울금(Curcuma longa L.)이 이상지질혈증 흰쥐의 혈액요소질소(BUN) 및 효소활성에 미치는 영향 (Effects of Turmeric (Curcuma longa L.) Supplementation on Blood Urea Nitrogen and Enzyme Activities in Dyslipidemic Rats)

  • The purpose this study was to investigate the influences of 5% turmeric (Curcuma longa L.) supplementation on enzyme activities such as aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), amylase, lipase and catalase in serum of dyslipidemic rats. Sprague-Dawley (SD) rats (24 male) were divided into four groups, namely the ND group (normal-nondyslipidemic diet), NT group (normal-nondyslipidemic diet+5% turmeric), DD group (control-dyslipidemic diet), and DT groups (dyslipidemic diet+5% turmeric). Serum concentrations of blood urea nitrogen (BUN), creatinine and uric acid were significantly decreased (p<0.05) by turmeric supplementation diet. The activities of AST, ALT, ALP, LDH, amylase and lipase in sera of turmeric diet group were significantly decreased (p<0.05). The catalase activity in serum of turmeric supplementation group was significantly increased than dyslipidemic diet (p<0.05). In vivo experiment with dyslipidemic rats showed that ingestion of turmeric were effective in kidney and hepatic functional enzyme activities. Which suggests that turmeric material could be used for further studies as a potential source for nutraceutical foods.

Effect of Beverage Containing Fermented Akebia quinata Extracts on Alcoholic Hangover

  • The present study was conducted to investigate the effects of beverages containing fermented Akebia quinata extracts on alcoholic hangover. For this study, 25 healthy young men were recruited. All participants consumed 100 mL of water (placebo), commercial hangover beverage A or B, fermented A. quinata leaf (AQL) or fruit (AQF) extract before alcohol consumption. After 1 h, all participants consumed a bottle of Soju, Korean distilled liquor (360 mL), containing 20% alcohol. Blood was collected at 0 h, 1 h, 3 h, and 5 h after alcohol consumption. The plasma alanine transaminase (ALT) activity was highest in the placebo group. Compared with the control group, the AQL and AQF groups showed decreased ALT activity at 5 h after alcohol consumption. Plasma ethanol concentration was increased after alcohol intake and peaked at 3 h after alcohol consumption. Compared with the control group, the A group showed a higher plasma ethanol concentration at 1 h (P<0.05). At 3 h after alcohol consumption, the AQF group showed the lowest mean plasma ethanol concentration compared to the other groups; however, there were no statistical differences. After 5 h of alcohol consumption, the AQL and AQF groups showed lower plasma ethanol concentrations compared with the B group. The sensory evaluation score for the fermented A. quinata fruit extract was lower than for the commercial hangover beverages. In conclusion, the present intervention study results suggest that fermented A. quinata extracts alleviate alcoholic hangover and reduce plasma ethanol concentrations.

Ameliorative Effects of Combinative Injection of Ginko biloba Leaves Extract and Vitamin C on Ischemia/Reperfusion Liver Damages Model

  • Hepatic ischemia-reperfusion injury (HIRI) is linked with high mortality rate. Several agents have been developed so far to reduce the risk of HIRI. In this study, we investigated the effects of combined treatment of Ginko biloba leaves extract and vitamin C (GLEVC) on hepatic ischemia-reperfusion injury. To explore the protective effects of GLEVC on HIRI rats model were tested. After the development of HIRI by using clamping method rats were then randomly divided into four groups. Different doses of GLEVC were administered in HIRI rat model. The level of ALT, AST, SOD and MDA content in serum were detected in HIRI groups. Moreover, the activity of SOD, content of MDA, and GSH in hepatic tissue were also examined. Bcl-2 and Bax protein expression were detected by immunohistochemical staining method. Compared with sham group, GLEVC has the protective effect on the HIRI-induced model. Level of ALT, AST, and MDA in blood were significantly lower in GLEVC group compared with HIRI-induced group. Moreover, SOD activity and GSH were increased in GLEVC group whereas MDA content was reduced by GLEVC treatment. Furthermore, HIRI-induced Bax protein was reduced upon GLEVC treatment, whereas Bcl-2 protein expression was enhanced. These results demonstrate that GLEVC treatment may provide potential ameliorative therapy by reducing damaged signaling mechanism in hepatic ischemia/reperfusion injury model.

