  • Biodegradability of the $poly({\beta}-hydroxybutyrate-co-3 -hydroxyvalerate)$ [PHB/V] containing 0, 10 and 15mol% hydroxyvalerate [HV] was studied. Bioderadability of PHB/V was evaluated at $30^{\circ}C$ for 58 days and $55^{\circ}C$ for 33 days by monitoring the time-dependent changes in weight loss(erosion) of aerobic conditions in a temperature-controlled microcosms containing the earthworm cast($30^{\circ}C$) and compost ($55^{\circ}C$). It was found that PHB/V biodegradability occurred with increasing HV monomer concentration from 0 mol% to 15 mol%.

Utilization of the Bulking Agents for Environment-friendly Toilet in Rural Village (농촌마을에서 자연발효형 화장실의 통기성 매질 이용에 관한 연구)

  • The biodegradability test of bulking agents in feces composting process is conducted to increase the management efficiency for environment-friendly rural toilet developed by National Rural Resource Institute in 2001. In the case study through references and field study, ash, fallen leaves, and wood chips are commonly used in the ecological toilet. Extension officers and farmers prefer rice straw, fallen leaves, and chaff among the rural resources as a bulking agents in the aspect of getting and storage as the result of questionnaire survey. As the result of biodegradability test including the temperature change, $CO_2$ gas production, and C/N ratio change of composting pile under the condition of aerobic composting apparatus, rice straw and chaff are good bulking agents for environment-friendly rural toilet. It is recommended that easy getting materials among the above mentioned materials are used for bulking agents as the region and season in environment-friendly rural toilet.

  • The authors investigated the biodegradability and toxicity of polyoxyethylene-alkyl citric diester-triethanolamine(PAT), which is one of new non-ionic detergents. 1) The PAT is very biodegradable, which 50% is biodegraded for one day and 100% for eight days by the activated sludge treatment and 40% for one day and 93% for eight days by the aerobic domestic sewage treatment. 2) $LD_{50}$ of PAT for chicken, mouse, and rat by oral adminstration are 7.1g/kg, 8.4g/kg and 14.0g/kg reactively. 3) $TL_{m}$ of PAT for goldfish is 64.0mg/l in 24bours. 4) No reaction for humun skin is determined by PAT.

Effect of Salt Concentration on the Aerobic Biodegradability of Sea Food Wastewater (수산물 가공폐수의 호기성 생분해도에 미치는 염분농도의 영향)

  • The study was performed to evaluate the effects of chloride concentrations on the ultimate aerobic biodegradability and to use the result as the fundamental data for sea food wastewater treatment. When the organic removal efficiency by chloride concentrations (1,400~18,000 mg/L) was evaluated, microbes adapted to the saline at ${\leq}$ 6,000 mg/L of chloride but treatment efficiency was not improved at ${\geq}$ 12,000 mg/L of chloride because of delayed reaction time. Functional coefficient $Y_I$ of non-biodegradable soluble organic and inert material production coefficient Yp by microbe metabolism increased as chloride concentrations increased. Soluble organic matter ratio by chloride concentration (0~18,000 mg/L) was 10.8~13.1%, inert material production efficiency by microbes metabolism was evaluated as 7.0~24.6%. $NH_3$-N removal efficiencies were 96.2, 96.5, 90.2 and 90.3% using original wastewater HRT 18 hr, 6,000 mg/L chloride concentration HRT 22 hr, 12,000 mg/L chloride concentration HRT 30 hr, and 18,000 mg/L chloride concentration HRT 45 hr, respectively. Nitrification process was more sensitive to salt concentration than organic matter removal to salt concentration. Under ${\geq}$ 6,000 mg/L chloride concentration, conversion rate from $NO_s$-N to $NO_2$-N was low.

Effects of Advanced Oxidation of Penicillin on Biotoxicity, Biodegradability and Subsequent Biological Treatment (고도산화공정 처리가 페니실린의 생독성, 생분해도 및 생물학적 분해에 미치는 영향)

  • Advanced oxidation processes (AOPs) composed of O3 and UV were applied to degrade penicillin (PEN). The degradation efficiency was evaluated in terms of changes in the absorbance (ABS) and total organic carbon (TOC). The combination of $O_3/H_2O_2/UV$ and $O_3/UV$ showed the best performance for the reduction of ABS (100% for 9 min) and TOC (70% for 60 min) values, although the mineralization was uncompleted under the experimental condition in this study. The change in biotoxicy was monitored with Escherichia coli susceptibility and Vibrio fischeri biofluorescence. The E. coli susceptibility was eliminated completely for 9 min by $O_3/UV$, and the toxicity to V. fischeri biofluorescence was 57% reduced by $O_3/H_2O_2/UV$. For the ultimate treatment of PEN, it is suggested that an AOP using $O_3/UV$ is followed by biological treatment, utilizing the enhanced biodegradability by the AOP. During 30 min of $O_3/UV$ treatment, the $BOD_5/COD$ ratio as an indication of biodegradability showed about 4-fold increment, compared to that of using a non-treated sample. TOC removal rate for AOP-pretreated PEN wastewater increased 55% compared to that of using the non-pretreated one through an aerobic biological treatment by Pseudomonas putida for artificial wastewater containing 20 mg/L of PEN. In conclusion, $O_3/UV$ process is recommended as a pretreatment step prior to an aerobic biological process to improve the ultimate degradation of penicillin.

