Variation of 7S and 11S Seed Protein Concentrations in Different Food Types of Soybean Seed

  • Soybean varieties derived from Korea are classified into four groups on the basis of their food types such as soybeans for vegetable, sprout, sauce and paste and soybeans with colored seed coat. This study was carried out to know the differences in storage protein concentrations among these four groups. There were differences in storage protein concentrations among four groups. In 7S protein, the $\alpha$'-and $\alpha$-subunit concentrations did not vary among four groups, while a $\beta$-subunit concentration greatly varied. 7S protein concentration was the highest(40.6%) in soybean for sauce and paste and the lowest(37.7%) in soybean for vegetable, while 11S protein concentration was the highest (62.3%) in soybean for vegetable and the lowest (59.4%) in soybean for sauce and paste. In view of the fact that 11S protein has much higher sulfur containing amino acids than 7S protein, it was shown the soybeans for vegetable may have higher nutrition value than other groups.

분리대두단백질이 첨가된 증편의 저장성에 관한 연구 (The Effects of SPI(Soybean Protein Isolate) on Retrogradation in Jeung-pyun)

  • This study examined Jeung-pyun(JP) Retrogradation in samples containing 3% whole protein, 7S protein, or 11S protein(w/w) that were stored at $4^{\circ}C$ for 6, 12, 24 and 72 hr. Rheometery and differential scanning calorimetry(DSC) were used in the analysis. The pH of the dough decreased during the fermentation process, but it increased after steaming. The JP prepared with soybean protein isolate(SPI) had higher pH than the control group. During storage the textural characteristics of the JP showed effects according to the additions of SPI. After 6 hr of storage, the JP samples containing soybean flour, whole protein, 7S protein, and 11S protein had lower hardness valuse. From 4 hr to 12 hr, higher springiness values were found in the samples containing whole protein, 7S protein and 11S protein. At 0 hr, the control group had the highest cohesiveness value, but after 24 hr it presented the lowest value. For gumminess, after 6 hr of storage, the control group offered the lowest value. Whereas after 12 hr of storage the whole protein group showed the highest value, and at 24 hr, the whole protein, 7S protein, and 11S protein groups had higher values. According to the DSC results, the 11S protein group had lower enthalpy values(${\bigtriangleup}H$) suggesting that adding 11S protein to JP might improve starch retrogradation. After 72 hr of storage, the control group had the highest onset temperature($T_{o}$) and peak temperature($T_{p}$) whereas the 7S and 11S protein JP samples had higher conclusion temperatures($T_{c}$). Therefore, based on the different analysis result between the control and treatment groups, the addition of SPI to Jp had effects on retrogradation.

  • 단백질 및 지방질의 주요한 공급원인 콩은 오래전부터 다양한 용도로 우리의 식생활에 이용되어 왔으며 앞으로도 안정적인 단백질 공급원을 위해서는 양질 고단백 콩 신품종 육성에 많은 노력을 기울여야 한다. 따라서 콩 단백질의 특성에 관한 연구내용들을 정리한 바 다음과 같이 요약할 수 있었다.1. 콩 종실 부위중 배아와 자엽은 전체 콩 종실무게의 2.4%와 90.3%를 차지하며 단백질도 건물중으로 40%이상 함유되어 있는 영양적으로 중요한 부분이 된다. 2. 콩 종실 단백질의 대부분은 Globulin이 차지한다. 3. 콩 종실 단백질의 Amino산 조성은 Glutamic acid, Aspartic acid, Arginin의 순으로 다량 함유되어 있으며 Cystine, Methionine, Thyrosine이 적게 함유되어 있다. 4. 콩 종실의 주요 단백질 분획은 7S와 11S로 되어 있으며 11S가 7S보다 함류황 Amino산 함유량이 많다. 5. 콩의 가용성 단백질 함량이 높을수록 두부의 수율도 증가했으며 이들의 상관계수는 0.96을 보였다.

