  • Capillary zone electrophoresis (CZE) which is highly efficient separation technique has been domestically established having optimum detection sensitivity. By applying 20∼35 kV of electric potential to the narrow (50 ${\mu}m$ i.d.) capillary tubing filled with running buffer, this technique can quickly (< 20 min) separate the small quantities of sample with high separation efficiency (number of theoretical plates : 200,000∼500,000). Factors affecting the separation efficiency and resolution in CZE were examined by analyzing adenine and catecholamine derivatives.

The effect of oxygen on NAD breakdown in salmonella typhimurium

  • The breakdown rate of NAD in Salmonella typhimurium was investigated both in aerobic and anaerobic conditions. After NAD is broken down to nicotinamide ring containing moiety, almost all the nicotinamide ring containing moiety recycles back to NAD pool. However almost none of the adenine containing moiety recycles back. We pulse-label the endogeneous NAD with [$\^$14/C]-adenine and [$\^$3/H]- niacin. The remaining [$\^$14/C]-radioactivity in NAD pool at each time was regarded as unbroken portion of NAD, WHERE AS THE OF [$\^$3/H] was served as a total amount of NAD to start with. Under aerobic condition, the half-life of NAD was around 2 hours. However, the breakdown rate was significantly reduced (around 3-5 fold) under anaerobic condition. The observation that under aerobic conditions, NAD turnover is considerably faster than under anaerobic conditions suggests that oxygen has important effect in NAD breakdown.

  • The photocycloaddition reaction of 8-methoxypsoralen with purine and/or pyrimidine bases is studied as a model for the charge transfer interactions of psoralens with DNA bases by the FMO method. The results indicate that, in the case of the molecular complex formation between psoralens and purine and/or pyrimidine bases, the most probable photocycloaddition should occur in the following order: Thy (5,6)<>(3,4) 8-MOP, Cyt(5,6)<>(3,4)8-MOP, Ade (7,8)<>(3,4)8-MOP, Gua(7,8)<>(3,4)8-MOP. The theoretical results for the photocycloaddition reaction are also correlated with the experimental results. The photoadducts between 8-methoxypsoralen and adenine are likely to be C4-cycloadducts through the cycloaddition of 3,4-pyrone double bond of 8-methoxypsoralen to 7,8-double bond of adenine.

  • The interaction of salicylic acid (S. A.), salicylamide (S,M) with nucleic acid base derivatives such as 9-ethyl adenine (A), 1-cyclohexyl uracil (U), 2', 3'-benzylidine-5' trityl-cytidine (C), gaunosine-2', 3', 5'-isobutylate (G) has been spectroscopically investigated to determine the binding mechanism. NMR and IR spectra were measured in nonpolar solvents. The association constant K of the formation of complex was calculated from the IR spectra. Compounds S. A. and A form a 1:1 or 1:2 cyclic hydrogen-bonded complex depending on the sample concentration. Compounds S. A. and U form a 1:1 or 1:2 hydrogen-bonded complex on the sample concentration. Compounds S. A. and C form a 2:1 hydrogen-bonded complex at low concentration (0.0016M). Compound S. A. binds compound G, but its binding does not completely break the self-association of compound G, Compound S. M. binds compounds A. U. C. G. very weakly.

  • It is the most important step to screen soluble and insoluble proteins when we attempt to improve the solubility of recombinant proteins through directed evolution approach. Here we show that the solubility of a recombinant protein in vivo can be examined by expressing the recombinant protein with beta-lactamase as a fusion partner. First we constructed an expression system which can produc a fusion protein with the C-terminal of beta-lactamase. Two soluble proteins, i.e. adenine deaminase and aspartate aminotransferase, and insoluble GlcNAc-2-epimerase were cloned into the developed expression vector, respectively. We investigated the effect of the expression of the three recombinant fusion proteins on the growth of E. coli, and confirmed that the solubilities of the recombinant proteins correlated with cell growth rates.

  • The antitumor properities of metal are not limited to Pt(II) complexes. Recently, it was reported that some Pt(II) and Pd(II) complexes with sulfur and nitrogen ligands had potential antitumor reactivity. Pt(II) complexes is toxic drug for anticancer. Therefore, the complexes of Pd(II) with adenine, uracil, cytosine and malonate ligands are interesting for anticancer drug. We synthesized new palladium complexes containing nucleosides. The reactions of Pd(II) with adenine, uracil, cytosine and malonate ligands have been studied in aqueous solutions. The complexes isolated from these reactions have the following formulas; $[Pd(en)(C_5H_5N_5)_2](NO_3)_2,\;[Pd(en)(C_4H_3N_2O_2)Cl],\;[Pd(en)(C_3H_2O_4)]\;and\;[Pd(en)(C_4H_5N_3O)_2](NO_3)_2{\cdot}(C_4H_5N_3O)$. The compounds have been identified by elemental analysis, mass spectra, infrared spectra and electronic spectra.

  • The molecular composition of the postsynaptic density (PSD) is largely hon. In this report, an electroelution protocol was demonstrated to be used for efficient isolation of PSD proteins with diverse molecular sizes. Using this protocol, a 31 kDa protein in the 1% n-octyl glucoside-insoluble PSD fraction (termed as PSD31) was purified from SDS-gels, and internal peptides were determined for amino acid sequences. The amino acid sequences of the PSD31 were highly homologous with the adenine nucleotide translocator 1 (ANTI). The association of ANTl with PSD suggests presence of a mechanism in synapses for releasing adenosine nucleotides into the extracellular space.
