Induced neural stem cells from human patient-derived fibroblasts attenuate neurodegeneration in Niemann-Pick type C mice

  • Background: Niemann-Pick disease type C (NPC) is caused by the mutation of NPC genes, which leads to the abnormal accumulation of unesterified cholesterol and glycolipids in lysosomes. This autosomal recessive disease is characterized by liver dysfunction, hepatosplenomegaly, and progressive neurodegeneration. Recently, the application of induced neural stem cells (iNSCs), converted from fibroblasts using specific transcription factors, to repair degenerated lesions has been considered a novel therapy. Objectives: The therapeutic effects on NPC by human iNSCs generated by our research group have not yet been studied in vivo; in this study, we investigate those effects. Methods: We used an NPC mouse model to efficiently evaluate the therapeutic effect of iNSCs, because neurodegeneration progress is rapid in NPC. In addition, application of human iNSCs from NPC patient-derived fibroblasts in an NPC model in vivo can give insight into the clinical usefulness of iNSC treatment. The iNSCs, generated from NPC patientderived fibroblasts using the SOX2 and HMGA2 reprogramming factors, were transplanted by intracerebral injection into NPC mice. Results: Transplantation of iNSCs showed positive results in survival and body weight change in vivo. Additionally, iNSC-treated mice showed improved learning and memory in behavior test results. Furthermore, through magnetic resonance imaging and histopathological assessments, we observed delayed neurodegeneration in NPC mouse brains. Conclusions: iNSCs converted from patient-derived fibroblasts can become another choice of treatment for neurodegenerative diseases such as NPC.

Famine exposure in early life and type 2 diabetes in adulthood: findings from prospective studies in China

  • BACKGROUND/OBJECTIVES: This study examined the relationship between famine exposure in early life and the risk of type 2 diabetes in adulthood during the 1959-1961 Chinese Famine. SUBJECTS/METHODS: A total of 3,418 individuals aged 35-74 years free of diabetes from two studies in 2006 and 2009 were followed up prospectively in 2009 and 2012, respectively. Famine exposure was classified as unexposed (individuals born in 1962-1978), fetal exposed (individuals born in 1959-1961), child exposed (individuals born in 1949-1958), and adolescent/adult exposed (born in 1931-1948). A logistic regression model was used to assess the relationship between famine exposure and diabetes after adjustment for potential covariates. RESULTS: During a three-year follow-up, the age-adjusted incidence rates of type 2 diabetes were 5.7%, 14.5%, 12.7%, and 17.8% in unexposed, fetal-exposed, child-exposed, and adolescent/adult-exposed groups, respectively (P < 0.01). Relative to the unexposed group, the relative risks (95% confidence interval) for diabetes were 2.15 (1.29-3.60), 1.53 (0.93-2.51), and 1.65 (0.75-3.63) in the fetal-exposed, child-exposed, and adolescent/adult-exposed groups, after controlling for potential covariates. The interactions between famine exposure and obesity, education level, and family history of diabetes were not observed, except for the urbanization type. Individuals living in rural areas with fetal and childhood famine exposure were at a higher risk of type 2 diabetes, with relative risks of 8.79 (1.82-42.54) and 2.33 (1.17-4.65), respectively. CONCLUSIONS: These findings indicate that famine exposure in early life is an independent predictor of type 2 diabetes, particularly in women. Early identification and intervention may help prevent diabetes in later life.

  • We examined the cause of a disease outbreak in adult olive flounder Paralichthys olivaceus, which occurred at a Korean aquaculture farm in Korea in 2011. The principal signs included an expanded abdomen and congested liver, with persistent mortality (a little over two months). At the beginning of the outbreak, farm administrators misjudged the disease as bacterial in origin, because of the aforementioned signs, persistent mortality, and the detection of bacterial species, including Vibrio spp. and Streptococcus spp. Moreover, the detection of viral hemorrhagic septicemia virus (VHSV) by reverse trasnscription-PCR analysis was complicated by use of the VHS-VN primer set, which has been in general use recently, because it produced weak bands in some samples. Therefore, we recommend the use of at least two different primer sets in the diagnosis of VHSV. Our histopathological findings indicate that necrotizing myocarditis could be considered a pathogenic sign of VHSV infection.

