쇠뜨기(Equisetum arvense L.)의 수용추출액이 몇가지 식물의 캘러스 생장, 종자발아, 유묘생장 및 항균활성에 미치는 영향 (Effect of Aqueous Extracts on the Callus Growth, Seed Germination, and Growth of Some Plants and Antibacterial Activity by Equisetum arvense L.)

  • 쇠뜨기에 다른식물의 생장을 저해하는 물질이 생성되는지를 알아보기 위하여, 수용추출액으로 몇가지 식물의 캘러스 생장, 종자발아, 유묘생장 및 항균활성에 미치는 영향을 조사하였다. 수용 추출액의 첨가농도가 높은 배지에서 벼와 배추의 캘러스 유기 및 발달을 억제하였으며, 8$0^{\circ}C$ 추출액이 $25^{\circ}C$ 추출액보다 유의하게 캘러스 유기 및 생장을 억제하였다. 20%추출액 첨가배지에서 공시식물의 캘러스 생장은 추출온도에 관계없이 도라지, 참깨, 배추, 벼 순으로 억제되었다. 냉동건조한 수용추출액 고형물은 저농도에서는 영향이 적었으나, 처리농도에 비례하여 벼, 참깨, 배추의 종자발아를 억제하였고, 특히 배추는 2,000$\mu\textrm{g}$/mι농도에서 대조구의 28.3%로 급격한 발아억제를 보였다. 유식물 생장은 500$\mu\textrm{g}$/mι의 저농도에서는 대조구보다 초장이 약간 촉진되는 경향을 보였으나, 처리농도가 증가함에 따라 심하게 억제되었으며, 특히 근장이 유의하게 억제되었다. 추출액 고형물의 항균활성은 500$\mu\textrm{g}$에서 E. carotovora에서만 약간의 억제대를 형성하였으며, X. oryzae pv. oryzae와 E. coli에 대해서 는 항균활성을 보이지 않았다.

Agrobacterium tumefaciens에 의한 벼 형질전환에 미치는 품종과 Acetosyringone의 영향 (Effects of Variety and Acetosyringone Influencing Transformation of Rice Mediated by Agrobacterium tumefaciens)

  • 배추에서 분리된 cytosolic glutathione reductase (BcGR1)유전자를 벼에 효과적으로 도입하기 위하여 몇 가지 실험을 수행하여 얻어진 결과를 요약하면 다음과 같다. 수정 후 3주된 미숙배에서 유도된 캘러스를 이용하여 형질전환 효율의 품종간 차이를 조사한 바 '대립벼'가 42.5%로 가장 높게 나타났다. 공동배양 배지 내에 acetosyringone (AS)이 첨가되지 않았을 경우 형질전환된 개체를 얻을 수 없었으나, 50$\mu$M을 첨가되었을 때 41.0%의 가장 높은 형질 전환효율을 나타내었다. PCR 증폭과 Southern blot 분석을 실시한바 BcGR1 유전자가 형질전환체의 게놈상에 정상적으로 삽입된 것이 확인되었으며, T$_1$세대 28개통 중 17개체의 hygromycin 저항성 유전형질이 3:1로 분리되었다.

Water Extract from Spent Mushroom Substrate of Hericium erinaceus Suppresses Bacterial Wilt Disease of Tomato

  • Culture filtrates of six different edible mushroom species were screened for antimicrobial activity against tomato wilt bacteria Ralstonia solanacearum B3. Hericium erinaceus, Lentinula edodes (Sanjo 701), Grifola frondosa, and Hypsizygus marmoreus showed antibacterial activity against the bacteria. Water, n-butanol, and ethyl acetate extracts of spent mushroom substrate (SMS) of H. erinaceus exhibited high antibacterial activity against different phytopathogenic bacteria: Pectobacterium carotovorum subsp. carotovorum, Agrobacterium tumefaciens, R. solanacearum, Xanthomonas oryzae pv. oryzae, X. campestris pv. campestris, X. axonopodis pv. vesicatoria, X. axonopodis pv. citiri, and X. axonopodis pv. glycine. Quantitative real-time PCR revealed that water extracts of SMS (WESMS) of H. erinaceus induced expressions of plant defense genes encoding ${\beta}$-1,3-glucanase (GluA) and pathogenesis-related protein-1a (PR-1a), associated with systemic acquired resistance. Furthermore, WESMS also suppressed tomato wilt disease caused by R. solanacearum by 85% in seedlings and promoted growth (height, leaf number, and fresh weight of the root and shoot) of tomato plants. These findings suggest the WESMS of H. erinaceus has the potential to suppress bacterial wilt disease of tomato through multiple effects including antibacterial activity, plant growth promotion, and defense gene induction.

