New Records of Two Stichotrichid Ciliates, Afroamphisiella multinucleata and Pseudokahliella marina (Ciliophora: Spirotrichea: Stichotrichida) from Korea

  • Two stichotrichid ciliates, collected from marine waters in Jeju Island, were identified as Afroamphisiella multinucleata Foissner et al., 2002 and Pseudokahliella marina (Foissner et al., 1982) Berger et al., 1985. They are recorded for the first time in Korea. The descriptions are based on examinations of living as well as protargol-impregnated specimens. These species are characterized as follows. Afroamphisiella multinucleata has a body size in vivo of $70-95{\times}20-35{\mu}m$; elongate rectangular in shape; contractile vacuole located slightly above mid-body. The adoral zone is bipartited into 3 distal and 13-17 proximal membranelles and occupies 28-35% of the body length. The frontal row comprises 1-4 cirri and one buccal cirrus. The amphisiellid median cirral row is composed of 14-21 cirri, 10-19 left marginal cirri, and 21-30 right marginal cirri. Cortical granules are yellowish. 11-20 globular/ellipsoidal macronuclear nodules arrange proximally along the cell margins. Pseudokahliella marina has a body size in vivo of $110-195{\times}40-110{\mu}m$ and broadly elliptical in shape. The adoral zone of the membranelles occupies 50-60% of the body length, and is composed of 41-70 membranelles. A prominent frontal scutum is present. The contractile vacuole is located below the mid-body. There are 11-13 frontoventral rows, including marginal rows. Caudal cirri and transverse cirri are absent. Three invariable non-fragmented bipolar dorsal kineties are present. The left and right marginal rows are composed of 22-35 and 28-40 cirri, respectively. Colourless cortical granules are present. 8-11 spherical/ellipsoidal macronuclear nodules are connected with each other by thread-like tructures, forming an inverted C-shape.

First Record of Two Cold-Water Jellyfishes Aurelia limbata and Parumbrosa polylobata (Scyphozoa: Semaeostomeae: Ulmaridae) in Korean Coastal Waters

  • Most scyphomedusae jellyfishes recorded in Korean waters are temperate and subtropical species. In the present study, two cold-water jellyfishes from Korean waters are first described. Scyphomedusae were collected from the coasts of Gangneung and Ulsan of the East Sea, and Boryeong, and the eastern area of the Yellow Sea from June 2006 to May 2015. Scyphomedusae collected in Ulsan and Gangneung were identified morphologically as Aurelia limbata Brandt, 1835. Their umbrella was 250-500 mm in width and 30-50 mm in height; the exumbrella was white and the subumbrella was dark brown in color. The vascular system was complex with anastomosed branches. Individuals of this species lived at about 6.6-9.9℃ and 30-50 m depth in Korean waters. Scyphomedusae collected from the center of the Yellow Sea and from Boryeong were identified as Parumbrosa polylobata Kishinouye, 1910. Their umbrella was disc-shaped, 80-200 mm in width and 20-40 mm in height. Its milky white gonads could be seen through the transparent exumbrella. The stomach cavity was round and flat with four interradial cavities connected by subgenital cavities, and the four oral arms were bifurcated and spear-head shaped. Individuals were found at about 6.4-10.0℃ and 40-90 m depth in Korean waters. The identities of both species were confirmed by molecular analysis using nuclear ribosomal DNA sequences.

Effect of $Ar/H_2$ Mixed Gas Sputtering on the Exchange Coupling of NiFe/WeMn Interface (스퍼터링 가스내 수소첨가에 의한 NiFe/FeMn의 교환결합력 향상에 관한 연구)

  • The effect of H$_2$ content in Ar sputtering gas on exchange coupling field(H$_{ex}$) for NiFe/FeMn interface was studied. When NiFe layer of Si(100)/Ta(50 $\AA$)/NiFe(60 $\AA$)/FeMn(250 $\AA$)Ta(50 $\AA$) was deposited at 8% H$_2$ in sputtering gas, the maximum exchange coupling field(H$_{ex}$) and minimum coercivity(H$_{c}$) were obtained. When Si(100)/Ta(50 $\AA$)/NiFe(60 $\AA$)/FeMn(250 $\AA$)/NiFe(70 $\AA$)/Ta(50 $\AA$) was deposited at 5% H$_2$ in sputtering gas, the maximum exchange coupling field(H$_{ex}$) of 148 Oe was obtained. The (111) preferred orientation and grain size of underlayer NiFe were increased and the internal stress was reduced by H$_2$ in sputtering gas. And the (111) preferred orientation and grain size of FeMn layer were also increased.d.ased.

