Effects of Intercrops on Growth and Yield of Fritizzaria thunbergii MIQ (간작물 도입이 패모의 생육 및 수량에 미치는 영향)

  • This experiment on intercropping system of Fritillaria thunbergii MIQ, using safflower, young radish, sesame, and periller as intercrops, had been carried out to increase the productivity of natural resources to the fullest and to use farm land most efficiently for 3 years from 2001 to 2003. Every intercrop could be grown up without any harmful effect on the growth of Fritillaria thunbergii MIQ. There was little difference in bulb yield plant of Fritillaria thunbergii MIQ. per among intercropping systems, but there was more bulb yield per the unit area in single cropping than in intercropping system. The income of cropping system containing Fritillaria thunbergii MIQ. was higher $45\%$ and $35\%$ in Fritillaria thunbergii MIQ.+young radish and Fritillaria thunbergii MIQ.+safflower+perilla than in Fritillaria thunbergii MIQ. mono-crop system respectively.

Study on practical usage of Fritillaria Bulb and Fritillaria Thunbergii Bulb in formulas (천패모(川貝母)와 절패모(浙貝母)의 처방활용(處方活用)에 관한 고찰(考察) - 동의보감(東醫寶鑑)과 방약합편(方藥合編)을 중심으로 -)

  • In this study, we considered the usecase of formulas containing either Fritillaria Bulb (川貝母) or Fritillaria Thunbergii Bulb (浙貝母). These two medicinal materials are distinguished in the Korea Pharmacopoeia and circulating the market separately. But only term "貝母" is traditionally written for describing formulas, and the proper material will have to be determined by a subjective point of view. So, we have comparatively studied the nature, flavor, meridian entry, effect and indication of two materials first. Then we have chosen 54 formulas that "貝母" was used as a primary material from the Donguibogam and the Bangyachappyun, and we have compared effects, indications of two materials and indication of 54 formulas. Finally, we have suggested proper materials for each formula by referring the results above and sentences, which is explaining formulas and the roles of materials in the combination for treatment. The analysis showed that Fritillaria Bulb is proper to 15 formulas, Fritillaria Thunbergii Bulb is proper to 31 ones. This number is 85.2% of 54 formulas and this result shows that we need to distinguish between two materials for "貝母" in the formulas from old traditional literature. For the rest 8 formulas (14.8%), we could not analyze which material is proper or not.

Plant Virome Analysis by the Deep Sequencing of Small RNAs of Fritillaria thunbergii var. chekiangensis and the Rapid Identification of Viruses

  • Thunberg fritillary (Fritillaria thunbergii), a perennial used in traditional Chinese herbal medicine, is a members of the family Liliaceae. The degeneration of germplasm is a severe problem in the production of Fritillaria thunbergii var. chekiangensis. However, no information about viral infections of F. thunbergii var. chekiangensis has been reported. In this study, we sequenced the small RNAs of F. thunbergii var. chekiangensis from leaves and bulbs, and viruses were identified using a phylogenetic analysis and BLAST search for sequence. In addition, multiplex reverse transcriptase-polymerase chain reaction (RT-PCR) was used to rapidly detect viruses in this variety. Our study first reported that five viruses infected F. thunbergii var. chekiangensis. Among them, fritillary virus Y (FVY), lily mottle virus (LMoV), Thunberg fritillary mosaic virus (TFMV), and hop yellow virus (HYV) had been reported in F. thunbergii, while apple stem grooving virus was first reported in the genus Fritillaria. A multiplex RT-PCR method was developed to rapidly test the four viruses FVY, LMoV, TFMV, and HYV in F. thunbergii var. chekiangensis. Our results provide a better understanding of the infection of F. thunbergii var. chekiangensis by viruses and a basic reference for the better design of suitable control measures.

Ultrastructural Difference and Intercellular Transport of Metabolites in Old and New Bulb of Fritillaria pallidiflora (Fritillaria pallidiflora의 신구인경에 있어서 대사물질의 세포간 이동과 미세구조의 차이)

  • The structure of amyloplasts and intercellular transport in the old and new bulbs of Frjtillaria pallidiflora were observed by means of electron microscope. The structure of internal membrane system was different between new and old amyloplasts. The active intercellular transport was observed in both new and old bulbs. The phenomena of encytosis and exocytosis always could be found in the cell membrane, and plasmodesmata established a symplasmic pathway for intercellular transport. Groups of vesicles often located at the ends of plasmodesmata, showing that they participated in the intercellular transport. These results laid a foundation for the further study on the mechanism of growth and development in Fritillaria pallidiflora.

Antitussive Effect of Fritillaria unibracteata Hsiao et K. C. Hsia

  • This experiment was conducted to clarify the antitussive effect of Fritillaria unibracteata Hsiao et K. C. Hsia. The antitussive effect in ethanol (75, 50, 30%) extracts from F. unibracteata was relatively high. The ethanol extracts of 20, 40 mg/kg concentrations were especially good antitussive effect. The ethanol extracts of F. unibracteata showed lower antitussive activities of 20~30% than sodium chloride injection used as a blank space control.

  • Amanita Pers. is a well-known monophyletic mushroom genus with a broad distribution. However, the diversity of Korean Amanita species has been underestimated, and most taxonomic studies conducted in Korea have only investigated their morphological characteristics. This approach is frequently insufficient for correct identification in fungal classification; therefore, we constructed a phylogeny of Amanita subgen. Lepidella in order to understand the phylogenetic placements of 16 Amanita specimens collected in Korea in 2012. The phylogeny constructed using the sequence data of the internal transcribed spacers and the partial large subunit of ribosomal RNA identified nine Amanita species (A. citrina, A. excelsa var. spissa, A. flavipes, A. fritillaria, A. oberwinklerana, A. pallidorosea, A. rubescens, A. subjunquillea, and A. volvata); of these, A. fritillaria, A. oberwinklerana, and A. pallidorosea are new to Korea.

