Bacillus vallismortis EXTN-1-Mediated Growth Promotion and Disease Suppression in Rice

  • Bacillus vallismortis EXTN-1, a biocontrol agent in cucumber, tomato and potato was tested in rice pathosystem against rice fungal pathogens viz. Magnaporthe grisea, Rhizoctonia solani and Cochliobolus miyabeanus. Apart from increasing the yield in the bacterized plants (11.6-12.6% over control), the study showed that EXTN1 is effective in bringing about disease suppression against all the tested fungal pathogens. EXTN-l treatment resulted in 52.11% reduction in rice blast, 83.02% reduction in sheath blight and 11.54% decrease in brown spot symptoms. As the strain is proven as an inducer for systemic resistance based on PR gene expression in Arabidopsis and tobacco models, it is supposed that a similar mechanism works in rice, bringing about disease suppression. The strain could be used as a potent biocontrol and growth-promoting agent in rice cropping system.

  • A total of 19 selections derived from 4 sources of peppers with resistance to gray leaf spot (KC43, KC47, KC220, and KC319) were tested for their nuclear genotype of the gene conferring the ability to restore the cytoplasmic male sterility. All the selections derived from KC220 and KC319 were maintainers with a genotype of Nrfrf, while all the selections from KC43 and KC47were restorers with a genotype of N(S)RfRf.

  • Phytoterpenoid isolated from Artabotrytis odoratissimus inhibited spore germination of some plant pathogenic as well as saprophytic fungi e.g. Alternaria alternata, A. solani, Cercospora sp., Curvularia maculans, C. pennisetti, Fusarium udum, Helminthosporium echinochlova, H. frumentacie, H. penniseti and Ustilago cynodontis. In Curvularia maculans and H. frumentacie, spore germination was completely inhibited at 2000 ppm. However, Curvularia maculans and C. pennisetti showed considerable sensitivity to this chemical even at 500 ppm.

  • The tetrahydropahnatine alkaloid was assayed against spore germination of some saprophytic and pathogenic fungi e.g., Alternaria solani, A. brassicicola, A. brassicae, A. alternata, Erysiphe pisi, Curvularia lunata, C. pallescens, C. maculans, Curvularia species, Colletotrichum species, C. musae, Helminthosporium echinoclova, H. pennisetti, H. spiciferum, and Heterosporium sp. It inhibite spore germination of all the fungi tested. Colletotrichum spp. Curvularia lunata, Helminthosporium spiciferum and Heterosporium sp. were most sensitive as complete inhibition of spore germination was observed at very low concentration(200 ppm).

Effect of 1-corydalmine, an Alkaloid Isolated from Corydalis chaerophylla Roots on Spore Germination of Some Fungi

  • 1-Corydalmine, an alkaloid isolated from roots of Corydalis ehaerophylla inhibited spore germination of some plant pathogenic as well as saprophytic fungi e.g. Alternaria brassicae, A. brassicieola, A. solani, Curvularia lunata, C. maculans, C. sp., C. pallseens, Erysiphe pisi, Fusarium udum, Helminthosporium species, H. penniseti and a Heterosporium species. 1-Corydalmine significantly inhibited spore germination of all the fungi at 100 to 1500 ppm. It was effective against all the fungi at 1500 ppm. C. lunata was highly sensitive to this chemical even at 250 ppm.

Antifungal Activity of Chaerophylline and Berberine Hydroxide Isolated from Corydalis Species

  • Chaerophylline and berberine hydroxide, both being alkaloids, isolated from Corydalis chaerophylla and C. longipes respectively were assayed against spore germination of some fungi, e.g. Alternaria solani, A. brassicola, A. brassicicola, Helminthosporium penniseti, Helminthosporium sp., Heterosporium sp., Curvularia penniseti, C. maculens and C. palliscens. While chaerophylline inhibited spore germination of most of the fungi at 1000 ppm, being also effective at 50, 100, 200 and 500 ppm, berberine hydroxide was significantly effective at much lower concentration, i.e., 400 ppm against several fungi. This compound was also effective against some fungi at 50, 100, 150, 200 ppm. There was 100% inhibition of spore germination in several fungi at highest concentration of both the compounds. Some of the fungi showed similar results even at lower concentrations.

Cloning, Characterization and Antifungal Activity of Defensin Tfgd1 from Trigonella foenum-graecum L.

  • Defensins are small cysteine rich peptides with a molecular mass of 5-10 kDa and some of them exhibit potent antifungal activity. We have cloned the coding region of a cDNA of 225 bp cysteine rich defensin, named as Tfgd1, from the legume Trigonella foenum-graecum. The amino acid sequence deduced from the coding region comprised 74 amino acids, of which the N-terminal 27 amino acids constituted the signal peptide and the mature peptide comprised 47 amino acids. The protein is characterized by the presence of eight cysteine resisdues, conserved in the various plant defensins forming four disulphide bridges, which stabilize the mature peptide. The recombinant protein expressed in E coli exhibited antifungal activity against the broad host range fungus, Rhizoctonia solani and the peanut leaf spot fungus, Phaeoisariopsis personata.

  • An isolate, 90-GT-302, was found to produce antibiotics inducing typical mycelial swelling in Magnaporthe grisea and Fusarium solani. This isolate formed yellow substrate and white rectiflexbiles aerial mycelia in the early stages of growth. The aerial mycelium gradually changed its color to white and finally formed a gray spore mass. Analysis of the cell wall acid hydrolysate revealed the presence of LL- and meso-diaminopimelic acids, glycine, and galactose, which indicated cell wall type X. This result placed our isolate in genus Kitasatosporia. A comparison of isolate 9O-GT-302 with reference strains of Kitasatosporia spp., which not only demonstrated several differences in their physiological properties but also novelty of the active compounds produced by this isolate, led us to designate the isolate as Kitasatosporia kimorexae.

Designing and Synthesis of Antifungal Active Macrocyclic Ligand and Its Complexes Derived from Diethylphthalate and Benzidine

  • Three novel complexes of Cu(II), Co(II) and Zn(II) using a macrocyclic ligand derived by the condensation of diethylphthalate and benzidine have been designed, synthesized and characterized by UV-Vis., IR, Mass and Elemental analyses data in order to find out their antifungal activities. The stoichiometry of the complexes has been found to be 1:1 (Metal: Ligand). The analytical data indicate that the complexes exhibit square-planar geometry. The antifungal activity of the macrocyclic ligand and its metal complexes has been screened in vitro against fungi such as Aspergillus niger, A. flavus, Trichoderma harizanum, T. viridae and Rhizoctonia solani.

  • This study is aimed to clarify on the influence of cultivated conditions and fungicides on development of Rhizoetonia diseases of zoysiagrass and creeping bentgrass and results are as follows. Infection rates of disease were observed more serious when inoculum amounts were higher than low. Effects of the watering intervals on the severity of disease with artificial inoculating conditions, infection rates were higher when two days watering interval for creeping bentgrass and one day interval for zoysiagrass. Disease severity of creeping bentgrass incited by Rhizoetonia solani AG 1 (1A) was grater under conditions of double amounts of nitrogen fertilization. A posi- tive correlation was founded between the pH and disease development. The pots with pH 5 and 8 were significantly more diseased grass than those in which the pH was in 6.5. Mepronil and myclobutanil were extremely effective against brown patch of creeping bentgrass and large patch of zoysiagrass and followed by capro, and tebeonazole. Fenari was inferior effect to both diseases.