Population Genetic Structure and Evidence of Demographic Expansion of the Ayu (Plecoglossus altivelis) in East Asia

  • Plecoglossus altivelis (ayu) is an amphidromous fish widely distributed in Northeastern Asia from the East China Sea to the northern Japanese coastal waters, encompassing the Korean Peninsula within its range. The shore lines of northeastern region in Asia have severely fluctuated following glaciations in the Quaternary. In the present study, we investigate the population genetic structure and historical demographic change of P. altivelis at a population level in East Asia. Analysis of molecular variance (AMOVA) based on 244 mitochondrial control region DNA sequences clearly showed that as the sampling scope extended to a larger geographic area, genetic differentiation began to become significant, particularly among Northeastern populations. A series of hierarchical AMOVA could detect the genetic relationship of three closely located islands between Korea and Japan that might have been tightly connected by the regional Tsushima current. Neutrality and mismatch distribution analyses revealed a strong signature of a recent population expansion of P. altivelis in East Asia, estimated at 126 to 391 thousand years ago during the late Pleistocene. Therefore it suggests that the present population of P. altivelis traces back to its approximate demographic change long before the last glacial maximum. This contrasts our a priori expectation that the most recent glacial event might have the most crucial effect on the present day demography of marine organisms through bottleneck and subsequent increase of effective population size in this region.

  • Purpose : The purpose of this study is to test the effect of the web-b ased alcohol preventive education for ndicate the model of alcohol preventive education for students spreading al over the family and the comunity.Methods : This study was performed on the 1 public and 1 private eleme ntary school students who are the six grade in Seongdong-gu, Seoul. The number of experimental gr oup was 117 and control group's number was 131, totaled 248. eeks. The pre- and the post-test for web-based alcohol preventive education were tested about knowle dge and attitude toward drinking by questionnaire. The data analysis was done using SPS/Win 12.0 p rogram.Results:The results of this study are as follows;1. Compared with control group, experimental group which was educated the web-based alcohol preventive icant diference in the knowledge toward drinking.2. Compared with control group, also the experimental group w as improved with significant diference in attitude toward drinking after the education.3. In control group, female students' knowledge of drinking w as more increased than male students. And the he group which experienced drinking.Conclusions : The web-based alcohol preventi ve education is worth as a int ervention to get positive influence for the health education. Especially in female studen ts who didn't met with drinking, early education for alcohol prevention is very important. Therefore i t will be helpful that coperate with district ofice ithin the comunity for becoming established the controled drinking pattern.

  • Recently, duc to highly increased consumption of' ngg~.egatc>s f o ~ . construction. studies have focused on the effective utilization of rock wastes abandoned so far. This study was designed, firstly, to determine t,hc petrological, g'ochemical and mechanical cha~,acte~istics of' crushed limestone aggregates in thc Samhwa district for suitable construction aggregates and, secondly, to offer basic data for cff'ective utilization of low grade limestones. Results of' the petrographic st,udy indicates that the crushed limestone aggregates in the Samhwa district can bo separate4 into two groups, namely f'inc-grained and cowlxcgrained limestones. Dominantly distributed fine-grained limestone containing some dolomite has higher Mgo and $SiO_2$ contents compared to the coarse-graincd limestonr. It, can be classified as medium strength rock by the physical and mcxhanical pi.opertics. I3ased on the size of' mineral grains and chemical compositions, it is suggested that the crushed limestone aggregates in t,his study area would bctkr be u s ~ i for asphalt concr.ctt., road pavement, or railroad ballast materials than for cement concrete.

Bacillus stearothermophilus 에서 부분 정제한 Cytosine Deaminase 의 특성

  • Cytosine deaminase (EC 3.5.4.1) from BaciNus stc~urorhermophilus was partially purified 7.2-fold with an overall yield of 52.7%. The partially purified enzyme deiiminated cytosine only.but not 5-methylcytosine and 5-fluorocytosine. The apparent Michaclis constant. Km valuefor cytosine was 5.9 mM. The enzyme was relatively stable in the range of pH 4.0 to 7.0.furthermore extremely thermo-stable : more than 75'X) of the activity was remained afterheating at 80$^{\circ}$C for I0 min at pH 6.5. The enzyme had a pH optimum at around pH7.0 to 7.5. and temperature optimum at 35 to 31$^{\circ}$C. And the activation energ (En value)determined from an Arrhenius plot was 26 Kcal/mol. The enzyme activity was stronglyinhibited by heavy metal ions such as Cd", Hg". Cut' at 1 mM, anJ by o-phenanthroline,and p-chloromcrcuribcnzoate at I mM. But the enrymc activity was activatetl increased byGMP, and CMP at 1 mM.ased by GMP, and CMP at 1 mM.

