The Z-DNA domain of human ADAR1 ($Z{\alpha}_{ADAR1}$) produces B-Z junction DNA through preferential binding to the CG-repeat segment and destabilizing the neighboring AT-rich region. However, this study could not answer the question of how many base-pairs in AT-rich region are destabilized by binding of $Z{\alpha}_{ADAR1}$. Thus, we have performed NMR experiments of $Z{\alpha}_{ADAR1}$ to the longer DNA duplex containing an 8-base-paired (8-bp) CG-repeat segment and a 12-bp AT-rich region. This study revealed that $Z{\alpha}_{ADAR1}$ preferentially binds to the CG-repeat segment rather than AT-rich region in a long DNA and then destabilizes at least 6 base-pairs in the neighboring AT-rich region for efficient B-Z transition of the CG-repeat segment.
RNA molecules which bind to the G-rich sequence in the 5'-UTR RNA which plays an important role in expression of N-ras, were selected. The secondary structures of five selected RNA aptamers including primer sequence were found by the CLC RNA workbench ver. 4.2 program (www.clcbio.com) and investigated with RNA structural probes such as RNase T1 which has specificity for a G in single-stranded region, RNase V1 specific for double strand and nuclease S1 specific for single strand. The generalized secondary structure model was proposed and characterized. It was composed of a central long double strand region flanked by single strand region at both end sides. The double strand region had an internal single-strand region and bulges. The single strand loop in the right side was composed of four or five nucleotides.
In this study high power Nd:YAG laser welding of structural steel was investigated. For the test steel blocks of $50{\times}50{\times}200mm$ were cut and machined, and bead-on-plate weld was made on the machined surface. Argon, nitrogen, helium, dry air or mixed gases were used to find the effect of shielding conditions on the bead formation. Results demonstrated that there were Fe I rich region and Fe II rich region in the laser induced plasma column based on the spectral analysis with S-2000 field spectrometer The Fe I region was located at the root of the column near keyhole opening. On the other hand, Fe II region was found at the middle of the plasma column. In the Nd:YAG laser welding, Fe I region emitted continuum which had peak value at wave length of around 710nm, and Fe II region had the peak at 580nm. In the welding of steel by $CO_2$ laser, however, no continuum was observed. There showed two groups of strong spikes in the $CO_2$ laser welding; the first group was displayed at the wave band of 450-560nm. This spike group emitted stronger intensity of light and sharper peaks than those group at 680-800nm.
The phase separation in ferrite phase of duplex stainless steel is the primary cause of thermal aging embrittlement of the LWR primary pressure boundary components. In this study the phase separation of simulated duplex stainless steel was detected by Mossbauer spectroscopy and magnetic property analysis by VSM(Vibrating Specimen Magnetometer). The simulated duplex stainless steels, Fe-Cr binary, Fe-Cr-Ni ternary, and Fe-Cr-Ni-Si quarternary allots, were aged at 370 and 40$0^{\circ}C$ up to 5,340 hours. It was observed from Mossbauer spectra analysis that internal magnetic field increases with aging time and from VSM that the specific saturation magnetization and Curie temperature increase with aging time. These result are indicative that phase separation into Fe-rich region and Cr-rich region is caused by thermal aging in the temperature range of 370~40$0^{\circ}C$ In cases of specimens containing Ni, the increase of specific saturation magnetization is much higher. This implies that Ni seems to promote Fe-Cr interdiffusion, which accelerates the phase separation into Fe-rich $\alpha$ phase and Cr-rich $\alpha$' phase.
Testis-specific protein Y(TSPY) is thought to play an important role during spermatogenesis in primates. The Arg and Lys-rich region of TSPY was implicated as a potential DNA binding site in the human and Japanese monkey. In the study, we investigated this further through looking at the putative amino acid sequences of Arg and Lys-rich region of TSPY gene from seven species of gibbons, five species of Old World monkeys and five species of New World monkeys. A comparison with those of the human and Japanese monkey revealed that the Arg and Lys-rich motif was hightly conserved in various primates. This finding suggests that a possible role for the Arg and Lys-rich motif in primate TSPY is DNA recognition. The functional implications for TSPY are discussed in the light of this and previous findings.

