  • Objectives : An aim of study is to investigate effects of curculiginis rhizoma in vitro (factor Xa (FXa) inhibitor assay, prothrombinase assay, prothrombin time (PT) assay, activated partial thromboplastin time (aPTT) assay) and in vivo experiment (blood clotting time, thromboxane B2 content assay in serum and weight of thrombus by AV-shunt rat model). Methods : We gained a human serum and used serum in vitro study such as factor X activity (FXa) inhibition, prothrombinase inhibition, prothrombin time (PT) and activated partial thromboplastin time. Fifteen SD rats were divided into three groups (intact control group and two experimental group treated with extract of Curculiginis Rhizoma(ECR)). Rats were orally administrated DW (intact control group), 600 mg/kg concertration of ECR and 200 mg/kg concertration of ECR. After one hour, we anesthetized rats and made arteriovenous (AV) shunt rat models to study weights of thrombus, took a hole blood to study content of thromboxane B2 and blood clotting time. Results : In vitro, ECR increased a inhibitory activity of FXa, prothrombinase and aPTT compared than intact control group. Especially ECR made significant increase of FXa and prothrombinase inhibitory activity (p<0.05, p<0.01). And PT were increased in ECR control group compared with intact control group. In vivo, a blood clotting time of experiment group treated with ECR 600 mg/kg were significantly increased compared with that of intact control group (p<0.05) and content of thromboxane B2 was significantly decreased in group treated with ECR 600 mg/kg in seum. The weight of thrombus were significantly reduced in group treated with ECR 600 mg/kg compared with intact control group (p<0.05). But in vivo experiment study, those of group treated with ECR 200 mg/kg were reduced compared with those of intact control group without statistical significance. Conclusions : ECR has a antithromboic activity in internal course with inhibitory activity of FXa and prothrombinase in vitro, it required to research more study for effective compounds.

  • An eight-week-old male ICR mouse, which was induced with acute alcohol and sub-acute alcohol poisoning condition, was administered with bohee tea(Camelia sinensis L) extract. After oral administration of bohee tea and inducement of acute alcohol poisoning condition, the mouses blood alcohol concentration became as low as that of the normal control group. Its decrease rate was 87.26%, in comparison with that of the positive control group. Moreover, its blood GOT activity decreased with a rate of 93.1 % until it reached the normal level, as opposed to that of the positive control group. In addition, the GOT activity, despite rising after the alcohol intake, decreased(p<0.05) significantly after administration of each sample and reached the normal level. The bohee tea group experienced a significant decrease in the GOT activity, compared with the A1codex group and the Drink group. The GPT activity of the Alcodex group decreased by 11 % compared with that of the positive control group. The CTP activity of the bohee tea group decreased by 8.2%, while that of the Drink group decreased by 6.5%(P<0.05). However, there were no significant differences between the results in the control group and those of the test group. The bohee tea group's hepatic ADH activity increased by 22.7% compared with that of the positive control group. On the other hand, the hepatic ADH activity of the Drink group increased by 33.6% while that of the A1codex group increased by 20.4%. On the contrary, the bohee tea extract, the hepatic ALDH did not manifest any significant difference as compared with the normal control group. However, its decrease rate was about 16.67% as compared with that of the positive control group. The Drink group, meanwhile, obtained a decrease rate of about 21.59%.

  • To investigate the effects of internal radiation of various intensityon the test crop(flax), seeds of four levels of activity($15.5{\times}10-4 $\mu$c/grain, 9.0{\times}10-2$\mu$c/grain, 2.2{\times}100$\mu$c/grain and 3.8{\times}10-1$\mu$c/grain)$ which had been obtained by immersing them into various concentrations of P-32 original solution (total activity: 90 mc, To: 3/21) for 24 hours at room temperature, were germinated, transplanted later into pots, and the rate of germination and the successive growth were observed, and the inorganic conents of the plant top were analyzed. The results are as follows: 1. At the early stage of germination, the plants manifested themselves both inhibitory and promotive effects at higher and lower activity levels respectively, compared those of the control. These difference of growth on account of different levels of activity appeared, however, to be gradually narrowed in the course of time after germination, except at the highest activity. 2. Two weeks after transplanting, the plants of the lowest activity showed more vigorous growth than those of control. The plants belonging to the other activity levels on the other hand, tended to be less growing, the higher the activity. However, this growth gap between treatments seemed to be progressively closed one month after transplanting. 3. Most of the leaves and stems of the plants belonging to the highest activity level ($3.8{\times}101$\mu$c/grain$) were withered during the early stage of growth, and this damage did not recover. 4. Practically no difference of growth was observed among treatments(excluding that of highest activity) one and half months after transplanting. 5. The fluorescence tended to be mroe delayed than the control, as the activity decreased. 6. There was a tendency that the number of pods harvested were larger in the plants treated with P-32 than that of the control. 7. The proportion of fiber in the plants at harvest appeared to be larger at lower activity and smaller at higher P-32 concentration than that of control. 8. As for the inorganic contents of the plant top harvested the floowing tendency was observed: (1) Nitrogen content was highest at the highest activity level at which the poorest growth ensued. (2) There was no clear difference of phosphorus content among treatments. (3) The contents of potassium and magnesium were higher than control at the medium levels of activity. (4) Calcium content of all treated blocks was found to be more than that of control.

