Investigation on the Current Labelling Trend of Tar Color Additives in Infant's Drugs Marketed in Korea (국내 시판중인 소아용 의약품에 함유된 타르색소의 표기실태 조사 연구)

  • Objective: This paper was aimed to investigate the current labelling trend of tar color additives in infant's drugs used in Korea and other countries. Method: The current labelling system of the tar color additives in infant's drugs was investigated using inserted papers for drug information of infant's drugs as well as three drug information internet sites popular in Korea including internet sites of foreign countries of USA, UK, Japan, and China for infant's drug information. Results: Among 325 drugs registered as syrup form for infant's drugs (sources, http://www.health.kr), 4 drugs indicated tar color related information on the internet and only 2 drugs indicated tar color additives' name. And from the investigated results of 76 inserted papers for infant's drug information, among 31 drugs containing tar dyes, in only 19 drugs the tar color additives' names were notated. Among 6 drugs which do not contain tar dyes, only 1 drug was notated as 'Dye-Free'. Conclusion: For advanced labelling system in Korea, we proposed the below guidelines. [1] The name of tar color additives used in infant's drugs should be notated even in drug information on internet sites. [2] All drug ingredients used in infant's drugs need to be separately notated as active and inactive species on both internet sites and inserted papers. [3] 'Dye-Free' notation should be mandatory for infant's drugs which do not contain tar color additives in drug information on the internet sites as well as inserted papers.

Comparative Analysis of Intracellular Trans-Splicing Ribozyme Activity Against Hepatitis C Virus Internal Ribosome Entry Site

  • Internal ribosome entry site (IRES) of the hepatitis C virus (HCV) is known to be essential for HCV replication and most conserved among HCV variants. Hence, IRES RNA is a good therapeutic target for RNA-based inhibitors, such as ribozymes. We previously proposed a new anti-HCV modulation strategy based on trans-splicing ribozymes, which can selectively replace HCV transcripts with a new RNA that exerts anti-HCV activity. To explore this procedure, sites which are accessible to ribozymes in HCV IRES were previously determined by employing an RNA mapping method in vitro. In this study, we evaluate the intracellular accessibility of the ribozymes by comparing the trans-splicing activ­ities in cells of several ribozymes targeting different sites of the HCV IRES RNA. We assessed the intra­cellular activities of the ribozymes by monitoring their target-specific induction degree of both reporter gene activity and cytotoxin expression. The ribozyme capable of targeting the most accessible site iden­tified by the mapping studies then harbored the most active trans-splicing activity in cells. These results suggest that the target sites predicted to be accessible are truly the most accessible in the cells, and thus, could be applied to the development of various RNA-based anti-HCV therapies.

Oviposition site preference in Black Soldier Fly, Hermetia illucens (Diptera: Stratiomyidae), in Artificial Rearing System

  • In natural conditions, the black soldier fly, Hermetia illucens (L.) (Diptera: Stratiomyidae), colonizes in warm temperate regions, and is active in Korea from May through October. Information on black soldier fly rearing, which is affected by seasonal factors in Korea, is limited. This species colonizes and oviposits in a wide variety of decomposing vegetable and animal matter. For mass rearing of black soldier flies, the egg deposition methods are dependent on sunlight and oviposition sites. In this study, we investigated the substrates and oviposition sites preferred by black soldier fly in artificial rearing system. Our results showed that as black soldier flies prefer mating under sunlight, they prefer ovipositing at sites where adequate sunlight and food substrate are available. Further detailed research is required to develop methods for artificially rearing black soldier fly throughout the year in Korea.

