AtERF73/HRE1, an Arabidopsis AP2/ERF Transcription Factor Gene, Contains Hypoxia-responsive Cis-acting Elements in Its Promote (애기장대의 AP2/ERF 전사인자인 AtERF73/HRE1의 프로모터에 있어서 저산소 반응 cis-조절 요소의 분석)

  • In a signal transduction network, from the perception of stress signals to stress-responsive gene ex- pression, binding of various transcription factors to cis-acting elements in stress-responsive promoters coordinate the adaptation of plants to abiotic stresses. Among the AP2/ERF transcription factor family genes, group VII ERF genes, such as RAP2.12, RAP2.2, RAP2.3, AtERF73/HRE1, and AtERF71/ HRE2, are known to be involved in the response to hypoxia stress in Arabidopsis. In this study, we dissected the HRE1 promoter to identify hypoxia-responsive region(s). The 1,000 bp upstream promoter region of HRE1 showed increased promoter activity in Arabidopsis protoplasts and transgenic plants under hypoxia conditions. Analysis of the promoter deletion series of HRE1, including 1,000 bp, 800 bp, 600 bp, 400 bp, 200 bp, 100 bp, and 50 bp upstream promoter regions, using firefly luciferase and GUS as reporter genes indicated that the -200 to -100 region of the HRE1 promoter is responsible for the transcriptional activation of HRE1 in response to hypoxia. In addition, we identified two putative hypoxia-responsive cis-acting elements, the ERF-binding site and DOF-binding site, in the -200 to -100 region of the HRE1 promoter, suggesting that the expression of HRE1 might be regulated via the ERF transcription factor(s) and/or DOF transcription factor(s). Collectively, our results suggest that HRE1 contains hypoxia-responsive cis-acting elements in the -200 to -100 region of its promoter.

상업용 가압경수로의 무붕산 운전 가능성 연구

  • 600MWe급 상업용 가압경수로의 무붕산운전 가능성을 핵설계 측면에서 검토하였다. 기본 원자로 핵연료 형태는 AP600 기준으로 무붕산 노심이 가능하도록 핵연료 농축도, BP 재질/갯수/위치/농도, 제어봉의 재질/위치/갯수등의 설계 변수등을 변화시켰다. 핵연료 농축도는 1.95w/o, 2.9w/o, 3.5w/o를 사용하였으며, BP의 갯수는 8에서 24개까지 사용하였으며, 재질은 Gd$_2$O$_3$, 농도는 축방향 출력분포를 평탄화 시키기 위하여 10w/o-14w/o를 사용하였다. 제어봉 설계에서 재질은 경제적이며, 제어봉가가 큰 B$_4$C를 사용하였으며, 반응도 제어에는 weak 제어봉을 사용하여 출력분포에 대한 영향을 최소화 하도록 설계하였다. 또한 출력분포제어에는 독립적으로 작동되는 고반응도가의 제어봉을 설정하여 노심 상부에서 동작하게 함으로써 반응도에 미치는 영향이 적도록 설계하였다. 설계 결과 아직 안전성 확보 여부에 대한 연구가 수행되지 않은 상태이나 주기초에서 주기말까지 잉여반응도를 1% 미만으로 유지하고, 축방향 BP zoning이 이루어지면 현재의 상업용 원자로에도 무붕산 운전이 가능할 것으로 판단된다. 또한 주기초 A.O.가 -14%, 제어봉의 삽입과 인출에 따른 국부 첨두치가 2.16으로 나타나 안전성 확보에도 큰 어려움은 없을 것으로 생각된다.

Optimizing the binding activity of the AP2/ERF transcription factor with the GCC box element from Brassica napus by directed evolution

  • In this study, we cloned the ERF-B3 subfamily transcription factor gene BnaERF-B3-hy15 from Brassica napus L. Huyou15. This 600 bp gene encodes a 199 amino acid classic ethylene responsive factor (ERF), which shown no binding or very weak binding GCC box-binding activity by the yeast one-hybrid assay. We used gene shuffling and the yeast one-hybrid system to obtain three mutated sequences that can bind to the GCC box. Sequence analysis indicated that two residues, Gly156 in the AP2 domain and Phe62 at the N-terminal domain were mutated to arginine and serine, respectively. Changes of Gly156 to arginine and Phe62 to serine increased the GCC-binding activity of BnaERF-B3-hy15 and the alter of Gly156 to arginine changed the AP2-domain structure of BnaERF-B3-hy15.

