Changes of Total Polyphenol Content and Electron Donating Ability of Aster glehni Extracts with Different Microwave-assisted Extraction Conditions (마이크로웨이브 추출조건에 따른 섬쑥부쟁이 추출물의 총 폴리페놀 함량 및 전자공여 작용 변화)

  • Aster glehni was extracted by microwave-assisted extraction(MAE) and reflux extraction(RE) methods and their extraction efficiencies were compared. A considerable reduction in extraction time was achieved by MAE. When 70% methanol, 50% methanol, 70% ethanol, or 50% ethanol was used, MAE extract contained nearly same amounts of soluble solid and total polyphenol contents as obtained by RE. The optimum MAE conditions for the extraction of Aster glehni were $120{\sim}150$ watts of microwave energy and $4{\sim}8$ minutes of extraction time. No significant changes were found in electron donating ability(EDA) over the variation of microwave energy or extraction time. The use of diluted methanol or ethanol resulted in improving extraction yield(24%), total polyphenol content(2.6%) and EDA(60%).

Anti-oxidant and anti-adipocyte differentiation of Aster glehni and Aster yomena (섬쑥부쟁이와 쑥부쟁이의 항산화 및 지방세포 분화 억제 효과)

  • Purpose: Aster glehnii (AG) and Aster yomena (AY) are medicinal plants that belong to the family Compositea and grow widely in Korea. Plants in the genus Aster have been used to treat snakebite wounds or bruises in oriental medicine. This study compared the effects of anti-oxidants and anti-adipocyte differentiation according to the species (the aerial parts of AG and AY). Methods: AG and AY were extracted using 70% ethanol (-E) and water (-W) at room temperature. The anti-oxidant activities were measured by total phenol contents (TPC), total flavonoid contents (TFC), DPPH and $ABTS^+$ assay. In addition, correlation analysis was performed for the anti-oxidant compounds and effect. The level of anti-adipocyte differentiation was assessed using an oil red O assay on pre-adipocytes. Results: AG-W showed higher TPC ($6.92{\mu}g/mL$) and AG-E presented higher TFC ($8.22{\mu}g/mL$) than the other extracts. Furthermore, AG-E exhibited higher radical scavenging activity in the DPPH and $ABTS^+$ assay ($IC_{50}$: 104.88 and $30.06{\mu}g/mL$). In the cytotoxicity assay, AG and AY extracts at concentrations less than $100{\mu}g/mL$ were non toxic. AG-W reduced the lipid accumulation of 3T3-L1 cells significantly after differentiation (70.49%) compared to the other extracts. Conclusion: These results show that the water extract of AG has anti-oxidant effects and reduces the differentiation of 3T3-L1 cells. Therefore, AG has utility as a functional food material for its anti-oxidant activities and ability to prevent lipid accumulation.

Elimination of Aster Yellows Phytoplasma from Dendranthema grandiflorum by Application of Oxytetracycline as a Foliar Spray

  • Aster yellows phytoplasma-infected chrysanthemums showing stunt, rosette, and excessive branching were treated with a foliar spray of 400 mg/I oxytetracycline at three-day interval for 1,2,3 and 4 months. Two months after the final treatment, new shoots from the recovered chrysanthemums showed the recurrence of the disease symptoms. However, cuttings from chrysanthemums treated with oxytetracycline did not express any photoplasma infection symptoms for more than 10 months. Also, chrysanthemums dipped in 100 mg/I oxytetracycline solution combined with a foliar spray of 400 mg/I oxytetracycline for 4 weeks showed the same results. Using an electron microscope, ultrathin sections of leaf midribs of chrysanthemum cuttings treated with oxytetracycline for 4 months did not show phytoplasma bodies 10 months after treatment. Nucleic acids from chrysanthemums, which did not express phytoplasma infection symptoms for more than 10 months, did not amplify 16S rRNA gene of phytoplasma by polymerase chain reaction. These results may have implications in the propagation of phytoplasma-free healthy stocks for a wide range of plant species.

Vegetation Structure of Peucedanum japonicum Thunb. Community in East Seaside of South Korea (우리나라 동해변 식방풍 군락의 식생구조)

  • This study was conducted to evaluate the vegetation structure and ordination of the Peucedanum japonicum Thunb. community by the Phytosociological method, floristic composition table on east coast of South Korea. The vegetation of Peucedanum japonicum Thunb. community was classified into 2 communities such as the Aster Spathulifolius community and the calystegia soldanella-Artemisia princeps community. Vegetation of the Peucedanum japonicum Thunb. community with the Aster spathulifolius community was shown southward, whereas vegetation of Peucedanum japonicum Thunb. community with the calystegia soldanella-Artemisia princeps community was shown northward from the base line of Pohang at North latitude of 36" 05'. Accordingly, the Peucedanum japonicum Thunb. community was grown commonly with the Aster spathulifolius community southward of the base line of Pohang, whereas it was grown commonly with the calystegia soldanella-Artemisia princeps community northward on the east coast. All environmental conditions of habitat taken together, the optimum habitat of Peucedanum japonicum Thunb. was at Ulsan geographically and the middle region of sea cliff topography, and was suitable for alkali. sandyloam.