Glutathione reductase 유전자 도입에 의한 유채의 형질 전환 (Transformation of Brassica napus with Glutathione Reductase Gene)

  • This study was conducted to construct of the transgenic plants wliich are resistant to oxidative stresses including ozone with B. mpestris cytosolic glutathione reductase cDNA using the binary vector system of Agrobacterium tumefaciens. The 1.8kb B. campestris cytosolic GR cDNA was subcloned into the unique Sma I site of the plant transformation vector pBKSI- I, downstream of the constitutive CaMV 35s promoter and upstream of the nos termination sequence, in place of the uidA (GUS) reporter gene. The resulting plant transformation vector, pBKS-GRI, was introduced into A. tumefaciens LBA4404 by two cycles of tkeze-thaw method. The B. nqus cotyledonary petioles were transformed by the Agrubaferium harboring pBKS-GRI. Transformed shoots were induced and selected on regeneration medium supplemented with kanarnycin. The shoot formation was increased remarkably by addition of Ag$NO_3$, in MS media. The transgenic plants were analyzed for the presence of the B. campestris GR gene by Southern blot analysis and it was confirmed that a foregin gene was stably integrated into the genomes of B. nqus plants.

Xanthomonas campestris에 의한 Xanthan gum 생산에 관한 연구 (A Study on the Production of Xanthan Gum by Xanthomonas campestris)

  • In the Xanthan gum fermentation by Xanthomonas campestris there are problems of the large energy consumption by long fermentation time, the mass transfer of oxygen and nutrients by high viscous fermentation broth. In this study, the media optimization and the fed batch fermentation were carried out to decrease fermentation time and increase Xanthan gum yield. The $O_2$ uptake rate (OUR) and $CO_2$ evolution rate(CER) which were obtained from the analysis of fermentation exit gas using a gas chromatograph were investigated. As a result, the fermentation time decreased at optimal assimilable nitrogen concentration but increased at poor or rich assimilable nitrogen concentration, the Xanthan gum biosynthesis was stimulated under the limited condition of assimilable nitrogen source and the optimum fermentation medium was obtained as follow; Glucose=30g / l, Peptone=8.0g / l, $K_2HPO_4=2.0g/l$, $MgS0_47H_2O=10g/l$, Sodium acetate=20g/l, Sodium pyruvate=0.5g/1. As the agitation speed and nitrogen concentration increased, the $O_2$ uptake rate and $CO_2$ evolution rate increased. The OUR and CER were 37.3mmol $O_2/\;l$ hr and 20.2 mmol $CO_2/\;L$ hr at peptone 11g / l and agitation speed 990RPM, respectively. In fed batch fermentation, the final concentration of Xanthan gum was enhanced up to 29g / l.

Toward Functional Genomics of Plant-Pathogen Interactions: Isolation and Analysis of Defense-related Genes of Rot Pepper Expressed During Resistance Against Pathogen

  • To understand plant-pathogen interactions, a complete set of hot pepper genes differentially expressed against pathogen attack was isolated. As an initial step, hundreds of differentially expressed cDNAS were isolated from hot pepper leaves showing non-host resistance against bacterial plant pathogens (Xanthomonas campestris pv. glycines and Pseudomonas syringae pv. syringae) using differential display reverse transcription polymerase chain reaction (DDDRT-PCR) technique. Reverse Northern and Northern blot analyses revealed that 50% of those genes were differentially expressed in pepper loaves during non-host resistance response. Among them, independent genes without redundancy were micro-arrayed for further analysis. Random EST sequence database were also generated from various CDNA libraries including pepper tissue specific libraries and leaves showing non-host hypersensitive response against X. campestris pv. glycines. As a primary stage, thousands of cDNA clones were sequenced and EST data were analyzed. These clones are being spotted on glass slide to study the expression profiling. Results of this study may further broaden knowledge on plant-pathogen interactions.

Geographic Information System (G.I.S)을 이용한 서부호주 밀재배 지역의 흰개미집 밀도와 분포에 있어서 식물상과 토양형이 미치는 영향평가 (Influence of Vegetation and Soil Types on the mound Density and Distribution of the Wheatbelt Termite in Westerm Australia: Using a Geographic Information System (G.I.S))

  • 서부호주 밀재배 지역의 Durokoppin 자연보호 구역내 흰개미(Drepanotemes tamminensis(Hill)) 집의 분포와 밀도에 있어서 토양형보다는 식물상이 주된 영향을 미치는 것으로 밝혀졌다. D. tamminensis의 집은 Woodland (우점종-Eucalyptus Capillosa)와 Casuarina (우점종-Allocasuarina campestris) 식물군에서 가장 높은 밀도를 보였다. 비록 Booram 토양형에서 개미집 밀도가 높은 경향을 보였으나, 개미집 분포에 있어서 토양형이 미치는 영향은 없었다.
