$3{\beta}$-Hydroxy-12-oleanen-28-oic Acid 유도체들의 PTP-1B 저해활성에 대한 CoMFA 분석과 활성 분자들의 예측 (CoMFA Analysis on Inhibitory Effect of $3{\beta}$-Hydroxy-12-oleanen-28-oic Acid Analogues on PTP-1B Activity and Prediction of Active Compounds)

  • 기질 화합물로써 $3{\beta}$-hydroxy-12-oleanen-28-oic acid 유도체들의 치환기($R_1{\sim}R_4$)가 변화함에 따른 protein tyrosine phosphatase(PTP)-1B의 저해활성에 관한 비교 분자장 분석(CoMFA) 모델을 유도하고 정량적으로 검토하였다. 최적의 CoMFA F1 모델은 가장 높은 예측성과 상관성($r^2_{cv.}=0.654$$r^2_{ncv.}=0.995$)을 나타내었다. 저해활성에 관한 CoMFA장 기여비율(%)의 순서는 입체장(53.0%), 정전기장(36.2%) 및 소수성장(10.8%) 이었다. 등고도 분석 결과로부터 저해활성은 기질 분자 내 $R_4$-치환기에 의존적이었으며 특히 melanin 저해활성이 높은 새로운 화합물(P1 및 P2)이 예측되었다.

히알루론산이 골 형성에 미치는 영향에 관한 실험적 연구 (THE EFFECT OF HYALURONIC ACID ON MOUSE CALVARIA PRE-OSTEOBLASTS OSTEOGENESIS IN VITRO)

  • Hyaluronic acid (HA) is an almost essential component of extracellular matrices. Early in embryogenesis mesenchymal cells migrate, proliferate and differentiate, in part, because of the influence of HA. Since the features of embryogenesis are revisited during wound repair, including bone fracture repair, this study was initiated to evaluate whether HA has an effect on calcification and bone formation in an in vitro system of osteogenesis. Mouse calvaria Pre-osteoblast (MC3T3-E1) cells were cultured in ${\alpha}-MEM$ medium with microorganism-derivative hyaluronic acid that was produced by Strep. zooepidemicus which of molecular weight was 3 million units. The dosages were categorized in each 0.5, 1.0 and 2.0 mg/ml concentration experimental groups. After 2 and 4 days cultures in expeirmental and control groups, the tendency of cell proliferation, MTT assay, protein synthesis ability, collagen synthesis and alkaline phosphatase activity were analysed and bone nodule formation capacity were measured with Alizarin Red S stain after 29 days cultures. The cell proliferation was increased in time, especially the group of 0.5 and 1.0 mg/ml concentration of HA were showed prominent cell proliferation. After 2 and 4 days culture, experimental groups in general were greater cell activity in MTT assay. The protein synthesis was increased in all experimental groups compared to control group, especially most prominent in 1.0 mg/ml concentration group. The collagen synthesis capacity were increased in HA experimental groups, especially prominent in 1.0 mg/ml group and the activity of alkaline phosphatase were increased, especially also prominent in 1.0 mg/ml group, compared to control group. Above these, the activity of mouse carvarial pre-osteoblast cells was showed greater bone osteogenesis activity in all applied HA experimental group, especially group of 1.0 mg/ml concentration of HA.

연산 오골계 물 추출물이 조골세포와 파골세포의 활성에 미치는 영향 (Effect of Gallus gallus var. domesticus (Yeonsan ogolgye) Extracts on Osteoblast Differentiation and Osteoclast Formation)

  • 본 연구는 연산 오골계의 물 추출물을 이용하여 MG-63 조골세포와 RAW 264.7 파골세포의 분화에 미치는 영향을 분석하였다. 조골세포의 분화에 미치는 영향을 관찰하기 위해 ALP 활성 분석과 alizarin red 염색을 통해 골 석회화를 측정하였다. 그리고 RAW 264.7 파골세포의 분화 억제 활성을 확인하기 위해 TRAP 활성과 염색을 분석 측정하였다. 연산 오골계 물 추출물을 농도별($250-1,000{\mu}g/ml$)로 처리한 결과 조골세포와 파골세포에서 세포독성을 나타내지 않았다. ALP 활성은 3년산 암탉 육질이 133.8%, 3년산 수탉 육질은 129.6%로 육질이 껍질보다 조골세포 분화력이 높았다. 그리고 육질에서는 3년산이 1년산보다 ALP 활성이 높으나 껍질에서는 1년산이 3년산보다 조골세포 분화능력이 높았다. 성별 간 ALP 활성은 전체적으로 암탉이 수탉보다 높은 ALP 활성을 보였다. 또한, 골 석회화 능력은 3년산 암탉 육질이 연령과 성별을 통틀어 124.3%로 가장 뛰어났으며 TRAP 활성은 3년산 수탉 육질이 31.8%로 연령과 성별을 통틀어 억제 활성이 가장 뛰어났다. 연산 오골계 물 추출물은 조골세포 분화능력이 뛰어나 골의 석회화를 촉진하는 능력이 뛰어났으며 파골세포의 분화를 억제하여 골 흡수를 억제하는 능력이 뛰어났다. 이에 연산 오골계는 골 기능 강화와 골 관련 질환에 대한 예방과 치료에 효과가 있을 것으로 보인다.

