1-MCP Improves Display Life in Begonia × hiemalis 'Blitz' and 'Carnival'

  • We investigated the effect of 1-methylcyclopropene (1-MCP) on ethylene production induced by simulated transport stress in $Begonia$ ${\times}$ $hiemalis$ 'Blitz' and 'Carnival' to improve the display life in potted plants. The simulated transportation conditions were imposed for 4 days in simulated export containers with darkness, vibration with continuous shaking ($150{\pm}20$ rpm) on a rotary lab shaker, and low temperature ($12^{\circ}C$). Plants were treated with 1-MCP at three concentrations (5, 25, or 125 $nL{\cdot}L^{-1}$) and for three different periods (0, 6, or 12 hours) before undergoing the simulated transport stress treatments. Treatment with 25 or 125 $nL{\cdot}L^{-1}$ 1-MCP inhibited the abscission of open flowers by more than 40% as compared to the untreated plants. One week after the treatments, the ethylene production decreased in the plants treated with 125 $nL{\cdot}L^{-1}$ 1-MCP for 'Blitz' and 25 $nL{\cdot}L^{-1}$ for 'Carnival'. Ethylene production was correlated with concentration and duration of 1-MCP treatment in 'Blitz', but not in 'Carnival'. To reduce flower abscission and ethylene production, thus improve the display life when plants are exposed to transportation stress, we recommend pre-treatment with 1-MCP before packaging, using concentrations and durations specific to each cultivar, 125 $nL{\cdot}L^{-1}$ for 6 h and 25 $nL{\cdot}L^{-1}$ for 12 hours for 'Blitz' and 'Carnival', respectively.

Comparison of 'Kyoho' and 'Campbell Early' Table Grape Fruit Quality in Wholesale Market (시장 출하 '거봉' 및 '캠벨얼리' 포도의 등급과 품질 조사)

  • A significant difference in table grape quality was found between harvest seasons, producers and cultivars. In general, 'Kyoho' grapes showed much greater difference in fruit quality than 'Campbell Early'. The ratio of 'Campbell Early' grapes with poor quality (below quality standard within grades), was higher in fruit harvested early in the season, mainly because of immature fruit harvest. In 'Kyoho', poor quality of fruit seemed to be derived from the deviation of cultural practice between producers. Major factors responsible for poor quality in both cultivars includes harvest of unripe cluster, poor sorting and grading, berry abscission, and poor coloration. It is recommended to introduce a new quality standards considering the market condition in 'Kyoho' or a fresh-cut technology of grape berries for niche market.

On Utilization of Canavanine and Activity of Canavanase during Germination and Growth of Canavailia lineata (L.) DC (해녀콩의 발아와 생장시 Canavanine의 이용과 Canavanase의 활성에 대하여)

  • Canavanine content of the cotyledons of Canavalia lineata decreased gradually during germination and growth of seedlings but continued to increase in roots and leaves. After abscission of cotyledons, canavanine content of leaves depleted competely. The activity of canavanase could be detected in leaves and roots, but not in cotyledons. High arginase activity was observed in the cotyledons of seeds at the earlyimbibition period. During the growth of seedlings, cotyledonary canavanine appeared to be transported to the growing of seedlings where it could be utilized through nitrogen metabolic pathways. In crude cell-free extracts of leaves, maximum activities of canavanase or arginase appeared in 30mM Tris-HCl buffer (pH 9.0) or 30mM NaHCO3 buffer (pH 10.0), respectively. The activities of these two enzymes differed from each other when treated with Co2+ or Mn2+. These results support the idea that canavanase and arginase might be different enzymes.

