Effect of Feelch on Blood Alcohol Concentration in Human and Hepatic Alcohol Metabolizing Enzyme Activity in Alcohol-Fed Rats (필치가 혈중 알코올농도와 간세포내 알코올대사 효소 활성에 미치는 영향)

  • To investigate the effect of Feelch on alcohol metabolism, we measured both blood alcohol concentration in human and hepatic alcohol metabolizing enzyme activity in rats. The blood alcohol concentration in Feelch-ingested group was significantly lower than that in water-ingested group at 0, 40, and 80 minute after alcohol intake. The blood alcohol concentration between male and female taken 300ml of $21\%$ alcohol showed the significant differences; the peak value of blood alcohol concentration in male and female were $0.083\pm0.014\%\;and\;0.108\pm0.018\%$, respectively. In alcohol-fed rats, aldehyde dehydrogenase (ALDH) activity was significantly increased, whereas alcohol dehydrogenase (ADH) activity was not changed. In both Feelch-fed group and Feelch plus alcohol-fed group, ADH and ALDH activity were significantly increased as compared with each control group. Feelch decreased phospholipase $A_2$ activity and lipid peroxidation in hepatic tissue and activities of serum aminotransferases as compared with control. These results suggest that Feelch may have a hepatoprotective effects and this is likely due to lower blood alcohol concentration via the increment of hepatic ADH and ALDH activity.

Activity of Alcohol Dehydrogenase and Ethanol, Acetaldehyde Levels in Normal Adults Blood (정상인의 혈중 알코올 탈수소효소 활성도 및 에탄올, 아세트알데히드 농도에 관한 연구)

  • To identify normal levels of ethanol, acetaldehyde and alcohol dehydrogenase(ADH) activity in blood of Koreans, ethanol and acetaldehyde levels, activity of ADH in blood of Koreans, ethanol and acetaldehyde levels, activity of ADH in blood were measured in 97 subjects(male : 36, female : 61), 45 subjects(male : 21, female : 24) were not exposed to organic solvents and any other chemicals. Fifty two subjects(male : 15, female : 37) were exposed to organic solvents including toluene and xylene. The results were summerized as follows : 1. The blood ADH was not detected in exposed and non-exposed group. 2. The average blood ethanol level of non-exposed group was 0.0490 mg/dl, and exposed group was 0.0363 mg/dl. They were statistically significant(p<0.05). 3. The blood acetaldehyde levels in exposed group were significantly higher than that of non-exposed group was not statistically significant (p>0.05). 4. The average blood ethanol level of males in both groups was significantly higher than that of females, however, they were not statistically significant (p>0.05).

Comparison of Lines from Anther and Maternally-derived dihaploids, Single-seed Descent and Bulk Breeding Method in Flue-cured Tobacco (Nicotiana tabacum L.) (연초(Nicotiana tabacum L.)의 반수체 육종법 1주1계통법, 집단육종에 의한 육성계통의 특성비교)

  • The present study was conducted to compare the relative efficiency of four different breeding methods in tobacco varietal development. A single Fl hybrid plant from cross of two flue-cured cultivars of Nicotiana tabacum L Bright Yellow 4(BY4) and NC95, was selfed. F2population above cross was screened for resistance against bacterial wilt caused by Pseudommonas solanacearum E.F.Smith under the naturally infested field conditions, and the 30 lines were developed from F2 individual plant by anther culture (ADH), maternal method utilizing Nicotiana afpicana (MDH), single- seed descent(SSD) and Bulk breeding method, respectively. All characters except content of total alkaloids of ADH and MDH which wore bred by haploid methods reduced more than that of lines bred by conventional methods(SSD & Bulk) : however, the yields were 8% lower than other lines. The total alkaloid content of ADH was higher than that of MDH, and yield was reduced about 4n even though the number of leaf was identical with the MDH. All other characters of ADH were also reduced significantly. In the lines bred by conventional methods, population developed by SSD showed significantly wider leaf width, shorter plant height, later days to flower, and lower in percent reducing sugar than those by Bulk. The populations derived from haploid method showed greater phenotypic variance and wider range of variation than conventionally developed ones. The results obtained indicate that selection will provide a significantly greater genetic gain for leaf number and leaf length in the ADU and MDH populations, and for plant height and days to flower in the SSD and Bulk populations.

