Properties of the High and Low Molecule of the Proteoglycan Extracted from Ganoderma lucidum IY009 (Ganoderma lucidum IY009 배양균사체 유래 단백다당류의 저분자와 고분자 분획의 특성)

  • To examine the structural properties of the proteoglycan (GMPG, Ganoderma lucidum mycelial proteoglycan) obtained from mycelia in Ganoderma lucidum IY009, we obtained the low and high molecular proteoglycan by ultrafiltration and sepharose CL-4B column chromatography. The physicochemical properties of these fractions were as follows. When the proteoglycan separated by ultrafiltration and sepharose CL-4B column chromatography, its was not fractionated completely. The molecular weight of high molecular proteoglycan by the gel column chromatography (CH) was 250 kD and 2,000 kD, and low molecular proteoglycan was 12kD. The total carbohydrate was consisted of 75.7% (UH) and 96.7% (CH), and the low fraction was 72.7% (UL) and 87.1% (CL), respectively. The sugar of high and low molecular proteoglycan composed of glucose, mannose, fructose, galactose, xylose, ribose and arabinose. Glucose contents of all fraction were ranged from $46.9%{\sim}82.4%$ of the total sugar and the ratio of ${\alpha}$\;and\;{\beta}-glucose$ was $0.84{\sim}1.14$, and its indicated the proteoglycan to be ${\beta}-glucan$. Amino acids pattern showed that the fractions contained a large amount of aspartie acid, glutamic acid, alanine and leucine. These fractions showed the characteristics of IR absorption for ${\beta}-glucan$ at $890\;cm^{-1}\;and\;^{13}C-NMR$ spectroscopy showed the presence of the ${\beta}-1,3-glucan$ and a ${\beta}-1,6-glucan$.

Developmemt of Rice Husk Pellets as Bio-filter Media of Bio Scrubber Odor Removal System (왕겨펠렛 생물담체 개발 및 이를 이용한 bio scrubber형 악취제거 시스템 성능평가)

  • The rice husk contains nutrients which can be easily utilized by microorganisms, and also has a water retaining ability, which played a crucial part in enabling it to become a biofilter media. In this study, we evaluated the applicability of rice husk pellet bio-scrubber as a microbiological carrier. The pelletization experiment of rice husk as a biological media was performed using PVA and EVA binder. Also, the feasibility tests of rice husk as a biological media for odor removal were carried out in order to know whether rice-husk contains useful components as a media for microbiological growth or not. Lastly, a combined test for odor gas absorption and biological oxidation was conducted using a lab scale bio-filter set-up packed with rice-husk pellets as wet-scrubber. The major components of the rice husk were carbon, hydrogen, nitrogen, and oxygen, while carbon acted as the main ingredient which comprised up to 23.00%. The C : N : P ratio was calculated as 45 : 1 : 2. Oxygen uptake rate, yield and decay rate of the rice husk eluent was calculated to be $0.0049mgO_2/L/sec$, 0.24 mgSS/mgCOD and 0.004 respectively. The most stable form of rice husk pellets was produced when the weight of the rice husk, EVAc, PVAc, and distilled water was 10 : 2 : 0.2 : 10. The prepared rice husk pellets had an apparent density of 368 g/L and a porosity of 59.00% upon filling. Dry rice husks showed high adsorption capacity for ammonia gas but low adsorption capacity for hydrogen sulfide. The bio-filter odor removal column filled with rice husk pellets showed more than 99.50% removal efficiency for NH3 and H2S gas. Through the analysis of circulation water, the prime removal mechanism is assumed to be the dissolution by water, microbial nitrification, and sulfation. Finally, it was confirmed that the microorganisms could survive well on the rice husk pellets, which provided them a stable supply of nutrients for their activity in this long-term experiment. This adequate supply of nutrients from the rice husk enabled high removal efficiency by the microorganisms.

