Changes of Germination Rate of Pulses Seed Germplasm after Long-term Conservation

  • The seeds of soybean (Glycine max), adzuki bean (Vigna angularis), mung bean (Vigna radiata), and kidney bean (Phaseolus vulgaris L.) were examined the germination rate after 10 years of long-term storage ($-18^{\circ}C$) conservation. For soybean seeds, 2,313 accessions were examined and germination rate of 1,082 accessions was decreased with below 15% of initial germination rate. For 227 accessions of soybean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 589 accessions was increased and showed no change for 415 accessions after 10 years of long-term storage. For adzuki bean seeds, 2,058 accessions were examined and germination rate of 739 accessions was decreased with below 15% of initial germination rate. For 63 accessions of adzuki bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 535 accessions was increased and showed no change for 721 accessions after 10 years of long-term storage. For mung bean seeds, 438 accessions were examined and germination rate of 139 accessions was decreased with below 15% of initial germination rate. For 5 accessions of mung bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 155 accessions was increased and showed no change for 139 accessions after 10 years of long-term storage. For kdney bean seeds, 366 accessions were examined and germination rate of 7 accessions was decreased with below 15% of initial germination rate. For 65 accessions of kidney bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 201 accessions was increased and showed no change for 93 accessions after 10 years of long-term storage.

Studies on the Trypsin Inhibitor in Raw Beans of Korea (한국산 두류의 Trypsin Inhibitor에 관한 생화학적 연구)

  • This study was undertaken establish the relationship between trypsin inhibitor in raw soybean and antinutritional effect of raw legumes. 1) Among legumes produced in Korea, Glycine max contains a relatively high amount of protein(higher than 40%) compared with kindey bean, sword bean and mung bean and, furthermore, soybean which contains a high amount of protein possesses high trypsin inhibitory activity. 2) Disc electrophoretic pattern exhibited pattern exhibited that the crude protein preparation from Glycine max produced about 9-12 protein bands, and the pattern of electrophoretic mobility was very similar to each other. However, only a few protein bands were observed from the crude protein preparation of yard long bean, sword bean, adzuki bean, mung bean and rice adzuki. From the eluate of the sliced gel, it was confirmed that among those bands, only the fastest moving band contains trypsin inhibitory activity. 3) In chicks fed the normal diet the body weight was increased steady from one week and reached to 40% increase for three weeks but in chick fed raw bean diet, there was no body weight gain until two weeks feeding and only 10-20% of body weight gain was observed at the end of three week feeding. On the other hand, in chicks fed raw bean diet the weight of pancreatic tissue per 100g body weight was increased to about two-fold for two or three weeks but there was no change in liver weight. 4) In the case of amylase secretion from the pancreatic fragment, very strong stimulation on amylase secretion from pancreatic tissue of chicks fed a normal diet was produced by one unit of cholecystokin-pancreozymin. However, no stimulation was observed from pancreatic fragment of chick fed raw bean diet.

Effect of different water levels on the photosynthetic pigments of crops

  • An excess soil water condition is one of the major problems for the field crops growing in paddy fields because of their poor drainage and less availability for oxygen uptake which leads to adversely affect the photosynthesis. Therefore, the current study was undertaken with aim to investigate the effects groundwater level on the photosynthetic response of soy bean (Urum), red bean (Arari), sesame (Geonbaek), perilla (Dayu) after the transplanting to the lysimeter to investigate the plant-water relation and their effect on photosynthesis. The chlorophyll content of the crops according to the humid conditions of the soy bean, sesame and the perilla was found to be 5%, 6.89 % and 13.7% higher than that of the groundwater treated at 40cm, respectively. On the other hand, the chlorophyll content of adzuki bean decreased 6.6% from the groundwater level of 40cm, and the sorghum decreased by 5.7%. As a result of investigating the Fv / Fm value of groundwater, the adzuki bean at 20cm above groundwater was lower than that of groundwater by 40cm immediately before flowering. The Fv / Fm value of soy bean and sesame at 40cm above groundwater were lowered by flowering under groundwater 20 cm and Fv / Fm value of sorghum is increased at 40 cm treatment immediately before flowering while the Fv / Fm values of the perilla had no significant difference in comparison to those at 20 cm and 40 cm of groundwater. In the case of chlorophyll fluorescence reaction, it is known that the when the absolute value is closer to 0.82, the stress is considered less. As a result of comparing the numerical values of the crops, it was found that the sorghum was the most stressed followed by adzuki bean and sesame, while the soy beans and perilla was found on the average, as they received less stress.

