본 연구에서는 대표적인 퇴행성 신경질환인 알츠하이머성 치매에 대한 마늘 물, 100% 메탄올, 디클로로메탄 추출물들의 acetylcholinesterase (AChE) 저해 및 신경세포 보호효과를 조사하였다. 마늘 디클로로메탄 추출물은 농도 의존적으로 AChE를 저해하는 것으로 나타났으며, $IC_{50}$은 $36.1{\mu}g/mL$로 나타났다. MTT reduction assay를 이용해 amyloid ${\beta}$ protein ($A{\beta}$) 유도성 신경세포 독성에 대한 신경세포 보호효과를 측정한 결과, 세 가지 마늘 추출물들은 대부분 40% 미만의 세포생존율을 보였고 이 결과는 $A{\beta}$ 유도성 신경세포 독성보다 상대적으로 더 높은 세포독성을 보여주었다. LDH assay에서는 마늘 물 추출물이 37%의 LDH 방출량을 나타내 $200{\mu}M$의 vitamin C과 유사한 세포막손상 보호효과를 보였다. 마지막으로 neutral red uptake assay를 실시한 결과, MTT reduction assay와 마찬가지로 모든 마늘 추출물들에서 세포생존율의 감소를 확인하였으며 특히 디클로로메탄 추출물의 경우 현저하게 낮은 세포생존율을 나타내었다. AChE 저해활성을 갖는 마늘 디클로로메탄 추출물로부터 얻은 column fractionations에 함유된 생리활성물질을 탐색하기 위해 HPLC 분석을 실시하였으며, 마늘 98:2 fraction의 LC-MS 분석을 통하여 allyl methyl disulfide, diallyl monosulfide, diallyl disulfide로 추정되는 물질군이 확인되었다.
본 연구에서는 눈개승마(Aruncus dioicus var. kamtschaticus)의 in vitro 산화방지활성 및 고당(intensive glucose)과 과산화수소($H_2O_2$)로 야기된 산화적 스트레스로부터의 뇌신경세포 손상에 대한 보호효과와 더불어 알파글루코사이드가수분해효소 및 아세틸콜린에스터가수분해효소 억제효과를 확인하였으며 또한 HPLC를 이용하여 주요 물질을 분석하였다. 눈개승마 아세트산에틸 분획물(ethylacetate fraction from Aruncus dioicus var. kamtschaticus: EFAD)은 매우 우수한 총 페놀화합물 함량(430.08 mg GAE/g)과 총 플라보노이드 함량(511.72 mg RE/g)을 나타냈으며, 높은 ABTS 라디칼 소거 활성과 과산화지방질생성물의 억제력을 확인하였다. 또한 고당(intensive glucose)과 과산화수소($H_2O_2$)로 야기된 뇌신경세포에서의 산화적 스트레스 및 이로 인한 뇌신경세포 사멸을 측정한 결과, 눈개승마 아세트산에틸 분획물(EFAD)의 유의적인 뇌신경세포 보호효과를 확인할 수 있었다. 추가적으로 다당류 분해 효소인 알파글루코사이드가수분해효소 억제효과 및 아세틸콜린 분해 효소인 아세틸콜린에스터가수분해효소 억제효과를 살펴봄으로써 혈당 상승 억제효과와 뇌신경전달물질의 세포 내 유지 효과를 확인하였다. 마지막으로 눈개승마 아세트산에틸 분획물(EFAD)의 주요 페놀성 물질을 확인하기 위해 HPLC분석을 한 결과 카페산으로 추정되었다. 본 연구 결과들을 고려할 때, 눈개승마는 고혈당 지연 또는 개선을 통한 고혈당 예방 소재로서의 가능성뿐만 아니라 이로 인해 야기되는 산화적 스트레스로부터 뇌신경 세포를 보호함으로써 고혈당에 의한 대사성 뇌신경질환 예방 소재로도 활용 가능성이 있을 것이라 판단된다.