A significant portion (about 8% in human genome) of mammalian mRNA sequences contains AU(Adenine and Uracil) rich elements or AREs at their 3' untranslated regions (UTR). These mRNA sequences are usually stable. ARE motifs are assorted into three classes. The importance of AREs in biology is that they make certain mRNA unstable. We analyzed the occurrences of AREs and Alu, and propose a possible mechanism on how human mRNA could acquire and keep A REs at its 3' UTR originated from Alu repeats. Interspersed in the human genome, Alu repeats occupy 5% of the 3' UTR of mRNA sequences. Alu has poly-adenine (poly-A) regions at the end that lead to poly -thymine (poly-T) regions at the end of its complementary Alu. It has been discovered that AREs are present at the poly -T regions. In the all ARE's classes, 27-40% of ARE repeats were found in the poly -T region of Alu with mismatch allowed within 10% of ARE's length from the 3' UTRs of the NCBI's reference m RNA sequence database. We report that Alu, which has been reported as a junk DNA element, is a source of AREs. We found that one third of AREs were derived from the poly -T regions of the complementary Alu.

A numerical analysis of reactive flow in a liftoff flame is accomplished to elucidate the characteristics of flame propagation velocity and volume integral of reaction rate with the variation of fuel injection velocity at the fuel rich region, fuel lean region and diffusion flame region. The increase of fuel injection velocity enhances flame propagation velocity, but its effect on the flame propagation velocity is not much greater under 4%. The increase of fuel injection velocity affects directly and linearly on the flame surface area in the fuel rich region and so enhances volume integral of reaction rate to accommodate the increment of fuel.
In this work, a 1.9-kb region of enterococcal plasmid p703/5 was isolated and the nucleotide sequence analysis of the region was performed. One major open reading frame (ORF) was identified encoding a polypeptide of 28 kDa. Database comparisons suggested that the protein showed some homology with other bacterial RepA proteins. Upstream of the ORF, a potential dnaA box, AT-rich region and 22-bp tandemly repeated sequences (DNA iterons), a feature typical for many replication ori sites, were recognized. Deletion analysis using Exonuclease III and several restriction enzymes indicated that the three elements and the gene product from the ORF were essential for replication and that the minimum unit of DNA required for replication resided on the 1.2-kb AvaII subfragment. Thus, this gene product was referred to as RepA.
The type of mixed gel network of $\kappa$-carrageenan/agar was determined by applying rheological principles. Apparent Young's modulus of the mixed gels was mathematically analyzed with (a) simply adding the moduli of two component gels, (b) phase-separated type's upper and lower bound models, (c) interpenetrating type's logarithmic model. The experimental data fitted the estimates from the operation (a). Whereas, as for the models (b), the experimental values in the agar-rich region fitted the estimates of the upper bound model, but in the $\kappa$-carrageenan-rich region slightly deviated from those of the lower bound model. It reflected an evidence of a phase-separated type, although it was not typical, that there must be data good-fit in the agar-rich and $\kappa$-carrageenan-rich regions with the upper and lower bound models, respectively. Experimental values disagreed with estimates of the model (c). Gel time was analyzed to evince the phase-separated type. As agar concentrations increased at a fixed amount of $\kappa$-carrageenan, gel time gradually decreased and then sharply increased and decreased again. The pattern of such change in gel time also represented a typical behavior of phase-separated type's mixed gels.

HOX11 encodes a homeodomain-containing transcription factor which directs the development of the spleen during embryogenesis. While HOX11 expression is normally silenced through an unknown mechanism in all tissues by adulthood, the deregulation of HOX11 expression is associated with leukemia, such as T-cell acute lymphoblastic leukemia. The elucidation of regulatory elements contributing to the molecular mechanism underlying the regulation of HOX11 gene expression is of great importance. Previous reports of HOX11 regulatory elements mainly focused on the 5'-flanking region of HOX11 on the chromosome related to transcriptional control. To expand the search of putative cis-elements involved in HOX11 regulation at the post-transcriptional level, we analyzed HOX11 mRNA 3'-untranslated region (3'UTR) and found an AU-rich region. To characterize this AU-rich region, in vitro analysis of HOX11 mRNA 3'UTR was performed with human RNA-binding protein HuR, which interacts with AU-rich element (ARE) existing in the 3'UTR of many growth factors' and cytokines' mRNAs. Our results showed that the HOX11 mRNA 3'UTR can specifically bind with human HuR protein in vitro. This specific binding could be competed effectively by typical ARE containing RNA. After the deletion of the AU-rich region present in the HOX11 mRNA 3'UTR, the interaction of HOX11 mRNA 3'UTR with HuR protein was abolished. These findings suggest that HOX11 mRNA 3'UTR contains cis-acting element which shares similarity in the action pattern with RE-HuR interactions and may involve in the post-transcriptional regulation of the HOX11 gene.