경북지역 일부 초등학생의 비만도에 따른 생화학 검사치와 식습관 (The Differences of Biochemical Status and Dietary Habits according to the Obesity Degree among Obese Elementary School Students in the Gyungbuk Area)

  • The prevalence of obesity in children has steadily risen during recent years in developed countries. Child obesity has become a major concern to health providers since it has grown to epidemic proportions over the past few decades. This study was conducted to investigate the biochemical status, dietary habits and life styles according to the obesity degree among obese children residing in the Gyungbuk area. The subjects were 148 elementary school students (boy 103, girl 45) and classified as mildly obese (n = 56), moderately obese (n = 61) and severely obese (n = 31) by the obesity index. The average body weight and height increased significantly according to the obesity degree (p < 0.05, p < 0.001). Serum cholesterol concentration came under the normal level in all groups. Serum AST activity increased according to the obesity degree but it was not significant. Serum ALT activity increased according to the obesity degree (p < 0.05). Dietary habit score was lower significantly according to the obesity degree (p < 0.01). Life style of the subjects was significantly different for the level of 'healthy state (p < 0.05)', 'body perception (p < 0.01)', 'watching TV and video (p < 0.05)', 'normal sleeping hour (p < 0.05)' and 'degree of stress (p < 0.05)' according to the obesity degree. A positive relationship between obesity index and blood pressure, ALT has been shown in the subjects. Obesity index and dietary habit scores were negatively associated. Therefore, these results suggest that a practical nutrition-exercise education program for the prevention of child obesity should be provided to elementary school children. Favorable Hepatoprotective Effects of Gongjin-dan on the Acute Ethanol-induced Liver Damaged C57BL/6 Mice

  • To observe the potential hepatoprotective effects of Gongjin-dan on the acute ethanol (EtOH)-induced liver damages in C57BL/6 mice with its possible action mechanisms. EtOH-mediated acute hepatic damages were induced by oral administration of EtOH total 3 doses. The changes on the body weight, liver weight, albumin, TG, AST, ALP, ALT, hepatic TG contents, hepatic antioxidant defense system, TNF-α, CYP 2E1 activity and mRNA expressions of hepatic lipogenic genes - SREBP-1c, SCD1, ACC1, FAS, PPARγ and DGAT2 or genes involved in fatty acid oxidation - PPARα, ACO and CPT1 were observed with final liver histopathological inspections after 15 days of continuous administration of silymarin 200 mg/kg, Gongjin-dan (GJD) 400, 200 and 100 mg/kg. The results were compared with silymarin 200 mg/kg treated mice. Marked decreases of body and liver weights, increases of serum AST, ALT, Albumin and TG levels, hepatic TG contents, TNF-α level, CYP 2E1 activity and mRNA expressions of hepatic lipogenic genes or decreases mRNA expressions of genes involved in fatty acid oxidation were observed with histopathological changes related hepatosteatosis increases of immunolabelled hepatocytes, as the results of a binge drinking of EtOH in the present study. Also destroys of hepatic antioxidant defense systems were demonstrated in EtOH control mice as compared with intact vehicle control mice, respectively. The results suggest that oral administration of 400, 200 and 100 mg/kg of GJD favorably protected the liver damages from acute mouse EtOH intoxications.

침술과 피어싱으로 발생한 만성 C형 간염 1예 (A Case of Chronic Hepatitis C Acquired through Ear Piercing and Acupuncture)

  • 산발성 C형 간염 환자의 약 40%에서 감염경로가 확실치 않다. 이러한 환자들에서 HCV 감염이 수혈, 투석, 수술, 수직감염 및 성적 접촉 등의 잘 알려진 감염 경로 이외에도 침, 귓불 천공 등의 경피적 경로를 통해 전파 될 수 있음을 간과하지 말아야 할 것이다. 저자들은 간염의 가족력이나 수술과 수혈 과거력이 없는 10세 여아에서 피어싱과 침술로 인해 발생한 만성 C형 간염을 진단하고, pegylated interferon과 경구 ribavirin으로 치료한 증례를 경험하였기에 문헌고찰과 함께 보고 한다.

Acetaminophen에 의해 유도된 흰주의 간손상에 미치는 Calcium channel Blocker의 효과 및 조직학적 소견 (Effect of Calcium Channel Blocker on Acetaminophen-induced Hepatotoxicity in Rats and Histopathologic Examination)

  • 해혈 및 진동 효과를 가진 APAP는 과량 복용시 CYP-450에 의해 독성 유발 물질인 NAPQI로 대사되어 간장과 세포막을 붕괴시켜 세포 내 calcium 유입을 증가시킴으로서 간세포의 괴사를 일으킨다. DIL은 CYP-450 작용을 억제하는 것으로 알려진 칼슘채널차단제이다. 따라서 본 연구에서는 APAP 300mg/kg을 경구 투여한 후 3, 6. 9 및 12시간에 DIL을 복상내로 투여하여 DIL이 APAP의 독성에 미치는 영향을 조사하였다. APAP 투여 12시간후 DIL 투여군에서 혈청과 간조직의 생화학 분석과 조직학적 관찰에서 간손상의 개선 효과가 확인되었으며, 이는 세포내로의 calcium 유입과 지질과산화의 억제 및 GST의 활성도 증가에 기인한 것으로 보인다. 그러므로 APAP 과량 복용 12시간 후의 DIL 투여는 간손상의 억제에 효과적일 것으로 사료된다.