  • The purpose of this study is to identify the characteristics of physical setting of adult day care as a place for the elders and adult and to develop an basic understanding of the architectural program and physical setting for adult day care for the cognitively-impaired in the U.S.A. The data was collected from 13 Adult Day Care Centers(ADCC) from 2001 to 2002 by interview and the documents about those facilities. Physical environment should be viewed as an element in the care of individuals with dementia, and that the principles used to plan and design environment should be consistent with the principles used in providing other aspects of care. The results of the study provided the information about the best condition of physical setting of ADCC for the elderly with dementia. Memory Loss Adult Day Center and St. Ann Center for Intergenerational Care have better architectural program than other facilities. It is the hope of this study to provide guideline for design and care professionals with a first draft of a "sense-making" template and to slow own the progression of the disease by the appropriate physical environment.

Echocardiograpyhic Assessment and Clinical Application of Cardiac Disease in Korea Jin-do Dog III. Detection of Heartworm (진돗개에서 심장초음파 측정치와 평가와 임상적 응용)

  • Echocardiography was performed to obtain the optimal planes for diagnosis of canine heartworm disease in 11 dogs. Imaging planes were taken the left ventricular outflow tract long-axis view, 4-chamber long-axis view, chordae tendineae level short-axis view and modified pulmonary arteries level short-axis view in the right parasternal window, and aptical 4-chamber long-axis view in hte left parasternal window. After echocaridography, the pulmonary artery valve pat was ligated with double 1-0 silk under the general anesthesia to prevent heartworm moving from pulmonary artery to right ventricle. The dogs were euthanized and examained for heartworms in heart and great vessels. Many adult heartworms were identified ultrasonographically within main pulmonary artery, bifurcation of pulmonary artery and right pulmonary artery, but not identified within right ventricle and atrium. At necropsy, adult heartworms were mainly found in pulmonary artery in 11 days, and also found in right ventricular outflow tract and right ventricle in 2 of 11 dogs. The modified pulmonary arteries level short-axis view was the best imaging plane for identifiying heartworms because the main pulmonary artery, branches of pulmonary artery and right pulmonary artery could be showed. The aortic root internal dimension (AOID) and right pulmonary atery internal dimension (RPAID) were measured from the modified pulmonary ateries level short-axis view and left ventricular outflow tract long-axis view. The RPAID and RPAID/AOID was higher in heartworm infected dogs than normal Korea Jin-do dogs in modified pulmonary arteries level short-axis view, and the AOID, RPAID and RPIAID/AOID was higher than in left ventricular outflow tract long-axis view, respectively. These results indicate that the pulmonary arteries were the major habitat of adult heartworm in canine heartworm disease and the modified pulmonary arteries level short-axis view was the best imaging planes for identifying heartworms because the main pulmonary arteries could be showed. Therefore the modified pulmonary arteries level short-axis view can be used for diagnosing heartworm disease and for monitoring dilation of pulmonary artery.

Mindfulness Meditation Experiences among Patients with Coronary Artery Disease: A Phenomenological Study (관상동맥질환자의 마음챙김 명상에 대한 경험: 현상학적 연구)

  • Purpose: This study was aimed to explore and describe the experiences of mindfulness meditation (mindfulness- based stress reduction, MBSR) among patients with coronary artery disease (CAD) as a method of cardiac rehabilitation. Methods: In-depth interviews was conducted with 7 participants with CAD post percutaneous coronary intervention. Main question was 'what is your experiences with MBSR?' Colaizzi's seven steps of phenomenological analysis was used for eliciting embedded meanings. Results: Five theme clusters were found; rediscovery of mindfulness meditation, obtaining self-control for stress management, self-worth, feeling of happiness, and looking into myself. Conclusion: This study provides a profound picture on the experiences of mindfulness meditation among participants with coronary artery disease. MBSR can help patients with CAD in reducing stress and to enhance their cardiac rehabilitations. It is hoped that mindfulness meditation can be used in clinical setting as a part of cardiac rehabilitation program.

  • Purpose: The purpose of this study was to investigate the effect of the asthma management educational program given to the allergic asthmatics receiving immunotherapy due to house dust mite on the disease related to knowledge, stress, and self-efficacy. Method: The subjects of this study were the patients received immunotherapy to house dust mite at a week interval after being diagnosed for house dust mite allergic asthma at the respiratory center of a hospital affiliated to the university. They were divided into the experimental group of 29 patients who received asthma management education, the control group of 32 patients, and 61 patients in total. Experimental treatment, as an asthma management educational program, was the group education of one time and the reinforcement education of three times with environmental therapy and immunotherapy to house dust mite. Results: The results revealed that the improvement in disease related knowledge, the improvement in self-efficacy and the decrease in stress, were significantly higher in the experimental group than the control group. Conclusions: The asthma management educational program had an effect on improving the disease related knowledge and self-efficacy, and decreasing the stress of the patients asthmatics allergic to house dust mite.
