Ansamitocin P-3 is a potent antitumor agent produced by A. pretiosum. A deletion mutant of A. pretiosum was constructed by deleting the asm2 gene, a putative transcriptional repressor. The deletion mutant showed a 9-fold enhanced ansamitocin P-3 productivity. The response surface method with central composite design was employed to further optimize the culture medium composition for ansamitocin P-3 production by the deletion mutant. The concentrations of four medium ingredients, dextrin, maltose, cotton seed flour, and yeast extract, which have been reported as major components for ansamitocin production, were optimized through a series of flask culture experiments. The optimum concentrations of the selected factors were found to be dextrin 6.0%; maltose 3.0%; cotton seed flour 0.53%; and yeast extract 0.45%. The maximum titer of ansamitocin P-3 was 78.3 mg/l with the optimized composition, about 15-folds higher than the unoptimized titer of 5.0 mg/l obtained with YMG medium.
Background: Texture analysis has been used as a method for quantifying image properties based on textural features. The aim of the present study was to evaluate the usefulness of magnetic resonance imaging (MRI) texture analysis for the evaluation of viable ovarian tissue on the perfusion map of ovarian endometriosis. Methods: To generate a normalized perfusion map, subtracted T1-weighted imaging (T1WI), T1WI and contrast-enhanced T1W1 with sequences were performed using the same parameters in 25 patients with surgically confirmed ovarian endometriosis. Integrated density is defined as the sum of the values of the pixels in the image or selection. We investigated the parameters for texture analysis in ovarian endometriosis, including angular second moment (ASM), contrast, correlation, inverse difference moment (IDM), and entropy, which is equivalent to the product of area and mean gray value. Results: The perfusion ratio and integrated density of normal ovary were 0.52±0.05 and 238.72±136.21, respectively. Compared with the normal ovary, the affected ovary showed significant differences in total size (p<0.001), fractional area ratio (p<0.001), and perfusion ratio (p=0.010) but no significant differences in perfused tissue area (p=0.158) and integrated density (p=0.112). In comparison of parameters for texture analysis between the ovary with endometriosis and the contralateral normal ovary, ASM (p=0.004), contrast (p=0.002), IDM (p<0.001), and entropy (p=0.028) showed significant differences. A linear regression analysis revealed that fractional area had significant correlations with ASM (r2=0.211), IDM (r2=0.332), and entropy (r2=0.289). Conclusion: MRI texture analysis could be useful for the evaluation of viable ovarian tissues in patients with ovarian endometriosis.
Process analysis with ASM3 (Activated Sludge Model3) was performed to offer basic data for the optimization of aerated biofilter (ABF) process design and operation. This study was focused on the simulation of the nitrification reaction in ABF which was a part of the advanced nutrient treatment process using bio-adsorption. The ABF process has been developed for the removal of suspended solids and nitrification reaction in sewage. A GPS-X (General Purpose Simualtor-X) was used for the sensitivity analysis and operation assessment. Sensitivity of ASM3 parameters on ABF was analysed and 4 major parameters ($Y_A$, $k_{sto}$, ${\mu}_A$, $K_{A,HN}$) were determined by dynamic simulation using 70 days data from pilot plant operation. The optimized values were 0.14 for $Y_A$, 3.5/d for $k_{sto}$, 2.7/d for ${\mu}_A$ and 1.1 mg/L for $K_{A,HN}$, respectively. Simulation with optimized parameter values were conducted and TN, $NH_4{^+}-N$ and $NO_3{^-}-N$ concentrations were estimated and compared with measured data at the range of 10 min to 4 hrs of hydraulic retention time (HRT). The simulated results showed that optimized parameter values could represent the characteristics of ABF process. Especially, the ABF showed relatively high nitrification rate (60%) under very short HRT of 10 min. As a consequence, the ABF was thought to be successfully used in the site which having high variation of influent loading rate.

