Wear Corrosion Behaviour of Nitrogen Ion Implanted Super Stainless Steel (질소이온주입된 초내식성 스테인리스강의 마모부식 특성)

  • The wear corrosion behaviour of a nitrogen ion implanted super stainless steel (S.S.S, 22Cr - 20Ni - 6Mo - 0.25N) was compared with those of S.S.S, 316L SS and TiN coated 316L SS. The Cr and Ni amounts won out from the materials were investigated using an electrothermal atomic absorption spectrometry. We observed that the Cr dissolution rate of the S.S.S was similar to that of 316L SS, however, the Ni release of the S.S.S was feater than 316L SS. The metal ions released from the nitrogen ion implanted S.S.S surface were significantly reduced. The wear corrosion behaviour of the stainless steels was not correlated with the results shown by a static metal ion release test.

Geochemical Characteristics of Mine Wastes in Abandoned Mines in Korea (휴/폐광 광산폐기물의 지구화학적 특성)

  • The objective of this study is to investigate geochemical characteristics of mine wastes including tailings in various abandoned mines in Korea. Tailings and wastes were sampled in and around 39 metalliferous mines, and analysed heavy metal concentrations including Cd, Cu, Pb and Zn extracted by 0.1N HCl and aqua regia by Atomic Absorption Spectrometry. Measurements of paste pH and lime requirement were carried out to examine a general geochemical characteristics of the materials. Lots of mine wastes were characterized by very low pH values of 2 to 4 and high lime requirement to control neutralization of the wastes. In addition, elevated levels of heavy metals were found in various mine wastes extracted by both 0.lN HCl and aqua regia. Because the mine wastes can be dispersed into the downstream by clastic movement and wind-blow, it is necessary to control the materials with a proper method for their reclamation.

  • A method has been described for the determination of Cu(Ⅱ), Pb(Ⅱ), Ni(Ⅱ), Cd(Ⅱ), Mn(Ⅱ) and Fe(Ⅲ) by flame atomic absorption spectrometry (FAAS) after preconcentration on Amberlite XAD-16 resin, using hexamethyleneammonium-hexamethylenedithiocarbamate (HMA-HMDTC) as a chelating agent, and NH3/NH4Cl buffer solution (pH 9). Influences of various analytical parameters such as pH, concentration of nitric acid, amount of analytes, diverse ions and sample volume were investigated. The relative standard deviation (RSD) and the detection limit (LOD) were found in the range of 0.8-2.9% and 0.006-0.277 ㎍/mL,respectively. Recoveries obtained by the column method were quantitative ( >95%) at optimum conditions.The method was applied to the determination of some metal ions in seawater and wastewater samples. A high preconcentration factor (about 150 for seawater and 75 for wastewater samples) and simplicity are the main advantages of this suggested method.

High Throughput Proteomic Approaches for the Dissection of Light Signal Transduction Pathways in Photosynthetic Cyanobacterium Synechocystis sp.PCC 6803

  • Light is an environmental signal that regulates photomovement and main energy source of photosynthesis in the cyanobacterium Synechocystis sp. PCC 6803 (Syn6803). Syn6803 is a popular model system for study of plant functional genomics. In this report, we adopted 2D gel based proteomics study to investigate proteins related with the light absorption and photo-protection in Syn6803. More than 700 proteins were detected on the SDS-gels stained with silver nitrate. Several proteins showing different expression level under various light conditions were identified with MALDI-TOF Mass spectrometry. As a comparison, we also conducted ICAT-based proteome study using WT and cphl (cyanobacterial phytochrome 1) mutant. A cphl deletion led to changes in the expression of proteins involved in translation, photosynthesis including photosystem and CO2 fixation, and cellular regulation. We are currently involved in TAP-tagging method to study protein-protein interactions in search for the molecular component involved in the light signal transduction of Syn6803 photomovement.

  • Ferritin is an iron storage protein found in most living organisms. For expression and industrial use, human light chain ferritin (L-ferritin) was cloned from human liver cDNA library and expressed in Pichia pastoris strain GS115. The recombinant L-ferritin in Pichia pastoris was glycosylated. In a fed-batch culture, the cell mass reached about 57 g/l of dry cell weight, and the L-ferritin in the cell was increased to about 95 mg/l after 150 h. In an atomic absorption spectrometry analysis, the intracellular content of iron in the L-ferritin transformant was measured as $1,694{\pm}85\;\mu\textrm{g}g/g$, which is 5.4-fold more than that of the control strain. This L-ferritin transformant could serve as iron-fortified nutrients in animal feed stock.

