Ameliorative Effect of Aster scaber Thunberg and Chaenoleles sinensis Koehne Complex Extracts Against Oxidative Stress-induced Memory Dysfunction in PC12 Cells and ICR Mice (PC12세포와 동물모델에서의 기억력 장애를 유도하는 산화적스트레스에 대한 취나물과 모과 복합추출물의 개선 효과)

  • Background: Oxidative stress plays an important role in neuro-degenerative disorders such as Alzheimer's disease. Oxidative stress is mediated by reactive oxygen species (ROS), which are implicated in the pathogenesis of numerous diseases, and account for the toxicity of a wide range of compounds. Methods and Results: In order to study the neuro-protective effect of the complex extracts of Aster scaber Thunberg (AS) and Chaenoleles sinensis Koehne (CSK) against hydrogen peroxide in PC12 cells, cell viability was evaluated by the MTT assay using tetrazole, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and the intracellular ROS levels were determined the by 2',7'-dichlorofluorescein diacetate (DCF-DA) assay. In order to examine the anti-amnesic effects of the complex extracts of AS and CSK, behavioral tests were performed on male ICR mice. The ameliorating effect of the complex extracts against Aβ1-42-induced learning and memory impairment was analyzed by y-maze and passive avoidance tests. The AS and CSK extracts showed neuro-protective activity both in vitro and in vivo, and the neuro-protective effect of their 60 : 40 (AS : CSK) mixture was better than that of the other mixtures. Moreover, the complex extracts synergistically inhibited acetylcholinesterase activity and rapid peroxidation. Conclusions: A mixture of the AS and CSK extracts could be used to develop functional foods and serve as raw materials for the development of therapeutics against Alzheimer's disease.

Land Cover Classification of Multi-functional Administrative City for Hazard Mitigation Precaution (행정중심복합도시 재해경감대책을 위한 토지피복분류)

  • In this study, land cover classification and NDVI evaluation for hazard mitigation precaution are carried out in surrounding areas of Yeongi-gun, Chungcheongnam-do ($132\;km^2$) where a project for multi-functional administrative city is promoted by government. Image acquired from KOMPSAT 2, LANDSAT and ASTER is utilized and comparative evaluation on limitation in classification based on resolution was carried out. The area mainly consists of arable land including mountains, rice fields, ordinary fields, etc thus special attention was paid to the classification of rice fields and ordinary fields. For the classification of image acquired from KOMPSAT 2, segmentation technique for classification of high-resolution image was applied. To evaluate the accuracy of the classification, field investigation was conducted to examine the sample and it was compared with the land usage and classification of land category in land ledger of Korea. Acquired results were made into theme map in shape file format and it would be of great help in decision making of policy for the future-oriented development plan of multi-functional administrative city.

Immunostimulating Activites of Polysaccharide Fractions isolated from Aster scaber Thunb. (참취에서 분리한 다당의 면역자극 활성)

  • ASW0 is a polysaccharide derived from the perennial herb Aster scaber Thunberg. We isolated ASW0, a fraction of crude polysaccharide, by means of ethanol precipitation and dialysis after hot water extraction to investigate its physicochemical properties and immunostimulatory effects. ASW0 contains neutral sugar (45.7%), acidic sugar (51.6%), protein (2.3%), and 2-keto-3-deoxy-D-manno-octonate (KDO) (0.4%). The neutral sugar in ASW0 (in mole percentage) was mainly composed of arabinose (34.5 mol%), glucose (31.1 mol%), galactose (14.9 mol%), and rhamnose (8.1 mol%), which are characteristic of pectic polysaccharides. ASW0 also contained small amounts of xylose, mannose, and fucose. The anti-complementary activity of ASW-0 was similar to that of polysaccharide K (used as positive control). ASW0 exhibited no cytotoxicity in RAW 264.7 macrophages and dramatically increased nitric oxide (NO) production in a dose dependent manner ($0.3{\sim}30{\mu}g/mL$). Also, macrophages stimulated with ASW0 showed enhanced production of immunostimulatory cytokines such as interleukin-6 (IL-6) and tumor necrosis factor alpha ($TNF-{\alpha}$) in a dose dependent manner. These results suggest that the ASW0 have a potent immunostimulatory effect and can be used as a natural immune health ingredient.

Inhibitory Effects of the EtOH Extract of Aster koraiensis on AGEs formation in STZ-induced diabetic rats and AGEs-induced Protein Cross-linking in vitro (벌개미취 에탄올추출물의 STZ-유도 당뇨 모델에서의 최종당화산물의 생성 및 교차결합에 미치는 효과)

  • Advanced glycation end products (AGEs) such as $N^{\varepsilon}$-(carboxy-methyl)lysine (CML) have been implicated in the development of diabetic nephropathy. The aim of this study was to investigate the inhibitory effects of ethanolic extract of Aster koraiensis (AKE) on AGEs formation and AGEs-collagen cross-linking in vitro and CMLs formation in streptozotocin (STZ)-induced diabetic rats. AKE significantly inhibited AGEs formation ($IC_{50}$ value of $18.74{\mu}g/mL$) and AGEs-collagen cross-linking ($IC_{50}$ value of 0.274 mg/mL) in vitro than the well-known glycation inhibitor aminoguanidine ($IC_{50}$ value of $72.12{\mu}g/mL$ and 1.99 mg/mL, respectively). AKE (100 mg/kg per day) was given to diabetic rats for 9 weeks. In STZ-induced diabetic rats, severe hyperglycemia was developed, and urinary CMLs and plasma CMLs were markedly increased. Immunohistochemical stain revealed that CMLs were accumulated within renal glomerulus in STZ-induced diabetic rats. However, AKE significantly reduced urinary CMLs and plasma CMLs in diabetic rats. CMLs accumulation was inhibited by AKE treatment in the renal glomerulus. These results suggest that AKE had an inhibitory effect of AGE accumulation in the glomeruli of diabetic rat and could be an inhibitor of AGE-induced protein cross-linking. The oral administration of AKE may significantly help to prevent the progression of diabetic nephropathy in patients with diabetes.