Heat Transfer in the Passive Containment Cooling System (수동형 격납용기 냉각계통에서의 열전달)

  • The objective of this work is to obtain the experimental data for the heat transfer processes occurring both on the inside and outside surfaces of containment steel wall with dry and wet outer surface conditions in the passive containment cooling system. The test model represented a 60$^{\circ}$ section of a containment vessel based on the AP 600 geometry. Major linear dimensions of the test model ore reduced tv a factor of ten. To simulate the decay heat a steam generator heated by electricity was placed in the test model. The maximum heat flux was 8.91 kW/$m^2$. Two types of tests were performed. The one was the tort on the natural convection of air without water film flow. The other was the evaporative heat transfer test with the falling water film flow and natural air draft. no test result shooed that the heat transfer capability by the natural convection from the containment to the air without oater film flow was limited at about 1.48 kW/$m^2$ heat flux. It was found that the heat removal capability was remarkably enhanced in the tests with the waster film flow and air draft. The obtained heat transfer data ore compared with the existing correlations.

Identification of Phellinus linteus by Comparison of Colony Shapes and Using PCR techniques (목질진흙버섯(Phellinus linteus)의 균총형태 비교 및 PCR 기법을 이용한 동정)

  • Twenty-two Phellinus strains were characterized using colony morphologies and polymerase chain reaction (PCR) to divide into Phellinus linteus. There were some differences in mycelial growth and colony shapes among the strains when they were grown on various media such as PDA, MCM, MEA and YM. Phellinus linteus was slowly growing, formed golden-yellow colony, and produced blue pigment on PDA media. When the regions of internal transcribed spacer (ITS) were amplified from ribosomal RNA (rRNA) coding genes of P. igniarius and P. linteus strains by means of PCR, two types of band (700 bp and 800 bp) were appeared, respectively. For the amplified intergenic region I (IGRI), P. igniarius strains showed a different band among 500, 600, 700 and 800 bp according to the strains, whereas P. linteus strains did one specific band of 700 bp. By polymorphism analysis after digesting the amplified products with 6 different restriction enzymes, a band specific to P. linteus was generated when the products for ITS region were digested with HaeIII, suggesting that the enzyme digestion could provide effective method to distinguish between P. igniarius and P. linteus. And also, the analysis of genetic relationship showed that the genetic similarities were 89% and 95% in P. igniarius and P. linteus strains, respectively. Random amplification polymorphic DNA (RAPD) analysis using multiple primer sets and arbitrarily primed PCR (AP-PCR) with ITS3 primer could also result in a reproducible way to identify P. linteus strains.

Effects of Whole Body Fatigue and Limited Visual Field on Postural Stability (전신 피로와 시야 제한이 자세균형 능력에 미치는 영향)

  • Loss of postural stability can possibly lead to slip and fall accidents in the number of workplaces and everyday life. This study was aimed to examine the effects of whole body fatigue and partially limited visual field on the ability of maintaining postural balance during quiet standing. A group of twelve healthy male subjects participated in the experiment. Before and after experiencing the whole body fatigue induced by bicycling exercises, the position coordinates of subject's center of pressure (COP) were obtained under the two levels of visual field condition (i.e., open visual field and limited visual field). Four levels of the whole body fatigue examined were rest, 300watt, 600watt, and 900watt. Position coordinates of COPs measured on a force plate were then converted into the total length of postural sway path in both the medio-lateral (ML) direction and the anterior-posterior (AP) direction. Two-way ANOVA result showed that the length of sway path in the AP direction became significantly larger as the whole body fatigue accumulated. Post-hoc test revealed statistically significant differences between rest and 900watt and between 300watt and 900watt. The significant increase of the sway length was also found when the visual field was partially obstructed by the boxes. In the ML direction, however, there was no statistically significant difference of the postural sway in both the AP and ML directions. The results imply that the ability of maintaining postural stability can be reduced significantly due to such tasks along with whole body fatigue. The postural balance can also be impaired by the limited visual field.