콩의 7S α' - subunit 단백질의 유전 (Inheritance of 7S α' - subunit Protein in Soybean Seed)

  • 인간과 가축의 영양을 위한 식물성 단백질의 주요 공급원은 콩이며 콩 단백질은 영양 및 기능성면에서 우수하여 소비가 점차 증가하고 있다. 그러나 콩 단백질에는 알러지를 일으키고 영양가치를 떨어뜨리는 성분도 포함되어져 있다. 7S 및 11S 글로블린은 콩 저장단백질의 대부분을 차지하며 7S는 영양가치가 떨어지고 7S의 함량을 줄어든 콩 계통 육성에 대한 관심이 높아지고 있다. 7S 성분중의 하나인 ${\alpha}^{\prime}$-subunit의 유전양상을 파악하기 위하여 진품콩2호와 PI506876의 교배로부터 98개의 F2 종자가 얻어졌다. SDS-PAGE로 각각의 종자를 분석한 결과 ${\alpha}^{\prime}$-subunit을 가진 종자가 70개였고 결핍된 종자가 28개였다. 이러한 유전양상은 단인자 유전원칙 (${\chi}^2=0.667$, P=0.414)과 일치하여 콩 종자에서 7S의 ${\alpha}^{\prime}$-subunit 단백질은 한 개의 유전자에 의 해서 좌우되었다. 이 결과는 7S 단백질 함량이 줄어든 콩 계통 선발에 유용하게 활용될 것으로 기대된다.

  • 분획(分劃)된 대두(大豆) 7S 및 11S 단백질(蛋白質)은 단백가수분해(蛋白加水分解) 효소(酵素)(alcalase 및 pronase)로 1시간(時間)동안 가수분해(加水分解)하였을 때 사용(使用)된 효소(酵素) 및 단백질(蛋白質) 종류(種類)에 관계없이 pH 5에서 용해도(溶解度), 열응고성(熱凝固性) 및 $Ca^{++}$에 대한 내침전성(耐沈殿性)은 상당히 증가(增加)에멀젼 활성 및 거품 안정성(安定性)은 감소(減少), 에멀젼 열안정성(熱安定性) 및 동점도(動粘度)는 거의 변화(變化)되지 않았다. 그러나 용해도(溶解度)에서 7S 단백질(蛋白質)은 pH 6에서 11S 단백질(蛋白質)은 pH 4에서 감소(減少)하였으며 또한 11S 단백질(蛋白質)은 유흡수성(油吸收性) 및 거품 형함능(形咸能)에서 가수분해(加水分解)에 의하여 증가(增加)하였으나 7S 단백질(蛋白質)은 거의 변화(變化)하지 않았다.

분리대두단백질 종류에 따른 증편의 품질특성 (A study on the Quality Properties of Jeung-pyun added SPI(Soybean protein isolate))

  • The quality properties of Jeung-pyun made with soybean protein isolate(SPI) were investigated. The SPI Jeung-pyun was manufactured using 3% whole protein, 7S protein or 11S protein(w/w). The redness values (a-values) of the Jeung-pyun were negative and the yellowness(b-value) of the control group was significantly lower compared to the other samples. The textural characteristics of the Jeung-pyun were influenced by the additions of SPI. The Jeung-pyuns containing soybean flour, whole protein, 7S and 11S protein had increases in springiness, but decreases in cohesiveness. In the sensory evaluation, overall desirability, sweetness, and moistness were highest in the whole protein group. The control group showed the highest scores for hardness and toughness, and the scores for after-taste and adhesiveness were the highest in the samples made with soybean flour. The 7S and 11S additions showed high scores in terms of color and springiness. In the analyses for consumer acceptance and sensory quality intensity, the whole protein addition had significantly higher scores for general acceptability and the Makgeolli flavor. Hardness was highest for the control group and the Makgeolli flavor was strongest with the soybean flour addition.

