Purification and Characterization of the $Exo-{\beta}-D-Glucosaminidase$ from Aspergillus flavus IAM2044

  • Chitosan-degrading activity induced by chitosan was founf in culture filtrate of Aspergillus flavus IAM2044. Aspergillus flavus IAM2044 had a higher level of chitosanolytic activity when chitosan was used as a carbon source, and yeast extract and peptone were supplemented as nitrogen sources. One of the chitosan-degrading enzymes was purified to homogeneity by ammonium sulfate precipitation followed by cation-exchange and gel filtration chromatographies. The enzyme was monomeric, and its molecular mass was 45 kDa. The optimum pH and temperature of the enzyme were 5.0 and $50^{\circ}C$, respectively. The activity was stable in the pH range of 3.5 to 7.0 and at a temperature below $50^{\circ}C$. Reaction products analyzed by the viscosimetric assay and thin layer chromatography clearly indicated that the enzyme was an exe-type chitosanase, $exo-{\beta}-D-glucosaminidase$, that released GlcN from the nonreducing ends of the oligosaccharide chains.

Screening of Volatile Organic Compound-Producing Yeasts and Yeast-Like Fungi against Aflatoxigenic Aspergillus flavus

  • Aflatoxin contamination in rice has been documented in a number of studies, and has a high incidence in Asian countries, and as such, there has been a growing interest in alternative biocontrol strategies to address this issue. In this study, 147 strains of yeasts and yeast-like fungi were screened for their potential to produce volatile organic compounds (VOCs) active against Aspergillus flavus strains that produce aflatoxin B1 (AFB1). Five strains within four different genera showed greater than 50% growth inhibition of some strains of A. flavus. These were Anthracocystis sp. DMKU-PAL124, Aureobasidium sp. DMKU-PAL120, Aureobasidium sp. DMKU-PAL144, Rhodotorula sp. DMKU-PAL99, and Solicococcus keelungensis DMKU-PAL84. VOCs produced by these microorganisms ranged from 4 to 14 compounds and included alcohols, alkenes, aromatics, esters and furans. The major VOCs produced by the closely related Aureobasidium strains were found to bedistinct. Moreover, 2-phenylethanol was the most abundant compound generated by Aureobasidium sp. DMKU-PAL120, while methyl benzeneacetate was the major compound emitted from Aureobasidium sp. DMKU-PAL144. On the other hand, 2-methyl-1-butanol and 3-methyl-1-butanol were significant compounds produced by the other three genera. These antagonists apparently inhibited A. flavus sporulation and mycelial development. Additionally, the reduction of the AFB1 in the fungal-contaminated rice grains was observed after co-incubation with these VOC-producing strains and ranged from 37.7 ± 8.3% to 60.3 ± 3.4%. Our findings suggest that these same microorganisms are promising biological control agents for use against aflatoxin-producing fungi in rice and other agricultural products.

홍삼(紅蔘) 사포닌이 Aspergillus flavus의 발육(發育)과 Aflatoxin 생산(生産)에 미치는 효과(效果) (Effects of Red Ginseng Saponins on Growth of and Aflatoxin Production by Aspergillus flavus)

  • A. flavus NRRL 3357의 발육(發育)과 발암물질(發癌物質)인 aflatoxin 생산(生産)에 최적 조건(條件)을 부여하여 홍삼(紅蔘) 사포닌을 첨가(添加)한 배지(培地)에서 배양한 균체(菌體)의 발육(發育)과 aflatoxin 생산능력(生産能力)을 조사하였다. Aflatoxin 정량은 HPLC를 이용하였고 균체발육(菌體發育)은 건조균체(乾燥菌體) 측정법(測定法)에 의하여 실시하였다. 홍삼(紅蔘) 사포닌 첨가배지(添加培地)에서 균체발육(菌體發育)은 대조군(對照群)과 큰 차이가 없었고 배양 4일째에 최대 발육을 나타내었다. Aflatoxin 생산능력(生産能力)은 홍삼(紅蔘) 사포닌 첨가배지(添加培地)에서 aflatoxin $B_1$$G_1$이 각각 대조군(對照群)의 31.6%, 2.12%로 억제효과(抑制效果)를 나타내었다. 균체발육(菌體發育) 및 독소생산(毒素生産)에 의한 배지(培地)의 변화(變化)는 거의 대등한 차이를 보이나 대조군(對照.群)보다는 높게 나타났다. 전기영동(電氣泳動)은 SDS/polyacrylamide gel을 이용하였으며 홍삼(紅蔘) 사포닌 첨가배지(添加培地)에 발육(發育)한 균체(菌體)단백질 패턴의 intensity는 대조군(對照群)보다 낮게 나타났다. A. flavus 균체(菌體)단백질은 비교적 낮은 분자량을 가진 단백질들의 생성이 많았다. 따라서 홍삼(紅蔘) saponin은 invitro 실험(實驗)에서 aflatoxin $B_1$$G_1$의 생성(生成)을 효과적(效果的)으로 억제(抑制)함을 알 수 있었다.

