Effect of Adipic Acid on Extending Shelf Life of Kimchi During Cooling Storage (Adipic Acid 첨가에 의한 김치의 저온 저장 중 저장 기간 증가에 관한 연구)

  • To delay the fermentation of Kimchi during storage and handling, adipic acid, was selected due to its low acid dissociation constant. Sensory evaluation was conducted to find maximum dosage of adipic acid which does not influence the raw taste of Kimchi and 0.12%(wt. of adipic acid/wt. of Kimchi) was selected. At $10^{\circ}C$, the optimum and maximum shelf life was respectively 3.2 and 2.3 times longer than that of control. At 15 and $20^{\circ}C$, extension of shelf life was as follows: maximum shelf life with adipic acid was extended about 2.3-2.7 times and optimum shelf life was 1.3-1.5 times.

  • Using a new conductometric method, dissociation constants of 3-cyano, 4-cyano, 3-amino and 4-aminopyridine were measured in the temperature range 15 ∼ 40${\circ}C$ and pressure up to 2500bar in aqueous media. This method is convenient to apply to the low dissociative acid and base but have to do tedious extrapolating procedure for the ionic conductance in elaborated temperatures and pressures and have to know any reference dissociation constant. The measured dissociation constants were increased as the temperature increase but decreased as the pressure increase. From the constants, various thermodynamic properties were evaluated and discussed for the dissociation reactions.

  • Mononuclear schiff base ligand N,N'-bissalicylidene-1,2-phenylenediamine(BSPD) and binuclear schiff base ligands N,N',N',N'''-tetrasalicylidene-3,3',4,4'-tetraaminodiphenyl-methane (TSTM), N,N',N'',N'''-tetrasalicylidene-3,3'-diaminobenzidine (TSDB) have been synthesized. Proton dissociation constants of the ligands were determined by potentiometric method. The synthesized ligands and complexes formed with Cu(II) ion. These complexes were investigated by cyclic voltammetry and differential pulse voltammetry. The results revealed two step diffusion controlled redox process. The mononuclear complex Cu(II)-BSPD and binuclear complexes $Cu(II)_2$-TSDB and $Cu(II)_2$-TSTM were used in the oxidation reaction of ascorbic acid. The reaction rates were in the order of $Cu(II)_2$-TSTM>$Cu(II)_2$-TSDB>Cu(II)-BSPD, indicating that the binuclear $Cu(II)_2$-TSTM complex had the fastest values.

Behavior of Burkholderia thailandensis (Burkholderia pseudomallei surrogate) in Acidified Conditions by Organic Acids Used in Ready-to-Eat Meat Formulations under Different Water Activities

  • This study evaluated the antimicrobial effects of meat processing-related organic acids on Burkholderia thailandensis (Burkholderia pseudomallei surrogate) with different water activities. B. thailandensis KACC12027 (4 log CFU/mL) was inoculated in microwell plates containing tryptic soy broth pH-adjusted to 4, 5, 6, and 7 with ascorbic acid, citric acid, and lactic acid and with water activities adjusted to 0.94, 0.96, 0.98, and 1.0 with NaCl, followed by incubation at $35^{\circ}C$ for 30 h. The optical density (OD) of the samples was measured at 0, 3, 6, 12, 24, and 30 h at 595 nm to estimate the growth of B. thailandensis. Growth of B. thailandensis was observed only at water activity of 1.0. In general, more bacterial growth (p<0.05) was observed at pH 6 than at pH 7, and the antimicrobial effects of the organic acids on B. thailandensis were in the following order: Ascorbic acid > lactic acid > citric acid after incubation at $35^{\circ}C$ for 30 h. These results indicate that organic acids in meat processing-related formulations should be useful in decreasing the risk related to an emerging high risk agent (B. pseudomallei).

THE RELATIONSHIP BETWEEN BUFFER INDEX CURVE AND FERMENTATION QUALITY OF SILAGE

  • The purpose of this study was to investigate the possibility of using the buffer index curve as a criterion of silage quality evaluation. The buffer index (or buffer capacity, $\beta$) value is the amount of 0.1N NaOH consumed in titrating from the beginning to the end of the chosen pH step. This value equals the slope of the titration curve at the chosen pH range (${\beta}={\Delta}NaOH/{\Delta}pH$). There were two patterns of buffer index curve. Poorly preserved silages had a peak at pH 5.0, whereas with well preserved silages the peaks tended to rise to the pH 3.75 or 4.00. Well preserved, wilted silages with high pH also appeared to have peak values at pH 3.75 or 4.00. There was a high positive correlationship between the concentration of lactic acid and peak value of good quality silage (r = 0.994, p < 0.01). Also a high positive correlationship (r = 0.899, p < 0.001) was found between the sums of concentration of acetic and butyric acids and the peak values of poor quality silages. The results of these experiments verified the value of the buffer index method as a criterion of silage quality evaluation.