Kinetic Mechanism of Nucleotide Binding to Escherichia coli Transcription Termination Factor Rho: Stopped-flow Kinetic Studies Using ATP and Fluorescent ATP Analogues

  • Escherichia coli transcription termination factor Rho catalyzes the unwinding of RNA/DNA duplex in reactions that are coupled to ATP binding and hydrolysis. Fluorescence stopped-flow methods using ATP and the fluorescent 2'(3')-O-( N-methylanthraniloyl) derivatives (mant-derivatives) of ATP and ADP were used to probe the kinetics of nucleotide binding to and dissociation from the Rho-RNA complex. Presteady state nucleotide binding kinetics provides evidence for the presence of negative cooperativity in nucleotide binding among the multiple nucleotide binding sites on Rho hexamer. The binding of the first nucleotide to the Rho-RNA complex occurs at a bimolecular rate of 3.6${\times}$10$\^$6/ M$\^$-1/ sec$\^$-1/ whereas the second nucleotide binds at a slower rate of 4.7${\times}$10$\^$5/ M$\^$-1/ sec$\^$-1/ at 18$^{\circ}C$, RNA complexed with Rho affects the kinetics of nucleotide interaction with the active sites through conformational changes to the Rho hexamer, allowing the incoming nucleotide to be more accessible to the sites. Adenine nucleotide binding and dissociation is more favorable when RNA is bound to Rho, whereas ATP binding and dissociation step in the absence of RNA occurs significantly slower, at a rate ∼70- and ∼40-fold slower than those observed with the Rho-RNA complex, respectively.

Blocking of Zeolite Pore by Loading Ni-Pt Nanoparticles for Maximization of Isomerization Selectivity

  • Zeolite HY is wet impregnated with Ni (0.1, 0.3, 0.4, 0.5 wt%), Pt (0.1 wt%) and reduced in presence of hydrogen to form nanosized particles of Ni and Pt. All the catalysts were characterized by XRD, TEM, ESCA, NH3-TPD, Pyridine adsorbed FT-IR and BET. Characterization results confirm that the Ni and Pt fractions effectively rehabilitated the physio-chemical properties of the zeolite HY catalysts. Further, all the reduced catalyst were screened with hydroisomerization of m-xylene at LHSV = 2.0 h-1 in the temperature range 250-400 ℃ in steps of 50 ℃ in hydrogen atmosphere (20 ml/g). The addition of Ni to Pt catalyst increases hydroisomerization conversion, as well as maximizes p-xylene selectivity by restricting the pore size. The increasing trend in activity continues up to 0.3 wt% of Ni and 0.1 wt% Pt addition over zeolite HY. The increasing addition of Ni increases the total number of active metallic sites to exposed, which increases the metallic sites/acid sites ratio towards the optimum value for these reactions by better balance of synergic effect for stable activity. The rate of deactivation is pronounced on monometallic catalysts. The results confirm the threshold Ni addition is highly suitable for hydroisomerization reaction for product selectivity over Ni-Pt bimetallic/support catalysts.

  • Supramolecules containing double hydrogen bonding sites at their both chain ends were self-polymerized to become solid state polymer and were utilized to improve the efficiency of solid state DSSCs. Hydrogen bonding sites were attached at the chain ends of PEG of Mw=2000, such as pyrimethamine and glutaric acid. The solar cell with the solid state supramolecular polymer electrolyte resulted in the overall energy conversion efficiency of 4.63 % with a short circuit current density $(J_{sc})$ of 10.41 $mAcm^{-2}$, an open circuit voltage $V_{oc}$, of 0.71 V and a fill factor (FF) of 0.62 at one sun condition ([oligomer]:[1-methyl-3-propyl imidazolium iodide (MPII)]:$[I_2]$ = 20 : 1 : 0.19, active area = 0.16 $cm^2$, $TiO_2$ layer thickness = 10 ${\mu}m$). The ionic conductivity of the sol id state electrolyte was $5.11{\times}10^{-4}$ (S/cm). The cell performance was characterized by electrochemical impedance spectroscopy and ionic conductivity.

