Immunogenicity of a Gamma-irradiat d Brucella Vaccine (Gamma선 조사로 만든 Brucella Vaccine의 생쥐에 대한 면역력)

  • A vaccine was prepared by $10^6$ r cobalt-60 irradiation of lyophilized virulent Brucella melitensis and tested in mice for immunogenic activity against a lethal challenge dose of the homologous strain. The vaccine(GIV) produced a high degree of immunity in mice, and comparative studies demonstrated it to be superior to vaccines prepared by heat or by chemical(ether, formalin, or phenol) treatment. Living vaccines, Brucella abortus. strain 19 and an R-form of Brucella melitensis were lethal for mice in larger doses. A comparison of seven adjuvant mixtures for use with the GIV showed no statistically significant difference, but. Freund's complete adjuvant and a mixture of aluminum-potassium sulfate and pectin appeared to enhance the activity of the GIV.

Analysis of lipopolysaccharides of Pasteurella multocida and several Gram-negative bacteria by gas chromatography on a capillary column (Gas chromatography의 capillary column을 이용한 Pasteurella multocida 및 기타 그람음성 세균의 lipopolysaccharide 분석)

  • Lipopolysaccharides (LPS) of Pasteurella multocida (P multocida) and several Gram-negative bacterial pathogens were analyzed by methanolysis, trifluoroacetylation and gas chromatography (GC) on a fused-silica capillary column. The GC analysis indicated that LPS prepared from a strain of P multocida by phenol-water (PW) or trichloroacetic acid (TCA) extraction were quite different in chemical composition. However, LPS prepared from Salmonella enteritidis by the two extraction methods were very similar. PW-LPS extracts from different Pasteurella strains of a serotype had essentially identical GC patterns. Endotoxic LPS extracted from 16 different serotypes of P multocida by PW or by phenol-chloroform-petroleum ether procedures yielded chromatograms indicating similar composition of the fatty acid moieties but minor differences in carbohydrate content. When the chemical composition of endotoxic LPS extracted from several Gram-negative bacteria (P multocida, Pasteurella hacmolytica, Haemophilus somnus, Actinobacillus ligniersii, Brucella abortus, Treponema hyodysenteriae, Escherichia coli, Bacteriodes fragilis, Salmonella abortus equi and Salmonella enteritidis) were examined, each bacteria showed a unique GC pattern. The carbohydrate constituents in LPS of various Gram-negative bacteria were quite variable not only in the O-specific polysaccharides but also in the core polysaccharides. The LPS of closely related bacteria shared more fatty acid constituents with each other than with unrelated bacteria.

Prevalence and co-infection status of brucellosis and tuberculosis in Hanwoo in Jeonnam province

  • Brucellosis and tuberculosis are major infectious and contagious bacterial diseases in cattle. These diseases are malicious diseases that must be inspected at the slaughterhouse of cattle in accordance with the practice of quarantine in Korea. Furthermore, both diseases lead to abortion, reproductive disorder, and calf disease, causing major difficulty in the breeding of Korean Native cattle (Hanwoo), a representative industrial animal currently being raised in Korea. Co-infections of these diseases intensify clinical symptoms such as abortion and have a particularly significant effect on increasing mortality. Thus, serological tests were performed in Hanwoo, to establish the association of co-infection between brucellosis and tuberculosis in cattle. ELISA and PCR tests were conducted on blood samples collected from a total of 102 cattle in Jeonnam province, Korea, to detect brucellosis and tuberculosis infections. The PCR results revealed that 41 samples tested positive for Brucella abortus (B. abortus) infection (40.20%), and 5 samples tested positive for Mycobacterium bovis (M. bovis) (4.90%) infection confirmed by PCR. Notably, 9.76% (4/41) of the cattle infected with brucellosis also tested positive for tuberculosis. In conclusion, this study highlights the co-infection of brucellosis and tuberculosis among Hanwoo cattle in Jeonnam province, which is expected to contribute to our understanding of disease transmission, pathogenicity, the establishment of future prevention strategies.

Development of PCR assay for the detection of Brucella spp in bovine semen (종모우 정액중 Brucella균 신속 검출을 위한 PCR기법 개발)

  • The diagnosis of brucellosis is currently based on serological and microbiological tests. However, the microbiological isolation and identification have several disadvantages such as time-consuming and laborious, and the serological methods have been reported to cross-react with antigens other than those from Brucella spp. To develop a sensitive and rapid diagnostic method for detection of Brucella species, the genus-specific primers were designed and synthesized from the sequence of gene encoding a 31kDa cell surface protein(BCSP) and a 36kDa outer membrane protein(OMPB) of B abortus. The amplified 711bp and 982bp DNA fragments were only visible in each species of Brucella by PCR method using the BCSP and OMPB primers, respectively. However, PCR product was not obtained with DNA from other Gram-negative bacteria. As little as 1pg of the B abortus genomic DNA could be detected by this PCR method. Using the PCR technique, semen samples from 185 bulls of Brucella-seronegative herds in Cheju island were examined for comparison of this PCR method with conventional methods in 1995. The semen samples from 5 bulls were positive by culture method and PCR, and one was positive and 5 were suspect by semen plasma agglutination test. However, the semen samples obtained from 177 bulls were negative by semen plasma agglutination, culture and PCR methods in 1996. The results of comparison tests suggested that PCR was a better test than agglutination test against semen of bulls. This study indicated that the PCR technique was a valuable for the diagnosis of bovine brucellosis, particulary in bull semens.

