호르몬 첨가가 돼지 난포란의 체외성숙과 체외수정에 미치는 영향 (Effect of Hormones on Pig Oocytes Maturation and Fertilization In Vitro)

  • This experiment was carried out to investigate in vitro maturation rate of pig follicular oocytes cultured from 30 to 48hr in TCM 199 supplemented with gonadotropins(FSH, LH) and estradiol-17$\beta$ and in vitro fertilization with ejaculated sperm preincubated in BO medium containing 2mM caffein and development of IVF oocytes. The results obtained in this experiments were as follows ; 1. In addition of hormones, in vitro maturation rate of follicular oocyte increased gradually from 36hr and 74.47% at 48hr in addition of hormones, but there was no differences among in vitro maturation rates after 36hr of culture. 2. Penetration rate of pig oocytes matured in FSH+LH+E2 and FSH+E2 was 71.8%, 71.0% and significantly increased by the addition of hormones. 3. Percentage of developed oocytes was 44.4% for oocytes matured in FSH+LH+E2-added medium and 48.7% for oocytes matured in FSH+E2-added medium, respectively. 4. Two to 16 cells stage embryos were obtained only when pig oocytes matuerd in vitro in hormones-added medium and 72hr after IVF. 5. From present results, it is concluded that gonadotropins and estradiol17$\beta$ can enhance in vitro fertilization and subsequent development as well as in vitro maturation pig follicular oocytes.

  • Alkaline hydrolysis to improve the hand of PET/acetate union fabric was studied in relation to skin saponification and retarding effect of salts on the alkaline hydrolysis of cellulose acetate fiber, accelerating effect of salts on the alkaline hydrolysis of PET fiber, and changes of total hand value(T.H.V) of PET/acetate union fabric treated with alkali/salt solution. It was found that the rate of saponification of acetate was delayed by the addition of salts such as LiCl, NaCl and CH$_{3}$COONa into NaOH solution below 10 minutes. The rate of shrinkage of acetate fabric treated with 5g/l NaOH solution at 9$0^{\circ}C$ and 60 minutes showed 20 % but it was decreased 8% by. the addition of 120g/l Concentration of inorganic salts. We could know that the rate of alkaline hydrolysis of filament yarn treated with alkali solution at 9$0^{\circ}C$ and 60 minutes was delayed about 3 % using 2,000T/M of hard twist yarn, especially acetate filament treated with alkali solution at 9$0^{\circ}C$ and 10 minutes was delayed about 10%. T.H.V. of PET/acetate union fabric was increased from 2.77 to 3.04~3.18 by the addition of salts into alkali solution.

당근과 케일에서 저장온도와 양념첨가에 의한 잔류 Captan의 제거효과 (Removal of Residual Captan in Carrot and Kale by Storage Temperatures and Addition of Condiments)

  • The removal of residual captan in carrot and by storage temperatures and the addition of condiments was investigates. The quantities of residual captan after sticking and drying of captan in carrot and kale wer 0.958 and 23.12 ppm, respectively. During storage of 20days at 15,3 and -17$^{\circ}C$, the levels of the residual captan in carrot decreased to 0.008 (removal rate: 99.2%) and 15.06 ppm (42.3%), respectively. The higher removal rate of residual captan was oberved at the higher storage temperatures, When the condiments of soy sauce, green onion, garlic and vinegar added to the carrot which contaminated with the captan and then stored at 15$^{\circ}C$ for 24hrs, the residual levels of captan decreased to 0.207 (removal rate: 78.4%), 0.196 (79.5$^{\circ}C$), 0.164 (82.8$^{\circ}C$) and 0.209 ppm (78.2%), respectively, showing the garlic was the most effective. However, the residual levels in tale were 2.27 (91.3%), 12.70 (51.4%), 16.42 (37.1%) and 13.70 ppm (47.5%), respectively under the same condition, indication the soy sauce was the most effective. The removal rates of residual captan in carrot and kale were significantly higher with the addition of the condiments than those of the controls that without the condiments.

무수석고-고로슬래그계의 수화반응 (Hydration in the System of Anhydrite II-Blastfurnace Slag)

  • In order to investigate the reaction in the system of anhydrite II-blast furnace slag, the paste hydration which made up with a liquid/solid ratio of 0.45 for 1, 3, 7, 14, 28days by the addition of accelerators to 10~30wt.% slag with natural gypsum calcined for 1hour at 500/$700^{\circ}C$ was studied by combined water determination, XRD, DTA, DSC and SEM. As a result of this experiment, it was found that hydration rate was faster in the system calcined at 50$0^{\circ}C$. Therefore the anhydrite was converted to calcium sulfate dihydrate in the hydration for 1day but the slag was not almost reacted. For the gypsum calcined at $700^{\circ}C$, the hydraton rate in the system of K2SO4 addition was faster than others in the earier period, but the activated effect of the system of Al2(SO4)3 addition was regarded as the highest over 3days. As the amount of slag was increased, they dydration rate was delayed and ettringite was observed in the case of K2SO4 system. However both Al2(SO4)3 and AlK(SO4)2 systems showed calcium sulfate dhydrate only as hydrated products.

