옥수수 불검화 추출물(Zea Mays L.)과 후박(Magnoliae cortex) 추출물 혼합물의 치주질환원인균에 대한 항균작용 및 치은섬유아세포 활성도에 미치는 영향 (Antimicorbial effect of Zea Mays L. and Magnoliae cortex extract mixtures on periodontal pathogen and effect on human gingival fibroblast cellular activity)

  • Zea Mays L. has been known to be effective for improving tissue health and Magnoliae cortex to have effective antibacterial and antimicrobial activity against pathogenic microbes. The purpose of this study was to examine the antimicrobial effects of Zea Mays L. and Magnoliae cortex extract mixtures on periodontal pathogens(Prevotella intermedia, Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, Streptococcus mutans )and to examine the effects on human gingival fibroblast cellular activity. Zea Mays L. and Magnoliae cortex extracts and their mixtures were prepared with various mixing ratios (0.5:1, 1:1, 1.5:1, 2:1). These extracts were loaded to periodontal pathogen cultured petri dish for antimicrobial test and also loaded to cultured human gingival fibroblast for cellular activity test. Each test was repeated 3 times and data were analyzed by one-way ANOVA with 95% confidence level. Mixture of these two extracts showed greater amount of inhibition area on periodontal pathogen and more improved gingival fibroblast activity as Zea Mays L. ratio reduced. So, mixture ratio 0.5:1 (Zea Mays L. : Magnoliae cortex) group showed statistical significance in antimicrobial activity and cellular activity among various mixtures(p < 0.05). In conclusion, 0.5:1 (Zea Mays L. : Magnoliae cortex) mixture possessed best gingival fibroblast cellular activity and antimicrobial activity toward periodontal pathogens.

수종의 상용 세치제들의 향균 및 향염효과 비교연구 (Comparative study on the antimicrobial and antiinflammatory activity of commercially available toothpastes)

  • It is known that some natural extracts from plants have a various range of antimicrobial and antiinflammatory activity. There are lots of clinical trials to develop toothpastes containing natural extracts for prevention of dental caries and gingival inflammation. The purpose of this study was to evaluate antimicrobial and antiinflammatory activity of magnolol containing toothpastes and other commercial toothpastes. Eleven kinds oftoothpastes were used. They include magnolol, sanguinarine, Myrrha, Mori radicis cortex,Cimicifugae rhizoma, sodium fluoride, aminocaprolactic acid etc. Six strains of bacteria were used for this test, ego Porphylomonas gingivalis, Prevotellain-termedia, Actinobacillus actinomy cetemcomitans, Streptococcus mutans, Stretococcus sanguis, and Actinomyces species. Antimicrobial activity was determined by an agar dillution method and a broth microdillution method. Antiinflammatory activity was assessed by the inhibition of $PGE_2$ production from gingival fibroblast with the addition of rHIL-1 and centrifuged solution of toothpastes. Control group was only rHIL-1 additive sample. $PGE_2$ enzyme immunoassay systemfAmersham, In. Buckinghamshire, U.K). $PGE_2$ level was measured by ELISA reader with 450 nm, The results from the study revealed that toothpastes containing natural extracts generally had high antimicrobial and antiinflammatory activity. Especially magnolol containing toothpaste showed higher antimicrobial activity than other toothpastes, and sanguinarine containing toothpaste showed particularly high antimicrobial activity in A. actinomicetemcomitans and A. viscosus. In some degree all toothpastes inhibited $PGE_2$ production, but magnolol containing toothpaste was potent inhibitor of $PGE_2$. Sodium chloride containing toothpaste had also effective result. The results suggested that toothpastes containing natural extracts were promising in plaque control and prevention of dental caries and gingivitis.

Identification of immunological parameters associated with the alveolar bone level in periodontal patients