Aspergillus flavus에 의한 Aflatoxin 생산능(生産能)에 Steroidal Carbamate Derivatives가 미치는 영향(影響) (Effects of Steroidal Carbamate Derivatives on the Aflatoxin Productivity by Aspergillus flavus)

  • Experiments were conducted to study effects of steroidal carbamate derivatives upon mycelial growth and aflatoxin production by Aspergillus flavus ATCC 15517. The basal medium was supplemented with various concentrations of these compounds and inoculated with spores. The developing cultures were incubated for 11 days at $28^{\circ}C$ without agitation. Aflatoxins were extracted with chloroform, separated by thin layer chromatography, and quantitated by ultraviolet spectrophotometry. At a concentration of 50 mg per 30 ml of medium., stigmasteryl-N-(2-chloroethyl) carbamate, cholesteryl- N - (2-chloroethyl) carbamate, $5{\alpha}-cholestan-3-one-oximino-N-(2-chloroethyl)$ carbamate and ${\beta}-sitosteryl-N-(2-chloroethyl)$ carbamate were the most effective in reducing aflatoxin production by Aspergillus flavus. However, cholest-4-ene-3-one-oximino-N-(2-chloroethyl) carbamate, at a concentration of 100 mg per 30 ml, significantly decreased aflatoxin production. There was no significant inhibition of mycelial growth by the addition of the various concentrations of these compounds.

몇몇 음식 부식 균류의 방사선 감수성에 대한 연구 (Radiation Sensitivity of Some Food Decay Fungi)

  • Five species of food decay fungl, Aspergillus flavus, Asp. uiger, Penicillum sp., Botrytis cinerea, and Rhizopus stolonifer, were examined for their radio sensitivity in several suspension media. Asp. flavus, Asp. niger and Penicillum sp. have almost the same sensitivity toward gamma rays. with D value in the range of 30 to 35 K rad, whereas Botrytis cinerea has a D value of approximately 55K rad and Rhizopus stolonifer, the most re4sistant fungus studied, has a crease in their radioresistnace when compared with spores irradiated in water. Asp. flavus and penicillium sp. spores irradiated in citrate buffer at pH3-7 showed almost no change in their radisensitiity with pH, but Botryis cinerea spores showed a distinct decrease in their radioresistnace at pH 6 and 7. Penicillum sp. spores irradiated in sucrose solutions showed no sinificant change in their radioresistance. Botrytis cinerea spores displayed a higher radioresistance when they were irradiated in sucrose solution than in water.

Molecular Characterization of Apple stem grooving virus Isolated from Talaromyces flavus

  • Talaromyces flavus mediates the transmission of Apple stem grooving virus (ASGV) to several host plants. The ASGV-F carried by T.flavus was partially purified from the fungus. Based on sequence analysis and homology searches, this is closely related to other ASGV strains isolated from host plants. The partially purified viral coat protein (CP) was separated on a 12% SDS-polyacrylamide gel and analyzed by Western blotting with an ASGV anti-serum. A single band at 28 kDa reacted with the ASGV anti-serum. The deduced amino acid sequence of the ORF-l showed conserved domains, including an NTP-binding helicase motif, GFAGSGKT. The amino acid sequences of the helicase and CP showed strong homology to other ASGV strains (98%). All ASGV isolated from plants and fungi had salt bridges composed of the CP and the GFAGSGKT motif of the helicase, which are commonly conserved in plant viruses. These results suggest that ASGV-F is one of ASGV strains isolated from T.flavus based on sequence similarity as well as the serological analysis of CP.

