Effects of vitamin C on the formation of aflatoxin B1-DNA adduct in rat livers treated with radiation and aflatoxin B1 (Vitamin C가 방사선과 Aflatoxin B1을 처리한 흰쥐의 간세포에서 Aflatoxin B1-DNA Adduct 형성에 미치는 영향)

  • The objective of this study was to examine the effects of vitamin C on the formation of aflatoxin $B_1$ ($AFB_1$)-DNA adduct and $AFB_1$-induing cellular oxidative damage in rat livers treated with radiation and $AFB_1$. Six-week-old male Sprague-Dawley rats were randomly divided into five groups: the control group, the $AFB_1$-treated group, the group treated with $AFB_1$ and vitamin C, the group treated with X-ray and $AFB_1$, and the group treated with X-ray and $AFB_1$ with vitamin C. On the first day of the experiment, only one dose of X-rays was exposed to the entire liver at 1,500 cGy. Next, vitamin C was injected at 10 mg/kg body weight via intraperitoneal injection, followed an hour later by the administration of 0.4 mg/kg of $AFB_1$ via intraperitoneal injection. These treatments were administered every three days for 15 days. On the 16th day, the animals were sacrificed. The $AFB_1$ contents of the rat sera were determined via indirect competitive ELISA. In the quantitative analysis of $AFB_1$ in the rat sera via ELISA, $5.17{\pm}0.34ng/mL$ of $AFB_1$ was detected in the $AFB_1$-treated groups, but the amount decreased more significantly to $3.23{\pm}0.76ng/mL$ in the groups treated with $AFB_1$ and vitamin C (p<0.01) than in the $AFB_1$-treated groups. The effect of vitamin C on $AFB_1$-DNA adduct formation was determined via ELISA. The values of $AFB_1$-DNA adduct formation were $9.38{\pm}0.41ng/mL$ in the $AFB_1$-treated groups, but the amount decreased more significantly to $5.28{\pm}0.32ng/mL$ in the groups treated with $AFB_1$ and vitamin C (p<0.01) than in the $AFB_1$-treated groups. Immunohistochemistry revealed that the accumulation of the $AFB_1$ was not observed in the normal liver tissue (G1). The $AFB_1$-positive materials were observed in the central vein and the portal vein of the liver tissue from the $AFB_1$(G2) treatment or the X-ray and $AFB_1$(G4) co-treatment, but the $AFB_1$-positive materials were observed weakly in the group treated with vitamin C (G3 and G5). These results indicate that vitamin C had ameliorating effects on the $AFB_1$ accumulation of liver tissue.

Effect of L-ascorbic acid on the degradation of aflatoxin $B_1$ (아스콜빈산에 의한 Aflatoxin $B_1$의 파괴에 관한 연구)

  • Large amount of aflatoxin $B_1(AFB_1)$ is disappeared in the presence of L-ascorbic acid(AA) in buffer solution at pH values from 1 to 7 during 5 days of Incubation at $37^{\circ}C$. $AFB_1$ was quite stable at pH's between 5 and 7 when AA was absent(control), however, $50{\sim}60%$ of APB, was degraded in its presence after 5 days. The rate of disappearance of $AFB_1$ increased with a decreasing of pH in the presence of AA, even though $AFB_1$ in the control degraded increasingly with the decrease in $pH(pH{\leq}4)$. The level of $AFB_1$, decreased as the reaction temperature increased when $AFB_1$ reacted with AA. The aflatoxin could not be detected at all after 3 days when the reaction occurred at $60^{\circ}C$, while the aflatoxin was stable at $5^{\circ}C$ thoughout the reaction period. $90{\sim}96%$ of $AFB_1$ was found to be degraded in a far when $AFB_1$ reacted with AA plus different concentrations of $CuSO_4{\cdot}5H_{2}O$, showing remarkably faster rate than the control; however, different concentrations of L-cysteine instead of $CuSO_4\;5H_{2}O$ protected the degradation of aflatoxin and no $AFB_1$ was degraded for a day and resulted in less $AFB_1$ disappeared than the control. The degradation of $AFB_1$ was dependent on AA concentration and the rate of disappearance as the concentration of AA decrease, but $AFB_1$ concentration did not influence the rate. The product formed when $AFB_1$ reacted with AA was identified to $AFB_{2a}$ by using HPLC chromatographic examinations, and by UV spectrum of $AFB_1$ reacted with AA. The disappearance of $AFB_1$ was correlated well in the appearance of $AFB_2a$. From the results, the degradation of $AFB_1$ in the presence of AA is probably due to one or more of the oxidative products of AA which was produced during the AA oxidation.