Effects of Red Bean (Vigna angularis) Protein Isolates on Rheological Properties of Microbial Transglutaminase Mediated Pork Myofibrillar Protein Gels as Affected by Fractioning and Preheat Treatment

  • Fractioning and/or preheating treatment on the rheological properties of myofibrillar protein (MP) gels induced by microbial transglutaminase (MTG) has been reported that they may improve the functional properties. However, the optimum condition was varied depending on the experimental factors. This study was to evaluate the effect of red bean protein isolate (RBPI) on the rheological properties of MP gels mediated by MTG as affected by modifications (fractioning: 7S-globulin of RBPI and/or preheat treatment (pre-heating; 95℃/30 min): pre-heating RBPI or pre-heating/7S-globulin). Cooking yields (CY, %) of MP gels was increased with RBPI (p<0.05), while 7S-globulin decreased the effect of RBPI (p<0.05); however, preheating treatments did not affect the CY (p>0.05). Gel strength of MP was decreased when RBPI or 7S-globulin added, while preheat treatments compensated for the negative effects of those in MP. This effect was entirely reversed by MTG treatment. Although the major band of RBPI disappeared, the preheated 7S globulin band was remained. In scanning electron microscopic (SEM) technique, the appearance of more cross-linked structures were observed when RBPI was prepared with preheating at 95℃ to improve the protein-protein interaction during gel setting of MP mixtures. Thus, the effects of RBPI and 7S-globulin as a substrate, and water and meat binder for MTG-mediated MP gels were confirmed to improve the rheological properties. However, preheat treatment of RBPI should be optimized.

콩 양질ㆍ고단백 품종 육성방향 (Perspectives of Breeding for High Protein Quantity and High Protein Quality of Soybeans)

  • Soybean grain is most widely used and soybean crop produces most high protein per area among crops. To meet rapid increase of human population and supply protein in safety. soybean has considered more and more important crop. And it has been emphasizing that high quality and high protein soybean breeding must be made efforts in future. Many papers related to soybean breeding for high quality and protein and soybean protein composition have suggested the problems to do in future. Soybean germplasm collection. classification and conservation should be continuously performed, and it is emphasized that wild type of soybeans (G. soja) be considered to use in breeding for high protein varieties. Selections would be better emphasized in first yield and therefore high yield of protein per area. Selection for high protein would be secondary criterion. High protein lines with high yielding potential could be selection from certain populations, and breeders should consider this phenomenon in procedure of selection. Heritability of protein percent is relatively high and genetic gain of increment of protein percent is large. Soybean protein which is comprised 70 to 90% of globulin is constituted mostly 11S and 7S proteins. Sulfur-containing amino acids, methionine and cysteine, are identified to contain more in 11S protein than 7S protein. High 11S germplasm should be desirable to use in crossing plan, and selection of high ratio of 11S/7S lines be better in development of high quality varieties.

클로람페니콜 내성 플라스미드 pKH7의 Pre 단백질의 염기서열 결정 (Nucleotide Sequence of Pre Protein in Chloramphenicol Resistance Plasmid pKH7.)

  • Partial nucleotide sequence (nt 1-1842) of chloramphenicol resistance plasmid pKH7 has been reported previously and residual nucleotide sequence (nt 1843-4118) of pKH7 was determined and then the complete nucleotide sequence of pKH7 was obtained. pKH7 consists of 4118 bp and has three ORFs. Besides Rep and CAT proteins described in previous paper, Pre protein which mediates site-specific recombination in Staphylococcus aureus was found to be on pKH7. R $S_{A}$, a site-specific recombination site of Pre protein, and palA, a specific lagging-strand conversion signal, was also found in pKH7. Amino acid sequence of Pre protein of pKH7 was compared with those of other antibiotic resistant Staphylococcus aureus plasmids.s.

분리 대두 단백질 첨가에 의한 증편의 이화학적 특성 (Rheological Properties of Jeung-pyun Prepared with SPI(Soybean Protein Isolates))

  • In this study the rheological properties of Jeung-pyun prepared with soybean protein isolate (SPI) were investigated. SPI Jeung-pyun samples were manufactured with 3% whole protein, 7S protein, or 11S protein (w/w). In terms of moisture content the Jeung-pyun samples prepared with soybean flour had greater moisture contents than the control group. With the addition of SPI water binding capacity solubility and swelling power increased. Dough pH decreased during the fermentation, but increased after steaming and the SPI Jeung-pyun samples presented higher pH levels han the control group. Foaming ability was significantly strong in the 7S, 11S and whole protein groups. The surface structures of the 7S, 11S and whole protein Jeung-pyun samples displayed small uniform pores when examined by SEM. Overall, the results suggest that SPI can contribute to quality improvements in Jeung-pyun through effects on dough fermentation.
