  • Normal phase or reversed phase liquid chromatographic separation of isoquinoline of heterocyclic compounds and structural isomers of external substituents, $COOCH_3$, CN and $CH_3$ has been carried out by using several different columns and various mobile phases. From this results, the order of elution of heterocyclic compounds appears to depend on the solvent effect with kinds of mobile phases. Retention mechanism of normal phase system for 2-methylindoline, 2-methylindole, benzoxazole and benzothiazole was also studied depending on adsorption strength between solute and stationary phase of column. However, retention factors of reversed phase system were found on hydrophobic interaction with solvophobic effect.

  • To improve the preservation of Kimchi, we isolated Lactobacillus plantarum lytic enzyme-producing strain from soil, and the enzyme was purified and characterized. From the observation of cultural and morpho- logical characteristics, the isolated strain was identified as Metarrisium anisopliae (Metschn.) Sorok. The enzyme was purified to 75-folds with 40% yields through affinity adsorption and CM-Sephadex C-50 column chromatog- raphy. The optimum pH and temperature for lytic activity are 4.0 and 40$\circ$C, respectively, and the enzyme acitvity is stable between pH 3.0 and 9.0, and up to 50$\circ$C. The enzyme is a monomeric protein with molecular weight of 40,000 daltons by SDS-PAGE and gel filtration. The enzyme is endopeptidase which breaks the peptide linkage of Lactobacillus plantarum peptidoglycan. The lytic action spectra confirmed that Leuconostoc mesenteroides, a useful strain for the fermentation of Kimchi, is not lysed by the enzyme. The enzyme activity is inhibited by N-bromosuccinimide (NBS), which probably indicates the involvement of tryptophan residue in active site of the enzyme, and also inhibited by Ag$^{+}$. The amino acid composition analysis showed that the enzyme contains more acidic amino acids than basic ones, and composition of alanine, glycine, proline and tyrosines was very high.

Cloning and Expression of Cyclodextrin Glycosyltransferase Gene from Paenibacillus sp. T16 Isolated from Hot Spring Soil in Northern Thailand

  • Gene encoding cyclodextrin glycosyltransferase (CGTase), from thermotolerant Paenibacillus sp. T16 isolated from hot spring area in northern Thailand, was cloned and expressed in E. coli (JM109). The nucleotide sequences of both wild type and transformed CGTases consisted of 2139 bp open reading frame, 713 deduced amino acids residues with difference of 4 amino acid residues. The recombinant cells required 24 h culture time and a neutral pH for culture medium to produce compatible amount of CGTase compared to 72 h culture time and pH 10 for wild type. The recombinant and wild-type CGTases were purified by starch adsorption and phenyl sepharose column chromatography and characterized in parallel. Both enzymes showed molecular weight of 77 kDa and similar optimum pHs and temperatures with recombinant enzyme showing broader range. There were some significant difference in pH, temperature stability and kinetic parameters. The presence of high starch concentration resulted in higher thermostability in recombinant enzyme than the wild type. The recombinant enzyme was more stable at higher temperature and lower pH, with lower $K_m$ for coupling reaction using cellobiose and cyclodextrins as substrates.

  • Monodisperse porous poly(styrene-co-divinylbenzene)(St-DVB) gel and poly(acrylonitrile-co-ethylene dimethacrylate)(AN-EDMA) gel have been prepared by seed polymerization using polystyrene seeds, which were prepared by dispersion polymerization. The St-DVB and AN-EDMA gels had a narrow size distribution and pores suitable for packing materials of HPLC. The columns packed with these gels were proved to have high efficiency for GPC or HPLC coluuns. Adsorption properties of $Cu^{2+}$, $Cd^{2+}$ and ${UO_2}^{2+}$ ions on AAN-EDMA gel prepared from amidoximation of AN-EDMA gel were also determined.

  • The antioxidative substances in black sesame and white sesame seed were investigated. The thiocyanate test indicated the presence of antioxidative substances in the extracts of black sesame seed as well as white sesame seed. The activity of white sesame seed was more strong than that of black sesame seed. The active components were discussed after purified using silica gel adsorption chromatography, Sephadex LH-20 chromatography, $C_{18}$ column chromatography, successively.