Activities of Antioxidation and Alcohol Dehydrogenase Inhibition of Methanol Extracts from Some Medicinal Herbs (약용식물 추출물에 대한 항산화성과 알코올 탈수소효소 저해성 연구)

  • The activities of antioxidation and alcohol dehydrogenase in hibitionin methanol extracts of thirty two medical herbs were tested using the method of DPPH activity, nitrite scavenging effect and alcohol dehydrogenase assay in vitro. In DPPH method, Eugenia caryophyllata, Thea sinensis, Paeonia suffruticosa, Alnus japonica showed over 90 % of free radical scavenging activities. The nitrite scavenging ability appeared Zanthoxylum bungeanum, Alnus japonica, Thea sinensis, Hovenia dulcis(cortex) and Illicium verum showed the high value. In connection with in vivo alcohol metabolism, thirteen medicinal herbs were screened for inhibition. As a reasult, we found significant inhibition of ADH by methanolic extracts of Glycyrrhiza uralensis, Pueraria thunbergiana(radix), Alnus japonica. These results indicate that the antioxidative effect was strongly related with alcohol dehydrogenase inhibitor; Thea sinensis and Alnus japonica.

  • Subcellular location of five isozymes in Populus species was studied by isozyme analysis with plant and mitochondria. Isozymes analyzed were ADH, 6-PGD, MDH, PGI, and Diaptrorase. ADH seems to be cytosolic isozyme encoded with nuclear genes at one locus, while 6-PGD seems to be mitochondrial one. MDH showed three hands in cytosol and four other bands in mitochondria. Three cytosolic hands were showed for PGI. Diaphorase showed one mitochodrial band and two cytosofic bands which seemed to the encoded by nuclear genes at one locus.

  • Alcohol dehydrogenase (ADH) and Aldehyde dehydroganase (ALDH) activities of turnip extracts with ultrasonification at $60,\;100^{\circ}C$ temperature were investigated. Ultrasonification extracts showed a increase as compared with the normal extracts. ALDH activities were high than ADH activities. So, turnip extracts was shown efficient plants in alcohol oxidation. Appraisers liked powder extracts better than liquid extracts in sensory score.

The effect of herbal mixture on alcohol metabolism in Sprague Dawley rats (알코올을 투여한 흰쥐의 알코올 대사에 미치는 한약재 혼합추출물의 영향)

  • Objectives : Alcohol hangover is a common phenomenon which basically occurs after heavy drinking. Moreover, heavy alcohol consumption leads to acute and chronic diseases. We investigated the effect of herbal mixture (SJ) on alcohol metabolism in serum or/and liver. Methods : 5W-old Sprague Dawley rats were used for the study. To overnight fasted rats, 0.9% saline or SJ extract was administrated per os before alcohol treatment. Then, 40% alcohol was orally administrated to all rats in 30 mins. Ethanol, acetaldehyde concentrations, alcohol dehydrogenase (ADH), acetaldehyde dehydrogenase (ALDH) activities were measured by assay kits. Aspartate aminotransferase (AST) and alanine transaminase (ALT) were measured by analyzer. Results : Ethanol and acetaldehyde concentrations lowered in SJ groups compared with CON group. Especially, acetaldehyde concentration significantly decreased in SJ groups compared with CON group. AST and ALT levels tended to increase in SJ groups compared with CON group but there was no significant difference between CON group and SJ groups. ADH activity in serum was higher in SJ groups than CON group but no significant difference in liver. ALDH activity in both serum and liver showed significantly increased in SJ groups compared with CON group. Conclusions : Treatment of SJ extract showed not only reducing concentration of alcohol and acetaldehyde but also increasing activities of ADH and ALDH. These results suggest SJ may influence in alcohol metabolism via control of metabolic enzyme activities and metabolite. Therefore, SJ, herbal mixture, might have a function of preventing hangover after drinking alcohol.