  • Purpose: The present study was performed to clarify the relationship between periodontal disease severity and selected immunological parameters consisting of serum IgG titer against periodontopathogenic bacteria, the expression of the helper T-cell cytokine by gingival mononuclear cells, and patients' immunoreactivity to cross-reactive heat shock protein (HSP) epitope peptide from P. gingivalis HSP60. Methods: Twenty-five patients with moderate periodontitis had their gingival connective tissue harvested of gingival mononuclear cells during an open flap debridement procedure and peripheral blood was drawn by venipuncture to collect serum. The mean level of interproximal alveolar bone was calculated to be used as an index for periodontal disease severity for a given patient. Each of selected immunologic parameters was subject to statistical management to seek their correlations with the severity of periodontal disease. Results: A significant correlation could not be identified between serum IgG titers against specific bacteria (Porphyromonas gingivalis, Prevotella intermedia, Fusobacterium nucleatum, Actinobacillus actinomycetemcomitans, and Streptococcus mutans) and the severity of periodontal disease. Expression of interleukin (IL)-10 by gingival mononuclear cells was statistically significant in the group of patients who had higher levels of alveolar bone height. However, a similar correlation could not be demonstrated in cases for IL-4 or interferon-$\gamma$. Patients' serum reactivity to cross-reactive epitope peptide showed a significant correlation with the amount of alveolar bone. Conclusions: It was concluded that expression of IL-10 by gingival mononuclear cells and patients' sero-reactivity to the cross-reactive HSP peptide of P. gingivalis HSP60 were significantly correlated with alveolar bone height.

관중의 항균작용 및 세포독성에 관한 연구 (Antibacterial Effects and Cytotoxicity of Crassirhizomae Rhizoma)

  • The purpose of this study was to evaluate the antimicrobial activity of Crassirhzimae rhizoma and its possible use as an oral antiseptics for prevention of periodontitis. Its antibacterial activity against periodontopathic microorganisms including Actinobacillus actiomycetem - comitans, Capnocytophaga ochracea, Streptococcus mutans, Porphyromonas gingivalis, Prevotella intermedia, Actinomyces viscosus, Fusobacterium nucleatumwas evaluated via modified stab culture method. The cytotoxicity against gingival fibroblasts and rat osteoblasts was investigated via [$^3H$]thymidine incorporation and cellular activity was investigated via MTT assay. Chlorhexidine was used as control group. Crassirhizomae rhizoma was prepared at concentrations of 0.2, 0.15, 0.1, 0.05%. Chlorhexidine was also prepared at the same concentration. Crassirhizomae rhizoma showed lower antimicrobial antivity against these microorganism than chlorhexidine, but this difference was not significant. And, Crassirhzomae rhizoma showed more cellular activity and less cytotoxicity than chlorhexidine on human gingival fibrablast and rat osteoblast. This study suggests that Crassirhzomae rhizoma might be a candidate for a safe oral antiseptic for the prevention and treatment of periodontal disease.

Cefixime의 치주병원성 세균 6종에 대한 항균 효과 (Antimicrobial effect of cefixime on 6 species of periodotopathogens)

  • The aim of this study was to determine the minimal inhibitory concentration(MIC) of cefixime, which is a 3rd generation of cefalosporin, against 6 species of putative periodontopathogens; Fusobacterium nucleatum, Actinobacillus actinomycetemcomitans, Prevotella intermedia, Prevotella nigrescens, Tannerella forsythia and Porphyromonas gingivalis. The efficacy of cefixime was examined by comparing it with that of several antibiotics(amoxicillin, $Augmentin^{(R)}$ ciprofloxacin, metronidazole, and tetracycline), which were used as the control. The MIC was measured using a microdilution method. The MIC of cefixime against the putative periodotopathogens, as a single use regimen, was relatively lower than that of the other antibiotics. The MIC of cefixime/metronidazole against P. intermedia ChDC KB14, P. nigrescens ChDC KB50, F. nucleatum ChDC PV-F37, F. nucleatum ChDC F130, and F. nucleatum ChDC F175, as a simultaneous regimen, was lower than that of the other antibiotics. The concentration of cefixime in the crevicular fluid of volunteers who received 250mg every 12 hours for 3 days was $9{\mu}g/ml$ after 9 hours. In conclusion, cefixime showed good antimicrobial activity in a single treatment or as a combined therapy with amoxicillin, $Augmentin^{(R)}$ or metronidazole against 6 periodontopathogens.

Seroprevalence of selected porcine respiratory pathogens in the pig herds in Chungcheong and Gyeongsang provinces in Korea