Adsorption of an uranyl ion onto a divinylbenzene amidoxime resin in sodium carbonate solutions (탄산염 용액에서 아미드옥심 수지에 대한 우리닐 이온의 흡착거동)

  • Distribution coefficients (Kd) of uranyl ion onto divinylbenzene amidoxime resins were measured in sodium carbonate solution and the Kd values were increased up to about 70 as the resin bead size was decreased. At a condition of 0.0044 M $Na_2CO_3$, the adsorption capacity for uranium was $3.4{\mu}mole$ U/g-resin. The Kd values in the 0.5 M $Na_2CO_3-NaHCO_3$ solution, ranging from pH 9 to pH 11, revealed that they were increased as the pH increased and revealed lower values than those in the pure sodium carbonate solution. The amidoxime resins were characterized by FTIR-ATR showing the absorption bands of the amidoxime functional groups. A species of the uranyltricarbonate complex, $UO_2(CO_3)_3^{-4}$, was confirmed by UV-Vis spectroscopy, revealing four absorption peaks between 400 and 500 nm. Uranium was separated from some fission products by a column operation. However, most of the uranium and fission products were eluted before an adsorption and only a small amount of uranium was adsorbed onto the resin due to the low capacity of the resin.

  • The object of this study was to characterize Bacillus strains GB-017 and GB-0356, which produce antifungal substances, especially for plant pathogens. In addition, this study was undertaken to characterize the culture conditions required for the production of antifungal substances and to document some of the properties of the antifungal substance produced by these soil-isolated strains. Strains GB-0365 and GB-017 were found to be bacillus-shaped, gram-positive and motile, and to inhibit Botrytis cineria, Fusarium sp., Pythium sp., and Rhizoctonia solani. Antagonistic activity was maintained up to pH 9.0, and the antifungal activity was stable to heat at 80$^{\circ}C$ for 1 h. Antifungal substances were separated and purified using ion exchange and adsorption columns including WK-I0(H$\^$+) (pH 7.0), HP20 column (pH 3.0) and IPA (pH 3.0). and IPA. Its UV absorption spectrum showed major peaks at 231 and 259 nm, corresponding to polyene and lactone. A fast atom bombardment mass spectrum (FAB MS) showed a highest peak at 441 m/z and major peaks at 192, 205, and 370 m/z.

Studies on Ion-exchange Chromatography of Elements in Special Nonferrous Alloys (비철특수합금에서 금속원소의 이온교환 크로마토그라프에 관한 연구)

  • The purpose of this study was to develop a separation method of Zn(II), Cu(II) and Mg(II), by ion exchange chromatography using cation exchange resion (Dowex 50w${\times}$8, 80-100 mesh) and anion exchange (Amberlite IRA-400). Ion exchange resions were packed into 25 ${\times}$ 2cm ID column and flow rate was controlled to 0.30 ml/min. Good eluents for separation of nonferrous metal ions such as Zn(II), Cu(II), Mg(II) were as follow: 0.5M $NaNO_3$ (pH 3.1), 0.2~0.5M HCl + 50~60% Acetone, and 1M HAc + 0.1M NaAcf(pH 3.7) aqueous solution. The mixed solution of 0.1M NaAc(pH 3.7), 0.5M HCl + 50% Acetone were found to be the best eluent for step elution. Analysis of metals were determined by atomic absorption spectrophotometer. In addition, separated Zn(II) fraction was obtained by eluted with 0.12N HCl and 1.5N $NH_4OH$ aqueous solution. This solution was titrated by the E. D. T. A.

  • In order to remove $CO_2$ from the DME plant process, we investigated the composite membrane with rubbery polymers as the separation layer and its separation performance of $CO_2$ and $H_2$. Hollow fiber membranes for supporting layer were prepared by solution spinning method. In case of using PDMS as a separation layer, the composite membranes showed the permeation rates of $CO_2$ were over 300 GPU and minimum $CO_2/H_2$ selectivitties were 4.3 and in case of using PEBAX as a separation layer, the composite membranes showed the permeation rates of $CO_2$ were over 120 GPU and minimum $CO_2/H_2$ selectivities were 5.