Grey Leaf Spot Caused by Stemphylium lycopersici on Tomato Plants (Stemphylium lycopersici에 의한 토마토 점무늬병)

  • Grey leaf spot on tomato plants was first observed in Sedo-myeon, Puyo-kun, Chungnam province of Korea in 1994. This disease which had not been reported before in Korea exhibited different symptoms from those of other leaf spot diseases on tomato plants. The symptoms were characterized by small irregular-shaped spots on leaves at the initial stage of the infection, subsequent spread and coalescence of the spots throughout the leaves with ultimate necrosis, and abscission from the plants. When healthy tomato plants were inoculated with a conidial suspension of the fungus isolated from the lesion of a diseased plant in a field, the same characteristic symptoms as those in the field were produced. Furthermore, the same pathogen could be reisolated from the lesions formed buy the inoculation. Conidial characteristics of the pathogen were as follows; oblong shape with constricted 3 transverse septa, round-shaped base, round- or point-shaped apex, size of 45~75$\times$12.5~17.5 ${\mu}{\textrm}{m}$, and 3.5 : 1 ratio of length to width. The pathogen was identified as Stemphylium lycopersici and thus this is the first report on the occurrence of grey leaf spot disease on tomato plants caused by s. lycopersici in Korea.

Studies on Quality Maintenance of Fresh Fruit and Vegetables Using Modified Atmosphere Packaging (MA포장 기술을 이용한 신선한 과실 및 채소류의 품질보존에 대한 연구)

  • The objective of this study was to investigate the methods of quality maintenance using modified atmosphere packaging with various quality preservatives at ambient and low temperatures. Ethylene(<1.0ppm) accumulated during modified atmosphere storage of Chinese cabbage using polyethylene film(60$\mu\textrm{m}$) caused quality deterioration such as yellowing and abscission. The addition of potassium permanganate reduced ethylene level and thus extended storage life of l00% at 20$^{\circ}C$ and 140% at 0$^{\circ}C$. To establish the storage conditions of minimally processed sit and vegetables, the experiment was conducted with various quality preservatives and packaging materials. Ceramic film(Zeolite 7%, 60$\mu\textrm{m}$) showed increasing storage life of 50%, reducing total microbial counts of 50% and keeping high appearance quality of minimally processed pears stored at 20$^{\circ}C$ and 0$^{\circ}C$. Minimally processed Chinese cabbage treated with l% CaCl2 at 20$^{\circ}C$, and 1% CaCl2 and 1% NaCl at 0$^{\circ}C$ has a longer storage life of 90% with good maketable quality. Treatment of 1% NaCl for cut Asian penis appeared increased storage life of 100%, decreased cut surface browning and the best overall acceptability by a sensory panel.

Improvement in Clonal Propagation of Hemidesmus indicus R. Br. through Adenine Sulphate

  • A protocol has been developed for rapid large scale clonal propagation of an aromatic endangered medicinal plant, Hemidesmus indicus R. Br. with the elimination of the problems such as premature leaf fall and callus formation during caulogenesis and rhizogenesis. Multiple shoots were induced from shoot tip and nodal explants on Murashige and Skoog (MS) medium supplemented with 1 mg/L Benzylaminopurine (BAP) and 0.5 mg/L Napthaleneaceticacid (NAA). Addition of 15 mg/L adenine sulphate to the above medium checked leaf abscission completely, reduced the time required for caulogenesis and restored morphogenetic potential after several subcultures. The in vitro grown propagules were rooted in 1/2 MS medium supplemented with 2 mg/L Indolebutyric acid (IBA) +1 mg/L NAA and sucrose 0.7% (w/v). Addition of charcoal at 100 mg/L to the rooting medium quickened root initiation with a complete check on callus formation. The effect of sucrose concentration on both caulogenesis and rhizogenesis was also studied. The resultant plantlets were acclimatized and grown in fields where ninety eight percent of the rooted shoots survived and grew normally. The estimation of the secondary metabolite content in the shoots of the regenerated plant and the mother plant indicated that the concentration of the three secondary metabolites lupeol, vanillin and rutin was similar.

Structural Differentiation of the Connective Stalk in Spirodela polyrhiza (L.) Schleiden

  • Structural differentiation of the connective stalk in giant duckweed, Spirodela polyrhiza, was examined to reveal the anatomical and ultrastructural characteristics within reduced shoot. The study focuses primarily on structural features of the connective stalk (CT), which connect offspring to their mother fronds. Photoautotrophic offspring fronds remained connected by stalks to mother fronds in the reproductive pockets until separation. The CT originated from the meristematic region of the abaxial frond and joined the fronds laterally with two abscission layers. The most notable features of the CT were polymorphic mitochondria, random occurrences of fibrillar structures in intercellular spaces, and great variability in cell wall thickness. Vascular tissues in CTs were highly reduced, demonstrating only a central vascular strand. Grana with 2 to 4 thylakoids and starch grains were found in the chloroplasts. A chlorophyll assay indicated high chlorophyll concentrations in daughter fronds and low concentrations in CTs. The frond and CT, while physically connected to each other, functioned independently. Despite great reduction in S. polyrhiza, the CT has proven to be very efficient for separating offspring from the mother frond, which lends to its capacity for rapid vegetative reproduction. The ultrastructural aspects of CTs in S. polyrhiza were characterized for the first time in this study.

