A combination of electrospray ionization and tandem mass spectrometry was used to characterize the scavenging reactions of acrylamide (AA) in the presence of glutathione (GSH) in vitro. In the presence of GSH, AA was deactivated effectively and scavenged by reactions consuming small amount of GSH. Reaction products and structural information were identified using collision-induced dissociation (CID) in an ion trap mass spectrometer. In the mixture of GSH and AA, significant increase in abundance of fragment ion peak was observed at m/z 233, which was identified as $[Cys-Glu]^+$, formed by the elimination of glycine moiety of GSH. GSH also contributes to the AA scavenging reaction by conjugating with AA through the sulfhydryl group in cysteine moiety. The probable scavenging reaction pathway of AA in the presence of GSH has been proposed based on the CID experimental data.
Based on the zymography analysis, Bacillus sp. DJ-4 (screened from Doen-Jang, a Korean traditional fermented food) secretes seven extracellular fibrinolytic enzymes (EFEs; 68, 64, 55, 45, 33, 27, and 13 kDa) in culture broth. These seven EFEs were analyzed by newly applied SDS-fibrin zymography combined with gradient polyacrylamide (SDS-FZGP). This improved gel system was used with a 5-20% acrylamide gradient in a fibrin zymogram gel for the separation of proteins with molecular masses from below 10kDa to over 100kDa on one gel plate. Using this system, high molecular weight bands (HMWBs) were clearly and sharply resolved. We also examined the relationship between an acrylamide concentration and the enzymatic activity of EFE using densitometric analysis.
Collagen is the major structural protein of connective tissues. It can be used as a prosthetic biomaterial applicable to artificial skin, tendon, ligaments, and collagen implants. The objective of this study is to investigate the possibility of realizing wound dressing medical products by the synthesis of composite materials with collagen and a biodegradable polymer, PLLA, via a surface modification process. Type I collagen was obtained from pig skin by a separation process. The structural characteristics of the extracted collagen were confirmed by SDS-polyacrylamide (PAcr) gel electrophoresis (PAGE) and FTIR. Also, PLLA-g-PAcr was synthesized by the radical polymerization of acrylamide initiated by AIBN in the presence of PLLA. The surface of PLLA was modified by the presence of the acrylamide residues. The structural characteristics of the copolymer were analyzed by FTIR, $^1H-NMR$ and contact angle measurements. The water uptake and WVTR of the collagen/PLLA-g-PAcr composite tended to increase with increasing collagen concentration and with decreasing EDC concentration.
This study evaluated the kinetics of acrylamide (AM) biodegradation by mixed culture bacteria and Enterobacter aerogenes (E. aerogenes) in sequencing batch reactor (SBR) systems with AQUASIM and linear regression. The zero-order, first-order, and Monod kinetic models were used to evaluate the kinetic parameters of both autotrophic and heterotrophic nitrifications and both AM and chemical oxygen demand (COD) removals at different AM concentrations of 100, 200, 300, and 400 mg AM/L. The results revealed that both autotrophic and heterotrophic nitrifications and both AM and COD removals followed the Monod kinetics. High AM loadings resulted in the transformation of Monod kinetics to the first-order reaction for AM and COD removals as the results of the compositions of mixed substrates and the inhibition of the free ammonia nitrogen (FAN). The kinetic parameters indicated that E. aerogenes degraded AM and COD at higher rates than mixed culture bacteria. The FAN from the AM biodegradation increased both heterotrophic and autotrophic nitrification rates at the AM concentrations of 100-300 mg AM/L. At higher AM concentrations, the FAN accumulated in the SBR system inhibited the autotrophic nitrification of mixed culture bacteria. The accumulation of intracellular polyphosphate caused the heterotrophic nitrification of E. aerogenes to follow the first-order approximation.
본 연구는 후렌치 후라이의 가열 온도 및 시간에 따른 아크릴아마이드 생성량과 관능적 품질 특성을 조사하였다. 가열 온도와 가열 시간의 변화에 따라 아크릴아마이드 생성 함량에 많은 영향을 미치는 것으로 나타났다. 일반적으로 후렌치 후라이는 160-$180^{\circ}C$ 온도에서 2-5분간 가열하는데 아크릴아마이드의 생성은 가열 온도 및 시간에 크게 영향을 받으므로 이를 조절함으로써 식품 내 아크릴아마이드 생성을 줄일 수 있다. 또한 아크릴아마이드 생성량을 줄이면서 관능적으로 우수한 후렌치 후라이를 제조하는 것이 중요하다. 따라서 후렌치 후라이 제조를 위해 $165^{\circ}C$에서 2분 30초 동안 가열하는 것이 아크릴아마이드의 생성을 저감화하면서 크리스피 및 관능적 측면에서 우수한 후렌치 후라이를 제조할 수 있을 것으로 사료된다.