Identification of 11 species of Paemo through each original plant and medicines (11종 패모(貝母)의 기원별 자연·약재상태 감별)

  • Objectives : Paemo is a phlegm-resolving drug with cold properties and classified 5 kinds which come from 11 species of original plant. All the more, according to literature record, 20 species of original plant were used. As a natural result, these are easily to confuse and there are a lot of counterfeit product. So we are to present a differential standard of Paemo. Methods : It was planed a differential standard form through outer appearance of the original plant and outer appearance in the form of each medicines which was collected local market or field for 11 species which is listed in Korea or China pharmacopeia. Results : It was possible to distinguish the orignal plant between Fritillaria and Bolbostemma through its stem shape. In Fritillaria of original plant, it was possible to distinguish through its width of leaf, number of leafy bracts, color and position of flower and shape of leaf apex. In outer appearance in the form of each medicines, there are difference in color and texture of medicine between Fritillaria and Bolbostemma and there are difference in size, shape, size of inner and outter fleshy leaf of bulb, pattern of surface and apex of fleshy leaf of bulb among 10 Fritillaria species. Conclusions : This study presents various differences in the outer appearance of the original plant and the outer appearance in the form of each medicines among Paemo. It will be helpful to further applied research.

Inhibitory Effect of Fritillaria Verticillata Willd. var. Thunbergii Bak Ethanol Extract on Melanin Biosynthesis (절패모(浙貝母) 에탄올 추출물의 멜라닌 생성 억제 효과)

  • This study was conducted to evaluate the depigmenting properties of ethanol extract from a Fritillaria verticillata Willd. (EFV) in B16F10 cells. Fritillaria verticillata Willd., a perennial herbaceous plant, has been used as a stimulator of mammary gland, expectorant, blood pressure depressant, antitussive agents in Korean herbal medicine. In the present study, we observed that melanin synthesis of B16F10 cells were significantly decreased by EFV without cytotoxicity. However, EFV could not suppress tyrosinase activity in B16F10 cells and mushroom tyrosinase activity. Furthermore, EFV did not effect the protein expression of tyrosinase, tyrosinase-related protein -1 (TRP-1), and TRP-2. These results suggest that EFV inhibited melanin synthesis and the hypopigmentary effect of EVF was not due to regulation of tyrosinase protein.

Anti-Inflammatory Effects of Ji-Pae-San Water Extract (지패산(芷貝散)의 항염증(抗炎症) 효능(效能)에 대한 연구(硏究))

  • Although inflammatory mediators such as nitric oxide(NO) and pro-inflammatory cytokines are involved in host defense mechanism, these overproduction contributes to the pathogenesis of several diseases such as otitis media, hearing loss, periodontitis, bacterial sepsis, rheumatoid arthritis, chronic inflammation and autoimmune diseases. We investigate the anti-inflammatory effects of water extract from Ji-Pae-San(JPSWE) fomulated with Angelica dahurica plus Fritillaria Verticillata, Angelica dahurica(ADWE), and Fritillaria Verticillata(FUVE) in vitro and in vivo. Each extract inhibited the production of inflammatory mediators(NO, $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, and prostaglandin $E_2$) and the expression of inducible NO synthase(iNOS) and cyclooxygenase-2(COX-2) in lipopolysaccharide(LPS)-stimulated RAW 264.7 macrophages in a dose-dependent manner. These inhibitory effects were synergistically increased by their combination. JPSWE also inhibited $TNF-{\alpha}$, $IL-1{\beta}$, IL-6. and $PGE_2$ production as well as COX activity in LPS-stimulated mice. Moreover, JPSWE significantly suppressed death by LPS-septic shock in mice(survival rate: 100%). These results suggest that Ji-Pae-San may be useful for therapeutic drugs against inflammatory immune diseases, probably by suppressing the production of inflammatory mediators.

Diversity and Distribution of Bulb-associated fungi of Fritillaria Cirrhosae Bulbus Source Plants used in Traditional Chinese Medicine

  • Diversity and community composition of bulb-associated fungi of Fritillaria Cirrhosae Bulbus source plants, which are used in traditional chinese medicine, in the eastern Himalaya-Hengduan Mountains, southwestern China, were estimated based on the internal transcribed spacer rDNA sequence analysis, using host plant species, geographic area, and plant phenology as variables. A total of 1,486 fungal sequences assigned to 251 operational taxonomical units (OTUs) were obtained from the bulbs. Fungal OTUs comprised 96.41% Ascomycotina, 3.52% Basidiomycotina, and 0.07% Zygomycotina. Sordariomycetes, Hypocreales, and Nectriaceae were the most frequent fungal lineages at each taxonomic rank. Fusarium, Ilyonectria, Tetracladium, Leptodontidium, and Tomentella were the top OTU-rich genera. Fusarium sp. 03, Ilyonectria rufa, Fusarium sp. 08, Ilyonectria sp. 03, and Leptodontidium orchidicola 03 represented the most frequent OTUs. Fusarium spp. were the most frequent general taxa. The distribution of fungal community exhibited preferences for host plant species, geographic area, and plant phenology. These findings are the foundation of our research on culturing and active metabolites of bulb-associated fungi of Fritillaria Cirrhosae Bulbus source plants.
