Effect of Sulforaphane on LPS-Induced Matrix Metalloproteinase-9 (MMP-9) Expression (Sulfolaphane이 lipopolysaccharide (LPS)에 의해 유도된 matrix metalloproteinase-9 (MMP-9) 발현에 미치는 영향)

  • Sulforaphane is a naturally occurring member of the iosothiocyanate family, which reveals chemopreventive capacities including anti-cancer, anti-inflammation and inhibition of MMP-9 activities. In this study, we investigated the effect of sulforaphane on the expression of matrix metalloproteinase-9 (MMP-9) in lipopolysaccharide (LPS)-induced Raw 264.7 cells. Sulforaphane strikingly suppressed the LPS-induced MMP-9 activity and mRNA expression in a dose-dependent manner. In addition, sulforaphane inhibited not only the LPS-induced MMP-9 promoter activity but also LPS-mediated activator protein-1 (AP-1) and nuclear factor-kB (NF-${\kappa}B$) promoter activity. Transient transfection by MMP-9 constructs, in which specific transcriptional factors were mutagenized, indicated that the effects of LPS and sulforaphane were mediated via AP-1 and NF-${\kappa}B$ response elements. We found that sulforaphane had the ability to suppress LPS-induced invasion in vitro. Taken together, these results demonstrated that sulforaphane effectively suppressed LPS-induced MMP-9 expression via modulation of promoter elements (AP-1 and NF-${\kappa}B$) in MMP-9 transcriptional activation.

A Study on the Change of Streptococcus Mutans in Dental Plaque after Use of $0.05\%$ NaF in Orthodontic Patients (NaF $0.05\%$ 양치액 사용시 고정성 교정장치 장착 환자에서의 Streptococcus mutans 변화에 관한 연구)

  • The purpose of this study was to evaluate the effectiveness of gargling solution with 0.05% NaF and 10% Xylitol in orthodontic patients with fixed appliance. The sample consisted of 20 patients who were classified into an experimental group and a control group, 10 patients each. Experimental group was used experimental gargling solution and the control group was used with placebo solution. The change of S. mutans was analysed by culture on MSB and BHI agar plate pre and post 3, 6, 9 weeks. The results were as follows. 1 There were significant reduction in the number of S. mutans C.F.U. between pre and post 3 weeks(p<0.01), 9 weeks(p<0.05) in experimental group 2. There were significant reduction in the ratio of S. mutans C.F.U. to total C.F.U. between pre and post 3, 6, 9 weeks(p<0.01) in experimental group. 3. S. mutans, which were reduced until 3 weeks, did not show significant change after 3, 6, 9 weeks. 4. S. mutans were strongly suppressed until 3 weeks after gargling solution with 0.05% NaF and 10% Xylitol.

A study on photoreflectance of GaAs surface treated with $Na_2S.9H_2O$ (황처리된 GaAs표면의 Photoreflectance에 관한 연구)

  • The surface of GaAs was treated by using the 0.1M solution of N $a_{2}$S.9 $H_{2}$O. The passivation of the surface in this sample was investigated by the photoreflectance(PR) experiment. The surface electric field( $E_{s}$) and built-in voltage( $V_{bi}$ ) discussed from Franz-Keldysh oscillation of PR signals. The density of surface states and Fermi level of GaAs treated with N $a_{2}$S.9 $H_{2}$O for 40min were determined 1.61*10$^{12}$ c $m^{-2}$ and 0.73eV. These values were about 15 and 10% smaller than those in untreated sample.e.

  • The phase-shifted version of the multichannel quantum-defect theory (MQDT) was reformulated by disentangling the interloper spectrum from the perturbed dense Rydberg series for a systems involving 2 closed and more than 1 open channel. The theory was applied successfully to Martins and Zimmermann's photoionization spectra of the Rydberg series Cu I $3d^9\;4s(^1D_2)\;nd^2G_{9/2}$ perturbed by the interloper, $3d^9\;4p^2\;^4F_{9/2}$, for which Cohen's 4 channel QDT had failed. The zero surface graphic of the perturbed Fano's asymmetry parameter q of the autoionization spectrum of dense Rydberg series by the interloper was determined by only two parameters for this system. It was used as a map to trace the transformation route of the 3 channel autoionization spectra to the 4 channel spectra when the channel coupling of the closed channels with a newly added open channel was turned on progressively.

  • Clustered regularly interspaced short palindromic repeats and CRISPR-associated protein 9 (CRISPR-Cas9) is an ancient prokaryotic defense system that precisely cuts foreign genomic DNA under the control of a small number of guide RNAs. The CRISPR-Cas9 system facilitates efficient double-stranded DNA cleavage that has been recently adopted for genome editing to create or correct inherited genetic mutations causing disease. Congenital heart disease (CHD) is generally caused by genetic mutations such as base substitutions, deletions, and insertions, which result in diverse developmental defects and remains a leading cause of birth defects. Pediatric CHD patients exhibit a spectrum of cardiac abnormalities such as septal defects, valvular defects, and abnormal chamber development. CHD onset occurs during the prenatal period and often results in early lethality during childhood. Because CRISPR-Cas9-based genome editing technology has gained considerable attention for its potential to prevent and treat diseases, we will review the CRISPR-Cas9 system as a genome editing tool and focus on its therapeutic application for CHD.

Predictions of zinc selenide single crystal growth rate for the micro gravity experiments

  • One predicts the crystal growth rate of ZnSe with a low vapor pressure system in a horizontal configuration based on one dimensional advection-diffusion and two-dimensional diffusion-convection model. The present results show that for the ratios of partial pressures, s = 0.2 and 2.9, the growth rate increases with the temperature differences between the source and crystal. As the ratio of partial pressure approaches the stoichiometric value, s = 2 from s = 1.5 (zinc-deficient case: s < 2) and 2.9 (zinc-rich case: s > 2), the rate increases sharply. For the ranges from 1.5 to 1.999 (zinc-deficient case: s < 2) and from s = 9 to 2.9 (zinc-rich case: s > 2), the rate are slightly varied. From the viewpoint of the order of magnitude, the one-dimensional model for low vapor pressure system falls within the 2D predictions, which indicates the flow fields would be advective-diffusive. For the effects of gravitational accelerations on the rate, the gravitational constants are varied from 1 g to $10^{-6}$ g for $\Delta$T = 50 K and s = 1.5, the rates remain nearly constant, i.e., 211 mg/hr, which indicates Stefan flow is dominant over convection.