  • Various methods (OECD 301B, ISO 9439 and ASTM 5864) for biodegradability test of lubricants were reviewed, and a standard procedure was developed. Most lubrication products are released in rivers or sea then is degraded by microbial action in aerobic condition. Most international method are based on $CO_2$ evolution test. Inoculum obtained from a sewage disposal plant and test compound are cultivated in a mineral medium. Organic carbon of the test compound is degraded and oxidized through the enzymatic actions of inoculum, and ultimately mineralized to carbon dioxide. Biodegradability test conditions of lubricant oils were optimized. The highest biodegradability was achieved when the same medium as in ASTM 5864 and inoculum concentration of $10^4{\sim}10^5$ cell/L were used. The optimum standard materials were selected as aniline and sodium acetate. Additionally the effects of inoculum type on microbial growth and biodegradability were examined. Finally the standard operating procedure (SOP) for biodegradability test method was proposed.

Estimation of the methane generation rate constant using a large-scale respirometer at a landfill site

  • The objective of this study is the evaluation of the performance of a large-scale respirometer (LSR) of 17.7 L in the determination of the methane generation rate constant (k) values. To achieve this objective, a comparison between anaerobic (GB21) and LSR tests was conducted. The data were modeled using a linear function, and the resulting correlation coefficient ($R^2$) of the linear regression is 0.91. This result shows that despite the aerobic conditions, the biodegradability values that were obtained from the LSR test produced results that are similar to those from the GB21 test. In this respect, the LSR test can be an indicator of the anaerobic biodegradability for landfill waste. In addition, the results show the high repeatability of the tests with an average coefficient of variance (CV) that is lower than 10%; furthermore, the CV for the LSR is lower than that of the GB21, which indicates that the LSR-test method could provide a better representation of waste samples. Therefore, the LSR method allows for both the prediction of the long-term biodegradation potential in a shorter length of time and the reduction of the sampling errors that are caused by the heterogeneity of waste samples. The k values are $0.156y^{-1}$ and $0.127y^{-1}$ for the cumulative biogas production (GB21) and the cumulative oxygen uptake for the LSR, respectively.

A Study on Groundwater Quality and Biodegradability at the Petroleum Contaminated Sites of Kangwon and Gyeonggi Provinces, Korea (강원도 및 경기도 소재 유류오염지역의 수질특성, 발현 생분해능과 오염물질 저감속도)

  • This study focused on the groundwater quality, biodegradability and attenuation rate at the petroleum contaminated sites of Kangwon and Gyeonggi Provinces, Korea. For groundwater quality, Kangwon site showed chemical compositions of $Ca-SO_4+Cl$, $Ca-HCO_3$ and $Na+K-HCO_3$ types, while Gyeonggi site showed chemical compositions of $Ca-SO_4$, $Ca-HCO_3$ and $Na-HCO_3$ types. $Na+K-HCO_3$ and $Na-HCO_3$ types were detected only in February. Among many biodegradation processes, the majority was attributed to biodegradation from denitrification in both area. In Kangwon site, biodegradation from denitrification occupied 63.5%, and in Gyeonggi site it was 39.45%. Biodegradation from the most efficient aerobic respiration occupied 7.12% in Kangwon site, while Gyeonggi site in it did 27.29%. Point attenuation rate of BTEX in Gyeonggi site (GW-22) was 0.0182 $day^{-1}$, half life of BTEX was 84 days, and thus 124 days (0.34 year) would be required to clean up this site. Mean of point attenuation rate of TPH in Kangwon site was 0.0088 $day^{-1}$, mean of half life was 257 days, and thus 462 days would be required to clean up the site. Mean of point attenuation rate of TPH in Gyeonggi site was 0.0387 $day^{-1}$, mean of half life was 55 days, and thus remediation time was calculated as 99 days.

Evaluation of Biodegradation in an Aquifer Contaminated with Petroleum Hydrocarbon

  • To evaluate the biodegradability of contaminants in an aquifer, computer modeling with RT3D model (Clement, 1997) was used. The RT3D model simulates the biodegradation of organic contaminants using a number of aerobic and anaerobic electron acceptors. The RT3D model was applied to a well-studied petroleum hydrocarbon plume in a shallow unconfined aquifer in Uiwang, Korea. The results of this study demonstrate tile importance of biodegradation processes in the monitored natural attenuation and in reducing contaminant concentrations in a shallow aquifer. The modeling results tell that the amount of electron acceptors is the key factor affecting biodegradation of TEX, the petroleum hydrocarbon contaminant in shallow groundwater

Biodegradation of Potential Diesel-Oxygenate Additive Including DBM(DiButyl Maleate) (DBM(DiButyl Maleate)을 포함한 잠재적 디젤첨가제 생분해특성)

  • In this study, we have evaluated biodegradability of diesel-oxygenates including DBM and gasoline-oxygenates having similar physio-chemical properties using indigenous aerobic microorganisms from a diesel-contaminated soil. Toluene and Ethanol have shown higher biological activity and the first-order degradation rate constants ranged around $0.11{\sim}0.3day^{-1}$. However, MTBE, gasoline-oxygenate has shown as a limited substrate. Moreover, As increased initial concentrations of DBM and TGME, degradation rates of those were decreased relatively. As a strategy to evaluate biodegradability of DBM and TGME, reduction of diesel-oxygenates, $CO_2$ production and toxicity by algae were monitored. This results indicated possible mineralization of diesel-oxygenates, But we could predict that residual byproduct produced even though complete consumption of diesel-oxygenates were observed if algal toxicity variation considered. In conclusion, it is the first report that diesel-oxygenates including DBM could be biodegraded effectively by indigenous soil microorganisms and this result increased the possibility of bioremediation technology to apply into oil-contaminated sites.