Ethanol Extract of Oenanthe javanica Modulates Inflammatory Response by Inhibiting NF-${\kappa}B$ Mediated Cyclooxygenase-2 Expression in RAW 264.7 Macrophage

  • Effect of Oenanthe javanica ethanol extract (OJE) on nuclear factor-${\kappa}B$ (NF-${\kappa}B$)-mediated inflammatory reaction in RAW 264.7 macrophage cells was investigated. The OJE dose-dependently inhibited secretions of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and prostaglandins $E_2\;(PGE_2)$ from lipopolysaccharide (LPS)-stimulated RAW 264.7 cells and blocked LPS-induced expression of cyclooxygenase-2. To clarify mechanistic basis for its inhibitions of NF-${\kappa}B$ and activator protein-1 (AP-1) activations, effects of OJE on activations of NF-${\kappa}B$ and AP-1 genes by luciferase reporter activity were examined. The LPS-stimulated activations of NF-${\kappa}B$ and AP-1 were significantly blocked by 400 and $600\;{\mu$}g/mL of OJE, implicating that OJE might regulate gene expression through more than one signaling pathway. Cytosolic degradation of I-${\kappa}B{\alpha}$ was inhibited by OJE dose-dependently, indicating that the nuclear translocation of p65 was inhibited by OJE. These findings suggest that the inhibition of LPS-stimulated COX-2 expression by OJE is due to its inhibition of NF-${\kappa}B$ activation by blocking I-${\kappa}B{\alpha}$ degradation, which may be mechanistic basis of anti-inflammatory effects of OJE.

Nuclear Design Feasibility of the Soluble Boron Free PWR Core

  • A nuclear design feasibility of soluble boron free(SBF core for the medium-sized(600MWe) PWR was investigated. The result conformed that soluble boron free operation could be performed by using current PWR proven technologies. Westinghouse advanced reactor, AP-600 was chosen as a design prototype. Design modification was applied for the assembly design with burnable poison and control rod absorber material. In order to control excess reactivity, large amount of gadolinia integral burnable poison rods were used and B4C was used as a control rod absorber material. For control of bottom shift axial power shape due to high temperature feedback in SBF core, axial zoning of burnable poison was applied to the fuel assemblies design. The combination of enrichment and rod number zoning for burnable poison could make an excess reactivity swing flat within around 1% and these also led effective control on axial power offset and peak pin power, The safety assessment of the designed core was peformed by the calculation of MTC, FTC and shutdown margin. MTC in designed SBF core was greater around 6 times than one of Ulchin unit 3&4. Utilization of enriched BIO(up to 50w1o) in B4C shutdown control rods provided enough shutdown margin as well as subcriticality at cold refueling condition.

  • In supervised learning, labeling of all data is essential, and in particular, in the case of object detection, all objects belonging to the image and to be learned have to be labeled. Due to this problem, continual learning has recently attracted attention, which is a way to accumulate previous learned knowledge and minimize catastrophic forgetting. In this study, a continaul learning model is proposed that accumulates previously learned knowledge and enables learning about new objects. The proposed method is applied to CenterNet, which is a object detection model of anchor-free manner. In our study, the model is applied the knowledge distillation algorithm to be enabled continual learning. In particular, it is assumed that all output layers of the model have to be distilled in order to be most effective. Compared to LWF, the proposed method is increased by 23.3%p mAP in 19+1 scenarios, and also rised by 28.8%p in 15+5 scenarios.

Effect of Bombyx mori, Antheraea Yamamai and Antheraea pernyi Silk Protein in Skin Fibroblast Cell Proliferation After Injury (피부세포 증식에 관여하는 실크단백질 탐색)

  • We have studied the effect of silk proteins to the cell proliferation of human skin fibroblast cells (CCD-986sk) after injury. Silk proteins were extracted treatment with enzyme or NaOH solution from raw silk and culled-cocoon shell of Bombyx mori, Antheraea yamamai and A. pernyi. The cell proliferation after in vitro injury are increased in treatment by Bombyx mori (BM-1,2), Antheraea yamami (AY-1,2) and A. pernyi (AP-1,2). The silk protein fractions-treated cells exhibited proliferation in a dose dependent between $0.1\;{\mu}g/ml$ and $10\;{\mu}g/ml$. But, the macrophage, RAW 264. 7 cell viability was unaffected by the silk protein fractions by MTT assay. The molecular weights of the silk protein fractions were from 300-600 to 900-1500. These results results that the silk protein fractions may function through skin fibroblast proliferation.
