  • A mixture of 20(R)- and 20(S)-ginsenoside Rg$_{3}$ was obtained under mild acidic hydrolysis from protopanaxadiol saponins, ginsenosides Rb$_{1}$, Rb$_{2}$, Rc and Rd. The product was acetylated to give the peracetates, which were further converted into 20(R)-ginsenoside Rg$_{3}$, 20(S)-ginsenoside Rg$_{3}$, 20(R)-ginsenoside Rh$_{2}$ and 20(S)-ginsenoside Rh$_{2}$ by the direct alkaline treatment depending upon two kinds of temperature conditions respectively. The structure and physicochemical properties of a prosapogenin, 20(R)-ginsenoside Rh$_{2}$, were investigated.

  • Four new acetylated ginsenosides were isolated from the processed ginseng (SG, sun ginseng). Their structures were determined to be $3{\beta},{\;}12{\beta}-dihydroxydammar-20(22),24-diene-3-O-{\beta}-D-glucopyranosyl(1{\rightarrow}2)-{\beta}-D-6"-O-acetylglucopyranoside;{\;}3{\beta},12{\beta}-dihydroxydammar-20(21),{\;}24-diene-3-O-{\beta}-D-glucopyranosyl(1{\rightarrow}2)-{\beta}-D-6"-O-acetylglucopyranoside;{\;}3{\beta},{\;}6{\alpha},12{\beta}-trihydroxydammar-20(22),24-diene-6-O-{\beta}-D-6'-O-acetylglucopyranoside{\;}and{\;}3{\beta},6{\alpha},12{\beta}-trihydroxydammar-20(21),24-diene-6-O-{\beta}-D-6'-O-acetylglucopyranoside$ based on spectroscopic evidences. The compounds were named ginsenoside $Rs_4,{\;}Rs_5,{\;}RS_6{\;}and{\;}Rs_7$, respectively.pectively.

  • We reported a new processed ginseng with increased biological activities which is named as “sun ginseng (SG)”. Study on the saponin constituents of SG led to the isolation of seven new ginsenosides named as ginsenoside Rk$_1$, Rk$_2$, Rk$_3$, Rs$_4$, Rs$\_$5/, Rs$\_$6/ and Rs$\_$7/. Ginsenoside Rk$_1$, Rk$_2$ and Rk$_3$ were the Δ$\^$20(21),24(25)/-diene dammarane compounds, while ginsenoside Rs$_4$, Rs$\_$5/, Rs$\_$6/ and Rs$\_$7/ were mono-acetylated compounds. Many other ginsenosides which were reported as minor constituents of red ginseng were also isolated, which include 20(S)-Rg$_3$, 20(R)-Rg$_3$, Rg$\_$5/, Rg$\_$6/, F$_4$, Rh$_4$, 20(S)-Rs$_3$ and 20(R)-Rs$_3$. The major ginsenosides of SG were 20(S)-Rg$_3$, 20(R)-Rg$_3$, Rk$_1$ and Rg$\_$5/.

  • Endothelial progenitor cells (EPC) are a population of cells that circulate in the blood stream. They play a role in angiogenesis and, therefore, can be prognostic markers of vascular repair. Ginsenoside $Rg_3$ prevents endothelial cell apoptosis through the inhibition of the mitochondrial caspase pathway. It also affects estrogen activity, which reduces EPC senescence. Sun ginseng (SG), which is heat-processed ginseng, has a high content of ginsenosides. The purpose of this study was to investigate the protective effects of SG on senescence-associated apoptosis in EPCs. In order to isolate EPCs, mononuclear cells of human blood buffy coats were cultured and characterized by their uptake of acetylated low-density lipoprotein (acLDL) and their binding of Ulex europaeus agglutinin I (ulex-lectin). Flow cytometry with annexin-V staining was performed in order to assess early and late apoptosis. Senescence was determined by ${\beta}$-galactosidase (${\beta}$-gal) staining. Staining with 4'-6-Diamidino-2-phenylindole verified that most adherent cells (93${\pm}$2.7%) were acLDL-positive and ulex-lectin-positive. The percentage of ${\beta}$-gal-positive EPCs was decreased from 93.8${\pm}$2.0% to 62.5${\pm}$3.6% by SG treatment. A fluorescence-activated cell sorter (FACS) analysis showed that 4.9% of EPCs were late apoptotic in controls. Sun ginseng decreased the apoptotic cell population by 39% in the late stage of apoptosis from control baseline levels. In conclusion, these results show antisenescent and antiapoptotic effects of SG in human-derived EPCs, indicating that SG can enhance EPC-mediated repair mechanisms.