  • The chromatographic separation of the MeOH extract of the aerial parts from Aster hispidus (Compositae) led to the isolation of eight compounds. Their structures were established by spectroscopic methods to be ${\beta}-amyrin$ (1), oleanolic acid (2), (2R)-1, 2-O-(9Z, 12Z, $15Z-dioctadecatrienoyl)-3-O-{\beta}-D-galactopyranosyl\;glycerol$ (3), trans-phytol (4), 9, 12, 15-octadecatrienoic acid (5), kaempferol (6), 3,5-dicaffeoyl quinic acid (7), 3,4-dicaffeoyl quinic acid (8) and kaempferol-3-O-rutinoside (9). Compounds 1, $3{\sim}6$ and 9 showed non-specific moderate cytotoxicity against five human tumor cell lines $(5.44{\sim}23.51\;{\mu}g/ml)$. The other compounds were of marginal activity against tested five human cancer cell lines $(9.05{\sim}>30.0\;{\mu}g/ml)$.

Cooking Properties of Buckwheat Noodles Added Aster scaber $T{HUNB}$ Juice (참취 즙액 첨가가 메밀국수의 조리 특성에 미치는 영향)

  • This study was aimed to determine the cooking properties of noodles when dfferent concentrations of Aster scaber THUNB(AST) juice were added to the buckwhenat and wheat flours. Also, physicochemical effects of the noodles and compositions of the noodles and compositions of the noodle soup after cooking were determined. Buckwheat flour and AST contained greater amounts of minerals and essential amino acids than wheat flour. The contents of chlorophyll and carotenoids in the buckwheat noodle added AST juice increased as the concentrations of AST juice increased. Hydration capacity of buckwheat flour was higher than that of wheat flour when AST juice was added to flours. The added amounts of AST juice did not affect the volume and the weight of the noodles, but those were increased as cooking time proceeded. The release of proteins and minerals from the buckwheat noodle added AST juice increased as cooking time progressed and also at the added concentrations of AST juice increased. Texture indices showed lower values as the amount of added juice increased. In sensory evaluation, the scores of color, flavor and overall acceptability of the buckwheat noodle added AST juice were higher than those of the control.

Partial Purification and Some Properties of Polyphenol Oxidase from Aster scaber (참취로부터 추출한 Polyphenol Oxidase의 부분정제 및 성질)

  • A polyphenol oxidase from the crude extract of Aster scaber was partially purified by (NH4)2 SO4 precipitation and subsequent Sephadex G-150 chromatography. The final preparation showed five peaks of enzyme activity. Optimum pH and temperature for the activity of polyphenol oxidase were 7.0 and $30^{\circ}C$, respectively. Enzyme activity was stable at $40^{\circ}C$ for 5min in pH 7.0 reaction mixture but ceased completely at $60^{\circ}C$ for 30min and $70^{\circ}C$ for 5min at pH 7.0. The olyphenol oxidase has good activity acid but was inactive on DL-depa. The apparent Km for catechol was about 17.6mM.

Effect of elicited by methyl jasmonate on the saponin contents of Codonopsis lanceolata (Elicitor처리가 더덕사포닌 함량에 미치는 영향)

  • The roots of Codonopsis lanceolata (Campanulaceae) contain several kinds of triterpenoid saponin with high medicinal values, which have been used in traditional medicines. This study investigates the impacts of methyl jasmonate (MeJA) - adding time on the saponin synthesis and the hairy root growth of C. lanceolata. A significant decrease in major saponin (lancemaside of three kinds) content of hairy roots was observed with MeJA treatments. Contents of lancemaside A, B and E decreased about 15% more than non-treated hairy roots. In contrast, minor saponin (foetidissimoside A and aster saponin Hb) accumulation was about 15% higher than the non-treated hairy roots. These results suggest that MeJA treatment could be used in the production of teriterpene saponins.

  • The experiment was carried out in order to the effects of NAA on rooting of stem cutting of leaf variegated Aster glehni, dwarf type of Achillea sibirica and Solidago virga-aurea var. asiatica native to Korea. Higher rooting percentage(85%) of Aster glehni, promoted root number and root length of Achillea sibirica were obtained by basal dip treatment with $1,000mg{\cdot}L^{-1}$ NAA for 3 hours on late June. Percent rooting of the Solidago was not enhanced, but root number and root length were promoted by basal dip treatment with $100mg{\cdot}L^{-1}$ NAA.

LANDCOVER CHANGE DETECTION USING MODIS TEMPORAL PROFILE DATA SUPPORED BY ASTER NDVI

  • MODIS images have a great advantage of high temporal resolution to monitor land cover changes in a large area. The moderate and low spatial resolution satellite images are incomparably economic than high resolution satellite images. As diverse satellite images are provided recently, strategies using satellite images are necessary for continuous, effective and long-term land monitoring. This research purposed to use MODIS images to monitor land cover in Korean peninsula for long-term and continuous change detection. To maximize the advantages of high temporal resolution, the change detection was based on the MODIS temporal profiles of the surface reflectance for one year. In this study as the reflectance patterns of year 2005 were compared with the reflectance patterns of year 2007, the changed pixels could be detected during two years. To set up the threshold value for the decision of change, ASTER images with the higher spatial resolution, 15m, were used for this study. The test area covered the suburban area of metropolitan city, Seoul, where the landcover changes have been frequently happened.