Materials and Methods : Experimental studies were perfonned for the cytotoxic effect on BALB/c mouse lung fibroblast cells, the proliferating effect of splenic lymphocyte, the expression of CD3e/CD4, CD3e/CD8, and B220 in peripheral blood mononuclear cells (PBMCs), the cytotoxic effect on A549, SK-OV-3, SK-MEL-2, MCF-7 cells, the inhibitory effect on the activity of DNA topoisomerase I, the T/C% in ICR mice bearing S-180, the inhibitory effect of Cell adhesive of A549 Cells and SK-OY-3 Cells to complex extracellular matrix, the inhibitory effect on lung colonies, the change of lung tissue, the antiangiogenic activity, and the effect on MMP-2 and MMP-9 gene expression in the RT1080 cell line. Results and Conclusion : The results were obtained as follows : 1. In the cytotoxic effect on BALB/C mouse lung fibroblast Cell, GHT didn't show the significant cytotoxic effect on BALB/C mouse lung fibroblast cell compared to the control group. 2. In thymidine uptake assay, GHT showed the significant proliferating effect of splenic lymphocyte in proportion to the concentration. 3. In the expression of CD3e/CD4, CD3e/CD8, and B220 in peripheral blood mononuclea cells (PBMCs) of mice, GRT had no significant change to the normal group in CD4. However, GRT showed an increase to the normal group in CD8 and GHT in the only $1\mu\textrm{g}/ml$ category showed an increase to the normal group in B220. 4. In the cytotoxic effect of GRT on A549, SK-OY-3, SK-MEL-2 and MCF-7 cells, there was no significant cytotoxic effect compared to the control group. 5. In the inhibitory effect on the activity of DNA topoisomerase I, GHT in the $10\mu\textrm{g}/ml$ category showed the inhibitory effect on the activity of DNA topoisomerase I in proportion to the concentration. 6. In the T/C% in ICRmice bearing S-180, GHTtreated group showed 123.7% of T/C% compared to the control group. 7. In the inhibitory effect of cell adhesive of A549 Cells and SK-OV-3 Cells to complex extracellular matrix, GRT in the only $100\mu\textrm{g}/ml$ category showed the significant inhibitory effect compared to the control group. 8. In the inhibitory effect on lung colonies, GHT showed the significant inhibitory effect on lung colonies compared to the control group. 9. In the change of lung tissue, GHT showed a significant decrease of lung cancer growth, interalveolar fibrosis and hyaline material compared to the control group. In the development of lymphocyte around lung cancer cells and lung parenchymal, GHT showed the significant inducement efficacy compared to the control group. 10. In CAM assay, the antiangiogenic activity of GHT showed 30%. 11. In the effect on MMP-2 and MMP-9 gene expression in the RT1080 cell line, GHT had no significant inhibitory effect on MMP-2 and MMP-9 gene expression compared to the control group. According to the above results, it could be suggested that GHT has an antitumor activity and antimetastatic activity.

  • In this study, the α-amylase inhibitory activity, α-glucosidase inhibitory activity, pancreatic lipase inhibitory activity, and Xanthine Oxidase inhibitory activity of the fruiting body extracts of 5 varieties of Agaricus bisporus (AB) were confirmed. First, the α-amylase inhibitory activity of AB12, AB13, AB18, AB34, and AB40 methanol extracts was lower than that of acarbose, a positive control, in all concentration ranges. The α-glucosidase inhibitory activity of the AB40, AB13, and AB12 methanol extracts at the extract concentration of 1.0 mg/mL was 80.5%, 81.3%, and 78.5%, respectively, similar to that of acarbose, a positive control. The pancreatic lipase inhibitory activity of the methanol extract of Agaricus bisporus fruiting body was significantly lower than that of the positive control orlistat in the concentration range of 50~1.000 (mg/mL). The Xanthine Oxidase inhibitory activity was 0.5~8.0 mg/mL of each extract, which was significantly lower than that of the positive control allopurinol in the same concentration range. However, the Xanthine Oxidase inhibitory activity of AB13 and AB40 at 8.0 mg/mL was about 70%, which was higher than that of other mushrooms. In conclusion, five kinds of Agaricus bisporus fruiting bodies seem to have inhibitory effects on enzymes such as α-amylase, α-glucosidase, pancreatic lipase, and Xanthine Oxidase that degrade starch and protein. In particular, it has an inhibitory effect and a reduction effect on xanthine oxidase that causes gout, so it is expected that it can be developed and used as a food or health supplement with health functional properties through future research.

