Rhododendron micranthum is an endangered species in Korea. In order to develop the strategy of gene diversity conservation, estimation of the amount of genetic diversity, the genetic variation and relationship in the native populations of Rh. micranthum was performed on the basis of AFLP and RAPD analysis. Analysis of 56 accessions derived from 6 populations of Rh. micranthum with four AFLP primer combinations and ten RAPD primers detected a total of 33 polymorphic AFLP fragments and 15 polymorphic RAPD fragments, respectively. By UPGMA cluster analysis with molecular markers, the 56 accessions were grouped into three major clusters at 73.3% genetic similarity; group I consists of most accessions of populations I, II, IV, V and Ⅵ, group II consists of 7 accessions of population III, and group III consists of only two accessions of population IV. The geographic locations of the most accessions derived from six populations were not related to their position in the UPGMA cluster analysis, except for several accessions of populations III and IV. The genetic similarity of among six populations measured by AFLP and RAPD markers ranged from 0.66 to 0.99. Among them, population Ⅵ showed the highest GS with means of 0.87, while population I showed the lowest GS with means of 0.78. This result will be useful for designing the strategy of conservation in the native populations of Rh. micranthum.
Genetic diversity among four populations of Chinese shrimp Fenneropenaeus chinensis from Narodo, Yeonggwang, Taean and Chinese Bohai Bay was assessed by amplified fragment length polymorphism (AFLP) DNA fingerprinting. Total numbers of AFLP bands generated (ranging from 251 to 254) and average percent of polymorphic bands (27.1 to 28.1 %) were similar in the four populations. Heterozygosity and genetic diversity within or among the populations were very low for the populations with average values ranging from 0.1177 to 0.1288 and from 0.1099 to 0.1194, respectively. Analyses of pairwise distance, Fst index and genetic similarity among the populations also revealed the similar results with very low genetic differentiation each other. These results suggest that all the wild populations tested in the present analysis may be belonging to the same genetic origin, and also that they may have a close relationship in genetic structure without any significant differentiation.
Korean spotted barbel Hemibarbus mylodon, is an endangered and endemic freshwater species in the Korean peninsula. Amplified fragment length polymorphism (AFLP) was used to analyze the genetic diversity and population genetic structure of three populations (Bukhan, Namhan and Imjin-river). Fifteen AFLP primer pairs produced 795 products of which 135 were polymorphic(17%). Percentages of polymorphic bands were similar among the three populations, with accounting 11.9%, 11.1%, and 13.4% for Bukhan, Namhan and Imjin-river populations, respectively. An average genetic similarity among the three populations was 0.969. The average heterozygosity (0.033-0.040) and genetic diversity (0.036-0.043) were significantly low. Pairwise distance and fixation index analyses of three populations also suggested quite a low genetic differentiation one another. These results would provide a fundamental baseline data to develop the effective strategy for the management and restoration of this endangered fish species.

We already reported four groups which contains some similar strains based on URP-PCR in the previous paper. The objective of this study was to confirm those strains by the amplified fragment length polymorphism (AFLP) and vegetative compatibility group (VCG). AFLP analysis showed no difference among these strains except ASI 2595 and 2183 in Weonhyeong group and ASI 2829 in Suhan group. They showed specific DNA bands only in the result of P + AG/M + AAG and P + GT/M + ATG primer combinations out of eight different combinations. The AFLP primers produced a total of 330 fragments between 80 and 1000 bp in length for 31 Pleurotus ostreatus strains. At a genetic similarity of 0.96, the UPGMA analysis separated the isolates into four distinct clusters. Each group was classified by similar strains. Confrontation test by vegetative compatibility groups (VCGs) also showed distinct line between strains from different groups, but no line between similar strains within the cluster. Our results indicate that most of similar strains was not distinctness. Thus, similar strains are considered to be very close on the genealogy of their parent or same strain with different name.