  • We studied the seroprevalence of four respiratory pathogens in Korean swine farms located in Chungnam, Chungbuk, Gyeongnam and Gyeongbuk provinces during the period of spring of 2007 to winter of 2008. Serological tests were performed using commercial ELISA kits. A total of 530 serum samples were tested for the antibodies against porcine reproductive and respiratory syndrome virus (PRRSV), porcine circovirus type 2 (PCV2), Mycoplasma hyopneumoniae (M. hyo) and Actinobacillus pleuropneumoniae (APP). Seroprevalence for four respiratory pathogens were estimated by ELISA-positive rates of the submitted samples. The overall seropositive rates of PRRSV, APP, M. hyo and PCV2 were 32.6%, 10.6%, 38.4% and 88.5%, respectively. By production stage, the seropositive rate for PRRSV was highest in nursery pig populations (46.2%). In contrast, the highest seropositive rates of APP and M. hyo were observed in sow and growing pigs. However, the seroprevalence of PCV2 was ranged from 85.7% to 89.6%, showing no significant difference among the production stages. In the seroprevalence by season, PRRSV, APP and M. hyo infections revealed typical seasonal patterns that the peaks of the seropositive rates were observed between early winter and late spring. In case of PCV2, no particular seasonal patterns were noticed. The pig herds in Gyeongbuk province where PMWS was endemic during the period of survey showed the highest seropositive rates for PRRSV (44.6%), M. hyo (47.5%), and PCV2 (92.7%). Seropositive rates for APP of four provinces were approximately 10%. These results might be valuable for control and prevention of the respiratory diseases and helpful to define strategies related to vaccine applications.

전북지역 도축돈 호흡기질병 조사 (Seasonal survey on the respiratory diseases of slaughtered pigs in Jeonbuk, Korea)

  • The present study was conducted to investigate the lesion of red internal organs in slaughtered pigs and provided assistant data for pig farms. During March to December 2015, a total of 1,160 lung samples out of 58 herds were collected randomly from pigs slaughtered in Jeonbuk province. In addition, 290 hilar lymph nodes from pig with pneumonic lung lesion (5 samples per herd) were screened for selected viral and bacterial pathogens. Gross lesions of lungs such as swine enzootic pneumonia (SEP), pleuritis, pleuropneumonia, pericarditis and liver white spots were examined. The overall prevalence of SEP was 64.3% (746/1,160). In the analysis of seasonal prevalence, there was an increase of occurrence during the spring months (287/400, 71.8%) and decrease during the fall months (93/200, 46.5%) among the whole herds. The mean number of SEP score per pig was $1.20{\pm}1.28$. The prevalence of pleuropneumonia, pleuritis, pericarditis, and milk spot was 25.5% (296/1,160), 44.1% (512/1,160), 3.8% (44/1,160) and 17.6% (204/1,160), respectively. The most frequent region with lung lesion was diaphragmatic lobes (left 17.1%, right 17.3%). In the detection of viral pathogens by PCR, porcine circovirus type2 (PCV2) was positive in 86.9% (252/290), while porcine reproductive and respiratory syndrome virus (PRRSV) was not detected, In the case of bacterial pathogens, 50 microorganisms were isolated by PCR and/or microbiological test. The most frequently isolated bacteria was Streptococcus suis (20, 34.4%), followed by Pasteurella multocida (17, 29.3%), Streptococcus spp. (11, 3.4%), Actinobacillus pleuropneumoniae (2, 8.9%).

재조합 lysozyme-HJP34 단백질의 다양한 병원성 세균에 대한 항균 효능 및 자돈 사료첨가제의 가능성 평가 (Antibacterial evaluation of recombinant lysozyme-HJP24 proteins against various bacterial pathogens, and of its possibility test as a feed additive in piglets)

  • The recombinant lysozyme-HJL34 proteins were expressed and purified using commercial Escherichia (E.) coli expression system. Stx2e+ F18+ E. coli, Actinobacillus pleuropneumoniae (APP), Streptococcus (S.) suis, and Clostridium (C.) perfringens strains were isolated from pigs. The minimum inhibitory concentrations (MICs) of the recombinant lysozyme-HJP34 proteins were examined by means of the microtiter plate method, according to the NCCLS recommendations. The possibility of its as the alternatives to antibiotics was tested in piglets. The MICs were determined as 75 ㎍/mL, 300 ㎍/mL, 75 ㎍/mL, 35.5 ㎍/m against Stx2e+ F18+ E. coli, APP, S. suis, C. perfringens, respectively. A total of 25 piglets were divied 5 groups. The piglets in group A~C were fed with commercial feed and those in groups D, E were fed with commercial feedstuff. All piglets in groups B~E were challenged with virulent Stx2e+ F18+ E. coli, APP, S. suis strains. Groups C and D were treated with antimicrobial from 24 h after challenge. All piglets in group B died within 3 days after challenge. Among 5 piglets in groups C and D piglets, 80% survived after challenge. Among group E piglets, 60% were alive until the end of this study. Therefore, this study indicates that recombinant lysozyme-HJP34 proteins is a suitable possibility as a feed additive for reduction of diseases by bacterial pathogens in piglet feed.