The growth and yield changes of foxtail millet (Setaria italic L.), proso millet (Panicum miliaceum L.), sorghum (Sorghum bicolor L.), adzuki bean (Vigna angularis L.), and sesame (Sesamum indicum L.) as affected by excessive soil-water

  • The objectives of this study were to investigate the effects of excessive soil-water on crop growth and to predict decrease of yields caused by excessive soil-water. The following five crops were selected for investigation: foxtail millet, proso millet, sorghum, adzuki bean, and sesame. These were planted in pots and a soil-water table was set to 10cm for 10 days. Crop susceptibility (CS) factors and stress-day indexes (SDI) were calculated for each crop to estimate effects of excessive soil-water. SDI models were calculated using CS and SDI data for each crop and predicted the yields of crops cultivated in paddy fields. All crops were cultivated in paddy fields with different soil water contents to evaluate the yield-SDI models. Results showed that yields decreased most when crops were affected by excessive soil-water at the early development stage. Decrease of yields was the greatest when the excessive soil-water treatment was applied at early growth stage. In the field experiment, crops from soils with the greatest soil-water content had the smallest yield, while ones from soils with the smallest soil water contents showed the greatest yields. Observed yields from the field and predicted yields from SDI models showed the least correlation for proso millet, foxtail millet, and adzuki bean and the greatest correlation for sesame. In conclusion, proso millet, foxtail millet, and adzuki bean were more susceptible to soil water than other crops, while sorghum and sesame were more suitable to cultivation in paddy fields.

Comparison of Antioxidant and yield Properties of of Adzuki Bean (Vigna angularis L.) Cultivars Under Different Air Temperatures and Sunshine Hours

  • This study investigated the antioxidant and yield properties of eight adzuki bean (Vigna angularis) cultivars under different mean temperature and sunshine hours in 2020 and 2021. The mean temperature in 2020 compared to 2021, which was normal, were lower during the pod extension and grain filling period. In addition, due to frequent rainfall during the vegetative perio d in 2020, there were fewer hours of sunshine compared to 2021. The Adzuki Bean yield in 2020 was about 13% lower than that in 2021 due to the decreased number of pods per plant and 100 seeds weight. The stability of yields was higher in 'Hongda' and 'Hongjin' than in 'Arari', which is the most cultivated in Korea. Also, the yields of these varieties were all higher than those of 'Arari' varieties for two years. Compared to 2021, in 2020, when weather conditions were not favorable, both antioxidant components and activity were higher. The cultivar 'Hongda' was a higher antioxidant component and activity than others for two years. The results suggested that the antioxidant component and activity were good in 2020 with about 13% decrease in yield compared to a normal year in 2021 due to mean temperature and sunshine hours.

Quality Characteristics of Adzuki Beans Sediment According to Variety (품종에 따른 팥 앙금의 품질 특성)

  • We evaluated the quality characteristics of adzuki bean sediment according to variety. The moisture, crude protein, and crude ash contents of the various adzuki bean varieties were 8.2~11.1, 15.4~20.6 and 3.3~3.6 g/100 g, respectively. The potassium contents of Chilbo-pat (CB) and Hongeon-pat (HE) were 875.1 and 873.1 mg/100 g, respectively. The calcium contents of Jungbu-pat (JB) and Kumsil-pat (KS)were 73.6 and 73.2 mg/100 g, respectively. A high level of magnesium (131.4 mg/100 g) was found in Yeonkeum-pat (YK). The yields of adzuki bean sediment according to variety were no different either wet (188.3~204.7%) or dry (62.1~66.0%). The L-values on sediment of YK and KS were 67.0 and 68.0, respectively; however, the CB L-value was low at 54.0. A high level of a- (6.6) and b-value (12.8) was found in YK; however, the values for CB were much lower at 3.8 and 5.9, respectively. There was no difference in particle-size distribution, water binding capacity, and solubility of adzuki bean sediment according to variety. High levels of peak (3.79 RVU), trough (3.75 RVU), final (7.33 RVU), and setback viscosity (3.54 RVU) were found in JB. The sensory properties of products in food processing are important, and the variety of adzuki bean sediment should be chosen depending on desired product characteristics.