구속스트레스를 가한 흰쥐에서 Cholate류의 항스트레스 작용에 관한 연구 (Anti-stress Effect of Cholic acid Derivatives in Restraint Stress Induced Rats)

  • This study was done to investigate whether cholic acid derivatives have anti-stress activity and what is a cause of this anti-stress effect. Seven cholic acid derivatives (cholic acid, taurocholic acid, ursodeoxycholic acid, tauroursodeoxychoic acid, chenodeoxy cholic acid, dehydrocholic acid, hyodeoxycholic acid) were used, silymarin and valproic acid were used as positive controls. Stress was induced by restraint immobilization technique plus water immersion (24hrs) and adrenal weight, spleen weight, adrenal ascorbic acid, serum cholesterol, lactate dehydrogenase (LDH), alkaline phosphatase (ALP), adrenal cholesterol, glucose and corticosterone levels were measured as stress indicators. Most cholic acid derivatives markedly decreased the adrenal weight, and TUDCA and DHCA increased the spleen weight. The restraint stress induced increments in serum LDH, ALP and cholesterol were attenuated by most cholic acid derivatives. Cholic acid, taurocholic acid and tauroursodeoxycholic acid only increased the content of adrenal ascorbate. While valproic acid showed an inhibitory effect against stress, silymarin did not. Our findings suggest that most cholic acid derivatives have anti-stress effect and that their anti-stress effect is, in part, related to choleretic activity.

Enzyme and Microbial Activities in Paddy Soil Amended Continuously with Different Fertilizer Systems

  • Soil enzyme and microbial activities are affected by fertilizer and compost applications and can be used as sensitive indicators of ecological stability. Microbial population and soil enzymes viz., dehydrogenase, urease, acid phosphatase and aryl-sulphatase were determined in the long-term fertilizer and compost applied paddy soil. Soil samples were collected from the four treatments (control, compost, NPK and compost+NPK). Long-term NPK+compost application significantly increased activities of urease, dehydrogenase and acid phosphatase than all other treatments. The compost application enhanced activities of urease, dehydrogenase and acid phosphatase than the NPK application. However, arylsulfatase activity was not significantly different between compost and fertilizer application. The highest microbial population was recorded in the NPK+compost treatment. The compost application also resulted in higher microbial population than the NPK application. The above results indicate that ecological stability could be maintained by application of compost alone or with NPK.

인삼이 장기의 Alkaline Phosphatase활성과 혈중 무기 인량에 미치는 영향 (Effects of Korean Ginseng on the Visceral Alkaline Phosphatase Activity and Blood Inorganic Phosphorus Level)

  • 인삼이 생쥐의 장기 alkaline phosphatase 활성에 미치는 영향을 아기 위하여 인삼의 알코올 추출물을 Tyrode solution에 희석하여 체중 10g당 0.1mg 또는 0.2mg 씩 매일 주사하여 7일간과 14일간 처리하였다. 공장, 신장, 간, 혈청등에서의 alkaline phosphatase 활성을 sodium ${\beta}-glycerophophosphate$를 기질로 하여 각 장기의 homogenate 또는 혈청을 반응시켜 효소의 unit를 결정하였으며, 혈중 무기 인의 함량도 측정하여 다음의 결과를 얻었다. 1 숫생쥐의 7일간 처리에서 인삼추출물(0.1mg/10g) 처리군이 Tyrode solution 처리군보다 공장, 신장, 간, 혈청에서 각각 15.28%, 12.86%, 19.05%, 11.70%의 alkaline phosphatase 활성증가를 나타내었다. 2. 암생쥐의 7일간 처리에서 인삼추출물(0.2mg/10g)은 공장, 신장, 간, 혈청에서 각각 3.46%, 6.94%, 20.37%, 4.03%의 활성증가를 나타내었다. 3. 숫생쥐의 14일간 처리에서 인삼추출물(0.1mg/10g)은 공장, 신장, 간, 혈청에서 각각 15.92%, 19.76%, 10.16%, -1.63%의 활성증가를 나타내었다. 4. 암생쥐의 14일간 처리에서 인삼추출물(0.2mg/10g)은 공장, 신장, 간, 혈청에서 각각 18.89%, 24.55%, 16.97%, 27.59%의 활성증가를 나타내었다. 5. 암수 다 같이 7일간 처리에서 간의 alkaline phosphatase 활성증가현상은 14일 처리에서 약간 감소하는 반면 공장, 신장, 혈청의 효소활성증가 현상은 7일간 처리에서보다 14일간 처리에서 훨씬 촉진되었다. 6. 인삼추출물의 7일간, 14일간 처리는 수컷의 혈중 무기 인량을 각각 22.20%, 20.96% 증가시켰으며, 암컷에서는 각각 22.38%, 17.57% 증가시켰다.

