Anticancer activity of subfractions containing pure compounds of Chaga mushroom (Inonotus obliquus) extract in human cancer cells and in Balbc/c mice bearing Sarcoma-180 cells

  • The Chaga mushroom (Inonotus obliquus) has been used in folk medicine to treat cancers. However, limited information exists on the underlying anticancer effects of the major component of I. obliquus in vivo. We hypothesize that the pure compounds ($3{\beta}$-hydroxy-lanosta-8,24-dien-21-al, inotodiol and lanosterol, respectively) separated from I. obliquus would inhibit tumor growth in Balbc/c mice bearing Sarcoma-180 cells (S-180) in vivo and growth of human carcinoma cells in vitro. To test this hypothesis, the growth inhibition of each subfraction isolated from I. obliquus on human carcinoma cell lines (lung carcinoma A-549 cells, stomach adenocarcinoma AGS cells, breast adenocarcinoma MCF-7 cells, and cervical adenocarcinoma HeLa cells) was tested in vitro. Then, after S-180 implantation, the mice were fed a normal chow supplemented with 0, 0.1 or 0.2 mg of subfraction 1, 2 or 3 per mouse per day. All of the subfractions isolated from I. obliquus showed significant cytotoxic activity against the selected cancer cell lines in vitro. Subfraction 1 was more active than subfraction 2 and subfraction 3 against the A549, AGS and MCF-7 cancer cell lines in vitro. In in vivo results, subfraction 1 isolated from I. obliquus at concentrations of 0.1 and 0.2 mg/mouse per day significantly decreased tumor volume by 23.96% and 33.71%, respectively, as compared with the control. Subfractions 2 and 3 also significantly inhibited tumor growth in mice bearing S-180 as compared with the control mouse tumor. Subfraction 1 isolated from I. obliquus showed greater inhibition of tumor growth than subfractions 2 and 3, which agrees well with the in vitro results. The results suggest that I. obliquus and its compounds in these subfractions isolated from I. obliquus could be used as natural anticancer ingredients in the food and/or pharmaceutical industry.

자양대보탕(滋養大補湯)의 발효 전·후 항염증, 항암, 항산화 효과 비교 연구 (Comparison of Jayangdaebo-tang before and after fermentation on antioxidant, anti-inflammatory and anticancer effects)

  • Objectives : In this study, it was investigated the anti-inflammatory, anticancer, and antioxidant effects of Jayangdaebo-tang (JDT) consisting of twelve herbs before and after fermentation. Methods : JDT extract was fermented using the Lactoplantibacillus plantanum (JDT-L), Bacillus subtilis (JDT-B), and L. plantanum plus B. subtilis (JDT-L+B). The effects of each extract were measured in LPS-stimulated RAW264.7 cells, MCF-7 breast cancer and A549 lung cancer cells, and H2O2-stimulated HepG2 cells. Results : The extracts of JDT-L, JDT-B and JDT-L+B at 1 ㎎/㎖ decreased significantly the levels of nitric oxide (NO) in LPS-treated RAW264.7 cells and also inhibited the expression of iNOS and COX-2, and the phosphorylation of ERK and NF-κB. The JDT-L+B extract decreased significantly the expression of apoptotic proteins, Bax, cleaved caspase-3, and PARP in MCF-7 and A549 cancer cells. The JDT-L, JDT-B and JDT-L+B extracts increased significantly the cell viability in H2O2-stimulated HepG2 cells and the JDT-L+B extract decreased significantly the expression of SOD, catalase, HO-1, and NRF-2. Among fermented JDT extracts, JDT-L+B was the best effective on response of macrophage inflammation, cancer cell apoptosis, and liver cell damage. Conclusions : Our results were suggested that the fermentation can be used as a useful way to enhance the biological activity of JDT.