  • Aflatoxin ($AFB_1$) is a potent hepatotoxic and hepatocarcinogenic mycotoxin in humans. It is also well-known to be accumulated in animal tissues via various metabolic pathways. This study was conducted to determine the effects of vitamin C on the residual $AFB_1$ in rat sera that were treated with radiation and $AFB_1$. Six week-old male Sprague-Dawley rats were randomly divided into five groups: a control group, $AFB_1$-treated group, the group treated with $AFB_1$ and vitamin C, the group treated with X-ray and AFB1, and the group treated with X-ray and $AFB_1$ with vitamin C. The $AFB_1$ contents of the rat sera of the groups treated with X-ray and $AFB_1$ did not significantly decreased with the administration of vitamin C. The $AFB_1$ content of the rat sera that was analyzed via HPLC showed a tendency similar to that of the content that was analyzed via ELISA. With regard to these data, vitamin C was very effective in reducing $AFB_1$ residue in rat sera.

Mutagenicity of Reaction Products of Aflatoxin $B_1$ and Ascorbic Acid (아스콜빈산과 Afatoxin $B_1$ 반응생성물의 돌연변이 유발성)

  • Aflatoxin $B_1$ ($AFB_1$) was reacted with ascorbic acid (AA) alone, with AA plue cysteine and with AA plus cupric ion for 5 days (at $37^{\circ}C$ and pH 5), and the mutagenicity of the reaction products was tested with Salmonella typhimurium TA 100. About 10% of AFBl induced mutagenesis was reduced when $AFB_1$ reacted with AA. This decreasing effect was more severe when $AFB_1$ reacted with AA plus cysteine. The mutagenicity of $AFB_1$ when reacted with AA plus cupric ion was almost completely inhibited, however, eupric ion itself was shown to enhance the mutagenicity of $AFB_1$. Therefore, $AFB_1$ may be degraded in the presence of AA under the given reaction condition and the reaction products was observed to have nonmutagenic effects on the bacterial mutagenecity trials.

Effect of Aflatoxin B1 on the Function of Peritoneal Macrophage from Mule Duck

  • This study was conducted to investigate the effect of aflatoxin $B_1$ ($AFB_1$) alone or mixed function oxidase (MFO)-activated $AFB_1$ on various functions of mule duck peritoneal macrophages. Duck peritoneal macrophages were incubated with $AFB_1$ 0, 5, 10, 20, 50 and $100 {\mu}g/ml$ for 12 h. The cell viability significantly declined as the concentration of $AFB_1$ increased and more obviously detrimental effects was noticed in MFO-metabolized $AFB_1$ treatments. Either in opsonized or unopsonized Candida albicans, phagocytotic ability of macrophages was decreased with the elevation of the concentration of $AFB_1$. Significantly higher levels of macrophages were damaged in MFO-metabolized $AFB_1$ than $AFB_1$ alone in concentrations above $20{\mu}g/ml$. The cytotoxicity activity was in the range of 41 to 33% after exposure to $AFB_1$ 5 to $100{\mu}g/ml$, and a significant higher TNF-like substance secretion by lipopolysaccharide (LPS) stimulation was obtained. When LPS was present in the medium, the percentage of cytotoxicity was higher than all treatments of $AFB_1$ both with and without MFO-activation in the absence of LPS. The results suggest that MFO-metabolized $AFB_1$ can alter cell viability and morphology of duck macrophages more than $AFB_1$ administered alone. Both with and without MFOactivation, $AFB_1$ has detrimental effects on phagocytotic ability and TNF-like substance secretion, increasing with level of $AFB_1$.

