TLR5 mRNA and protein expression was well maintained in senescent fibroblasts and aged tissues, whereas TLR4 mRNA and protein were diminished in those cells and tissues. To determine the mechanism of caveolin-1-dependent TLR5 expression, we examined TLR5 expression in caveolin-1 deficient mice. Interestingly, TLR5 mRNA and protein levels were decreased dramatically in tissues from caveolin-1 knockout mice. Moreover, overexpressed caveolin-1 in vitro enhanced TLR5 mRNA through the MAPK pathway and prolonged TLR5 protein half-life through direct interaction. These results suggest that caveolin-1 may play a crucial role in maintaining of TLR5 by regulating transcription systems and increasing protein half-life.