  • Hemolymph SHBP (hJHBP) was partially purified from last instar larvae of Bombyx zori by gel filtration and their optimal reaction conditions of dextrin coated charcoal binding assay were determined. Dissociation constant (KD) of hJHBP for JH III was calculated to be 1.45 $\times$ 10-7 M at $4^{\circ}C.$ The molecular weight of hJHBP was estimated to be 30 kDa by gel filtration on a calibrated Sephadex G-100 column and 33 kDa by SDS-PAGE. These results indicate that hSHBP consists of a single polvpeptide chain. Isoelectric point of hJHBP was found to be pH 5.1 and 19 of the first 20 amino acid residues were determined from N-terminus of purified hJHBP.

  • The $pK_a$ values of benzoic acid and meta, para-halogen substituted benzoic acids in MeOH-$H_2O$ mixtures (0∼80% of MeOH) have been determined at 25$^{\circ}$C using a conductometric method on the basis of the Fuoss-Kraus equation, and further verified using modified conductometric method of Gelb. The dependence of $pK_a$ on halogen substituents has been discussed in terms of substituent-constant (${\sigma}$), which is devided into electron-withdrawing inductive contribution (${\sigma}_1$) and electron-donating ${\pi}$-resonance one (${\sigma}_R$). The linear-dependence of ${\sigma}_1$'s on $D^{-1}$ with positive slope and that of ${\sigma}_R$'s on $D^{-1}$ with negative slope have been interpreted on the basis of field effect and through-space interaction of ${\pi}$-lone pair of halogen substituent and ionization center via ${\pi}$-system of benzene ring.

Interaction of ${\alpha}$-Ketoglutarate Dehydrogenase Complex with Allosteric Regulators Detected by a Fluorescence Probe, 1,1'-bi(4-aniline)naphthalene-5,5'-disulfonic acid, an Inhibitor of Catalytic Activity

  • The interaction of ${\alpha}-ketoglutarate$ dehydrogenase complex (${\alpha}-KGDC$) with a hydrophobic fluorescent probe [1,1'-bi(4-aniline)naphthalene-5,5'-disulfonic acid] (bis-ANS) was studied. The punfied ${\alpha}-KGDC$ was potently inhibited by bis-ANS with an apparent half maximal inhibitory concentration ($IC_{50}$) of 9.8 ${\mu}m$ at pH 8.0. The catalytic activities of both the E1o and E2o subunits were predominantly inhibited while that of the E3 component was hardly affected. The binding of bis-ANS to the enzyme caused a marked enhancement and blue shift from 523 nm to 482 nm in the fluorescence emission spectrum. The dissociation constant ($K_d$) and the number of binding sites (n) were calculated to be 0.87 mM and 158, respectively. Allosteric regulators such as purine nucleotides and divalent cations further increased the fluorescence intensity of the $bis-ANS-{\alpha}-KGDC$ binary complex. These data suggest that the binding of these allosteric regulators to ${\alpha}-KGDC$ may cause the conformational changes in the enzyme and that bis-ANS could be used as a valuable probe to study the interaction of the multi-enzyme complex and its allosteric regulators.

  • Characteristics of poly(vinyl alcohol)-poly(methacrylic acid) system (PVA.-PMAA system) for fixation of radioactive contaminant on sand were studied. Dissociation of carboxyl group in PMAA was found to be suppressed by PVA Permeability of sand layer treated with PVA-PMAA solution is directly proportional to the PMAA concentration when the [PMAA] is below 0.082 M and the empirical proportional constant (k) is -8.95$\times$10 ̄$^4$cm$^{5}$ /mole. The change of permeability can be explained by the formation of an intermacromolecular complex between PVA and PMAA The polymer bridge formed on a sand surface combines sand yams more firmly. The PVA-PMAA system is more effective than the PVA system for the fixation of deposited condensational radionuclides.