Analyses of the OMI Cloud Retrieval Data and Evaluation of Its Impact on Ozone Retrieval (OMI 구름 측정 자료들의 비교 분석과 그에 따른 오존 측정에 미치는 영향 평가)

  • The presences of clouds significantly influence the accuracy of ozone retrievals from satellite measurements. This study focuses on the influence of clouds on Ozone Monitoring instrument (OMI) ozone profile retrieval based on an optimal estimation. There are two operational OMI cloud products; OMCLDO2, based on absorption in $O_2-O_2$ at 477 nm, and OMCLDRR, based on filling in Fraunhofer lines by rotational Raman scattering (RRS) at 350 nm. Firstly, we characterize differences between $O_2-O_2$ and RRS effective cloud pressures using MODIS cloud optical thickness (COT), and then compare ozone profile retrievals with different cloud input data. $O_2-O_2$ cloud pressures are significantly smaller than RRS by ~200 hPa in thin clouds, which corresponds to either low COT or cloud fraction (CF). On the other hand, the effect of Optical centroid pressure (OCP) on ozone retrievals becomes significant at high CF. Tropospheric ozone retrievals could differ by up to ${\pm}10$ DU with the different cloud inputs. The layer column ozone below 300 hPa shows the cloud-induced ozone retrieval error of more than 20%. Finally, OMI total ozone is validated with respect to Brewer ground-based total ozone. A better agreement is observed when $O_2-O_2$ cloud data are used in OMI ozone profile retrieval algorithm. This is distinctly observed at low OCP and high CF.

Studies on the Absorption and Accumulation of Fenitrothion in Rice Plants (수도에 살포한 Fenitrothion의 부위별 잔류에 관한 연구)

  • The experiment It·as conducted to find out the distribution of fenitrothion in rice plants at the preheading stage and its residue in harvested grains. The results obtained were as follows; 1. Distribution of fenitrothion in rice plants were measured 15.32 ppm attached to the leaf surface immediately after spray and 6.73 ppm was penetrated into tissues after 24 hours. 2. Residual amounts of fenitrothion in rice grains was 0.04 ppm at chaff and 0.01 ppm at unpolished rice, respectively.

Catalytic Technologies for Nitric Acid Plants N2O Emissions Control: In-Duct-Dependent Technological Options (질산제조 플랜트 N2O 제거용 촉매기술: 적용위치별 기술옵션)

  • A unit emission reduction of nitrous oxide ($N_2O$) from anthropogenic sources is equivalent to a 310-unit $CO_2$ emission reduction because the $N_2O$ has the global warming potential (GWP) of 310. This greatly promoted very active development and commercialization of catalysts to control $N_2O$ emissions from large-scale stationary sources, representatively nitric acid production plants, and numerous catalytic systems have been proposed for the $N_2O$ reduction to date and here designated to Options A to C with respect to in-duct-application scenarios. Whether or not these Options are suitable for $N_2O$ emissions control in nitric acid industries is primarily determined by positions of them being operated in nitric acid plants, which is mainly due to the difference in gas temperatures, compositions and pressures. The Option A being installed in the $NH_3$ oxidation reactor requires catalysts that have very strong thermal stability and high selectivity, while the Option B technologies are operated between the $NO_2$ absorption column and the gas expander and catalysts with medium thermal stability, good water tolerance and strong hydrothermal stability are applicable for this option. Catalysts for the Option C, that is positioned after the gas expander thereby having the lowest gas temperatures and pressure, should possess high de$N_2O$ performance and excellent water tolerance under such conditions. Consequently, each de$N_2O$ technology has different opportunities in nitric acid production plants and the best solution needs to be chosen considering the process requirements.

Separation and Characteristics of Antioxidative Substances in Fermented Soybean Sauce (양조간장으로부터 항산화성 물질의 분리 및 그 특성)

  • Fermented soybean sauce was fractionated by gel filteration chromatography and the physico-chemical properties of each fraction were investigated for the study on the separation and characteristics of antioxidative substances in the soybean sauce. The brown coloured melanoidin fractions(MF-I) collected by Sephadex G-10 column were found to contain nitrogeneous compounds and to have strong antioxidative activity and reducing power. The freeze dried MF-I was further fractionated with Sephadex G-50 and G-100 successively and then MF-II and MF-III fractions were observed to be the most effective antioxidant and reducing power among the fractions. The UV-VIS and IR absorption spectra of the each fraction were also determined and discussed.