Investigation of Root Morphological and Architectural Traits in Adzuki Bean (Vigna angularis) Cultivars Using Imagery Data

  • Roots play important roles in water and nutrient uptake and in response to various environmental stresses. Investigating diversification of cultivars through root phenotyping is important for crop improvement in adzuki beans. Therefore, we analyzed the morphological and architectural root traits of 22 adzuki bean cultivars using 2-dimensional (2D) root imaging. Plants were grown in plastic tubes [6 cm (diameter) × 40 cm (height)] in a greenhouse from July 25th to August 28th. When the plants reached the 2nd or 3rd trifoliate leaf stage, the roots were removed and washed with tap water to remove soil particles. Clean root samples were scanned, and the scanned images were analyzed using the WinRHIZO Pro software. The cultivars were analyzed based on six root phenotypes [total root length (TRL), surface area (SA), average diameter (AD), and number of tips (NT) were included as root morphological traits (RMT); and link average length (LAL) and link average diameter (LAD) were included as root architectural traits (RAT)]. According to the analysis of variance (ANOVA), a significant difference was observed between the cultivars for all root morphological traits. Distribution analysis demonstrated that all root traits except LAL followed a normally distributed curve. In the correlation test, the most important morphological trait, TRL, showed a strong positive correlation with SA (r = 0.97***) and NT (r = 0.94***). In comparison, between RMT and RAT, TRL showed a significantly negative correlation with LAL (r = -0.50***); however, TRL did not show a correlation with LAD. Based on RMT and RAT, we identified the cultivars that ranked 5% from the top and bottom. In particular, the cultivar "IT 236657" showed the highest TRL, SA, and NT, while the cultivar "IT 236169" showed the lowest values for TRL, SA, and NT. In addition, the coefficient of variance for the six tested root traits ranged from (14.26-40%) which suggested statistical variability in root phenotypes among the 22 adzuki bean varieties. Thus, this study will help to select target root traits for the adzuki bean breeding program in the future, generating climate-resilient adzuki beans, especially for drought stress, and may be useful for developing biotic and abiotic stress-tolerant cultivars based on better root trait attributes.

Effect of Enhancement on Functionality of Germinated Adzuki Bean (Angularis angularis var. nipponensis) with High Hydrostatic Pressure (HHP) Treatment (고압처리에 의한 발아팥의 기능성 향상 효과)

  • We investigated the influence of germination and high hydrostatic pressure (HHP) treatment conditions on the conversion of functional compounds and antioxidant activity in adzuki bean. The adzuki bean germinated at $25^{\circ}C$ for three- or six-days, and was later subjected to HHP at 0.1, 50, 100, or 150 MPa for 24 h. The highest polyphenol content (5.36 mg gallic acid equivalents (GAE)/g) and flavonoid content (0.91 mg catechin equivalents (CE)/g) were observed after germination for six days and HHP treatment at 100 MPa for 24 h, respectively. The total phenolic acid contents increased with increasing applied pressure from 88.86 to $208.26{\mu}g/g$ (100MPa, 24h). Phenolic acids are divided into two categories; those that exhibit increased content upon HHP treatment, and those that exhibit decreased content. The increasing phenolic acids were gallic acid, chlorogenic acid, (+)-catechin, ${\rho}-coumaric$ acid, ferulic acid, heperidin, salicylic acid, protocatechuic acid, cinnamic acid, naringenin. The total anthocyanin content decreased with increasing applied pressure from 22.42 mg/100 g to 6.28 mg/100 g (150 MPa, 24 h). The highest ABTS radical scavenging activity (8.02 mg eq AA/g) and DPPH radical scavenging activity (1.22 eq Trolox/g) were observed after germination for six days and HHP treatment at 100MPa for 24h, respectively. These results suggested that the combination of HHP and germination can lead to improved functionality in adzuki bean.