The Effects of Antioxidant Vitamins Via Comet and Mitogenic Assay in Mice Exposed to $AFB_1$ ($AFB_1$에 노출된 마우스에서 Comet Assay와 Mitogenic Assay에 의한 항산화 비타민의 효과)

  • The objective of this study was to examine the effects of antioxidant vitamins on the cellular oxidant damage by observing the mitogenicity in the mouse spleen and the strand breaks of DNA in mouse blood induced by $AFB_2$. Intraperitoneal(i.p.) injections of vitamin C(VC) of 10 mg/kg and vitamin E(VE) of 63.8 mg/kg were repeatedly administered to male ICR mice of 6 weeks old at intervals of 4 times every 2 days. After one hour vitamin treatments, $AFB_1$ of 0.4 mg/kg was injected into the $AFB_2$ plus vitamin treated groups in the same way. On the other hands, into the $AFB_2$ only treated group, only $AFB_2$ was injected without vitamins in the same method as above. The results of the experiment are as follows ; as regard to comet assay, DNA strand breaks were clearly present and they formatted a typical comet tail in the mice blood of the $AFB_2$ only treated groups. However, comet tails apparently disappeared in $AFB_2$ plus antioxidant vitamins treated groups since oxidant damage was controlled in an almost similar level to the control group. Mitogenicity of the spleen also showed a similar tendency as before, and these differences were more remarkably observed in the reaction against Con-A, which is a T-cell mitogen. In these data, the statistical significance was p<0.01. The LDL and VLDL levels were 408.72, 504.47 mg/dl respectively in the $AFB_2$ only treated groups. Compared with the $AFB_1$ only treated groups, those of $AFB_2$ plus antioxidant vitamin treated groups decreased to 272.06(VC), 305.28 mg/dl(VE), respectively. On the other hand, HDL levels were diminished to 32.60, 29.60 mg/dl in $AFB_2$ only treated groups, compared to 42.23, 41.14 mg/dl in the $AFB_2$ plus antioxidant vitamins treated groups. But, blood glucose levels were not statistically significant.

Production of Group Specific Monoclonal Antibody to Aflatoxins and its Application to Enzyme-linked Immunosorbent Assay

  • Through the present study, we produced a monoclonal antibody against aflatoxin B1 (AFB1) using AFB1-carboxymethoxylamine BSA conjugates. One clone showing high binding ability was selected and it was applied to develop a direct competitive ELISA system. The epitope densities of AFB1-CMO against BSA and KLH were about 1 : 6 and 1 : 545, respectively. The monoclonal antibody (mAb) from cloned hybridoma cell was the IgG1 subclass with ${\lambda}$-type light chains. The $IC_{50}s$ of the monoclonal antibody developed for AFB1, AFB2, AFG1 and AFG2 were 4.36, 7.22, 6.61 and 29.41 ng/ml, respectively, based on the AFB1-KLH coated ELISA system and 15.28, 26.62, 32.75 and 56.67 ng/ml, respectively, based on the mAb coated ELISA. Cross-relativities of mAb to AFB1 for AFB2, AFG1 and AFG2 were 60.47, 65.97 and 14.83% in the AFB1-KLH coated ELISA, and 59.41, 46.66 and 26.97% in the mAb coated ELISA, respectively. Quantitative calculations for AFB1 from the AFB1-Ab ELISA and AFB1-Ag ELISA ranged from 0.25 to 25 ng/ml ($R^2$ > 0.99) and from 1 to 100 ng/ml ($R^2$ > 0.99), respectively. The intra- and inter-assay precision CVs were < 10% in both ELISA assay, representing good reproducibility of developed assay. Recoveries ranged from 79.18 to 91.27%, CVs ranged from 3.21 to 7.97% after spiking AFB1 at concentrations ranging from 5 to 50 ng/ml and following by extraction with 70% methanol solution in the Ab-coated ELISA. In conclusion, we produced a group specific mAb against aflatoxins and developed two direct competitive ELISAs for the detection of AFB1 in feeds based on a monoclonal antibody developed.