Jute is the principal coarse fiber for commercial production and use in Bangladesh. Therefore, the development of a high-yielding and environmental-stress tolerant jute variety would be beneficial for the agro economy of Bangladesh. Two molecular fingerprinting techniques, random-amplified polymorphic DNA (RAPD) and amplified-fragment length polymorphism (AFLP) were applied on six jute samples. Two of them were cold-sensitive varieties and the remaining four were cold-tolerant accessions. RAPD and AFLP fingerprints were employed to generate polymorphism between the cold-sensitive varieties and cold-tolerant accessions because of their simplicity, and also because there is no available sequence information on jute. RAPD data were obtained by using 30 arbitrary oligonucleotide primers. Five primers were found to give polymorphism between the varieties that were tested. AFLP fingerprints were generated using 25 combinations of selective-amplification primers. Eight primer combinations gave the best results with 93 polymorphic fragments, and they were able to discriminate the two cold-sensitive and four cold-tolerant jute populations. A cluster analysis, based on the RAPD and AFLP fingerprint data, showed the population-specific grouping of individuals. This information could be useful later in marker-aided selection between the cold-sensitive varieties and cold-tolerant jute accessions.
Radiation induced and somaclonal variations were investigated in the regenerates from gamma irradiated and controlled embryogenic callus (EC) of sweetpotato cvs., Yulmi and White Star by morphological, RAPD and AFLP analysis. Most (approx. 90%) of the EC produced somatic embryos developed into plantlets after being transferred to the auxin-free medium. The frequency of morphological variants derived from the irradiated callus ranged from 3 to 7.8% compared to 0.1-1.1% of that derived from the non-irradiated. Morphological variants were selected from the regenerates and analyzed by RAPD and AFLP procedures. RAPD polymorphisms of Yulmi and White Star regenerates from irradiated calli were 8.8% and 6.1%, respectively. However, the polymerphisms among regenerates from the non-irradiation treatment in these two cultivars were non-detectable and 3%, respectively. AFLP polymorphisms of Yulmi and White Star regenerates from irradiated calli were 29.9% and 28.6%, respectively. while the frequencies for those form non-irradiated calli were 8.5% and 5.6%, respectively. Collected germplasms of five representative dandelion species (Taraxacum ohwianum, T. platycarpum, T. platypecidum, T. officinale, and T. coreanum) were 104 lines from different habitates in Korea and China. Their genetic diversity was analyzed by genomic fingerprinting method using amplified fragment length polymorphism (AFLP). AFLP results of 6 primer combinations were revealed 1,176 total DNA fragments and 523 polymorphic bands with a 44.4% ratio of polymorphism. On the basis of similarity coefficient analysis by unweight pair group method with arithmetic averages (UPGMA), 104 dandelion germplasm lines were ranged from 0.64 to 0.99 and clustered distinct five group depending on the species. Furthermore, a principal coordinate analysis (PCA) by the application of multi-variate analysis indicated significantly greater differences among species than geographical origins.

Late blight caused by Phytophthora infestans is historically a serious epidemic disease in potato and tomato cultivations. Accession L3708 (Solanum pimpinellifolium), a new source for late blight resistance was identified in AVRDC, and carries the resistance gene, Ph-3, incompatible to P. infestans race 3. The AFLP markers linked to Ph-3 were previously developed from the L3708 accession (Chunwongse et al. 2002). To facilitate tomato breeding with the Ph-3 gene, an attempt was made to convert AFLP markers to sequence-characterized amplified region (SCAR) markers. Among 6 AFLP markers, only one AFLP marker, L87, was successfully converted to SCAR marker. The resistance-specific 230 bp AFLP fragment was cloned and sequenced, and the PCR primer amplifying a 123 bp fragment was designed. This SCAR marker could discriminate resistant and susceptible individuals with high stringency. The developed SCAR marker could be used for the marker assisted-selection in tomato breeding programs.
Although sea slaters Ligia have a significant role in rocky shore habitats, their taxonomic entities have not been clearly understood. In this study, we investigated whether genetic variation inferred from a nuclear genetic marker, namely amplified fragment length polymorphism (AFLP), would conform to that of a mitochondrial DNA marker. Using both the mitochondrial DNA marker and the AFLP marker amplified by the six selective primer sets, we analyzed 95 Ligia individuals from eight locations from East Asia. The direct sequencing of mitochondrial 16S rRNA gene revealed three distinct genetic lineages, with 9.8-11.7 Kimura 2-parameter genetic distance. However, the results of AFLP genotyping analysis with 691 loci did not support those of mitochondrial DNA, and revealed an unexpectedly high proportion of shared polymorphisms among lineages. The inconsistency between the two different genetic markers may be explained by difference in DNA evolutionary history, for example inheritance patterns, effective population size, and mutation rate. The other factor is a possible genomic island of speciation, in that most of the genomic parts are shared among lineages, and only a few genomic regions have diverged.
A recent study on the mitochondrial genetic variation of the Carassius auratus population in South Korea suggested that there are 3 distinct mitochondrial lineages in the country, and that they are geographically separated between westward rivers and southward rivers, respectively. In this study, the population genetic structure of amplified fragment length polymorphism (AFLP) of Carassius auratus was investigated. The results of analysis of molecular variance (AMOVA) supported the geographic distinction between westward and southward river populations, but only 3.66% of total genetic variance lies among these populations. The panmicticity of the AFLP genetic variation is backed up by the results of the neighbor-joining dendrogram drawn from a linearized pairwise $F_{ST}$ matrix and Bayesian clustering analysis. The discordance of genetic structure between mitochondrial and AFLP genetic variation may come from difference in effective population size between these markers and/or gene flow between westward and southward river populations through river capture events.