  • Recently as large sized urban development and the city ward drifting of population are caused, the urban surface temperature is raised very seriously and rapidly. These artificial developments have destroyed the inner and outer landscapes such as topography and have changed complex local climate such as a sudden rise in temperature, the change of wind field and air pollution. In order to clarify this problem visually, the studies on extracting the thermal infrared and the characteristic analysis of local climate in urban area had been performed by using the sixth band of Landsat TM and ETM+. However, there is a need to alternate Landsat TM and ETM+ because these satellite images are not applied any more. Therefore, in this paper it is proposed to use 2 Aster image (2004.4.17 daily 2b03, 2004.10.10 night 2b03) of EOS AM and to extract the surface temperature. Also, the pattern of surface temperature in urban area and the application possibility in local climate study are proposed by verifying the correlation with A WS data. Also, IKONOS image was used to figure out the artificial development area in visual.

Slow Development of Diabetic Cataract in Streptozotocin-induced Diabetic Rats via Inhibition of Aldose Reductase Activity and Sorbitol Accumulation by Use of Aster koraiensis Extract (알도즈 환원효소 활성억제와 소비톨 축적 억제로 인한 벌개미취 추출물의 당뇨병성 백내장 발병 지연 효능)

  • Diabetic cataract is a major complication of diabetes mellitus. Excess accumulation of sorbitol plays an important role in the pathogenesis of diabetic complications such as cataract formation. In this study, we investigated the inhibitory effect of the extract of the aerial parts of Aster koraiensis (AK) on diabetic cataractogenesis. To examine this further, we evaluated sorbitol accumulation during cataract development using streptozotocin-induced diabetic rat, an animal model of type 1 diabetes. Diabetic rats were treated orally with AK (100 mg/kg and 200 mg/kg body weight) once a day orally for 9 weeks. In vehicle-treated diabetic rats, lens opacity was increased, and lens fiber swelling and membrane rupture were observed. In addition, sorbitol accumulation in diabetic lens was markedly enhanced. However, AK treatment delayed the progression of diabetic cataract through the inhibition of sorbitol accumulation, and prevented lens fiber degeneration in a dose-dependent manner. These observations suggest that AK treatment can delay the progression of lens opacification in the diabetic rats during the early diabetic cataractogenesis.

Assessing Soil Fertility Status of Edible Wild Plants Fields in Ulleung Island

  • The perennial edible wild plants such as Aster glehnii, Solidago virgaurea subsp. gigantean, Allium ochotense, Athyrium acutipinnulum, Aruncus dioicus var. kamtschaticus and Codonopsis lanceolata have cultivated as the main income crops introduced into the fields about 30 years ago in Ulleung island. Soil samples were collected from 190 fields and assessed the effects of management practices on soil chemical properties at wild edible plant fields under no-till system. The strong acidic soils of pH 5.4 or less were detected in 45% of the soil samples. The level of soil organic matter was being held at mean $63{\pm}28g\;kg^{-1}$, 2.7 times higher than upland soils in Korea. Available phosphate and exchangeable potassium showed more than recommended levels of upland crops as $680{\pm}489mg\;kg^{-1}$ and $1.94{\pm}1.7cmol_c\;kg^{-1}$, respectively. The fields of Solidago and Aster showing strong soil acidity and high level of available phosphate and water soluble $NO_3{^-}$ were distinguished from other crops in analysis of variance and principal component analysis of soil chemicals. These results suggested that high frequency of acidic soil and high levels of available $P_2O_5$, exchangeable $K_2O$ and water soluble $NO_3{^-}$ were accompanied with the use of urea and NPK-fertilizer based on nitrogen in the field. However, further research is needed to understand the appropriate management of fertilization and the prevention of soil acidification for wild edible plants.

Effects of Shading Net Treatment and Planting Density on Growth and Fresh Leaf Yield of Aster glehni (섬쑥부쟁이의 차광망 처리에 재식거리에 따른 생육 및 수량변화)

  • The study was conducted to find out the ways of increasing fresh leaf yield under conditions of the optimum shading net treatment and planting densities of Aster glehni. Although the most effective shading net treatment to acclimatize the plant was 75% shading in 1993, as growing years were prolonged, effects of shading net treatment was not recognized. Ca contents in 75% shading net treatment was much higher than other shading net treatments. Shading effect tend to promote their growth and fresh leaf yield in 1993 even though 75% shading net treatment did not increase fresh leaf yield in 1994 and 1995. Planting density. $30\times20cm$. showed the effectiveness fresh leaf yield for 1994 and 1995 except for 1993.