  • Tile present study was carried out to investigate the incidence of bovine brucellsis in Cheju-do during the period from 1985 to 1990. The results were summarized as follows. 1. In the total 239,238 cattles tested. 1180(0.49%) were positive by standard tube agglutination test during the period from 1985 to 1990. 2. The major causes of incidence on brucellosis was grazing with carriers and repeated incidence in a herd. 3. The 13 Brucella abortus biotype 1 isolated from 10(50%) of 20 cattles slaughtered on brucellosis in 1990.

  • 브루셀라증(brucellosis)은 세포내 기생하는 브루셀라균에 의해 발생하는 인수공통감염질환으로써, 결핵과 유사하게 만성육아종성 염증을 일으키고 장기간의 병합 항균요법을 필요로 하는 질환이다. 그럼 음성 단간균(coccobacilli)인 브루셀라균은 bartonella 및 리켓챠 등과 함께 proteobacteria의 2종에 속하며 전통적으로는 주로 침범하는 숙주에 따라 6종으로 분류되는데, 그 중 Brucella melitensis(양, 염소), B. abortus (소), B. suis (돼지) 및 B. canis (개) 등의 4종이 사람 브루셀라증을 일으키는 것으로 알려져 있다.

Associations of Polymorphisms in Four Immune-related Genes with Antibody Kinetics and Body Weight in Chickens

  • Four biological candidate genes, natural resistance associated macrophage protein 1 (SLC11A1 or NRAMP), prosaposin (PSAP), interferon Gamma (IFNG), and toll-like receptor 4 (TLR4), were examined to identify single nucleotide polymorphisms (SNP) and associations of the SNP with antibody response kinetics in hens. An $F_2$ population was produced by mating $G_0$ highly inbred (<99%) males of two MHC-congenic Fayoumi lines with highly inbred Leghorn hens. The $F_2$ hens (n = 158) were injected twice with SRBC and whole, fixed Brucella abortus (BA). Blood samples were obtained before each immunization, at 7 d after primary immunization, and at several time points after secondary immunization. Minimum titers (Ymin) and the time needed to reach them (Tmin), and maximum (Ymax) titers and the time needed to reach them (Tmax), were estimated from the seven post-secondary immunization titers using a nonlinear regression model. The $F_2$ hens were genotyped for the four candidate genes by using PCR-RFLP for one SNP per gene, which identified the parental allele. General linear models were used to test associations of SNP genotypes with antibody response parameters and BW measured at 4 ages. The IFNG SNP was highly significantly (p<0.0125) associated with primary response to SRBC, Tmin to BA, Ymin to BA, and 12-week BW. The current study demonstrated that the novel IFNG promoter SNP was associated with antibody kinetics for BA and SRBC in laying hens, and also with BW, suggesting that this cytokine may play a pivotal role in the relationship between immune function and growth.

Seroprevalence of specific Brucella infection of cattle in Bangladesh Agricultural University Veterinary Clinics and its surrounding areas

  • A cross sectional survey was conducted to determine the seroprevalence of brucellosis in cattle in Bangladesh Agricultural University (BAU) Veterinary Clinics, in BAU Dairy Farm and Vabokhali from June 2008 to November 2008. A total of 200 serum samples were collected from BAU Veterinary Clinic, from BAU Dairy Farm and Vabokhali. Among the serum samples 143 sera samples were collected from BAU Veterinary Clinic, 42 serum samples from BAU Dairy Farm and 15 serum samples from Vabokhali. Sera were separated from blood samples and tested with specific Brucella abortus antigen (BAA) test and B. melitensis antigen (BMA) test. The overall seroprevalence of brucellosis in cattle was 5% in BAA and 0.5% in BMA. It was observed that, a significant higher prevalence of B. abortus was found in female than male. An insignificant higher prevalence of brucellosis was found in adult cattle (aged above 5 years), in cross breed cattle, in cattle with grazing, cattle breed by natural breeding, and in pregnant cows. Although insignificant but a higher prevalence of brucellosis was found in aged cattle than young cattle, cross bred cattle, pregnant cattle than non pregnant cattle, cattle with grazing. A higher prevalence of brucellosis was found in female cattle than male.