Based on the activated sludge model(ASM), a mathematical model which represents the aerobic sludge digestion by sequencing batch reactor(SBR) combined with ultrasonic treatment was composed and performed in this study. Aerobic digestion using sequencing batch reactor(SBR) equipped with ultrasound treatment was also experimented for the purpose of parameter calibration. Most of the presented kinetic parameters in ASM or ASM2 could be used for the aerobic digestion of sludge but the parameters related in hydrolysis and decay rate needed modification. Hydrolysis rate constant of organic matter in aerobic condition was estimated at $0.3day^{-1}$ and the maximum growth rate for autotrophs in aerobic condition was $0.618day^{-1}$. Solubilization reactions of particulate organics and nitrogen by ultrasonication was added in this kinetic model. The solubilization rate is considered to be proportional to the specific energy which is defined by specific ultrasound power and sonication time. The solubilization rate constant by ultrasonication was estimated at $0.202(W/L)^{-1}day^{-1}$ in this study. Autotrophs as well as heterotrophs also decomposed by ultrasonic treatment and the nitrification reaction was limited by the lack of autotrophs accumulation in the digester.
Addition effects of free fatty acids (FFA), glycerol, monoacylglycerol (MAG), sesamol, and aqueous extracts of sesame seed meal (ASM) on the changes of sesamol and sesamolin were determined in thermally oxidized sesame oil (SO) at 180$^{\circ}C$ for 90 min. Sesamol and sesamolin in SO were analyzed by high performance liquid chromatography (HPLC). As the concentration of FFA and MAG in SO increased up to 10% (w/w), the concentration of sesamol increased significantly by 0.94 and 0.70 mM, respectively (p < 0.05) whereas sesamol in control samples increased by 0.09 mM for 90 min oxidation. Sesamolin in 10% MAG and FFA added SO significantly decreased by 15 and 18%, respectively (p < 0.05) compared to control samples. Sesamolin in SO with addition of 1.5 and 2.5 mM sesamol were not significantly different (p > 0.05). Addition effects of ASM on the changes of sesamol and sesamolin in SO were not constant during thermal treatment. Conversion of sesamol from sesamolin in SO during thermal treatment seemed to be influenced by the presence of FFA and MAG.
We present our analysis results for an updated orbital ephemeris for the dipping low mass X-ray binary 4U 1624-49, using the light curve collected by the All Sky Monitor (ASM) on board the Rossi X-ray Timing Explorer (RXTE) and the Monitor of All-Sky X-ray Image (MAXI). To make clear dip profiles, the light curve from the ASM and the MAXI were divided into ten 500d segments and four 400d segments for ASM and MAXI light curves, respectively, and folded with the linear ephemeris proposed by Smale et al. (2001). The phases of dip centers were determined by the method adopted from Hu et al. (2008). The phase drift was then fitted with a linear function. We obtained an updated orbital period of 0.869896(1) d and a phase zero epoch of JD 2450088.6618(57). No clear orbital period derivative is detected with a 2-sigma upper limit of $1.4{\times}10^{-6}(yr)^{-1}$ from a quadratic curve fitting of the dip phase evolution.

In frequency hopping spread spectrum(FHSS) communication systems, exact frequency synchronization is required due to the random hopping of carrier frequencies between transmitter and receiver even under harsh channel conditions. For synchronization of FHSS communications, multi-frequency hopping synchronization(MFHS) method has been used in which a small set of frequencies are repeatedly sent several times for long duration. But this long duration resulted in being easily detected by the unauthorized users as well as long duration of acquisition time. In this paper, motivated by these problems, an adaptive synchronization method(ASM) is proposed. ASM is technics to reduce the synchronization time where the number of synchronization frequencies and repetition numbers is adaptively changed (increased or decreased) according to the channel conditions. The performance analysis showed that the time duration of synchronization was reduced to 0.2sec, and the influence of jamming or interference was decreased to 46% in ASM.