아그로박테리움을 이용한 Actinobacillus pleuropneumoniae ApxIIA (ApxII toxin) 유전자 발현 옥수수 형질전환체 개발 (The development of transgenic maize expressing Actinobacillus pleuropneumoniae ApxIIA gene using Agrobacterium)

  • 돼지 흉막폐렴백신을 개발하기 위해 옥수수 HiII genotype 으로부터 유도한 type II형의 배발생캘러스를 식물발현벡터 pMYV611, pMYV613, pMYV616, V621, V622 및 V623로 형질전환시킨 Agrobacterium (C58C1)과 공동배양 하였다. 이들 식물발현벡터는 paromomycin 항생제 저항 유전자인 NPTII 선발마커와 표적 유전자로서 흉막폐렴균의 여러 가지 혈청을 생산하는 apxIIA유전자로 재조합하여 구축하였다. 식물발현벡터pMYV611, pMYV613, pMYV616, V621, V622 및V623의 경우 각각 4,120개, 5,959개, 7,581개, 52,329개, 48,948개 및 56,188개의 캘러스 클론을 Agrobacterium과 공동한 후 NPTII assay kit에 의해 nptII유전자의 발현빈도를 조사한 결과 각 벡터별로 2.3-4.4%의 캘러스 클론에서 항체결합 양성반응을 보였고, 이들 중 최종적으로 선발된 형질전환 캘러스 클론은 pMYV611에서 3개 (0.07%), pMYV613에서 4개 (0.07%), pMYV616에서 2개 (0.02%), V621에서 51개 (0.1%), V622에서 72개 (0.15%) 및 V623에서 102개 (0.18%)를 각각 얻었다. 형질전환된 캘러스 클론으로부터 재분화된 식물체에서 유전자 도입여부를 Southern 분석으로 통해 확인한 결과 pMYV613에서 2개 식물체 및 V623에서 얻은 2개 식물체에서 각각 확인되었다.

정상인과 정신지체인, 다운증후군 환자에서 치주질환 원인균의 출현율 (PERIODONTOPATHIC BACTERIA IN SUBGINGIVAL PLAQUE OF NORMAL AND HANDICAPPED PERSON)

  • 정상인과 정신지체인 그리고 다운증후군 환자에서 치주상태 및 치주질환 원인균의 출현율을 비교 평가하고자 정상 학생 65명과 정신지체를 가진 학생 34명, 다운증후군을 가진 학생 28명, 총 127명을 조사대상으로 치태지수와 치은지수를 측정하고 치은연하 치태에 존재하는 Porphyromonas gingivalis, Tannerella forsythia(B. forsythus), Treponema denticola, Fusobacterium nucleatum, Actinobacillus actinomyce temcomitans 균을 중합효소연쇄반응을 이용하여 검사하고 다음과 같은 결과를 얻었다. 1. 치태지수와 치은지수는 정신지체인군과 다운증후군 환자군에서 정상인군보다 높았다. 2. 정신지체인군과 다운증후군 환자군은 정상인군에 비해 치주질환 원인균의 출현율이 높았으며 특히 P. gingivalis와 T. denticola. A. actinomycetemcomitans가 유의한 차이를 보였다. 다운증후군 환자군과 정신지체인군의 비교시 치주질환 원인균의 출현율은 유의한 차이를 보이지 않았다. 3. 연령별 비교시 정상인군의 P. gingivalis는 14세이상군에서 크게 증가하는 양상을 보였다. 정신지체인군과 다운증후군 환자군에서 P. gingivalis는 정상인군보다 조기에 더 높은 비율로 출현하였으며 연령의 증가에 따라 증가양상을 보였다. T. denticola, A. actinomycetemcomitans는 정상인군보다 정신지체인군과 다운증후군 환자군에서 모든 연령대에서 높은 비율을 보였다. 4. 치태지수는 T. denticola와 유의한 관련성이 있었으며, 치은지수는 T. denticola, A. actinomycetemcomitans와 높은 관련성을 보여주었다. 이상의 결과를 요약해 보면 정신지체인군과 다운증후군 환자군은 정상인군보다 치태지수, 치은지수가 더 높았으며, P. gingivalis, T. denticola, A. actinomycetemcomitans의 출현율에서 유의한 차이를 보였다. 다운증후군 환자군과 정신지체인군은 치주질환 원인균이 어린 시기부터 매우 높게 출현하였으며, 두 군간에는 유의할 만한 차이를 보이지 않았다.