순창군 장류로부터 분리된 황국균의 동정 및 특성 (Identification and Characterization of Aspergillus oryzae Isolated from Soybean Products in Sunchang County)

  • 본 연구에서는 순창지역에서 만들어지는 장류에서 곰팡이를 분리하고 동정하여 보다 안전하고 기능성이 높은 발효제품을 위한 균주를 확보하고자 하였다. 순창지역에서 생산되는 장류 제품으로부터 곰팡이를 분리하여 ${\beta}$-tubulin 유전자 분석 통해 10개의 균주가 Aspergillus oryzae/flavus complex임을 알 수 있었다. 보다 정확한 동정을 위하여 아플라톡신 클러스터 유전자 중에 하나인 omtA의 염기서열을 증폭하여 A. oryzae와 A. flavus 표준 균주의 omtA 서열과 함께 계통 분류한 결과, A. oryzae의 표준 균주와의 유연관계가 높음을 알 수 있었다. 또한 norB-cypA 사이의 염기서열을 증폭한 결과 500 bp이 증폭 산물이 확인되었는데 이는 표준 균주인 A. oryzae의 norB-cypA 사이의 염기서열 증폭 산물과 동일한 크기임을 확인할 수 있었다. A. oryzae로 확인된 10균주를 활용하여 코지를 제조하고 ${\alpha}$-amylase 활성과 protease 활성을 측정하였다. Protease 활성은 6, 13, 17, 27, 37, 그리고 38 균주로 제조된 코지는 대조구(시판되고 있는 종균으로 제작한 코지)보다 2배 정도 높은 protease 활성을 보였으며, ${\alpha}$-amylase 활성은 257~320 U/mL로 측정되었다. 식품안전성을 위한 아플라톡신 분비 확인 결과, 63번 균주로 제조된 코지를 제외한 모든 코지에서 아플라톡신을 만들지 않는 것으로 확인되어, 순창에서 분리된 A. oryzae는 추후 메주 접종균으로 개발할 수 있음을 보여주었다.

한국전통 식품의 원료인 메주와 누룩에서 분리된 황곡균에 대한 연구 (Studies on the Yellow Fungal Isolates (Aspergillus species) Inhabiting at the Cereals in Korea)

  • The yellow fungal isolates inhabiting at the cereals (Hwang-Kuk, HK-fungi) were widely collected from the mejus and nuluks in Korea; the meju is a raw material for Korean traditional foods for soysauce and soypaste, and the nuluk is a raw material for Korean traditional rice wine. These isolates, well known as an Aspergillus oryzae producing amylase for Korean rice wine or producing protease for soybeans, were compared with Aspergillus species known. All isolates were microscopically observed to be a species of A. oryzae or its related, but to be difficult to be identified. Thus, RAPD-DNA techniques were applied for these isolates and analyzed with nummerical values using NT-system, or Ecological programs or Factorial analyses. Several common bands of RAPD-DNA in the 28 isolates were synthesized with the different OPD primers and speculated to be used for identification of HK fungi. The HK-fungi isolated were revealed to belong to the group of A. flavus previously defined. Particularly, the isolates collected from mejus were analyzed to be more closed to A. flavus, The species of A. flavus, A. oryzae and A. sojae were grouped at the values lower than those indicating the diversity of species. In other words, these three fungal species were not distinguishable and all isolates known as a HK-fungus were very closed to A. flavus, All isolates were not diversified at groupings of RAPD-DNA, and considered to be not the natural flora at the mejus or nuluks.   • Two Rice cultivars , a high yield variety of Cheongcheong and a Korea native kind of Chucheong were grown in paddy. 1$\mu\textrm{g}$ and 10$\mu\textrm{g}$ of aflatoxin B1 and Aspergillus flavus (106 conidia/ml) were inoculated at milk stage. At harvest, kernels from the inoculated plant showed slightly lower ripening rates and 100-grain weight than did those of controls. The nutrient and fatty acid composition of unpolished rice of inoculated group were similar to those of controls and difference between the control and inoculated group were not significantly different.

Gamma-Irradiation Sensitivity of Aspergillus flavus Contaminated in Semi-Dried Beef Jerky

  • This study was conducted to investigate the adequate irradiation dose to eliminate harmful fungi inoculated if beef jerky with a 10% higher moisture content and improved textural property. Aspergillus flavus (approximately $10^6\;CFU/cm^2$) was tested in broth, spore suspension, and inoculated jerky. $D_{10}$ values of A. flavus were 0.36 kGy in the broth and suspension, and 0.47 kGy in the jerky. The results indicate that gamma irradiation can be effectively used to control the fungus growth in beef jerky with an improved quality and higher moisture content.