The purpose of this study was to analyze the effects of combined exercise on body composition and cardiovascular disease risk factors in sarcopenic obesity elderly women. The subjects for the study were 21 obesity elderly women over 65 years old . They were divided into two groups, the sarcopenic obesity group(n=9) and non-sarcopenic obesity group(n=12). The variables of body composition and cardiovascular fitness were measured in all the subjects before and after 16-week combined exercise. All data was analyzed by two-way ANOVA with repeated measures, and paired t-test by using SPSS 18.0. The findings of this study were as follows; 1. Effects on body composition The relationship between sarcopenia and body composition were only significant for the group ${\times}$ time interaction in FFM. In the sarcopenic obesity elderly women, % body fat significantly decreased and sarcopenia reduced by 30%, but there were no significant changes in the FFM and ASM. On the other hand, the non-sarcopenic obesity elderly women significantly decreased % body fat and significantly increased FFM and ASM. 2. Effects on cardiovascular fitness Both groups significantly increased V˙ O2peak. The most important finding in this study was that sarcopenic obesity elderly women showed delayed effects of a 16-week combined exercise on FFM and ASM compared to the non-sarcopenic obesity elderly women. However, the further studies might need to confirm these results with both genders, different age groups, and various exercise types.

BACKGROUND/OBJECTIVES: The measurement of body composition, including muscle and fat mass, remains challenging in large epidemiological studies due to time constraint and cost when using accurate modalities. Therefore, this study aimed to develop and validate prediction equations according to sex to measure lean body mass (LBM), appendicular skeletal muscle mass (ASM), and body fat mass (BFM) using anthropometric measurement, serum creatinine level, and lifestyle factors as independent variables and dual-energy X-ray absorptiometry as the reference method. SUBJECTS/METHODS: A sample of the Korean general adult population (men: 7,599; women: 10,009) from the Korean National Health and Nutrition Examination Survey 2008-2011 was included in this study. The participants were divided into the derivation and validation groups via a random number generator (with a ratio of 70:30). The prediction equations were developed using a series of multivariable linear regressions and validated using the Bland-Altman plot and intraclass correlation coefficient (ICC). RESULTS: The initial and practical equations that included age, height, weight, and waist circumference had a different predictive ability for LBM (men: R2 = 0.85, standard error of estimate [SEE] = 2.7 kg; women: R2 = 0.78, SEE = 2.2 kg), ASM (men: R2 = 0.81, SEE = 1.6 kg; women: R2 = 0.71, SEE = 1.2 kg), and BFM (men: R2 = 0.74, SEE = 2.7 kg; women: R2 = 0.83, SEE = 2.2 kg) according to sex. Compared with the first prediction equation, the addition of other factors, including serum creatinine level, physical activity, smoking status, and alcohol use, resulted in an R2 that is higher by 0.01 and SEE that is lower by 0.1. CONCLUSIONS: All equations had low bias, moderate agreement based on the Bland-Altman plot, and high ICC, and this result showed that these equations can be further applied to other epidemiologic studies.
GMT secondary mirror system consists of 7 segmented adaptive mirrors. Each segment consists of a thin shell mirror, actuators and a reference body. The thin shell has a few millimeters of thickness so that it can be easily bent by push and pull force of actuators to compensate the wavefront disturbance of light due to air turbulence. The one end of actuator is supported by the reference body and the other end is adapted to this thin shell. One of critical role of the reference body is to provide the reference surface for the thin shell actuators. Therefore, the reference body is one of key components to succeed in development of GMT ASM. Recently, Korea Research Institute of Standards and Science (KRISS) and University of Arizona (UA) has signed a contract that they will cooperate to develop the first set of off-axis reference body for GMT ASM. This project started August 2021 and will be finished in Dec. 2022. The reference body has total 675 holes to accommodate actuators and 144 pockets for lightweighting. The rear surface has a curved rib shape with radius of curvature of 4387 mm with offset of 128.32mm. Since this reference body is placed just above the thin shell so that the front surface shape needs to be close to that of thin shell. The front surface has a concave off-axis asphere, of which radius of curvature is 4165.99 mm and off-axis distance is about 1088 mm. The material is Zerodur CTE class 1 (CTE=0.05 ppm/oC) from SCHOTT. All the actuator holes and pockets are machined normal to the front surface. It is a very complex challenging optical elements that involves sophisticated machining process as well as accurate metrology. After finishing the fabrication of reference body in KRISS, it will be shipped to UA for final touches and finally sent to Adoptica in Italy, in early 2023. This paper presets the development plan for the GMT ASM Reference Body and relevant fabrication and metrology plans.