  • A method has been investigated for the determination of selenium and tellurium in Ginseng, Ganoderma and Garlic using hydride generation atomic absorption spectrometry(HG-AAS). The concentration effects of hydrochloric acid and sodium tetrahydroborate on the hydride generation for the determination of selenium and tellurium were investigated. The method of sample decomposition was also investigated using various mineral acids, such as nitric, perchloric and sulfuric acid in the closed system, and foreign ion effects containing in the samples were studied. The calibration curves of selenium and tellurium were obtained in the range of 0~40 ppb. The detection limits(S/N=2) of selenium and tellurium are 0.1 and 0.2 ppb. Analytical data of selenium and tellurium in Garlic, Ganoderma and Ginseng are 289, 296 and 198 ng/g for selenium and 146, 127 and 110 ng/g for tellurium, respectively.

Studies on the Germanium Components of Zingiberis Rhizoma (생강중 게르마늄 성분에 관한 연구)

  • Germanium (Ge) content in Zingiberis Rhizoma was determined by flameless atomic absorption spectrometry with a graphite tube atomizer. The Ge content of the cortical layer was 65.mu.g/g, the central cylinder, $98\mu$g/g and the total, $87\mu$g/g. In the effect of the extraction of Ge by various solvents, polar solvent was more effective than the nonpolar one, Especially, water was the most effective solvent. The water extract of Zingiberis Rhizoma, separated by sephadex G-25 gel filtration, was fractionated into 2 peaks at 254nm. Both the peaks showed atomic absorptions of Ge. It may be concluded that the Ge components of Zingiberis Rbizoma exist as the form of an aqueous organic compounds or associated forms with proteins.

  • In order to investigate harmful trace in commercial teaproducts. The contents of copper, lead, cadmium, zinc, chromium, manganese and iron were studied in this paper. The samples studied were Youlmoo tea(7) Arrow root tea(9) Ginger tea(4) collected from markets in Seoul area and analysis of seven metals by means of Automic absorption spectrometry. The results obtained were as follows : Lead contents is ranged from 0.06 to 0.72 ppm, cadmium from ND to 0.17 ppm, copper from 0.8 to 16.7 ppm, zinc from 10.4 to 40.4 ppm, manganess from 2.7 to 16.5 ppm, chromium from 0.04 to 4.7 ppm, iron from 7.9 to 55.7 ppm.

  • A simple and inexpensive colorimetric spectrometer for determining total cholesterol has been developed,comprising a diode laser as the light sourcoptical fibers for the light guide and a photodiode as the detector.The stabilty and performance of the new system was investigated by obtaining the calibration curve for stan-dard cholesterol solutions. The total cholesterol in humanserum was also measured by the analyzer and com-pared with the value obtained by a conventional spectrometer. The results showed that the developed spectrometer was useful for the determination of cholesterol levels. The visible diodelaser used in the study exhibited good spectroscopic and operational properties for colorimetric absorption spectrometry and could be a key component for the development of a simple and economical analyzer.

Identification of Natural dyes used in 16th pink Dallryeong (Official's robe in Joseon Dynasty) Excavated from Cheonan, Chungnam (충남 천안시 출토 16세기 분홍 단령에 사용된 염재 동정)

  • Aim of this study is to identify dyestuff of the Dallryeong(official's robe in Joseon Dynasty, 16th century) excavated from Yuryang-dong, Cheonan, in 1996. For this purpose, extracted dyestuffs from Dallryeong fabric and from natural dyestuffs for red color(safflower, Sapanwood, Madder) which are presumed to have been used in the Dallryeong, are analyzed and compared by high performance liquid chromatography(HPLC). As a result, HPLC chromatogram of extracts of the Dallryeong's dyestuff and safflower are showed a peak at 17.5 minutes. The UV/Vis spectra of the samples are showed the maximum absorption wavelength at 519nm. This result is identical with the analysis of the previous studies on red dyestuff of safflower. In addition, the analysis of Mass Spectrometry(MS) showed the identical result of the peak with m/z 910. Following these results, excavated pink Dallryeong were considered to have been dyed with safflower.