A Mechanistic Study on Addition Reactions of Alicyclic Amines to 3-Butyn-2-one

  • Second-order rate constants have been measured spectrophotometrically for the addition reaction of a series of alicyclic amines to 3-butyn-2-one to yield their respective enamines at 25.0 'C. The reactivity of the amines increases with increasing the basicity of the amines. However, the Bronsted-type plot obtained exhibits a downward curvature as the basicity of the amines increases, i.e. βnuc decreases from 0.3 for low basic amines (pKa < 9) and to 0.1 for highly basic amines (pKa > 9). Such a curvature in the Bronsted-type plot is clearly indicative of a change in the reaction mechanism or transition state structure. From the corresponding reactions run in D2O, the magnitude of kinetic isotope effect (KIE) has been calculated to be about 0.8 for highly basic amines and 1.21 for weakly basic amines. The difference in the magnitude of KIE also supports a change in the reaction mechanism or transition state structure upon changing the basicity of the amines. Furthermore, the small KIE clearly suggests that H+ transfer is not involved in the rate-determining step, i.e. the addition reaction is considered to proceed via a stepwise mechanism in which the attack of the amines to the acetylene is the rate-determining step. The curvature in the Bronsted-type plot has been attributed to a change in the degree of bond formation between the amine and the acetylene.

  • Vinylsulfilimine(VSI) 유도체($p-OCH_3$, H, p-Cl 및 p-Br)에 대한 thioglycolic acid의 친핵성 첨가반응속도를 자외선 분광법으로 측정하여 넓은 pH 범위에서 적용될 수 있는 속도식을 구하였다. pH에 따른 속도상수의 변화, general base 및 치환기 효과 등을 바탕으로 반응 메카니즘을 제안하였다. 즉 pH3.0 이하에서는 sulfilimine의 질소에 양성자가 먼저 첨가된 다음 중성 thioglycolic acid 분자가 탄소 이중결합에 첨가되고, pH 3.0-9.0 영역에서 thioglycolic acid의 중성분자와 음이온의 첨가가 경쟁적으로 일어나며, pH 9.0 이상에서는 황화 음이온이 첨가되는 전형적인 Michael type의 반응이 진행됨을 알았다.

  • ${\beta}$-nitrostyrene 유도체(p-H, p-Cl, p-CH$_3$, p-OCH$_3$, p-NO$_2$)에 대한 thiourea의 친핵성 첨가반응속도를 자외선 분광분석법으로 측정하여 pH에 따르는 반응속도 상수의 변화, general base 및 치환기 효과등으로부터 넓은 pH 범위에서 실험치와 잘 일치하는 반응속도식을 구하였고, 실험사실에 잘 맞는 반응 메카니즘을 제안하였다. 즉, pH 9.00 이상에서의 반응속도는 hydroxide ion의 농도에 비례하는 sulfide anion이 첨가되는 전형적인 Michael type의 반응이 일어나며, pH 9.00 ~ 7.00에서는 thiourea의 중성분자와 그의 anion들이 경쟁적으로 첨가되며 pH 7.00 이하에서는 thiourea의 중성분자만이 첨가됨을 알았다.

  • ${\beta},\;{\beta}$-diethoxycarbonylstryene 유도체(H, p-OCH$_3$, 3,4,5-(OCH$_3)_3$), 3,4,5-methylenedioxy)에 대한 3-mercaptopropionic acid의 친핵성 첨가반응 속도상수를 자외선 분광법으로 측정하여 넓은 pH 범위에서 적용될수 있는 속도식을 구하였다. pH에 따르는 속도상수의 변화, 일반염기촉매작용등을 바탕으로 이 첨가반응 메카니즘을 제안하였다. 즉 pH 6.0 이하에서는 중성인 3-mercaptopropionic acid 분자의 첨가로 이 반응이 시작되며 pH 6.0∼8.0영역에서는 이 중성분자와 황화음이온의 첨가가 경쟁적으로 일어나며, pH 8.0이상에서는 황화음이온에 의해서만 첨가반응이 진행된다.

  • 여러 가지 용매에서 phenylvinylketone에 대한 아민(piperidine 및 diethyl-amine)의 친핵성 첨가반응 속도 상수를 $25^{\circ}C$에서 분광광도법으로 측정한 결과 그의 첨가반응 속도상수는 용매의 극성에 비례함을 알았고 이것을 Kirkwood식으로 잘 설명할 수 있었다. 즉, 이 반응은 고리형 구조의 zwitter ion 중간체를 거쳐 일어남을 알 수 있었다. 또 phenylvinylketone에 대한 아민의 첨가반응에서 3차 아민은 1차 및 2차 아민에서 보다 반응이 훨씬 느리게 진행되었으며 이것은 1차 및 2차 아민에서는 고리형 구조의 zwitter ion 중간체를 형성할 수 있고 또한 음전하의 비편재화 및 엔올형의 구조까지 가능하지만 3차 아민에서는 위의 구조들이 불가능하기 때문에 1차 및 2차 아민에서 보다 느리게 진행된다고 생각된다.

  • This research presets the method of determining basic capacity of signalized intersection using the concept of ideal saturation flow rate. Vehicle discharge rates during green time were collected and studied as part of the preparation of Korean Highway Capacity Manual. From the result of this study the ideal saturation flow rate and saturation queue position were determined. In addition, based on the ideal saturation flow rate two geometric adjustment factors(lane width and grade adjustment factors) were studied. The results were presented in this paper.