The Effect of Antioxidant Vitamins on Aflatoxin $B_{1}-DNA$ Adduct the Formation in Aflatoxin $B_1$ Administered Mice Liver (항산화비타민이 Aflatoxin $B_1$ 투여 마우스의 간세포에서 Aflatoxin $B_{1}-DNA$ Adduct 형성에 미치는 영향)

  • The objective of this study was to examine the effects of antioxidant vitamins on the formation of $AFB_{1}-DNA$ adduct and $AFB_{1}-inducing$ cellular oxidative damage. Intraperitoneal(i.p.)injections of 10 mg/kg vitamin C(VC) and 63.8 mg/kg vitamin E(VE) were repeatedly administrated 4 times with 2 days interval to 6 week old male ICR mice. After one hour of vitamin treatments, 0.4 mg/kg $AFB_1$ was injected in $AFB_1$ plus vitamin treated groups by same way. On the other hands, $AFB_1$ treated group was only injected with $AFB_1$ by the same method described above without vitamins. According to quantitative analysis of the $AFB_1$ in mice serum by indirect competitive ELISA, 12.28 and 18.78 ng/mL were detected in $AFB_1-treated$ groups, but 7.60 and 4.85 ng/mL in $AFB_1$ plus VC and VE treated groups, respectively. 23.78, 25.48 ng/mL of $AFB_1-DNA$ adduct were detected in mice liver of $AFB_1$treated groups, while 5.26, 7.81 ng/mL in $AFB_1$ plus VC and VE treated groups, respectively. Consequently, the differences in the concentrations of $AFB_1$ related materials between vitamin treated and non-treated groups were significant. Immunohistochemistry revealed brownish infiltration of $AFB_1$ around central vein and sinusoid in $AFB_1-treated$ group. This manifestation was distinctly reduced in $AFB_1$ plus VC and VE treated groups.

Immunosuppressive Effect of Aflatoxin B1 (Aflatoxin B1의 면역억제작용)

  • Aflatoxin B1 (AFB1) has been reported to directly suppress the immune responses. In the present study, the effect of AFB1 on immune functions was investigated. Splenic lymphocytes were treated with various doses of the mitogens (lipopolysaccharide, concanavalin A) in the presence of AFB1. AFB1 pretretment decreased the number of plaque forming cells (PFC) in a dose-dependent manner. Antibody production of IgM and IgG class was significantly decreased in AFB1-treated splenic cells. In addition, when animals were exposed to AFB1, the susceptibility of bacterial infection as well as the growth of tumor cells was increased. These data suggest that AFB1 affected the immune function and humoral immunity impaired by AFB1 treatment contributed to pathological process.

Effects of Coffee and Aflatoxin B1 on the Pancreatic Exocrine Function and Structure (Coffee와 Aflatoxin B1이 췌장의 외분비 기능 및 조직에 미치는 영향)

  • Coffee is known to increase pancreatic secretion of digestive enzymes. The mutagen, aflatoxin B1(AFB1) is contained in fermented foods and known to increase the specific activities of pancreatic chymotrypsin, trypsi, amylase, and lipase. Nowadays, coffee intake is increased among Koreans who have consumed relatively high amount of traditional fermented foods. Therefore, this study was performed to examine the effect of coffee and AFB1 on pancreatic exocrine function and structure. Rats were divided into 10 experimental groups. The first five groups were W(control group), LD(0.2g decaffeinated coffee/Kg B.W), HD(3g decaffeinated coffee/Kg B.W), LC(0.2g coffee/Kg B.W), and HC(3g coffee/Kg B.W). The second five groups were WA, LDA, HDA, LCA, HCA, same as first five groups in caffieine level but treated with AFB1. The result of this experiment showed that the caffeine intake did not influence significantly on the growth and feed efficiency. But water intake was increased by caffeine intake and AFB1 treatment. The weights of pancreas and liver were increased as the caffeine intake was increased. Trypsin activities were tend to increase in concentrated coffee groups(HD, HC). AFB1 treated groups showed the higher trypsin level than the AFB1 untreated groups. Amylase activities were tend to increase in concentrated coffee groups(HD, HC) of AFB1 untreated animals. AFB1 treated did not show the additional effect on the stimulated amylase secretion by coffee. Lipase activities were tend to decrease in concentrated coffee groups(HD, HC) of AFB1 untreated animals. Lipase activities were increased in the order named WA group, coffee groups, decaffeinated coffee groups in AFB1 treated animals. AFB1 treated groups showed the higher lipase level than AFB1 untreated groups. In the histologic observation of pancreas HCA group showed more dense compound tubuloalveolar glands and proliferation of nuclei than normal. The result suggested a development of a atypia which is ongoing phase to a cancer.
