Isolatin and Quantitative Determination of 20-Hydroxyecdysone from Achyranthis Radix (우슬로부터 20-Hydroxyecdysone의 분리 및 함량분석)

  • Separation and quantitative determination of 20-hydroxyecdysone from Achyranthis Radix has been conducted by using HPLC method. 20-hydroxyecdysone in a methanol extract from the raw drug was separated on a reverse phase column using a $CH_3CN-H_2O\;(18:82)$ solvent system and the average content is $0.0931{\pm}0.0048%$. For the preparation of authentic standard, we isolated 20-hydroxyecdysone from the roots of Achyranthes fauriei by $SiO_2$ and RP-18 column chromatography.

Review on Original Plane of Oriental Medicines Used in Korea (국내 유통 한약재의 기원식물에 관한 고찰)

  • To clarify the botanical origins of oriental medicines which have been argued or confused for plant origins, species of original plants were investigated through the textural research for oriental medicines and the comparison of Chinese, Korean, Japanese and North Korean Pharmacopoeia. Twenty oriental medicines were studied; Angelicae Gigantis Radix, Rhei Rhizoma, Atractylodis Rhizoma Alba, Aconiti Lateralis Radix Preparata, Acanthopanacis Cortex, Osterici Radix, Cnidii Rhizoma, Saposhnikovae Radix, Magnoliae Cortex, Paeoniae Radix, Liriopis Tuber, Zanthoxyli Fructus, Achyranthis Radix, Sinomeni Caulis et Rhizoma, Polygonati Rhizoma, Cinnamomi Cortex, Visci Herba et Loranthi Ramulus, Fritillariae Thunbergii Bulbus, Pogostemonis Herba, and Curcumae Longae Radix.

Establishment of content criteria of marker compounds through the monitoring of Achyranthis Radix collected from Korea and China (한국 및 중국 지역에서 수집된 우슬의 모니터링을 통한 지표성분의 함량기준 설정)

  • Two marker compounds of Achyranthis Radix, ecdysterone and inokosterone, were analyzed by HPLC on an ODS column ($250{\times}4.6$ mm, 5 ${\mu}m$) with a mobile phase of 15% acetonitrile containing 0.08% formic acid at a flow rate of 1.0 mL/min and a detection wavelength of UV 254 nm. The method was validated by ICH guideline and applied to the monitoring of marker compounds in 93 samples of Achyranthis Radix collected at various areas in Korea and China. The new content criteria of ecdysterone and inokosterone, established using linear regression method were 0.033% and 0.020%, respectively. When the new content criteria were applied to the quality control test of commercial Achyranthis Radix, 95.4% of total samples including 100% of Korean and 92.6% of Chinese samples were passed the test. Application of new content criteria could protect the Korean products and decrease the distribution of Chinese products with lower quality.

Studies on Antioxidant and Anti-hyperlipidemic Effects in Induced Hypercholesterolemic Rats of Steamed Achyranthis Bidentatae Radix (우슬(牛膝)의 증숙(蒸熟)에 따른 항산화 효능 및 고지혈증 유발 흰쥐에 대한 고지혈증 개선 효능에 대한 연구)

  • Objectives : This study was conducted to compare the effect of Achyranthis Bidentatae Radix on antioxidant changes and antihyperlipidemics in hypercholesterolemic rats according to changes in the number of steams. Methods : The experimental animals were divided into six groups: normal (Nor), high cholesterol diet (Veh), high cholesterol diet with Simvastatin 5 mg/kg (Sim), high cholesterol diet with AR0 extract 200 mg/kg (AR0), high cholesterol diet with AR6 extract 200 mg/kg (AR6) and high cholesterol diet plus AR9 extract 200 mg/kg (AR9). I measured HDL, LDL TC, TG contents and ROS from each serums and compared them. I analyzed antioxidant-related protein expressions and cholesterol-related protein expressions in each liver tissues. I also performed optical microscopic analysis through liver tissue staining. Results : The Antioxidant activity of Achyranthis Bidentatae Radix was increased by steaming processes. In vivo, AR0 decreased liver weight in hyperlipidemic rats. LDL TC, TG, AI and CRF were decreased but HDL was increased in AR0. AST, ALT, creatinine, and BUN were decreased in AR0. ROS was decreased in AR0, AR6, and AR9. SOD, catalase and GPx were decreased by steaming processes. p-AMPK was decreased but SREBP-2, p-ACC, and HMGCR were increased by steaming processes. Histologic analysis showed that AR0 decreased liver fat accumulation. Conclusion : In conclusion, AR0 can be developed as a treatment for hyperlipemia. And it is required to research on antioxidant effect changes among different times steamed Achyranthis Bidentatae Radix.

