  • Toxicology screening following treatment with astemizole, a histamine receptor antagonist, at oral doses of 0, 10, 30 and 60 mg/kg was carried out in male cynomolgus monkeys (Macaca fascicularis). No dose-related changes in mortality, clinical signs, body weight changes, food consumption, or urine analysis occurred in any animal compared to the vehicle control. However, the high-dose group showed a decrease in BUN and ALP compared to vehicle control group. In addition, the levels of TG, AST, ALP and CK increased. Although astemizole did not produce significant toxicological changes at any dose tested, we predict that it can cause toxicological changes of the liver and heart based on the changes in the serum parameters related to the heart and liver. The Action Potential Duration (APD) was prolonged in the heart of 60 mg/kg treatment group compared to the control group. The APD increase in 60 mg/kg treatment group along the other related changes in toxicological parameters imply that astemizole has major cardiotoxic effects in the cynomolgus monkey. This study is a valuable assessment for predicting the general toxicity and cardiotoxic effects of antihistamine drugs using nonhuman primates.

  • Since some cyanobacterial isolates under selective culturing conditions are lacking of characteristic specialized cells or showing altered morphology, the morpho-taxonomic criteria are not accurate enough to discriminate between species. Instead of morphological parameters, a method based on the single or the combination of repetitive oligonucleotides in a single PCR, repetitive oligonucleotides-primed PCR (ROP-PCR), was applied to generate DNA profiles for members of the cyanobacterial genera Anabaena and Oscillatoria, both of which are responsible for causing poisonous blooms in various freshwater systems. ROP-PCR performed on 10 isolates of the cyanobacteria with ERIC and REP sequences from gram-negative bacteria, STRR1A and LTRR sequences derived from cyanobacterial genome, and eukaryotic repetitive sequences, led to the identification of distinct genotypes, and provided specific and repeatable DNA fingerprints for cyanobacterial isolates. Grouping analysis of cyanobacterial isolates showed a signifiant difference depending on the primer used in PCR.

  • This study was conducted to effect of extruded pellets (EPs) and moist pellet (MP) diet on growth and fish health parameters in farm cultured olive flounder Paralichthys olivaceus. Two replicate groups of 2,600 fish per each tank (initial weight of $30.1{\pm}0.2$ g) were fed one of the three EPs (EP1, EP2 and CEP) and one MP for 13 months. In field feeding experiment conducted in commercial flounder farm, survival, growth (30-600 g) and condition factor of fish fed experimental EP1 and EP2 were comparable to those of fish fed MP, but considerably higher feed efficiency and protein efficiency ratio were observed in fish fed experimental EPs. Viable cell counts from diet and water from MP diet were considerably higher than those from EP diet, and lysozyme activity from EP diet was higher than that from MP diet. GOT and GPT of fish fed the EP diet were considerably lower than those from MP group, but total protein and glucose concentration of fish fed the EP groups were not significantly different from that of fish fed the MP. Therefore, these results strongly suggest that EP diets could be developed to replace MP diet without adverse effects on growth performance and to enhance the immunity and protective ability of olive flounder against fish disease such as Vibro, Edwardsiella and Strptococcus.

  • Purpose: Eosinophilia is common in premature infants, and its incidence increases with a shorter gestation period. We investigated the clinical significance of eosinophilia in premature infants born at <34 weeks gestation. Methods: We analyzed the medical records of premature infants born at <34 weeks gestation who were admitted to the neonatal intensive care unit at Ewha Womans University Mokdong Hospital between January 2003 and September 2010. Eosinophilia was defined as an eosinophil percentage of >3% of the total leukocytes. Perinatal parameters and clinical parameters were also analyzed. Results: Of the 261 infants born at <34 weeks gestation, 22.4% demonstrated eosinophilia at birth. The eosinophil percentage peaked in the fourth postnatal week at 7.5%. The incidence of severe eosinophilia increased after birth up to the fourth postnatal week when 8.8% of all patients had severe eosinophilia. Severity of eosinophilia was positively correlated with a lower gestational age, birth weight, and Apgar score. Respiratory distress syndrome, bronchopulmonary dysplasia, nephrocalcinosis, intraventricular hemorrhage, and sepsis were associated with a higher eosinophil percentage. The eosinophil percentage was significantly higher in infants with bronchopulmonary dysplasia from the first postnatal week and the percentage was the highest in the fourth postnatal week, with the maximal difference being 4.1% (P<0.001). Conclusion: Eosinophilia is common in premature infants and reaches peak incidence and severity in the fourth postnatal week. The eosinophil percentage was significantly higher in bronchopulmonary dysplasia patients from the first postnatal week. Severe eosinophilia was significantly associated with the incidence of bronchopulmonary dysplasia even after adjusting for other variables.

