Novel Purification Method of Kv 4.2 Potassium Channel from Rat Brain Membrane

  • Kv 4.2 ion channel protein has an ability to open at subthreshold membrane potentials and to recover quickly from inactivation. That is very important for neuronal signal transmission in vertebrate brain. In order to purify Kv 4.2 protein, the novel purification methods were experimented. The purification procedure utilized chromatography on DE-52 ion exchange column and affinity chromatography on a WGA-Sepharose 4B, and Kv 4.2 affinity column chromatography. It was found that 0.5% (wt./vol.) Triton X-100 detergent in lysis buffer worked well for Kv 4.2 protein solubilization from rat brain membrane. Protein quantitative determination was conducted by BCA method at 562 nm for each purification step to avoid determination interference of protein at 280 nm by detergent. The confirmation of Kv 4.2 existence and amount is performed using by SDS-PAGE/immunoblotting or 96-well dot blotting. The Kv 4.2 without interacting protein that contains carbohydrate, was purified from novel biochemical 3-steps purification method for further research.

  • FcBP consisting of 13 amino acids specifically binds to Immunoglobulin G Fc domain. Initially, we utilized this peptide for preparation of antibody chip as a PEG composite for enhanced solubility. After then, the peptide conjugate was immobilized on agarose resin, resulting in highly efficient affinity column for antibody purification. The efficiency was comparable to commercial Protein A column. Recently, this peptide was conjugated with cell penetratingpeptide (CPP) on a backbone of GFP, affording antibody transducer, which carries antibody into live cells by simple mixing of antibody and the transducer in cell culture media. Antibody transduction into cells was monitored by live cell imaging. More recently, the FcBP was fused to ferritin cage, which consists of 24 ferritin protein molecules. The FcBP-ferritin cage showed greatly increased binding affinity to human IgG. Its binding was analyzed by QCM and SPR analysis. Finally, it was selectively delivered by Herceptin to SKBR3, a breast cancer cell, over MCF10A, non-tumorigenic cells.

  • To increase detection sensitivity for multi-DDT residues (o,p-/p,p-DDT, o,p-/p,p-DDE, o,o-/o,p-DDD) analysis, a highly selective sample clean-up method was introduced prior to GC/MS analysis using immunoaffinity column. The immunoaffinity matrix was prepared by coupling IgG fraction of DDT antiserum to cyanogens bromide activated Sepharose 4B. Three DDT antisera (DDA-1, DDHP-2, DDCP-3) were test for affinity column ligand that obtained by imunizing respective DDT immunogen to rabbits, and IgG was purified using protein A affinity purification.   • Gromwell colorants were extracted with methanol and dried. Four fractions were obtained by silica gel adsorption column chromatography using step-wise elution method. Relative ratio of four fraction is 1.00:0.07:0.22:0.30(Fl:F2:F3:F4) and gromwell colorants mainly consist of Fl, F3 and F4. IR analysis shows that each fraction has similar structure. Main component of gromwell extracts is acetyl derivative of naphthoquinone, and the rest are isobutyl derivative and isovaleryl derivative etc., in order. Gromwell colorants exhibit relatively good affinity to protein and polyamide fibers, but low affinity to cellulose and regenerated cellulose fibers.

  • For the investigations of the usefulness of immunoglobulin Y (IgY), we attempted to produce and isolate IgY from egg yolk of white Leghorn hens immunized with fetuin as a model antigen. We used three methods to optimize the recovery of IgY: water dilution, polyethylene glycol, and carrageenan. The daily yield of anti-fetuin IgY from egg yolk was also examined, and it was isolated using a fetuin-affinity column at a yield of IgY of 2.7% from total IgY Furthermore, peroxidase-labeled antifetuin IgY was prepared, and was used to determine the minimum sensitivity against fetuin, which was found to occur to a fetuin concentration of 1 ng/$m\ell$.

  • For the production of $B^{30}-homoserine$ insulin analog as a novel anti-diabetic drug, the fermentative study was attempted for the maximal gene expression of HTS-fused $B^{30}-homoserine$ insulin precursor in the recombinant Escherichia coli cells. In a batch fermentation, the maximal production of insulin precursor as much as 38.95 mg/L-h, which occupied more than 12.8% of total cell protein. was achieved when the gene expression was induced by 0.5 mM IPTG at the middle logarithmic growth phase. The HTS-fused $B^{30}-homoserine$ insulin precursor was recovered from a batch culture through the processes of cell harvest, collection of insoluble fraction after sonication and purification by nickel affinity column chromatography. The isolated insulin precursor was 14 mg/L with a recovery yield of 35.9% of expressed gene product. The insulin A and B chain mixture was recovered after the insulin precursor was subjected to CNBr cleavage and purified by nickel affinity column chromatography. The isolated insulin chains were then sulfitolyzed with sodium thiosulfat and sodium tetrathionate, and reconstituted to insulin analog with ${\beta}-mercaptoethanol$, followed by purification with CM-Sepharose C-25 column chromatography.

난백 내 Ovotransferrin의 분리방법에 관한 연구 (Development of the Purification Method of Ovotransferrin in Egg White)

  • 난백 단백질 중 ovotransferrin을 gel chromato- graphy와 heparin affinity chromatography를 통하여 분리 하였다. 1차 gel filtration의 경우에 샘플인젝션 후 65-70min(fraction No. 14) 이후에는 ovalbumin이 ovotransferrin과 혼입되어 분리되고 오히려 ovalbumin의 농도가 더 높은 분포를 보였다. 고순도의 ovotransferrin을 분리하기 위하여 다시 fraction No. 12-14을 농축한 뒤 gel filtration을 실시한 결과 ovotransferrin이 완전히 분리되지 않았는데 이는 gel filtration만의 반복을 통해서 순수한 ovotransferrin을 얻는 것이 비효과적임을 의미하는 것으로 판단된다. Ovotransferrin을 heparin affinity chromatography을 이용하여 분리한 경우 칼럼에 Fe2+를 고정시킨 후 50mM EDTA를 흘려 주었는데 ovalbumin이 5-10분경에 용출이 되었고 10-15분경에 ovalbumin과 ovotransferrin이 같이 용출되었다. 그 후에 50mM Phosphate buffer (pH 7.2, 0.15M salt)를 흘려주었는데 여전히 ovalbumin의 밴드가 보여 순수하지 않음을 확인하였다. Fe3+를 컬럼에 고정시킨 후 50mM EDTA를 흘려주었을 때 ovalbumin이 10-15분경에 용출이 되었고 15-20분경에 ovalbumin과 ovotransferrin이 같이 용출되었지만 50mM Phosphate buffer (pH 7.2, 0.15M salt free)를 흘려주었을 때 156-165분경에 ovalbumin이 혼입되지 않은 매우 순수한 ovotransferrin이 용출되는 것을 확인하였다. 위의 결과를 종합해볼 때 gel chromatography를 반복적으로 실시한 경우 보다는 heparin affinity chromatography를 이용하여 분리하고 컬럼에 Fe2+를 고정시킨 경우보다 Fe3+를 고정시켰을 때 더욱 순수한 ovotransferrin을 분리해낼 수 있었다.