감자칩을 대신할 고구마칩용으로 적합한 품종을 선별하기 위해 고구마 육색이 크림색인 신율미와 신천미, 주황색인 주황미와 신황미, 자색인 연자미와 신자미 및 새롭게 육종한 대유미, 다호미, 건황미, 풍원미로 고구마 칩을 제조하여 아크릴아마이드 함량과 품질 특성을 비교하였다. 고구마 칩의 아크릴아마이드 함량은 연자미(1.07 mg/kg)을 제외하고는 0.48 mg/kg로 낮은 값을 나타냈다(p<0.05). 색도는 품종 간에 유의적인 차이를 보였지만 생고구마와 거의 유사하였다. 고구마 칩의 기계적 텍스처 특성치인 경도는 주황미가 482.7 g, 신황미가 490.2 g으로 유의적으로 낮은 값을 보였으며 대유미는 1332.1 g으로 가장 높은 값을 보였고, 깨짐과 부서짐성에서도 주황미와 신황미가 가장 낮은 값 대유미에서 높은 값을 보였다(p<0.05). 관능평가 결과 칩의 품질은 경도, 부서짐성과 바삭함 뿐만 아니라 맛과 향미에도 영향을 받았다. 풍원미 칩 품질에 대한 선호도가 유의적으로 가장 높은 점수를 보였고 그 다음은 건황미와 다호미 순이었다(p<0.05). 위의 결과로부터 고구마 칩 제조에는 주황색의 풍원미, 건황미, 다호미가 적합한 품종으로 확인되었고, 자색은 신자미, 크림색은 대유미가 칩의 용도로 좋은 점수를 받았다. 고구마칩의 품질이나 산화안정성도 주황미와 신황미가 가장 낮아 칩과 같은 튀김에는 적합하지 않은 것으로 판단되었다.
본 연구에서는 백미, 현미, 가바현미, 귀리 및 보리를 이용하여 누룽지를 제조하고 각 곡류 별 누룽지의 품질특성, 아크릴아마이드 생성 정도 및 항산화 활성을 측정하였다. 백미로 제조한 누룽지의 수분 함량은 1.48%로 가장 낮았고, 보리로 제조한 누룽지의 수분 함량은 6.53%로 가장 높게 나타났다. 조단백질, 조지방 및 회분 함량은 곡류의 종류에 따라 다르게 나타났으며 귀리 누룽지는 조단백질 및 조지방 함량이 높은 것으로 나타났다. 가용성 고형분은 보리 누룽지가 1.90°Brix로 가장 높았고, 현미 누룽지는 1.50°Brix로 가장 낮았다. 명도(L*)는 백미 누룽지에서 가장 높았고, 가바현미 누룽지에서 가장 낮았다. 적색도(a*)는 백미 누룽지에서 다른 곡류들에 비해 비교적 높았고, 백미 이외의 다른 곡류에서는 통계적인 차이는 없는 것으로 나타났다. 아크릴아마이드는 백미 누룽지에서 37.24 ㎍/g로 가장 적게 검출되었고, 현미, 보리 및 가바현미로 제조한 누룽지에서는 70.41-99.99 ㎍/g, 귀리로 제조한 누룽지는 255.50 ㎍/g로 가장 많이 검출되었다. 총 폴리페놀 및 총 플라보노이드 함량은 백미 누룽지에서 가장 낮았고, 귀리와 가바현미 누룽지에서 비교적 높았다. 백미를 제외한 곡류(귀리, 가바현미, 현미, 보리)로 제조한 누룽지의 항산화 활성이 백미 누룽지에 비해 높은 값을 보였다. 이상의 결과를 통해, 백미 누룽지에 비해 현미, 가바현미, 귀리 및 보리로 제조한 누룽지의 총 폴리페놀, 총 플라보노이드 및 항산화 활성이 높은 것을 확인하였고, 누룽지 제조를 위한 곡물은 항산화 활성 및 아크릴아마이드 형성량에 대한 종합적인 고려를 통한 적절한 선택이 필요할 것으로 판단된다.