  • This study investigated the effects of a flavonoid-rich ethanol extract of persimmon leaf (PL), an ethanol extract of Citrus junos Sieb (CJS), and a PL-CJS mixture (MPC) on mice fed a high-fat diet (HFD). We sought to elucidate the mechanisms of biological activity of these substances using measurements of blood coagulation indices and lipid metabolism parameters. C57BL/6J mice were fed a HFD with PL (0.5% (w/w)), CJS (0.1% (w/w)), or MPC (PL 0.5%, CJS 0.1% (w/w)) for 10 weeks. In comparison with data obtained for mice in the untreated HFD group, consumption of MPC remarkably prolonged the activated partial thromboplastin time (aPTT) and prothrombin time (PT), whereas exposure to PL prolonged aPTT only. Lower levels of plasma total cholesterol, hepatic cholesterol, and erythrocyte thiobarbituric acid-reactive substances, hepatic HMG-CoA reductase, and decreased SREBP-1c gene expression were observed in mice that received PL and MPC supplements compared with the respective values detected in the untreated HFD animals. Our results indicate that PL and MPC may have beneficial effects on blood circulation and lipid metabolism in obese mice.

  • One of the problems for implementing the spot detection phase in the 2-DE gel image analysis program is the eliminating noises in the image. Remained noises after the preprocessing phase cause the over-segmented regions by the segmentation phase. To identify and exclude the over-segmented background regions, if we use the fixed thresholding method that is choosing an intensity value for the threshold, the spots that is invisible by the eyes but mean a very small amount proteins which have important role in the biological samples could be eliminated. This paper propose an adaptive thresholding method that come from an idea that is got on statistical analysing for the prominences of the peaks. The adaptive thresholding method works as following. Firstly we calculate an average prominence value curve and fit it to exponential function curve, as a result we get parameters for the exponential function. And then we calculate a threshold value by using the parameters and probability distribution of errors. Lastly we apply the threshold value to the region for determining the region is a noise or not. According to the probability distribution of errors, the reliability is 99.85% and we show the correctness of the proposed method by representing experiment results.

  • In this study, we investigated the effects of ostrich extract combined with Korean herb medicine(OEKHM) on the serum parameters in carageenan induced paw edema in rats. Carageenan causes enhanced edema formation which contributes to the pathophysiology of inflammation. Male Sprague-Dawley rats were used. There were four groups of treatments: OEKHMs and the control. After treatment with oral doses of 3 mL/180 g BW. 3 kinds of OEKHMs were processed which were ostrich bone with flesh meat with herb medicine(BFH), ostrich bone with flesh meat(BF), and ostrich bone with internal organs(BI). The contents of crude fat of the BFH was lower, but crude protein and crude ash of the BFH were significantly higher than those of the BF and the BI. In case of the levels of minerals(Ca, P, Mg, Fe, Cu and Zn ) of the BFH were significantly higher than those of BF and BI. And the heavy metals(Pb and Cd) were not detected in the OEKHM. The serum level of triglyceride in the BFH treated group decreased compared to that of the edema induced group. The serum levels of HDL-cholesterol concentration in the BFH supplemented group increased compared to that of the other groups. The serum levels of minerals(Ca, Mg, Fe, Cu and Zn) of the BFH treated group significantly increased than those of other groups(p<0.05).