도라지의 급이가 고콜레스테롤혈증 흰쥐의 간기능 및 간조직의 지질조성에 미치는 영향 (Effect of the Feeding Platycodon grandiflorum on Lipid Components of Liver and Liver Function in Hypercholesterolemia Rats)

  • 도라지의 급이가 고콜레스테롤혈증 흰쥐의 간기능과 지질대사에 미치는 영향 구명하기 위하여 SD계 숫흰쥐에게 동결건조한 도라지 분말 5%와 도라지 분말 5%에 해당하는 식물섬유, saponin, 에탄올 추출물을 각각 급이하여 3주간 실험 사육한 후 혈청효소 및 간장의 지질성분 분석, 간세포의 형태를 검토하였다. 혈청 중 AST, ALT효소활성은 대조군에 비해 전 실험군이 유의적으로 감소하였고 그 중 saponin 급이군이 가장 낮았고 그 다음이 식물 섬유군이었다. LDH효소 활성도 전 실험군에서 유의적으로 낮게 나타났으며 22년근 도라지의 식물 섬유군과 saponin군이 낮은 값을 보였다. 간장 중의 총 콜레스테롤 함량은 4년근과 22년근 도라지 분말 및 식물섬유 급이군이 비교적 낮았으며, 중성지질 함량은 대조군이 비해 전 실험군에서 낮았으나, 각 실험군간의 차이는 크지 않았고, 인지직 함량은 전 실험군에 있어 비슷한 수준이었다. 간장 조직세포의 전자현미경 관철 결과, 대조군은 지방구가 증가되어 있는 반면 도라지 급이군에서는 감소되었으며, 22년근 도라지 급이군은 간장 글리코겐 축적량이 증가되어 있었다.

대금음자가미와 이(梨)추출물 배합제제가 알코올로 유발된 간손상에 미치는 영향 (Effects of combination pear extract with Daekumeumjagami medication on hepatic injury induced by alcohol in mice)

  • Objectives : The effect of pear extract with Daekumeumjagami and vitamin C medication(PDV) on alcohol metabolism and hepatic injury was assessed following hepatic injury induced by alcohol in mice. Methods : The model of alcoholic hepatic injury was established by orally administration with 3 g/kg 25% alcohol in mice. PDV was orally administrated once a day for 5 days. Mice were randomly divided into 5 groups : normal group, control group, and PDV groups (PDV-A, PDV-B and PDV-C). The activities of aspartate amino transferase (AST) and alanine amino transferase (ALT) and alcohol dehydrogenase (ADH) in serum, superoxide dismutase (SOD) and catalase in liver were determined after alcohol exposure. Results : Compared with control group, treatment with PDV-B and PDV-C significantly elevated activities of ADH. Moreover, the index of hepatic injury in serum was significantly decreased by treatment with PDV-B and PDV-C in ALT activity and PDV-C in AST activity. Additionally, enhanced catalase activities in liver was found in PDV-C treated mice after exposure to alcohol. Also, WBC in blood was significantly lower by treatment with PDV-B and PDV-C. Conclusions : This study suggests that PDV treatment could enhance alcohol metabolism, and prevent hepatic injury after alcoholic hepatic injury and that this effect is likely related to its modulation on the alcohol metabolizing and antioxidant enzymes.

환경 Hormone에 대한 삼백초의 Glutathione 및 항산화 활성 효과 (Effects of Saururus chinensis Baill on Glutathione and Antioxidative Activity Against TCDD-treated Rats)

  • 삼백초가 TCDD 투여로 유발된 rat의 간손상에 미치는 효과를 알아보기 위해 NTT, TTA, STT군의 3군으로 각각 10마리씩 나누어 TCDD를 투여하고 4주 동안 물질을 투여하였다. 4주 후 희생시켜서 생체 내 간 조직 활성 효과 및 항산화 효과를 관찰한 결과 1. AST 활성도는 STT군이 TTA군과 비교해서 70.05%,의 억제 효과를 보였다. 반면에, ALT활성도는 STT군이 TTA군에 비해 24.00% 감소하였으며 64.80%의 억제 효과를 보였다. 2. SOD는 TTA군이 NTT군보다 137.78% 감소하였으며 , STT군은 TTA군에 비해 81.94%로 상승되었다. Catalase는 TTA군은 NTT군 보다 199.13% 감소하였으며, STT군의 catalase 활성도는 TTA 군과 비교하여 52.48% 증가하였고 55.45%의 회복율을 보였다. 3. TTA군의 GSH함량은 NTT군 보다 291.29% 감소하고 STT군은 TTA군보다 비교하여 74.20% 증가하였으며, 98.72%로 회복하였다. TTA군은 GSSG함량은 NTT군 보다 46.80% 증가하였고 STT군의 간조직중의 GSSG 함량은 TTA군과 비교하여 61.08% 감소하였으며, 81.01%로 억제되었다.