Study on Practical Prescription and It's Drug Composition for the Treatment of Diabetic Nephropathy (당뇨병성신병증(糖尿病性腎病證)의 활용 처방 및 약물에 관한 연구)

  • Based on the theories and clinical data of oriental medicine, traditional medicinal prescriptions known to be effective to diabetic nephropathy were collected. The prescriptions were carefully examined and analyzed in order to be used as fundamental material for experiments and in clinicals. More than 40 publications related to diabetes were arranged and analyzed. Of the complications, diabetic nephropathy part of the publications were specifically focused during analysis. Data were analyzed and classified according to the quantity, prescription, differentiation of symptoms, signs and addition and subtraction of each medicine. Frequently used medicines were statistically analyzed. The most frequently used prescription was Yukmigihwang-tang(六味地黃湯) based medicine and prescriptions of its addition and subtraction, which was 15% of the total prescriptions cited. The most frequently used medicine was Astragali Radix, mentioned 192 times in the prescriptions. Hoelen was next with 180 citations, followed by Dioscoreae Rhizoma with 147 times, Rehmanniae Radix with 140, Corni Fructus with 131, Salviae Miltiorrhizae Radix with 106, Angelicae Gigantis Radix with 101, Alismatis Rhizoma with 95, Rhei Rhizoma with 90, Atractylodis Rhizoma Alba with 84, Rehmanniae Radix Preparata with 78, Leonuri Herba with 74, Moutan Cortex Radicis with 66, Aconiti Lateralis Preparata Radix with 66 Cnidii Rhizoma with 65, Pseudostellaria heterophylla with 62, Liriopis Tuber with 55, Lycii Fructus with 52, Rhei Rhizoma with 49, Codonopsis Pilosulae Radix with 44, Paeoniae Radix Rubra with 44, Schisandrae Fructus with 42, Polyporus with 42, Achyranthis Radix with 41 and Euryales Semen with 40. Medicines prescribed more than 30 grams a day included Astragali Radix, Imperatae Rhizoma, Benincasae Pericarpium, Leonuri Herba, Dioscoreae Rhizoma, Plantaginis Semen, Polyporus, Hoelen, Halloysitum Rubrum, Achyranthis Radix, Arecae Pericarpium, Phaseoli Angularis Semen, Coicis Semen, Rhei Rhizoma, Salviae Miltiorrhizae Radix, Sargassum, Ostreae Concha, Pseudostellaria heterophylla, Epimedii Herba, Rehmanniae Radix, Scrophulariae Radix and Polygonati Rhizoma. No scientific reports on the traditional medicinal aspects of diabetic nephropathy was searched. This analysis report would be able to provide the basis of developing new drug candidates for the treatment of diabetic nephropathy, as well as securing the EBM for the traditional medicines already being prescribed to the patients.

Effects of Herbal medicine on Physiological Responses in Ovariectomized Rats and SAM P6 Mice (한약이 난소제거 흰쥐와 SAM P6 생쥐의 생리활성에 미치는 영향)

  • The effect of herbal medicine on osteoporosis was studied using ovariectomized rats as an animal model of Type I osteoporosis and SAM P6 mice as that of Type II. Each traditiona l boiling water extract of Achyranthis Radix, Psoraleae Radix, Rehmanniae Radix Preparat, Corni Fructus and Mycelia of Ganoderma, and systemic water fraction of Astragali Radix was given 5g(dried herbal weight)/kg/day p.o. for 30 days in each group of ovariectomized rats, SAM R1 and SAM P6. The extract of Cervi parvum Cornu was given for 14 days only. One ml of blood was taken by tail vein at day 0, 7, 14, 21 and 30 days after administration of the extract. Plasma levels of alkaline phosphatase, calcium, creatinine,inorganic phosphate, blood urea nitrogen, cortisol, total $T_3\;and\;total\;T_4$ were measured. In ovariectomized rats, administration of Achyranthis Radix or Corni Fructus decreased in alkaline phosphatase and that of Achyranthis Radix or Psoraleae Radix decreased in calcium comparing to the control (p<0.05). The administration of Psoralese Radix decreased in calcium and increased in urea comparing to day o(P<0.05)(Table I). There were not much changes in plasma calcium, inorganic phosphate, and alkaline phosphatase concentrations after uptake of these herbal medicine used in SAM P6(Table III). However, administration of Astragali Radix altered plasma inorganic phosphate and creatinine levels in SAM R1(p<0.01)(Table UU). The administration of Corni Fructus or Psoralease Radix induced the changes in plasma concentrations of cortisol, total $T_3$ and total $T_4$ in Type I(p<0.05) (Table IV). The uptake of Cervi parvum Cornu increased in total $T_3$ concentration and that of Mycelia of Ganodtragali Radix in SAM P6. However, the uptake of Mycelia of Ganoderma induced changes in cortisol and $T_4$ concentrations in SAM R1(p<0.05). Thus, there were significant differences in responses of herbal medicine in different types of osteoporosis.