Zeolite Filtration for Ammonium Nitrogen Removal in Drinking Water Treatment (정수처리에서 암모니아성질소 제거를 위한 제올라이트 여과)

  • This study was conducted to evaluate the feasibility of ammonia removal by zeolite adsorption in drinking water treatment. In generally, drinking water treatment process is conducted coagulation/flocculation, sedimentation, sand filtration and disinfection. We tested feasibility with two method, one is powdered zeolite dosing to coagulation tank and the other is to substitute granular zeolite for sand of sand filter. In powdered zeolite test, raw water is used tap water with putting of 2 mg/l of NH$_4$$\^$+/-N. Filtration of granular zeolite was conducted with 80 cm of effective column high and 120 m/d of flow rate. At above 100 mg/1 of zeolite dosage, ammonia concentration was decreased below 0.5 mg/l of NH$_4$$\^$+/-N in powdered zeolite test. But, turbidity was increased to 30 NTU by powdered zeolite dosage. That turbidity was scarcely decreased in generally coagulant using condition in drinking water treatment. In granular zeolite test, ammonia was not detected in treated water until 8 days. This result suggest that using of granular zeolite in sand filter could be removal ammonia in winter. But we need regeneration at zeolite filtration for ammonia removal. So, it is to make clear that zeolite regeneration ability was compared KCl with NaCl. The result reveal that KCl was more excellent than NaCl. Optimum regeneration concentration of KCl was revealed 100 mM. Regeneration efficient was not increased at pH range 10∼12.5.

Adsorption and Recevery of Cu(II) and Zn(II) Ions by Algal Biomass (해조류를 이용한 Cu(II) 및 Zn(II) 이온의 흡착 및 회수)

  • Algal biomass was used in our study in order to remove some metals. After packing of 40~60 mesh algae powder into column for use of metal adsorbent, the metal solution of 1mL/min of flow rate was eluted to adsorb in algae. More amount of Cu(II) or Zn(II) ion in green algae, Ulva pertusa Kjellman than in brown algae, Sargassum horneri (Turner) C. Agarch were adsorbed and Cu(II) ion was more adsorbed in both algae than Zn(II) ion. Recovery of metal from algae is showing higher in acidic or neutral than in alkalic conditions. Cu(II) ion is recovered relatively higher than Zn(II) ion in our system.

Analysis and Comparison of Volatile Flavor Components in Rice Wine Fermented with Phellinus linteus Mycelium and Regular Commercial Rice Wine

  • This study identified and compared the volatile flavor components of two commercial rice wines: one fermented using the mycelium of Phellinus linteus and a regular commercial rice wine. The volatile flavor components were isolated from the infusions by Porapak Q (50-80 mesh) column adsorption. The concentrated aroma extracts were then analyzed and identified by GC and GC-MS. Thirty-four kinds of flavor components were identified in the mycelium-fermented rice wine, including 11 alcohols, 8 esters, 3 ketones, 6 acids, 3 hydrocarbones, and 4 others. In the regular commercial rice wine, 36 kindss of flavor compounds were identified, including 9 alcohols, 6 esters, 4 ketones, 6 acids, 9 hydrocarbones, and 2 others. Therefore, the data indicate that the primary flavor components in the rice wines were alcohols and esters.

  • Bacillus macerans cyclodextrin glucanotransferase(EC 2.4.1.19: 1, 4-${\alpha}$-D(1, 4-${\alpha}$-glucano)-transferase, CGTase) was purified by the technique of starch adsorption and DEAE-cellulose column chromatography. The molecular weight of the enzyme was 67,000, consisting of a subunit. The enzyme converted starch into ${\alpha}$-, ${\beta}$-, and ${\gamma}$-CD in the relative amounts of 1:1.68:0.32, respectively. In the early reaction period, maltohexose was formed mainly by the coupling reaction of ${\alpha}$-CD with D-glucose and then other oligosaccharides. Maltotetrose was formed mainly from ${\alpha}$-CD in the initial stage of hydrolysis of the enzyme and then small amount of other oligosaccharides. Maltotriose was a good substrate for the enzyme and maltosyl or D-glucopyranosyl group can be transfered from this sugar. In this work, D-glutosyl transfer was premiered.

Solid-Phase Extraction of L-Muscone from Aqueous Samples with Amberlite XAD-4 for Gas Chromatographic Assay

  • An efficient analytical method was devised for the accurate L-muscone assay in aqueous samples. It involves solid-phase extraction of L-muscone in adsorption mode using XAD-4 as the sorbent and dichloromethane modified with 10% (v/v) methanol as the eluting solvent. The gas chromatographic analysis of the eluate residue dissolved in toluene on a DB-5MS capillary column provided complete resolution of L-muscone from the co-extracted interferences. The overall method showed excellent linearity ($r^2{\geq}$ 0.9994) in the range of 0.1 to 2.0 $\mu\textrm{g}$/mL with good intra- and inter-day precisions (% RSD = 2.5~7.3) and with high extraction recovery rates ($\geq$ 98.1 %). When the present method was applied to a L-muscone herbal drink product, the within-batch RE (%) in the labeled concentration (1.5 $\mu\textrm{g}$/mL) for the three randomly chosen bottles were -2.4, -1.3 and -3.3 with high precision (% RSD $\leq$ 3.1). The present method is considered to be suitable for quality control evaluation on liquid drinks and other complex formulations fortified with L-muscone.