Determining the Specific Status of Korean Collared Scops Owls

  • The collared scops owl that occurs in Korea is a protected species but its exact specific status has been questioned. To resolve the species status, a molecular phylogenetic analysis was conducted using two fragments of mitochondrial DNA, cytochrome b (cyt b, 891 bp) and NADH dehydrogenase subunit 2 (ND2, 627 bp) genes. Phylogenetic trees of cyt b revealed that all Korean specimens formed a monophyletic group with Japanese scops owl Otus semitorques with very low sequence divergence (d=0.008). We obtained a similar ND2 tree as well (d=0.003); however, the genetic distance between Korean individuals and O. lempiji from GenBank (AJ004026-7, EU348987, and EU601036) was very high and sufficient enough to separate them as species (cyt b, d=0.118; ND2, d=0.113). We also found that Korean species showed high differentiation from O. bakkamoena (AJ004018-20 and EU601034; cyt b, d=0.106; ND2, d=0.113) and O. lettia (EU601109 and EU601033, cyt b, d=0.110; ND2, d=0.117) as well. Therefore, we suggest that the Korean collared scops owl should be designated as Otus semitorques.

Phylogeographic and Feeding Ecological Effects on the Mustelid Faunal Assemblages in Japan

  • Phylogeographic and feeding ecological studies of seven terrestrial mustelid species (Carnivora, Mustelidae), the Japanese marten Martes melampus, the sable Martes zibellina, the Japanese badger Meles anakuma, the ermine or the stoat Mustela erminea, the Japanese weasel Mustela itatsi, the least weasel Mustela nivalis, and the Siberian weasel Mustela sibirica, representing four biogeographic patterns in the Japanese archipelagos (Hokkaido, Honshu-Shikoku-Kyushu, Tsushima, and Hokkaido-Honshu), were reviewed in order to clarify causes for the faunal assemblage processes of those mustelid species in Japan. Here, three main constraints were extracted as important factors on the mustelid assemblage. First, fundamental evolutionary differences maintained by niche conservatism in each ecologically diversified lineage ("evolutionary constraint") would enable the species to co-occur without any major problem (coexistence among Martes, Meles, and Mustela species). Second, "ecological constraints" would force two closely related species to be allopatric by competitive exclusion (Mu. itatsi and Mu. sibirica) or to be sympatric by resource partitions (Mu. erminea and Mu. nivalis). Third and most importantly, "geological constraints" would allow specific species to be embraced by a particular geographic region, primarily deciding which species co-occurs. The allopatric distribution of two Martes species in Japan would have been established by the strong effect of the geological separation in Tsugaru Strait. Elucidating both phylogeny and ecology of co-existing species in a community assemblage is important to know which species possess distinct lineage and which ecological traits are adapted to local environments, fulfilling the requirement of the field of conservation biology that endemism and adaptation should both be considered. The Japanese archipelagos would, therefore, provide valuable insight into the conservation for small carnivoran species.

Redescription of Australocirrus shii and First Report of Afrokeronopsis aurea (Ciliophora: Spirotrichea: Sporadotrichida) from South Korea

  • Two hypotrich ciliates, Australocirrus shii (Shi et al., 1997) Kumar & Foissner, 2015 and Afrokeronopsis aurea (Foissner & Stoeck, 2008) Foissner et al., 2010 isolated from freshwater habitats in Korea and were studied based on the specimens from live and after protargol impregnation. Australocirrus shii is redescribed based on morphology and 18S rRNA gene sequence, whereas Af. aurea is the first record for Korea. Main morphological features of the Korean population of Au. shii are as following: body size $100-200{\times}40-80{\mu}m$ in vivo; elongate to ellipsoidal or slightly elongate obovate, dorsoventrally flattened; transverse cirri arranged in (3+2) pattern, anterior pretransverse ventral cirrus distantly anterior of the first transverse cirrus; eight or nine dorsal kineties; and three caudal cirri. Main morphological features of the Korean population of Af. aurea are as following: body size $230-375{\times}70-145{\mu}m$ in vivo; shape elongate obovate or ellipsoidal, widest at the mid-body; undulating membranes in Australocirrus pattern with a buccal depression; and three caudal cirri. The Korean population of A. shii is similar in morphology with previous descriptions except for the presence of indentation at the posterior end in the Korean population. The Korean population of A. aurea is slightly shorter than the South African population and has slightly less marginal and mid-ventral cirri. The phylogenetic analysis of present two Korean hypotrichs and relevant species based on 18S rRNA gene sequences generated almost similar tree topologies compared with previous studies.