Cytotoxic Constituents from Notopterygium incisum

  • The MeOH extract of Notopterygium incisum showed a strong cytotoxicity against B16 murine melanoma cell line. From this extract three furanocoumarins including bergamottin, isoimperatorin, notopterol and one polyacetylenic compound (falcarindiol) together with one phenylpropanoid (caffeic acid methyl ester) and one triterpenoid (pregnenolone) were isolated. The isolated compounds were evaluated for cytotoxic activity against four kinds of cancer cell lines, e.g. P388 (murine lymphocytic leukemia), B16 (murine melanoma), A549 (human lung carcinoma) and SK-OV-3 (human ovarian cancer). Among the isolates, falcarindiol and caffeic acid methyl ester expressed a significant antiproliferative activity against all tested cell lines.

페렴구균 ClpP의 면역 교차 반응과 방어효과 (Cross-reactivity and Protective Immunity of Streptococcus pneumonieae ClpP)

  • ClpP is a stress-inducible protein and proteolytic subunit of the ATP-dependent Clp protease in prokaryotes and eukaryotes. Although its physiological roles in bacterial virulence were widely studied in various organsims, including Streptococcus pneumoniae, until now the immunological effect has not been investigated. Here, we have examined the cross reactivity of S. pneumoniae ClpP antibody with other organisms's cell lysate proteins. Although the protein sequence of S. pneumoniae ClpP was highly conserved among various organisms including human, the antibody rasised by S. pneumoniae ClpP was not cross-reacted with other organism's cell lysates, which were Saccharomyces cerevisiae , human lung A549 cell, Bacillus subtilis, Pseuomonas aeruginosa, E. coli, and Salmonella typhi. It was only reacted with S. pneumoniae and Lato-bacillus thermophilus. Thus we examined the immunoprotective effect of ClpP by immunizing mice with the purified ClpP. The mean survival time of mouse was significantly increased with the ClpP immunization. These results suggest that S. pneumoniae ClpP could be used as a vaccine candidate for prevention of S. pneumoniae infection.

  • Design and synthesis of novel 4-phenyl-1-arylsulfonylimidazolones 3 and 4-phenyl-3-arylsulfonylimidazolones 4 and evaluation of their cytotoxic activity against eleven human cancer cell lines and two murine leukemia cell lines in vitro were performed. As a result, a series of 4-phenyl-1(N)-arylsulfonylimidazolones (3) has been found to be the potential anticancer agent. Compounds 3b, 3c, and 3d exhibit strong activity as indicated by their $IC_{50}$ values 0.39, 3.19, $0.31{\mu}g/mL$ against A549 and 0.80, 0.48, $0.0007{\mu}g/mL$against SK-Mel-2, respectively. These compounds also possess much more potent activity (10-1000 times) than LY186641 against eleven other cell lines.

Antitumor Activity of LB42907, a Potent and Selective Farnesyltransferase Inhibitor: Synergistic Effect in Combination with Other Anticancer Drugs

  • Inhibitors of farnesyltransferase (FT), a key enzyme in the post-translational modifications of Ras proteins, have been extensively studied as novel anticancer agents in the preclinical stages, some of which are currently in clinical development. Previously, it has been reported that a novel FT inhibitor LB42907 inhibits Ras farnesylation in the nanomolar range in vitro. The aim of this study was to assess the antitumor efficacy of LB42907 in vitro and in vivo. Anchorage-independent growth of various human tumor cell lines was potently inhibited by treatment with LB42907, comparable to other FT inhibitors in clinical development. In the nude mouse, oral administration of LB42907 demonstrated potent antitumor activity in several human tumor xenograft models including bladder, lung and pancreas origin. Interestingly, significant tumor regression in EJ (bladder) and A549 (lung) xenografts was induced by LB42907 treatment. The effectiveness of LB42907 was also investigated in simultaneous combination with paclitaxel, vincristine, cisplatin or gemcitabine against NCI-H460, A549, and HCT116 cells in vitro using median-effect analysis. LB42907 markedly synergized with most anticancer drugs tested in this study in NCI-H460 cell. In contrast, LB42907 displayed antagonism or partial synergism with these drugs in A549 and HCT116 cells, depending on the class of combined drugs and/ or the level of cytotoxicity. Our results demonstrate that LB42907 is an effective antitumor agent in vitro and in vivo and combination of LB42907 with other chemotherapeutic drugs results in synergistic or antagonistic effects mainly in a cell line-dependent manner. Further preclinical study is warranted.