Selection of the optimal herbal composition of pomegranate concentrated powder from aqueous extracts of Eucommiae Cortex and Achyranthis Radix to treat osteoarthritis in rats

  • Objectives: We investigated whether a mixture of the main component of pomegranate concentrated powder (PCP) with appropriate proportions of Eucommiae Cortex (EC) and Achyranthis Radix (AR) could act synergistically as an effective treatment for osteoarthritis (OA). Methods: In order to evaluate the effects of PCP, EC, and AR against OA, knee thicknesses, maximum extension angle of each knee, anti - inflammation effects, the transcript levels of chondrogenic genes mRNA expressions in femur and tibia articular cartilage (AC) with synovial membrane (SM) were analyzed. In addition, the histopathology and immunohistochemistry of the femur and tibia AC or SM were performed. Results: The surgically-induced OA signs in rats were significantly inhibited by 28 days of continuous treatment of PCP, EC and AR single formulas, and PCP with EC:AR mixed formulas. Especially, PCP with EC:AR 4:1, 2:1 and 1:1 mixed formula treatment constantly showed significantly more favorable inhibitory activities, as compared with those of single formula of PCP, EC and AR treated rats. Conclusion: PCP and EC:AR 4:1 mixed formula showed similar OA refinement effects through potent anti-inflammatory pathways as compared with those of diclofenac treatment, and showed additional chondrocyte proliferating effects on the both femur and tibia AC.

  • Quantitative methods for the marker compounds of Cimicifugae Rhizoma, Achyranthis Radix, Artemisiae Capillaris Herba, Moutan Cortex Radicis and Arecae Semen, respectively, were developed using HPLC-DAD. Using the established methods, each extract of the natural medicines were evaluated. In addition, long term and accelerated stability test in the extracts were examined for six months. No significant change in content of the marker compounds of each extract observed during the time of investigation.

Inhibitory Effects of Achyranthis Bidentatae Radix on Osteoclast Differentiation and Bone Resorption (우슬의 파골세포 분화 억제와 골 흡수 억제효과)

  • Objectives : Achyranthis Bidentatae Radix (ABR) has been used for treating of many symptoms especially osteoporosis and rheumatoid arthritis. In this study, we determined the effects of water extract of ABR in RANKL (Receptor Activator for Nuclear Factor $\kappa$ B Ligand)-induced osteoclast differentiation culture system. Methods : We assayed mRNA expression levels of NFATc1, c-Fos, TRAP, OSCAR, $FcR{\gamma}$, DAP12 and GAPDH in bone marrow macrophages (BMMs) treated with ABR. The protein expression levels of NFATc1, c-Fos, MAPKs and $\beta$-actin in cell lysates treated with ABR were analysed by Western blotting. In addition we determined the effects of water extract of ABR on LPS-induced bone-loss mouse. Results : Water extract of ABR showed remarkable inhibition on RANKL-treated osteoclast differentiation without cytotoxicity. ABR down-regulated the induction of c-Fos and NFATc1 by RANKL. ABR suppressed phosphorylation of JNK, p38 and I-${\kappa}B$. ABR rescued bone erosion by LPS induction in vivo study. Conclusions : These results demonstrate that ABR may be a useful remedy for curing of bone-loss disease such as osteoporosis.

Effect of ethanol extract from Achyranthis Radix on hair growth (우슬의 에탄올 추출물이 모발 성장에 미치는 영향)

  • Objective : As more and more people are interested in appearance in modern society, the increasing number of hair loss population can have an important impact on psychological and social problems such as depression and inappropriate interpersonal symptoms. Therefore, much research is being done on treatments for alopecia using herbal extracts with relatively few side effects. This study was investigated about the effect of Achyranthis Radix (AR) extract with ethanol solvent on hair growth. Methods : We determined the promoting efficacy of AR-ethanol extract compared with minoxidil (MNXD) on the growth of human hair dermal papilla cells (HDPCs). Cell viability was measured by MTT assay and cell proliferation was confirmed by cell cycle analysis from flow cytometry in HDPCs. Also, we monitored the safe concentration range through MTT assay. And protein expression of hair growth-related genes (insulin-like growth factor 1 (IGF-1), Wnt3a, Protein kinase B (Akt), Extracellular signal-regulated kinase (Erk)) was monitored by western blot. Results : On cell cycle analysis, the G2/M phase was higher than that of the DW group in AR ethanol extract group at 0.05 and 0.1 mg/㎖. All protein expression levels of HDPCs were increased in AR ethanol extract groups and the MNXD group, compared to the DW group, respectively. Conclusion : As mentioned above, AR extract increased cell proliferation and the protein expression of IGF-1, Wnt3a, Akt, Erk in HDPCs. These results suggest that AR ethanol extract has promoted hair growth and it might be potential hair growth supplement.