The Variation of Isoenzymes and Morphological Characteristics of Needle, Cone and Seed According to Resin Duct Index in Pinus densiflora and Pinus thunbergii (소나무 및 곰솔의 수지구지수(樹脂溝脂數)에 따른 침엽(針葉), 구과(毬果) 및 종자(種子)의 형태적(形態的) 특성(特性)과 동위효소(同位酵素)의 변이(變異))

  • The variation of needle, cone and seed characteristics and of allelic frequency in isoenzyme, ADH, ME and PGI, according to different resin duct index in Pinus densiflora, Pinus thunbergii and their hybrids was analyzed. The results obtained were as follows : 1. With increase of number of resin ducts, morphological characteristics such as needle length, needle sheath length, cone size, seed size, seed wing size, 1000 seeds weight, etc. tended to he increased, while number of stomata row in needle to be decreased. 2. As the results of discriminant analysis for the morphological characteristics of needle, cone and seed, most individuals are generally coincided with number of resin duct mostly in Pinus densiflora were not. 3. According to the canonical discriminant function obtained from the morphological characteristics in Pinus densiflora, Pinus thunbergii and their hybrids including introgressive hybrid, the resin duct index, 1000 seeds weight, cone size and neelde sheath length characterized fairly their species. 4. With increase of resin duct index, hybrid index tended to be higher. The results obtained from the discriminant analysis and the hybrid index were nearly same each other. 5. With increase of number of resin duct, the allelic frequencies for isoenzyme, ADH-$B_2$, ME-$A_2$ and PGI-$B_1$. $B_2$ tended to increase but those of ADH-$B_3$, ME-$A_4$, and PGI-$B_3$, to decrease. It is thought that this increase of frequency for the former four isoenzymes was resulted in introgressive gene flow from Pinus thunbergii to Pines densiflora and accordingly the frequency of latter three isoenzymes tended to decrease.

Effect of Protein and Fiber Levels on Ethanol-Induced Brain Damage in Rats

  • The purpose of this study was to investigate the effect of protein and dietary fiber levels on the activities of ehanol metabilizing enzymes of the brain in acute and chronic ethanol-treated rats. Male Sprague-Dwley rats were fed on diets containing two levels of protein(7%, 20%)) with two levels of fiber(5%, 105) for 5 weeks. Rats were orally administered 40% (v/v) ethanol(5g/body weight) 90 min before decapitation in the acute ethanol-treated groups and 25% (v/v) ethanol (5g/kg body weight) once a day for 5 weeks in the chronic ethnol-treated groups. Cytosilic alcohol dehydrogenase (ADH) activities were higher than those of mitochondrial ADH. The ADH activities were increased by 20% protein and %% fiber levels in the diet in two fractions , but were decreased by chronic ethanol treatment. Mitochondrial aldehyde dehydrogenase (ALDH) activities did not change by ethanol treatment but were increased by the 20% protein level. However, cytosilic ALDH activities were decreased by chronic ethanol treatment at the 5% fiber level and did not change with protein levels. Both ALDH activities were higher in the 10% fiber groups than the 5% fiber groups. Cytochrome P-450 contents were significantly increased in the chronic ethanol-treated groups but xanthine oxidase (XO) activities did not change. P-450 contents and XO activities were significantly decreased in both the low protein and fiber groups.

Inflammation and hyponatremia: an underrecognized condition?

  • Timely diagnosis of hyponatremia is important for preventing potential morbidity and mortality as it is often an indicator of underlying disease. The most common cause of eurvolemic hyponatremia is the syndrome of inappropriate antidiuretic hormone (SIADH) secretion. Recent studies have demonstrated that proinflammatory cytokines such as interleukin (IL) $1{\beta}$ and IL-6 are involved in the development of hyponatremia, a condition that is associated with severe inflammation and is related to antidiuretic hormone (ADH) secretion. Serum sodium levels in hyponatremia are inversely correlated with the percentage of neutrophils, C-reactive protein, and N-terminal-pro brain type natriuretic peptide. Additionally, elevated levels of serum IL-6 and IL-$1{\beta}$ are found in inflammatory diseases, and their levels are higher in patients with hyponatremia. Because it is significantly correlated with the degree of inflammation in children, hyponatremia could be used as a diagnostic marker of pediatric inflammatory diseases. Based on available evidence, we hypothesize that hyponatremia may be associated with inflammatory diseases in general. Understanding the mechanisms responsible for augmented ADH secretion during inflammation, monitoring patient sodium levels, and selecting the appropriate intravenous fluid treatment are important components that may lower the morbidity and mortality of patients in a critical condition.