In Vitro Anticancer Activities of Anogeissus latifolia, Terminalia bellerica, Acacia catechu and Moringa oleiferna Indian Plants

  • The present study was designed to evaluate in vitro anti-proliferative potential of extracts from four Indian medicinal plants, namely Anogeissus latifolia, Terminalia bellerica, Acacia catechu and Moringa oleiferna. Their cytotoxicity was tested in nine human cancer cell lines, including cancers of lung (A549), prostate (PC-3), breast (T47D and MCF-7), colon (HCT-16 and Colo-205) and leukemia (THP-1, HL-60 and K562) by using SRB and MTT assays. The findings showed that the selected plant extracts inhibited the cell proliferation of nine human cancer cell lines in a concentration dependent manner. The extracts inhibited cell viability of leukemia HL-60 and K562 cells by blocking G0/G1 phase of the cell cycle. Interestingly, A. catechu extract at $100{\mu}g/mL$ induced G2/M arrest in K562 cells. DNA fragmentation analysis displayed the appearance of a smear pattern of cell necrosis upon agarose gel electrophoresis after incubation of HL-60 cells with these extracts for 24h.

된장 숙성기간중의 항산화 및 암세포 생육 억제효과 (Antioxidant and Anticarcinogenic Effects of Traditional Doenjang during Maturation Periods)

  • 장기간 자연숙성시킨 전통된장의 항산화 효과는 숙성기간이 길어질수록 대체로 감소되었으며, 숙성기간 1년 된장에서 methanol 분획물이 가장 우수하였으며, 다음으로 hexane과 물 분획물의 순이었다. 지용성 및 수용성 추출물은 숙성기간이 길어질수록 항산화 효과가 점진적으로 오히려 증가되었으나, 분획물에 비하여 낮은 수치를 나타내었다. 전통된장의 수소공여능은 수용성 색소 추출물과 methanol 및 butanol 분획물에서 대체적으로 높은 수치였으나, chloroform과 ethylacetate 분획물은 매우 낮았다. 전통된장의 항암효과에서 인체 폐암 세포주(A549)는 물 분획물에서 가장 높았고, 인체 유방암 세포주(MCF-7)는 methanol 분획물에서 가장 높은 수치를 나타내었으며, 특히 전통된장의 숙성기간이 길어질수록 암세포 성장억제효과는 높게 나타났다.

  • 본 연구에서 국내 연근해에 서식하는 해면 Sarcotragus sp. (Dictyoceratida속)에서 분리 추출된 7종의 furanoterpenoid계 화합물〔sarcotin A, epi-sarcotin A, ircinin-1, epi-sarcotrine B, sarcotin I, (8E,13Z,20Z) -strobilinin/ (7E,13Z,20Z) -felixinin and (7E,12E,18R,20Z)-variabilin〕의 항암 활성을 비교하기 위하여 A549 인체폐암세포를 대상으로 그들의 세포독성을 조사하였고, 이와 연관된 세포증식 억제 및 apoptosis 유발에 관여할 것으로 예상되는 중요한 유전자 몇 가지의 발현에 미치는 영향을 조사하였다. 조사된 7종의 화합물 모두 처리 농도 의존적으로 A549 폐암세포의 증식을 억제하였는데, 그중 sarcotin A 및 (7E,12E,18R,20Z)-variabilin이 비교적 높은 세포독성을 나타내었다. 이러한 세포증식의 억제는 종양억제 유전자 p53 의존적 또는 비의존적으로 Bcl-2 유전자에 대한 Bax의 발현 증가와 연관된 apoptosis 유발과 관련이 있었으며, epi-sarcotin A, ircinin-1 및 epi-sarcotrine B 처리군에서 이러한 현상은 두드러지게 관찰되었다. 또한 epi-sarcotin A와 ircinin-1은 COX-1의 발현에는 아무런 영향을 미치지 않았으나, COX-2의 발현은 선택적으로 저해하였다. 이러한 결과는 해양 해면동물에서 유래된 furanoterpenoid계 화합물이 선택 적으로 강력한 항암효과를 가질 수 있다는 것을 의미한다.