Screening of Personalized Immunostimulatory Activities of Saengsik Materials and Products Using Human Primary Immune Cell (사람 면역세포를 활용한 생식 원료 및 제품의 체질 맞춤형 면역 활성 탐색)

  • Sasang constitutional medicine is a Korean traditional medicine in which individuals are classified into four constitutional types, Taeyangin, Taeumin, Soyangin, and Soeumin. To develop new functional Saengsik products, we investigated the immunostimulatory activities of raw materials of Saengsik (milled rice, barley, Job's tears, soybean, adzuki bean, millet, eggplants, radish, carrot, Lentinus edodes, Agaricus bisporus, and Auricularia auricula-judae) and Saengsik products (Mix 1~6) in primary immune cells (lymphocytes and macrophages) isolated from blood of Taeumin, Soyangin, and Soeumin types. Increased cell proliferation as well as NO and TNF-${\alpha}$ protein production by immune cells treated with extracts of materials of Saengsik and Saengsik products were measured as immunostimulatory parameters. We chose milled rice, adzuki bean, radish, and Lentinus edodes to study the immunostimulatory activity of Teaumin, barley, soybean, eggplants, and Agaricus bisporus to study the immunostimulatory activity of Soyangin, as well as Job's tears, adzuki bean, carrot, and Auricularia auricula-judae to study the immunostimulatory activity of Soeumin. The mixtures (Mix 1~6) were made with different kinds and ratios of Saengsik materials based on the immunostimulatory activities of Saengsik materials. The immunostimulatory activity of Taeumin was highest in Mix 2 (rice 60%, adzuki bean 20%, radish 10%, and Lentinus edodes 10%) extract-treated immune cells among all six mixtures. The immunostimulatory activity of Soyangin responded best to Mix 4 (barley 60%, soybean 20%, eggplants 10%, and Agaricus bisporus 10%) among the six extracts (Mix 1~6). The increased immunostimulatory activity of Mix 6 (Job's tears 60%, adzuki bean 20%, carrot 10%, and Auricularia auricula-judae 10%)-treated immune cells was higher than the other five extracts (Mix 1~5). Accordingly, Mix 2, Mix 4, and Mix 6 may be useful as mixtures for Saengsik products having personalized immunostimulatory activities.

Changes in Physicochemical Characteristics and Antioxidant Activity of Adzuki Bean and Adzuki Bean Tea Depending on the Variety and Roasting Time (팥 품종별 볶음시간에 따른 팥과 팥차의 이화학적 특성 및 항산화활성의 변화)

  • This study investigated the changes in physicochemical characteristics of adzuki bean (Vigna angularis var. nipponensis) tea depending on the variety and roasting times, for the development of functional foods. The levels of total polyphenol, flavonoids, and tannin contents were 12.72 mg gallic acid equivalent (GAE)/g, 3.01 mg catechin equivalent (CE)/g, and 3.56 mg tannic acid equivalent (TAE)/g, respectively, for the Jeolgangsung-ipat sample roasted for 16 min. The highest DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity was 7.81 mg Trolox equivalent (TE)/g in Bulgeun-ipat sample that was roasted for 14 min. The highest total polyphenol content in the hot water-leached liqueur of adzuki bean tea was $26.55{\mu}g$ GAE/100 mL in Yungum-pat that was roasted for 12 min. The total flavonoids and tannin contents of Whin-ipat and Jeolgangsung-ipat roasted for 10 min were $15.10{\mu}g$ CE/100 mL and $14.60{\mu}g$ TAE/100 mL, respectively. The highest DPPH and ABTS (2,2'-azino-bis-3-ethylbenzo-thiazoline-6-sulfonic acid) radical scavenging activities of hot water-leached liqueur of adzuki bean tea were 488.72 and 728.25 mg TE/100 mL, respectively, in Yungumpat roasted that was for 12 min. The results of this study show that roasted adzuki bean tea has notable antioxidant activity and is considered to have significant health benefits.