Identification of the Most Accessible Sites to Ribozymes on the Hepatitis C Virus Internal Ribosome Entry Site

  • The hepatitis C virus (HCV) is a major causative agent of chronic hepatitis and hepatocellular carcinoma. The development of alternative antiviral therapies is warranted because current treatments for the HCV infection affect only a limited number of patients and lead to significant toxicities. The HCV genome is exclusively present in the RNA form; therefore, ribozyme strategies to target certain HCV sequences have been proposed as anti-HCV treatments. In this study, we determined which regions of the internal ribosome entry site (IRES) of HCV are accessible to ribozymes by employing an RNA mapping strategy that is based on a trans-splicing ribozyme library. We then discovered that the loop regions of the domain IIIb of HCV IRES appeared to be particularly accessible. Moreover, to verify if the target sites that were predicted to be accessible are truly the most accessible, we assessed the ribozyme activities by comparing not only the trans-splicing activities in vitro but also the trans-cleavage activities in cells of several ribozymes that targeted different sites. The ribozyme that could target the most accessible site identified by mapping studies was then the most active with high fidelity in cells as well as in vitro. These results demonstrate that the RNA mapping strategy represents an effective method to determine the accessible regions of target RNAs and have important implications for the development of various antiviral therapies which are based on RNA such as ribozyme, antisense, or siRNA.

  • A total of 460 palaeomagnetic samples was collected from the Tertiary Chongja-Ulsan basins and surrounding area in the southeastern part of Korean peninsula. All samples were stepwise demagnetized by either alternating field or thermal method. It was found that most sample-sites have ChRM declination which has been rotated clockwise from the north-south reference direction of Tertiary East Asia, although other two extrusive sample-sites within the Chongja sedimentary basin show counterclockwise rotation of ChRM declination. Fold tests for the site-mean ChRMs of the latter two sites reveal insignificant result and negative result with 95% confidence level, respectively. The amount of the clockwise deflection of declination varies from about $20^{\circ}$ upto about $80^{\circ}$ according to the block to which each sample-site belongs. The amount of the counterclockwise deflection is about $20^{\circ}$. It is concluded that the clockwise ChRM rotation has been caused by dextral simple shearing accompanied by NNW-SSE spreading of the East Sea which has been active until about 16Ma, and that the counterclockwise rotation is a result of sinistral simple shearing associated with WNW-ESE contraction in the Korean Strait-SW Japan region at about 15 Ma.

Catalytic Oxidation of Trichloroethylene over Pd-Loaded Sulfated Zirconia

  • The oxidative decomposition of trichloroethylene (TCE) was investigated using palladium catalysts supported on pure and sulfated zirconia. The reactions were performed under dry and wet conditions in the temperature between 200 and $550^{\circ}C$ keeping GHSV of 14,000 $h^{-1}.$ The products such as $C_2Cl_4,\;C_2HCl_5,\;CO\;and\;CO_2$ were observed in the reaction. The addition of water in the feed affected the distribution of reaction product with dramatically improved catalytic activity. The spectroscopic investigations gave an evidence that the strong acid sites play an important role on controlling the catalytic activity. Among the catalysts investigated, the Pd-loaded sulfated zirconia catalyst with 1 wt% Pd was found to exhibit the highest catalytic activity in the presence of water vapor having the stability for 30 h of the reaction at $500^{\circ}C$. The successful performance of the catalyst might be attributed to promotional effect of Pd active sites and strong acid sites induced from surface sulfate species on zirconia.

  • Web 2.0 which is characterized by openness, sharing with others and users' active participation has a strong impact on Individual and business environments. New technology such as open-API, RSS, collective intelligence gives various benefits to the website user. Recently, the Web 2.0 paradigm has also affected the public service. Many public service organizations are also adopting Web 2.0 applications to improve their ability for serving users more effectively. However, there is a contradiction between value of public service and that of web 2.0. Web 2.0 which pursues openness and freedom of expression can impair the accuracy and reliability of public information in the public sites. We conducted a case study on EBS which reflects the Web 2.0 strategy for websites renewal. To cope with environmental changes, EBS began reorganization of the web site and applied Web 2.0 strategy for leveraging an easy to use and user satisfaction. This research shows that how to Implement the Web 2.0 strategy and what is the limitation of it. The findings of this study suggest practical implications for Web 2.0 strategy in public web sites, especially for educational fields.
