Extraction, Purification and Property of the lipid from Scenedesmus sp. (Scenedesmus sp.로 부터의 지방 추출, 정제 및 특성)

  • Lipid from Scenedesmus sp. was extracted, fractionated and purified by silicic acid column chromatography. Total lipid content of extract was $21.38{\pm}1.03$ wt%, and triacylglycerol, an index lipid for biodiesel production was detected. Ten species of $C_{16}{\sim}C_{22}$ with saturated and unsaturated fatty acid were identified showing the very adequate fatty acid profiles for biodiesel production with a good flow property at low temperature. The fractions of glycolipid, neutral and phospholipid were 52.64, 28.10 and 19.26% of total lipid, respectively. The triacylglycerol of 12.63% and chlorophyll a of 49.47% was fractioned using stepwise elution of n-hexane-diethyl ether (95:5, v/v) solvent. The high content of chlorophyll was considered as a potential source of value-added co-product.

Influence of Different Strains of Agrobacterium rhizogenes on Hairy Root Induction and Rosmarinic Acid Production in Agastache rugosa Kuntze (Agrobacterium rhizogenes strains이 배초향 모상근 유도와 Rosmarinic acid 생산에 미치는 영향)

  • Rosmarinic acid, an ester of caffeic acid with 3,4-dihydroxyphenyl lactic acid, is one of the main active constituents of Agastache rugosa Kuntze and has an astringent property, antioxidant capacity, anti-inflammatory activity, antimutagenic ability, antimicrobial capacity, and an antiviral property. Five different strains of Agrobacterium rhizogenes differed in their ability to induce Korean mint (Agastache rugosa Kuntze) hairy roots and also showed varying effects on the growth and rosmarinic acid production in hairy root cultures. A. rhizogenes R1601 is the most effective strain for the induction (72.90%), growth (13.50 g/l) and rosmarinic acid production (22.60 mg/g) in hairy root of Korean mint. Our results demonstrate that use of suitable strains of A. rhizogenes may allow study of the regulation of rosmarinic acid biosynthesis in hairy root cultures of Agastache rugosa.

  • Lithium dual complex grease was prepared through the second continuous saponification reaction of a complex gellant system whose essential components comprised of a selected hydroxy fatty acid, lithium hydroxide monohydrate and boric acid to have a fiber structure of chemical, thermal and mechanical stability at high temperatures. An optimum amount of complex gellant was found to be 14% (NLGI #2), and an addition of castor wax of 1.5% provided an excellent performance properties, especially. The oil separation, oxidation stability, water wash-out property, shear stability, extreme pressure and wear property of thus prepared were tested by the ASTM and KS methods, and a characteristic result was obtained.

  • The high viscous ${\gamma}$-polyglutamic acid(${\gamma}$-PGA) from alkalophilic Alcaligenes sp. was purified and its solution property was investigated. The intrinsic viscosities for Na+ form and H+ form of ${\gamma}$-PGA were 31.1 and 0.38d$\ell$/g, respectively. The viscosity of H+-PGA was not influenced by pH or salts while that of Na+-PGA was influenced. The intrinsic viscosity of Na+-PGA solution decreased remarkably at the alkaline or acidic pH and showed the sharp decrease when salts were added. ${\gamma}$-PGA exhibited the property of the polyelectrolyte showing the .sharp decrease of intrinsic viscosity by the addition of NaCl, and intrinsic viscosity of dilute solution with the low concentration of NaCl was exponentially dependent on temperature and its temperature dependency increased with increasing NaCl concentration. The chain stiffness, coil overlap parameter and critical concentration of Na+-PGA were 0.08, 5.25 and 0.07g/d$\ell$, respectively.

Effect of Storage and Marketing Condition on Biochemical Property Changes of Garlic (Allium sativum L.) (마늘(Allium sativum L.)의 생화학적 변화에 대한 저장 및 유통조건의 영향)

  • Biochemical property changes of garlic during various storages and marketing after storage were investigated. Content of enzymatic pyruvic acid increased by room and low temperature storage but decreased by CA and MA storage. Fructan contents decreased rapidly by low temperature storage, but restrained decrement by CA and MA storage. Free sugar increased during storage, but did slowly by room temperature storage. Green pigment development was observed when garlics stored for 90days at low temperature were processed into crushed form. This discoloration was small for garlics stored in CA and MA, and never occurred for room temperature stored garlics. When marketed after storage, content of enzymatic pyruvic acid decreased in garlic stored in room and low temperature storage, but increased in garlics which decreased during CA and MA storage. Fructan contents deceased but free sugar contents continuously increased with marketing period. Green pigment development decreased in crushed garlic after 30days at room temperature marketing, but increased in low temperature marketing with marketing time progress.