성장 발육에 따른 흰쥐 장기내 phytase의 분포성 (Distribution of Phyrase in The Development Rat Organs)

  • Phytase(myo-inositol hexakisphosphate phosphohydro-lase;EC 3.1.38)활성은 소화기관인 소장 점막에서나 나타났고, 그 외 다른 장기에서는 alkaline phosphatase활성만을 측정할 수 있었다. 그리고 anti-90kDa phytase항혈청을 이용한 면역조직학적 조사 결과 소장 이외의 장기에서는 본 효소의 단백질 밴드가 검출되지 않았다. 이와 같은 결과 phytase는 소장에만 특이적으로 존재하며, 소장 특이적 효소의 생리적 역할로서 Phytic acid(inositol=hexakisphos-phate)를 가수분해한다. 흰쥐의 성장 발육과 더불어 phytase의 활성은 증가한다. 출생 후부터 이유기 전까지는 70kDa phytase외에도 90kDa phy-tase가 출현한다. 이 90kDa phy-tase는 이유기에 합성되는 것으로 추정된다.

Spermatogenic index and hormonal profile in the rats received chromatographic fractions of ethanol extract of Crotalaria juncea L. seeds

  • The ethanol extract of the Crotalaria juncea seeds, which showed promising antispermatogenic and antiandrogenic activities in albino mice, was taken up further for the isolation of the active fractions present in it. Two fractions that were obtained from thin layer chromatography were subjected for testing to know their antispermatogenic and antiandrogenic activities. After preliminary trials the fraction I showed maximum antifertility activity at the dose level of 200 mg/kg body weight when administered orally to the rats for 50 days. The fraction I was found to affect spermatogenesis as well as the endocrine functions of the testis as indicated by gravimetric, histopathological and biochemical changes. Further this fraction has caused degenerative changes in the seminiferous tubules and Leydig cells of the testis. The accessory reproductive organs like epididymis, seminal vesicles, vas deferens, prostrate, Cowper's gland and Levator Ani muscle showed significant malfunction. Cauda epididymal sperm count and sperm motility were reduced significantly. The treatment has also resulted in increase in the cholesterol level and alkaline phosphatase activity, and decrease in protein, glycogen, sialic acid contents and acid phosphatase activity in testis. It is noteworthy that RIA studies have shown significant reduction in serum FSH, LH and testosterone. Scanning electron microscopic observations revealed abnormalities in sperm structure.

발생과정중 흰쥐 간 Non-parenchymal Cell의 미세구조 및 수종 Phosphatase의 활성에 관하여 (Studies on Ultrastructure and Several Phosphatase Activity in the Non-parenchymal Cell of the Developing Rat Liver)

  • A number of recent ultrastructural studies have shown marked differences between the two lining cell types in adult liver sinusoids, endothelial cells and Kupffer cells. In the present study, the ultrastructural features and electron microscopic cytochemistry of sinusoidal lining cells in the fetal liver were studied through fetal period to neonate in the rat. At fetal period, the sinusoid, which contains various blood component, in lined by the endothelial cells, the Kupffer cells and the fat storing cells that located in the space of Disse. As gestation proceeded, these eel's are arranged as adult liver sinusoids. The sinusoidal wall appears to be discontinuous with open fenestration between endothelial cells, but no basal lamina can observed. It seems to be morphologically and functionally distinct at the early gestation between the endothelial cells and the Kupffer cells, the latter showing marked phagocytized activity. The fat storing cells, which contain several fat droplets, are located in the space of Disse. Ultrastructural localization of the acid and alkaline phosphatase activity were noted on the sinusoidal lining cells.

참두릅 잎의 Protein Tyrosine Phosphatase 1B와 α-Glucosidase 저해 활성 (Inhibitory Activity of Aralia elata Leaves on Protein Tyrosine Phosphatase 1B and α-Glucosidase)

  • Anti-diabetic potential of the leaves of A. elata through the inhibitory activity on PTP1B and ${\alpha}$-glucosidase has not been reported. In this study, the EtOAc fraction of methanolic extract from the leaves of A. elata showed potent inhibitory activity against the PTP1B and ${\alpha}$-glucosidase with $IC_{50}$ value of $96.29{\pm}0.3$ and $264.71{\pm}14.87{\mu}g/mL$, respectively. Three known triterpenoids, oleanolic acid, oleanolic acid-28-O-${\beta}$-D-glucopyranoside and oleanolic acid-3-O-${\beta}$-D-glucopyranoside were isolated from the most active EtOAc fraction. We determined the chemical structure of these triterpenoids through comparisons of published nuclear magnetic resonance (NMR) spectroscopic data. Furthermore, we screened these triterpenoids for their ability to inhibit PTP1B and ${\alpha}$-glucosidase over a range of concentrations ($12.5-50{\mu}M$). All three terpenoids significantly inhibited PTP1B in a concentration dependent manner and oleanolic acid effectively inhibited ${\alpha}$-glucosidase. In addition, these compounds revealed potent inhibitory activity with negative binding energies toward PTP1B, showing high affinity and tight binding capacity in the molecular docking studies. Therefore, the results of the present study clearly demonstrate that A. elata leaves and its triterpenoid constituents might be beneficial in the prevention or treatment of diabetic disease.