Method : The effects of the JWN extract on (1) $IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ mRNA of PC-12 cells treated with LPS; (2) amyloid precursor proteins(APP), acetylcholinesterase(AChE), and glial fibrillary acidic protein(GFAP) mRNA, the AChE activity and the APP production of PC-12 cell treated with CT-105; (3) the behavior; (4) expression of $IL-1{\beta}$, $TNF-{\alpha}$, MDA, $IL-1{\beta}$ mRNA, and $TNF-{\alpha}$ mRNA, (5) the infarction area of the hippocampus, and brain tissue injury in Alzheimer's diseased mice induced with ${\beta}A$ were investigated. Result : 1. The JWN extract suppressed the expression of $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ mRNA in THP-1 cells treated with LPS. 2. The JWN extract suppressed the expression of APP, AChE, and GFAP mRNA in PC-12 cells treated with CT-105. 3. The JWN extract suppressed the AChE activity, and the production of APP significantly in PC-12 cells treated with CT-105. 4. For the JWN extract group a significant inhibitory effect on the memory deficit was shown for the mice with Alzheimer's disease induced by ${\beta}A$ in the Morris water maze experiment, which measured stop-through latency, and distance movement-through latency. 5. The JWN extract suppressed the over-expression of $IL-1{\beta}$ protein, $TNF-{\alpha}$ protein, MDA, $IL-1{\beta}$ mRNA, $TNF-{\alpha}$ mRNA, and CD68/GFAP, in the mice with Alzheimer's disease induced by ${\beta}A$. 6. The JWN extract reduced the infarction area of hippocampus, and controlled the injury of brain tissue in the mice with Alzheimer's disease induced by ${\beta}A$. Specifically, the effects of the HSS extract on (1) $IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ mRNA of PC-12 cells treated with LPS; (2) amyloid precursor proteins(APP), acetylcholinesterase(AChE), and glial fibrillary acidic protein(GFAP) mRNA of PC-12 cells treated with CT-105; (3) the AChE activity and the APP production of PC-12 cell treated with CT-105; (4) the behavior; (4) expression of $IL-1{\beta}$, $TNF-{\alpha}$, $IL-1{\beta}$ mRNA, $TNF-{\alpha}$ mRNA, and reactive oxygen species(ROS); (5) the infarction area of the hippocampus, and brain tissue injury in Alzheimer's diseased mice induced with ${\beta}A$ were investigated. The results were summarized below ; 1. The HSS extract suppressed the expression of $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ mRNA in THP-l cells treated with LPS. 2. The HSS extract suppressed the expression of APP, AChE, and GFAP mRNA in PC-12 cells treated with CT-105. 3. The HSS extract suppressed the AChE activity, and the production of APP significantly in PC-12 cells treated with CT-105. 4. 본 연구에서는 다래 순 아세트산에틸 분획물(ethyl acetate fraction from Actiniadin arguta sprout; EFAS)의 in vitro 산화방지활성 및 고포도당으로 인한 산화적 스트레스로부터의 뇌신경세포 보호 효과를 검증하였다. EFAS은 상대적으로 우수한 총 폴리페놀(246.25 mg GAE/g) 및 총 플라보노이드(571.59 mg RE/g) 함량을 나타내었으며, 이로 인한 ABTS 라디칼 제거 활성($IC_{50}=252.00{\mu}g/mL$) 및 MDA 생성 억제 활성($IC_{50}=42.70{\mu}g/mL$)을 보여주었다. EFAS의 in vitro 항당뇨 활성은 알파글루코사이드 가수분해효소와 최종당화산물 생성 억제 활성을 통해 측정하였으며 각각 99.18과 $85.19{\mu}g/mL$의 $IC_{50}$ 값을 나타냈고, 뇌신경 전달물질인 아세틸콜린의 세포 내 유지 효과를 아세틸콜린 분해효소 억제 활성을 통해 측정하였을 때 $164.20{\mu}g/mL$의 $IC_{50}$ 값을 나타냈다. 또한 인간 유래 MC-IXC 뇌신경세포에 과산화수소와 고포도당을 처리하여 산화적 스트레스를 유발시켜 뇌신경세포 사멸에 대해 우수한 보호효과를 보여주었다. 마지막으로 HPLC를 통하여, EFAS의 주요 생리활성 물질을 분석한 결과는 루틴으로 유추되었으며, 그 함량은 $123.76{\mu}g/mL$ of dried weight로 분석되었다. 이상의 결과로부터 다래 순 분획물(EFAS)이 알파글루코사이드 가수분해효소 및 최종당화산물 생성 억제를 통해 식후 혈당의 급격한 상승을 예방할 수 있는 항당뇨 소재로의 가능성뿐만 아니라, 고포도당에서 유래되는 산화적 스트레스로부터 뇌신경세포 보호 및 뇌신경전달물질 체계 개선을 통한 당뇨성 퇴행성뇌질환 등의 합병증을 예방할 수 있는 고부가가치 식품 소재로의 활용 가능성을 확인하였다.
Amyloid ${\beta}-peptide$에 의해 유도되는 세포사멸을 보호하는 물질을 검색하기 위하여 250여 식물 재료 및 식품성분으로부터 스크리닝한 결과 가장 효과가 있는 시금치 추출물을 이용하여 뇌신경세포사멸(neuronal cell death)을 어느 정도 보호할 수 있는지를 알아보았다. 시금치 추출물이 항산화 활성과 acetylcholinesterase 활성에 대한 저해효과는 시금치 추출물 처리농도가 높을수록 유의적으로 높게 나타났다. 과산화수소와 amyloid ${\beta}-peptide$에 의해 유도된 SH-SY5Y 세포주의 세포사멸에 대한 시금치추출액의 억제효과를 살펴본 결과, 과산화수소에 의한 세포사멸에 대하여 시금치 추출물은 억제효과를 나타내었으나, amyloid ${\beta}-peptide$의 경우는 세포사멸억제효과를 나타내지 않았다.

① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는

- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