Effects of Spinning Speed and Heat Treatment on the Mechanical Property and Biodegradability of Polylactic Acid Fibers (제사속도와 열처리에 따른 polylactic acid 섬유의 물성 및 생분해성 변화)

  • This study was carried out to suggest the optimal spinning process condition which provides a proper range of tenacity and biodegradability as textile fibers. The effects of the melt spinning speed and heat treatment on the mechanical property and biodegradability of polylactic acid fiber were investigated. Polylactic acid(PLA) was spun in a high spinning speed of $2000{\sim}4000m/min$. Each specimen was heat-treated at $100^{\circ}C$ during 30min. Mechanical properties such as breaking stress and the degree of crystallinity were evaluated using WAXS. Biodegradability was estimated from the decrease of breaking stress, weight loss, and the degree of crystallinity after soil burial. Experimental results revealed that heat treated specimens showed higher breaking stress than untreated specimens, but the increase was not so high as was expected from the remarkable change of crystallinity by heat treatment. It was concluded that breaking stress was more influenced by spinning speed than heat treatment. In the soil burial test, however biodegradability calculated from weight loss was more influenced by heat treatment than spinning speed.

  • A paulownia wood has been widely used with various advantages as its low weight, permeability, convenient workability and aesthetic patterns for a long time. However, the related empirical researches and simultaneous evaluations of functionality are insufficient compared with acid-free archival boxes for now. In this study, the indoor and outdoor temperature and relative humidity control and heat release rate were evaluated under the controlled and uncontrolled circumstance in 2018. The paulownia wood storage box showed superior control effect of relative humidity than the acid-free archival box in constantly uncontrolled environment. Also, the possibility of the flame diffusion from the surface of the materials was higher for the paulownia materials, and the acid-free archival box showed more dangerous patterns in the early stages of the fire.

  • The thermal treatment for carbon nanotube was applied to remove the water, metal catalyst and other impurities in carbon nanotube. The surface of carbon nanotube was changed into open structure with acid treatment by mixed solution of $HNO_3$ and $H_2SO_4$. The dispersion property of the functionalized and modified carbon nanotube was assessed with naked eyes by dispersing it in DMF. Carbon nanotube mixd polyurethane film was made to estimate the dispersion property by reflectance of the film with UV-Vis spectrometer. Also the internal structure of carbon nanotube was observed with SEM and TEM and thermal pyrolysis property of the carbon nanotube was measured by TGA and DSC. The surface modification of carbon nanotube by thermal and acid treatments improved the dispersion property of carbon nanotube/polyurethane mixed materials.

Sensory Property Improvement of Jokbal (Korean Pettitoes) Made from Frozen Pig Feet by Addition of Herbal Mixture

  • This study was conducted to improve sensory quality of Jokbal (Korean Pettitoes) made from frozen pig feet by addition of herbal mixture (glasswort, raspberry and Sansa powders). After adding herbal mixture, lipid oxidation (2-thiobarbituric acid values, TBARS), sensory property, and textural property were determined. Herbs were individually added into cooking soup at concentration of 6% (low concentration treatment, LCT) or 12% (high concentration treatment, HCT) of raw pig feet. Refrigerated pig feet were used as control. Thawed feet without any herbal mixture were used as freezing treatment (FT). TBARS in LCT or HCT were lower than that in FT, and showed the similar to that in Control. Addition of the herbal mixture was effective in improving the flavor and textural property of thawed feet by inhibiting lipid oxidation and protein denaturation in a dose-dependent manner.

  • Physical and chemical properties of thexylboronic acid and its derivatives such as thexylboroxine and ethylene glycol or diethanolamine thexylboronic ester have been studied. Thexylboronic acid can be extracted from an organic solution with an aqueous sodium hydroxide solution as an “te”complex. It is readily converted into thexylboroxine in the presence of anhydrous magnesium sulfate in pentane. It reacts with simple alcohols only slowly; however, it reacts readily with excess diethanolamine in the presence of anhydrous magnesium sulfate to give the corresponding ester.