  • The antioxidant activities of the bread made from waxy black rice flour(WBRF) and wheat flour were investigated. The waxy black rice bread was manufactured with 10, 20, 30, or 40% waxy black rice flour(w/w). As the additive amount of WBRF were increased, DPPH(1,1-diphenyl-2-picryl-hydrazyl) radical scavenging activity and TEAC(trolox equivalent antioxidant capacity) were also increased. WBRF group showed the higher activity than the control group in DPPH radical scavenging activity and TEAC. The group with 10% WBRF added was not significantly different from the control group in metal chelating activity, but over 20% of addition, it had the higher metal chelating activity than that of the control group.

  • In order to study the effect of acupuncture stimulation on bone mineral density (BMD), using the ovariectomized (OVX) rat model, we assessed the degree of osteopenia by dual-energy X-ray absorptiometry, measured the level of locomotor activity using a metabolism measuring system, and performed histological studies of bone tissue. Twenty-four female Wistar rats (8 weeks old, 160 - 180 g)were divided into three groups. Rats in the OVX-A group underwent ovariectomy followed by acupuncture stimulation. The OVX rats in the Vehicle control group were not treated with acupuncture as a control. The rats in the control group received neither ovariectomy nor acupuncture. Acupuncture stimulation for 12 weeks in the OVX-A group inhibited the reduction in BMD of the femoral bones caused by ovariectomy. Moreover, in the two OVX groups, there was no clear difference in the level of locomotor activity between the active and resting phases prior to acupuncture stimulation in each rat, and the pattern of locomotor activity was irregular. After acupuncture stimulation of the OVX-A rats, the pattern of locomotor activity became diphasic with clear active and resting phases, as was observed in the Control group. On histological studies, the continuity of trabecular bone was maintained more favorably and bone mass was higher in the OVX-A group than in the vehicle control group. These results suggest that the increased locomotor activity that had been induced by acupuncture stimulation increased the BMD.

  • Although since 1963 QC activity for Korean industrial standard registered mark, many problems have been disclosed that the basic Quality Control principles. In practice, many manufacturing Companies are so typical and so impetuous of the tangible effect that it is worried to be inclined to out form rather than contents and quality. To prevent this point it is recommended Company Wide Quality Control activity be promoted contineously and on the basis of the original principle of C.W.Q.C. activity and small group activity of QC and Factory Seamaul Undong in Korea to obtain practical and real results. So the effective approach model is designed for Company wide Quality Control activity in Korea industry.

  • This study compared the heart rate variability tests of healthy college students and chronic fatigue patients for 2 years. Study subjects were idiopathic chronic fatigue patients who carried out the HRV tests and students conducted HRV test when health screening test. Study subjects were 250 people, 104 people and men (41.6%), women were 146 patients (58.4%). In patient group, the autonomic nervous system activity was decreased, the activity of the sympathetic nerve and the parasympathetic nerve were significantly reduced compared to the control group(Patient:84.17±16.27, Control:98.33±17.28). Regardless of gender, patient group's autonomic nervous system activity was decreased, the activity of the sympathetic nerve and the parasympathetic nerve were significantly reduced, compared to the control group(Patient Female:84.17±16.27, Patient Male:84.07±14.96/Control Female:98.33±17.28, Control Male:96.45±16.92). Even though same ages, patient group's autonomic nervous activity was reduced compared to control group(Patient:89.36±12.43, Control:97.39±16.91). Fatigue patients' activity of the sympathetic nerve and the parasympathetic nerve were significantly reduced, regardless of the fact that activity of the sympathetic nerve is increased and the parasympathetic nerve is decreased in stress state like an impassioned injury(七情傷). Therefore HRV test is useful to diagnose Fatigue from this study.

  • Purpose: This study compared the total antioxidant status (TAS) and superoxide dismutase (SOD) activity in the saliva of periodontally compromised patients before and after scaling and root planing (SRP) to assess their diagnostic utility. Methods: Severe chronic periodontitis patient (test group) and subjects with no attachment loss, sites showing a 3 mm or more probing depth and a sulcus bleeding index < 10% (control group) were enrolled in this study. Saliva sampling and clinical examination were performed at one week, one month and 3 months after SRP. The TAS and SOD activity in each patient's saliva was measured for the comparative analysis between the groups. Results: In the test group, the TAS decreased directly after SRP. With time, it increased slightly and was relatively unchanged compared to the baseline. In the control group, the TAS also decreased immediately after SRP but increased gradually with time until 3 months. The SOD activity in the test and control subjects decreased immediately after SRP until 1 month. At 3 months, the SOD activity had increased. Both groups had a similar profile of SOD activity. However, the SOD activity of the control group was significantly higher than that of the test group at each point in time (P < 0.05). Conclusions: There was a significant difference in the total salivary antioxidant level between the periodontitis and healthy or gingivitis (control) group during the experiment period. The total antioxidant level in the saliva was higher in the patients with severe chronic periodontitis than the healthy or gingivitis control before SRP. The SOD activity of the periodontitis patients was lower than the control at each time point. These findings conclusively reveal the possible use of saliva as a diagnostic tool for periodontal health.