Meningitis Occurred during Continuous Lumbar Epidural Block -A case report- (지속적 요부 경막외 차단 중 발생한 뇌막염 -증례 보고-)

  • Postpuncture headache is the most common complication of epidural block, others include abscission of the tip of catheter, epidural abscess and subarachnoid infection, etc. A 69-year-old female patient visited the Neuro-Pain Clinic of Seran General Hospital for treatment of lower back pain and both sciatica. She received continuous epidural block, psoas compartment block, lumbar facet joint block and lumbar facet thermocoagulation. During the epidural block procedure the dura was accidently punctured and auto-logous blood patch was performed. Three days later, she manifested fever, nausea, vomiting, mild neck stiffness and mental deterioration. Meningitis was suspected as the cause of these signs. The CSF study reported: protein 400 mg/dl, sugar 14 mg/dl, WBC $468/mm^3$. She was recovered from the meningitis after adequate antibiotic therapy.

Changes of Cheese Components and Texture Characteristics in Cheese Ripening by Fusant Developed by Lactic Acid Bacteria (융합주에 의한 치즈 숙성시 성분변화와 조직 특성)

  • This study was carried out to elucidate the utilization of the fusant for shortening the ripening time by making an observation of the microstructure and the profile of component change. In ripening cheese, moisture content of the sample treated with tested strain is not a remarkable difference among the test samples. With an increase of the ripening time, L. helveticus showed the highest increase in protein content, followed by fusant, and then L. bulgaricus. The fat content of all starters was gradually decreased while it was it was rapidly decreased after 7 days. The pH of all starters was gradually decreased when the ripening time increased. The titratable acidity was greatly increased between a 9th day and a 15th day ripening. In investigating the light microscopic microstructure of ripened cheese samples, the sample treated with fusant indicated little difference from the other starters in decomposition of protein and fat components by microbial enzymes. In SEM observation, the structure of all cheese samples was uniform and the rough texture was converted into smooth texture by the interaction of cheese components and the abscission of single bond in casein matrix when the ripening time is increased. The fusant showed similar results in the examination of component change and its microstructure compared with the other starters. Therefore, it was revealed that the fusant can be partially used as a cheese starter instead of conventional starters by replacing them or combining them together with the other starters for shortening the ripening time.

Leaf Thinning and Fruit Quality of 'Hongro'/M.9 Apple Trees by Foliar Application of Cu-EDTA and Fe-EDTA

  • Hand leaf thinning for improvement of fruit coloration and quality is one of the most labor-consuming works in Korean apple cultivation. The study was aimed at investigating whether foliar application of two chelate compounds (Cu-EDTA, Fe-EDTA) could defoliate effectively 'Hongro'/M.9 apple leaves and enhance fruit coloration and quality. At 30 days before harvest, foliar spray with two chelate compounds defoliated significantly the apple leaves. Chelate treatments did not affect the leaf chlorophyll content. High concentration of chelates decreased drastically photosynthetic rate of true leaves, and then it stayed in low levels, whereas low levels of chelates reduced sharply the photosynthetic rate but it was gradually restored over time. Cu-EDTA regardless of its concentration triggered higher defoliation compared to Fe-EDTA. Consequently, higher defoliation improved light interception of fruits and accumulated more anthocyanin. Particularly, Cu-EDTA could target mainly fruit cluster leaves which affect directly light interception of the fruits, i.e. more selective defoliants compared to Fe-EDTA. However, 3% Cu-EDTA rather defoliated excessively, accompanying with reduction of fruit weight and soluble solids. Therefore, our results suggest that 1% Cu-EDTA which defoliates properly the fruit cluster leaves could improve coloration and quality of 'Hongro'/M.9 apple fruits.
